Pyrrole-2-carboxaldehyde (Py-2-C) derivatives have been isolated from many natural sources, including fungi, plants (roots, leaves, and seeds), and microorganisms. The well-known diabetes molecular ...marker, pyrraline, which is produced after sequential reactions in vivo, has a Py-2-C skeleton. Py-2-Cs can be chemically produced by the strong acid-catalyzed condensation of glucose and amino acid derivatives in vitro. These observations indicate the importance of the Py-2-C skeleton in vivo and suggest that molecules containing this skeleton have various biological functions. In this review, we have summarized Py-2-C derivatives based on their origins. We also discuss the structural characteristics, natural sources, and physiological activities of isolated compounds containing the Py-2-C group.
Intestinal microbiota-derived metabolites have biological importance for the host. Polyamines, such as putrescine and spermidine, are produced by the intestinal microbiota and regulate multiple ...biological processes. Increased colonic luminal polyamines promote longevity in mice. However, no direct evidence has shown that microbial polyamines are incorporated into host cells to regulate cellular responses. Here, we show that microbial polyamines reinforce colonic epithelial proliferation and regulate macrophage differentiation. Colonisation by wild-type, but not polyamine biosynthesis-deficient, Escherichia coli in germ-free mice raises intracellular polyamine levels in colonocytes, accelerating epithelial renewal. Commensal bacterium-derived putrescine increases the abundance of anti-inflammatory macrophages in the colon. The bacterial polyamines ameliorate symptoms of dextran sulfate sodium-induced colitis in mice. These effects mainly result from enhanced hypusination of eukaryotic initiation translation factor. We conclude that bacterial putrescine functions as a substrate for symbiotic metabolism and is further absorbed and metabolised by the host, thus helping maintain mucosal homoeostasis in the intestine.
Recently, considerable research has focused on personal assistant robots, and robots capable of rich human-like communication are expected. Among humans, non-verbal elements contribute to effective ...and dynamic communication. However, people use a wide range of diverse gestures, and a robot capable of expressing various human gestures has not been realized. In this study, we address human behavior modeling during interaction using a deep generative model. In the proposed method, to consider interaction motion, three factors, i.e., interaction intensity, time evolution, and time resolution, are embedded in the network structure. Subjective evaluation results suggest that the proposed method can generate high-quality human motions.
The new era of cognitive computing brings forth the grand challenge of developing systems capable of processing massive amounts of noisy multisensory data. This type of intelligent computing poses a ...set of constraints, including real-time operation, low-power consumption and scalability, which require a radical departure from conventional system design. Brain-inspired architectures offer tremendous promise in this area. To this end, we developed TrueNorth, a 65 mW real-time neurosynaptic processor that implements a non-von Neumann, low-power, highly-parallel, scalable, and defect-tolerant architecture. With 4096 neurosynaptic cores, the TrueNorth chip contains 1 million digital neurons and 256 million synapses tightly interconnected by an event-driven routing infrastructure. The fully digital 5.4 billion transistor implementation leverages existing CMOS scaling trends, while ensuring one-to-one correspondence between hardware and software. With such aggressive design metrics and the TrueNorth architecture breaking path with prevailing architectures, it is clear that conventional computer-aided design (CAD) tools could not be used for the design. As a result, we developed a novel design methodology that includes mixed asynchronous-synchronous circuits and a complete tool flow for building an event-driven, low-power neurosynaptic chip. The TrueNorth chip is fully configurable in terms of connectivity and neural parameters to allow custom configurations for a wide range of cognitive and sensory perception applications. To reduce the system's communication energy, we have adapted existing application-agnostic very large-scale integration CAD placement tools for mapping logical neural networks to the physical neurosynaptic core locations on the TrueNorth chips. With that, we have successfully demonstrated the use of TrueNorth-based systems in multiple applications, including visual object recognition, with higher performance and orders of magnitude lower power consumption than the same algorithms run on von Neumann architectures. The TrueNorth chip and its tool flow serve as building blocks for future cognitive systems, and give designers an opportunity to develop novel brain-inspired architectures and systems based on the knowledge obtained from this paper.
Expressing emotions through various modalities is a crucial function not only for humans but also for robots. The mapping method from facial expressions to the basic emotions is widely used in ...research on robot emotional expressions. This method claims that there are specific facial muscle activation patterns for each emotional expression and people can perceive these emotions by reading these patterns. However, recent research on human behavior reveals that some emotional expressions, such as the emotion "intense", are difficult to judge as positive or negative by just looking at the facial expression alone. Nevertheless, it has not been investigated whether robots can also express ambiguous facial expressions with no clear valence and whether the addition of body expressions can make the facial valence clearer to humans. This paper shows that an ambiguous facial expression of an android can be perceived more clearly by viewers when body postures and movements are added. We conducted three experiments and online surveys among North American residents with 94, 114 and 114 participants, respectively. In Experiment 1, by calculating the entropy, we found that the facial expression "intense" was difficult to judge as positive or negative when they were only shown the facial expression. In Experiments 2 and 3, by analyzing ANOVA, we confirmed that participants were better at judging the facial valence when they were shown the whole body of the android, even though the facial expression was the same as in Experiment 1. These results suggest that facial and body expressions by robots should be designed jointly to achieve better communication with humans. In order to achieve smoother cooperative human-robot interaction, such as education by robots, emotion expressions conveyed through a combination of both the face and the body of the robot is necessary to convey the robot's intentions or desires to humans.
Abstract
Synchronization between humans is often observed in our daily lives, for example in breathing, in hand clapping in crowds, and in walking. It has been reported that pedestrian gait ...synchronization maximizes walking flow efficiency. As increasingly more mobile robots are being developed for practical use, it is important to consider how robots may impact pedestrian flows. While there is research on synchronization phenomena between humans and robots, gait synchronization between humans and robots has yet to be studied, particularly synchronization occurring with wheeled humanoid robots while moving. In this paper, we investigated the gait phase synchronization between humans and a wheeled mobile humanoid robot, which moved its upper body in three distinct types of motion patterns: (1) no-motion, (2) arm-swinging (as is common for typical mobile humanoids), and (3) arms-swinging in addition to periodic vertical-oscillation similar to the human upper body movement while walking. Rayleigh test was performed on the distribution of the obtained gait phase differences under each condition and a significant distributional bias was confirmed when participants were walking with the robot that performed both arm-swinging and vertical-oscillation of the upper body. These results suggest that humans can spontaneously synchronize their gaits with wheeled robots that utilize upper body oscillating. These findings can be important for the design of robot-integrated urban transportation systems, such as train stations and airports, where both humans and robots are mobile and a highly efficient flow is required.
We investigated whether multiple food allergies could be safely prevented by simultaneously administering very small amounts of multiple foods.
Infants 3–4 months old with atopic dermatitis from 14 ...primary care pediatric clinics in Japan were enrolled in this randomized, placebo-controlled trial. The infants were administered either mixed allergenic food powder (MP) containing egg, milk, wheat, soybean, buckwheat, and peanuts, or placebo powder (PP). The amount of powder was increased in a stepwise manner on weeks 2 and 4, and continued until week 12. The occurrence of food allergy episodes after powder intervention was assessed at 18 months old. This trial was registered with the University Hospital Medical Information Network Clinical Trials Registry (number UMIN000027837).
A total of 163 participants were randomly allocated to either the MP group (n = 83) or the PP group (n = 80). The incidence of food allergy episodes by 18 months was significantly different between the MP and PP groups (7/83 vs. 19/80, respectively; risk ratio 0.301 95% CI 0.116–0.784; P = 0.0066). Egg allergies were reduced in the MP group. In addition, food allergy episodes from any of the other five foods were significantly reduced, although the reductions in those due to individual foods were not significant.
Gradually increasing the intake of very small amounts of multiple foods in early infancy can safely reduce the incidence of egg allergies. Other foods may also suppress food allergies, but no definitive conclusions could be reached.

The follicle-associated epithelium (FAE) covering mucosa-associated lymphoid tissue is distinct from the villous epithelium in cellular composition and functions. Interleukin-22 binding protein ...(IL-22BP), provided by dendritic cells at the sub-epithelial dome region, inhibits the IL-22-mediated secretion of antimicrobial peptides by the FAE. The Notch signal from stromal cells underneath the FAE diminishes goblet cell differentiation. These events dampen the mucosal barrier functions to allow luminal microorganisms to readily gain access to the luminal surface of the FAE. Furthermore, receptor activator of nucleic factor-kappa B ligand (RANKL) from a certain stromal cell type induces differentiation into microfold (M) cells that specialize in antigen uptake in the mucosa. Microfold (M) cells play a key role in mucosal immune surveillance by actively transporting external antigens from the gut lumen to the lymphoid follicle. The molecular basis of antigen uptake by M cells has been gradually identified in the last decade. For example, GPI-anchored molecules (e.g., glycoprotein 2 (GP2) and cellular prion protein (PrP
)) and β1-integrin facilitate the transport of specific types of xenobiotics. The antigen transport by M cells initiates antigen-specific mucosal immune responses represented by the induction of secretory immunoglobulin A (S-IgA). Meanwhile, several invasive pathogens exploit M cells as a portal to establish a systemic infection. Recent findings have uncovered the molecular machinery of differentiation and functions of M cells.
This manuscript reviews seismic isolation and response control methods of buildings, which are able to make buildings resilient against earthquakes and have become popular during the last three ...decades in Japan. Seismic isolation is a method of protecting a building from major earthquakes by installing isolators and energy absorbing devices under the superstructure. The manuscript describes three types of laminated rubber bearings and three kinds of damping devices. Seismic isolation provides not only structural safety, but also safety and security for people and properties in the building. Seismic isolation is also used for the retrofit of historic buildings. Response control methods utilize various kinds of dampers that are installed into a building and absorb vibration energy. The manuscript explains three foremost response control dampers: the steel hysteretic damper, the viscoelastic damper and the viscous fluid damper. The effects of seismic isolation and response control methods were verified through shaking table tests, structural health monitoring and earthquake response analyses.
