Health conditions in the Huainan Coal Mine have improved greatly since 1950. In particular, the work and living environments, the underground dust concentration, the water supply and waste disposal ...systems, and the nutritional value of miners' food have become much better. Thus the rates of occupational, infectious, and parasitic diseases and nutritional deficiency diseases have decreased considerably among miners. The decreases in the prevalence of anthracosilicosis and hookworm have been especially striking.
To study the effects of losartan (Los) and captopril (Cap) treatment on expression of cardiac angiotensin II (Ang) AT1 receptor mRNA in rats following myocardial infarction (MI).
Twenty-four rats ...with MI after coronary ligation for 7 d were randomly divided into 4 groups: A) Cap in drinking water, ad lib (2 g.L-1), B) i.g. Los 10 mg.kg-1.d-1, C) i.g. Los 30 mg.kg-1.d-1, and D) placebo for 6 wk. Sham-ligation rats (group E) served as controls. The levels of cardiac Ang AT1 receptor mRNA expression in each group (n = 6) were examined by Dot blot using digoxigenin-labeled cDNA probes.
Comparing with reflected peak areas of hybridization positive signals in group D (2640 +/- 201 micron 2), the expression of the cardiac Ang AT1 receptor mRNA was much lower in the 3 treated groups (group A 1360 +/- 134 micron 2, group B 1430 +/- 244 micron 2, group C 1310 +/- 95 micron 2) (P < 0.01). But no difference was found between the 3 treated groups and sham-ligation group (1230 +/- 233 micron 2) (P > 0.05).
Los and Cap attenuated the increase of cardiac Ang AT1 receptor mRNA expression in rats following MI.
A recombinant human tumour necrosis factor (rhTNF) cDNA was constructed. The TNF gene was isolated from a human genomic gene library. There are four exons in the TNF gene. The fourth exon codes for ...140 amino acids of the TNF matured protein which is composed of 157 amino acids. A major portion of the fourth exon was isolated and then ligated to a synthesized DNA fragment coding for the remaining amino acids. The partial synthetic hTNF (rhTNF) cDNA thus generated was subcloned into a vector and successfully expressed in E. coli. 5-1 fermentator was used to produce rhTNF. About 20 g (wet weight) of bacterial pellet per liter medium and 10(6)-10(7) units of cytotoxicity to L929 cells per milliliter medium were obtained. rhTNF was purified by HPLC and dried with a freeze dryer. rhTNF with a purity of about 95% in the form of white powder was obtained. The sequence of ten amino acids at the amino terminus of the rhTNF was determined. The result showed that it was identical with that of the natural human TNF.
An experimental study of the effect of acute biliary hypertension on hemodynamics and activity of the major splanchnic nerve was conducted in Japanese big ear white rabbits. A catheter with an ...inflatable rubber bag fixed to its anterior end was inserted into the extrahepatic biliary duct via the duodenum, and a biliary high pressure of 20 kFa (150 mmHg) was created and maintained for 2 h by inflating the bag with water. The right major splanchnic nerve was isolated and the impulse frequencies of the nerve were recorded during the study. Arterial blood pressure was also dynamically monitored. A significant fall of the arterial blood pressure (P less than 0.01) and an increase in impulse frequency of the nerve (P less than 0.01) were found in this "simple type" acute biliary high pressure without infection. Biliary decompression immediately eliminated these abnormalities (P less than 0.01). The drop in blood pressure was much less prominent if the right splanchnic nerve was blocked prior to biliary high pressure.
