Here, in the present study, we evaluated the role of the TAZ domain-containing gene in response to various heavy metals. Initially, we found a total of 3, 7, 8, 9, 9, 9, 7, 14, 6, 10, and 6 proteins containing TAZ domain in stiff brome, millet, sorghum, potato, pepper, maize, rice, apple, peach, pear, and tomato genome that could trigger the plant resistance against various heavy metals, respectively. Various in-silico approaches were applied such as duplication, phylogenetic analysis, and gene structure, to understand the basic features of the TAZ domain-containing genes in plants. Gene expression analyses were also performed under heavy metals (Cr, Zn, Ni, Cd, Co, Fe, Mn, and Pb). The results of quantitative real-time PCR analysis indicated that the TAZ gene family members were differentially expressed under different heavy metals. We further characterized the functions of the TAZ domain-containing gene under the heavy metal stresses by overexpressing the OsTAZ4 gene in Arabidopsis. The TAZ genes could promote plant resistance against various heavy metals by interacting with OsMYB34 and OsFHA9 transcription factors. The results will contribute to elucidate the relationship of TAZ proteins with heavy metals stresses and also ascertain the biological function in plant growth and development.

Liquid Chromatography tandem mass spectrometry (LC-MS/MS) is used for the diagnosis of more than 30 inborn errors of metabolisms (IEMs). Accurate and reliable diagnosis of IEMs by quantifying amino ...acids (AAs) and acylcarnitines (ACs) using LC-MS/MS systems depend on the establishment of age-specific cut-offs of the analytes. This study aimed to (1) determine the age-specific cut-off values of AAs and ACs in Bangladesh and (2) validate the LC-MS/MS method for diagnosis of the patients with IEMs. A total of 570 enrolled healthy participants were divided into 3 age groups, namely, (1) newborns (1-7 days), (2) 8 days–7 years, and (3) 8–17 years, to establish the age-specific cut-offs for AAs and ACs. Also, 273 suspected patients with IEMs were enrolled to evaluate the reliability of the established cut-off values. Quantitation of AAs and ACs was performed on an automated LC-MS/MS system using dried blood spot (DBS) cards. Then the specimens of the enrolled clinically suspected patients were analyzed by the established method. Nine patients came out as screening positive for different IEMs, including two borderline positive cases of medium-chain acyl-CoA dehydrogenase deficiency (MCAD). A second-tier test for confirmation of the screening positive cases was conducted by urinary metabolic profiling using gas chromatography- mass spectrometry (GC-MS). Out of 9 cases that came out as screening positive by LC-MS/MS, seven cases were confirmed by urinary GC-MS analysis including 3 cases with phenylketonuria, 1 with citrullinemia type II, 1 with methylmalonic acidemia, 1 with isovaleric acidemia and 1 with carnitine uptake defect. Two borderline positive cases with MCAD were found negative by urinary GC-MS analysis. In conclusion, along with establishment of a validated LC-MS/MS method for quantitation of AAs and ACs from the DBS cards, the study also demonstrates the presence of predominantly available IEMs in Bangladesh.
Background. Night eating syndrome is common practice in the community. The purpose of this study is to determine the prevalence of night eating syndrome among medical students and its correlation ...with depression at a public sector medical university in Karachi, Pakistan. Methodology. It is a cross sectional study and the participants were selected by using multi-stage cluster sampling. Night Eating Syndrome Questionnaire (NES-Q) and Beck depression scale were used to determine the night eating syndrome and depression. Students of age group between 20 and 25 years were included. Comparison of night eating syndrome and depression between groups of students was determined through chi square and Mann-Whitney U test. Spearmen correlation was used to determine the correlation of night eating syndrome with depression. P value <0.05 was considered significant. Results. The mean age of participants was 21.48±4.08 years. Among students 49.36% and 27.84% were suffering from night eating syndrome and depression respectively (p < 0.05). Students were suffering form night eating syn-drome had significantly higher Beck Depression Inventory (BDI) score compared to Non NEQ group with p value 0.03. A significant correlation was found between night eating syndrome scores and Beck depression inventory scale with p value was ≤ 0.05. Conclusions. Results of this study found that medical students had high prevalence rate of night eating syndrome and depression. Depression was found to be a predictor of night eating syndrome.
The modern trend of research is highly focused on finding new bioactive molecules from medicinal plants. As a functional bicyclic monoterpene, Bornyl acetate (BA) has displayed antioxidant and ...anti-inflammatory properties in different types of cells and tissues. The purpose of this research was to evaluate the probable hypotensive effect of BA, an underlying mechanism(s) backboned by in-silico studies. Mean arterial pressure and heart rate were recorded via invasive blood pressure measuring technique in normotensive Sprague-Dawley rats following the administration of BA (1-80mg/kg). Docking studies were carried out with various targets involved in the pathophysiology of hypertension.RO5 and ADMET properties were also evaluated. In the current study dose-dependent reduction in systolic, diastolic and mean arterial pressure was observed. Pretreatment with atropine and captopril significantly (p<0.001) reduced the hypotensive effect produced by BA. On the other hand docking studies showed pronounced interactions with M2 mAch receptor in an agonistic way and ACE protein in an antagonistic way. BA justified all cut-off limits of RO5 and had an acceptable predicted computational toxicity profile. Results postulate that dose-dependent hypotensive effect of BA is mediated through the muscarinic pathway and ACE inhibitory activity corresponding well with findings of in-silico studies.
Background: Otalgia, one of the most common complain reported in OPD of otorhinolaryngologist, is agonizing and affects all age groups. The objective of the study was to highlight the use of ...Multidisciplinary team approach to the management of otalgia patients for speedy recovery. Methods: A cross sectional study was conducted on patients having ear pain. These patients presented to the out-patient department (OPD) of otolaryngology of a tertiary care hospital. Data was collected through questionnaire prepared in advance. Informed oral consent was taken from participants before administration of the questionnaire. Results: A total of 216 patients were included in the study. Out of these only 33(15.2%) were less than 18 years of age. Majority 139(64.35%) of subjects belonged to a low socioeconomic class. Out of these patients 119(55.1%) had ear infections who has to be seen by otolaryngologists. 63(29.2%) of patients had temporomandibular joint pain, referred to the Maxillofacial Surgeons and 34(15.7%) of patients suffering from neck pain, referred to Orthopaedic Surgeon. Conclusion: Multidisciplinary team approach was found effective in treating otalgia patients visiting the otorhinolaryngology clinics for the symptom, should be closely examined and referred to concerned departments if required for complete recovery.
