Enhancing crop productivity and their nutritional quality are the key components and primary focus of crop improvement strategy for fulfilling future food demand and improving human health. Grain ...filling and endosperm development are the key determinants of grain yield and nutritional quality. GRAIN WIDTH and WEIGHT2 (GW2) gene encodes a RING-type E3 ubiquitin ligase and determines the grain weight in cereal crops. Here we report GW2 knockout (KO) mutants in Indica (var. MTU1010) through CRISPR/Cas9 genome editing. The endosperm of GW2-KO mutant seed displays a thick aleurone layer with enhanced grain protein content. Further the loss of function of OsGW2 results in improved accumulation of essential dietary minerals (Fe, Zn, K, P, Ca) in the endosperm of rice grain. Additionally, the mutants displayed an early growth vigour phenotype with an improved root and shoot architecture. The hull morphology of GW2-KO lines also showed improved, grain filling thereby promoting larger grain architecture. Together, our findings indicate that GW2 may serve as a key regulator of improved grain architecture, grain nutritional quality and an important modulator of plant morphology. The study offers a strategy for the development of improved rice cultivars with enriched nutritional quality and its possible implementation in other cereals as well.
Objectives The objective of this work is to evaluate the hemodynamic performance of a new Y-graft modification of the extracardiac conduit Fontan operation. The performance of the Y-graft design is ...compared to two designs used in current practice: a t-junction connection of the venae cavae and an offset between the inferior and superior venae cavae. Methods The proposed design replaces the current tube grafts used to connect the inferior vena cava to the pulmonary arteries with a Y-shaped graft. Y-graft hemodynamics were evaluated at rest and during exercise with a patient-specific model from magnetic resonance imaging data together with computational fluid dynamics. Four clinically motivated performance measures were examined: Fontan pressures, energy efficiency, inferior vena cava flow distribution, and wall shear stress. Two variants of the Y-graft were evaluated: an “off-the-shelf” graft with 9-mm branches and an “area-preserving” graft with 12-mm branches. Results Energy efficiency of the 12-mm Y-graft was higher than all other models at rest and during exercise, and the reduction in efficiency from rest to exercise was improved by 38%. Both Y-graft designs reduced superior vena cava pressures during exercise by as much as 5 mm Hg. The Y-graft more equally distributed the inferior vena cava flow to both lungs, whereas the offset design skewed 70% of the flow to the left lung. The 12-mm graft resulted in slightly larger regions of low wall shear stress than other models; however, minimum shear stress values were similar. Conclusions The area-preserving 12-mm Y-graft is a promising modification of the Fontan procedure that should be clinically evaluated. Further work is needed to correlate our performance metrics with clinical outcomes, including exercise intolerance, incidence of protein-losing enteropathy, and thrombus formation.
Glutamine synthetase (GS) is a key enzyme involved in the nitrogen metabolism of higher plants. Abiotic stresses have adverse effects on crop production and pose a serious threat to global food ...security. GS activity and expression is known to be significantly modulated by various abiotic stresses. However, very few transgenic overexpression studies of GS have studied its impact on abiotic stress tolerance. GS is also the target enzyme of the broad spectrum herbicide Glufosinate (active ingredient: phosphinothricin). ) and chloroplastic GS2 (
) genes in transgenic rice on its tolerance to abiotic stresses and the herbicide Glufosinate. Our results demonstrate that the co-overexpression of
and
isoforms in transgenic rice plants enhanced its tolerance to osmotic and salinity stress at the seedling stage. The transgenic lines maintained significantly higher fresh weight, chlorophyll content, and relative water content than wild type (
) and null segregant (
) controls, under both osmotic and salinity stress. The
co-overexpressing transgenic plants accumulated higher levels of proline but showed lower electrolyte leakage and had lower malondialdehyde (MDA) content under the stress treatments. The transgenic lines showed considerably enhanced photosynthetic and agronomic performance under drought and salinity stress imposed during the reproductive stage, as compared to
and
control plants. The grain filling rates of the transgenic rice plants under reproductive stage drought stress (64.6 ± 4.7%) and salinity stress (58.2 ± 4.5%) were significantly higher than control plants, thereby leading to higher yields under these abiotic stress conditions. Preliminary analysis also revealed that the transgenic lines had improved tolerance to methyl viologen induced photo-oxidative stress. Taken together, our results demonstrate that the concurrent overexpression of
and
isoforms in rice enhanced physiological tolerance and agronomic performance under adverse abiotic stress conditions, apparently acting through multiple mechanistic routes. The transgenic rice plants also showed limited tolerance to the herbicide Glufosinate. The advantages and limitations of glutamine synthetase overexpression in crop plants, along with future strategies to overcome these limitations for utilization in crop improvement have also been discussed briefly.

The availability of orthophosphate (Pi) is a key determinant of crop productivity because its accessibility to plants is poor due to its conversion to unavailable forms. Weed's competition ...for this essential macronutrient further reduces its bio‐availability. To compensate for the low Pi use efficiency and address the weed hazard, excess Pi fertilizers and herbicides are routinely applied, resulting in increased production costs, soil degradation and eutrophication. These outcomes necessitate the identification of a suitable alternate technology that can address the problems associated with the overuse of Pi‐based fertilizers and herbicides in agriculture. The present review focuses on phosphite (Phi) as a novel molecule for its utility as a fertilizer, herbicide, biostimulant and biocide in modern agriculture. The use of Phi‐based fertilization will help to reduce the consumption of Pi fertilizers and facilitate weed and pathogen control using the same molecule, thereby providing significant advantages over current orthophosphate‐based fertilization.
Objectives In this study, with the use of computational fluid dynamics, we evaluate the postoperative hemodynamic performance of the first cohort of patients undergoing a handcrafted Y-graft Fontan ...procedure and validate simulation predictions of hepatic blood flow distribution against in vivo clinical data. Methods An 18-12 × 2–mm handcrafted Y-graft modification of the Fontan procedure was performed in 6 patients. Early (at the time of discharge) and 6-month postoperative 3-dimensional magnetic resonance imaging data were collected. Patient-specific models were constructed for flow simulations. Results Hepatic blood flow distribution varied among patients. Lung perfusion data (n = 3) showed good agreement with simulations. Postoperative asymmetry in hepatic blood flow distribution was reduced 6 months postoperatively. In 1 patient, low wall shear stress was found in the left limb of the Y-graft, corresponding to the location of subsequent thrombosis in the patient. Conclusions The credibility and accuracy of simulation-based predictions of postoperative hepatic flow distribution for the Fontan surgery have been validated by in vivo lung perfusion data. The performance of the Y-graft design is highly patient-specific. The anastomosis location is likely the most important factor influencing hepatic blood flow distribution. Although the development of thrombosis is multifactorial, the occurrence in 1 patient suggests that simulations should not solely consider the hepatic blood flow distribution but also aim to avoid low wall shear stress in the limbs.
In this study, an attempt is made to check whether a hybrid composite made up of both synthetic and natural fibers (Carbon and Cordia Dichotoma respectively) can be made bio-degradable, at least to ...some extent, without much compromising on the mechanical properties. Hybrid composites were prepared by reinforcing alkali treated Cordia dichotoma and carbon fibers into epoxy resin using hand lay-up method. By varying the number of layers of fibers in the composite specimen and fixing 20% fiber weight for all composites, nine distinct combinations of specimens were prepared. The maximum tensile strength of 386.68 MPa, flexural strength of 647.08 MPa, and impact energy of 4.82 J were obtained for composites produced with pure carbon fiber, whereas hybrid composite exhibit tensile strength of 367.76 MPa, flexural strength of 646.41MPa and impact energy of 4.74 J. The interfacial bonding between the fibers and matrix of tested specimens was studied using a scanning electron microscope (SEM), as well as the arrangement of fibers within the matrix for the manufactured composite. Thermogravimetric analysis (TGA) was used to investigate thermal stability, and it was found that it was thermally stable up to 415°C. 9. Crystallinity value increases from 20% carbon fiber to 20% alkali fiber.
Water pollution by industrial and anthropogenic actives has become a serious threat to the environment. World Health Organization (WHO) has identified that lead and fluoride amid the environmental ...pollutants are most poisonous water contaminants with devastating impact on the human race. The present work proposes a study on economical bio-adsorbent based technique using exhausted coffee grounds in the removal of lead and fluoride contaminants from water. The exhausted coffee grounds gathered from industrial wastes have been acid-activated and examined for their adsorption capacity. The surface morphology and elemental characterization of pre-and-post adsorption operations by FESEM, EDX and FTIR spectral analysis confirmed the potential of the exhausted coffee ground as successful bio-sorbent. However, thermodynamic analysis confirmed the adsorption to be spontaneous physisorption with Langmuir mode of homogenous monolayer deposition. The kinetics of adsorption is well defined by pseudo second order model for both lead and fluoride. A significant quantity of lead and fluoride is removed from the synthetic contaminated water by the proposed bio-sorbent with the respective sorption capabilities of 61.6 mg/g and 9.05 mg/g. However, the developed bio-sorbent is also recyclable and is capable of removing the lead and fluoride from the domestic and industrial waste-water sources with an overall removal efficiency of about 90%.

Tributyl phosphate (TBP) is commercially used in large volumes for reprocessing of spent nuclear fuel. TBP is a very stable compound and persistent in natural environments and it is not removed in conventional wastewater treatment plants. In this study, cultivation of aerobic granular biofilms in a sequencing batch reactor was investigated for efficient biodegradation of TBP. Enrichment of TBP-degrading strains resulted in efficient degradation of TBP as sole carbon or along with acetate. Complete biodegradation of 2mM of TBP was achieved within 5h with a degradation rate of 0.4μmolmL−1h−1. TBP biodegradation was accompanied by release of inorganic phosphate in stoichiometric amounts. n-Butanol, hydrolysed product of TBP was rapidly biodegraded. But, dibutyl phosphate, a putative intermediate of TBP degradation was only partially degraded pointing to an alternative degradation pathway. Phosphatase activity was 22- and 7.5-fold higher in TBP-degrading biofilms as compared to bioflocs and acetate-fed aerobic granules. Community analysis by terminal restriction length polymorphism revealed presence of 30 different bacterial strains. Seven bacterial stains, including Sphingobium sp. a known TBP degrader were isolated. The results show that aerobic granular biofilms are promising for treatment of TBP-bearing wastes or ex situ bioremediation of TBP-contaminated sites.
Objectives A novel Y-shaped baffle has been proposed for the Fontan operation with promising initial results. However, previous studies have relied either on idealized models or a single ...patient-specific model. The objective of this study is to comprehensively compare the hemodynamic performance and hepatic blood flow distribution of the Y-graft Fontan baffle with 2 current designs using multiple patient-specific models. Methods Y-shaped and tube-shaped grafts were virtually implanted into 5 patient-specific Glenn models forming 3 types of Fontan geometries: Y-graft, T-junction, and offset. Unsteady flow simulations were performed at rest and at varying exercise conditions. The hepatic flow distribution between the right and left lungs was carefully quantified using a particle tracking method. Other physiologically relevant parameters such as energy dissipation, superior vena cava pressure, and wall shear stress were evaluated. Results The Fontan geometry significantly influences the hepatic flow distribution. The Y-graft design improves the hepatic flow distribution effectively in 4 of 5 patients, whereas the T-junction and offset designs may skew as much as 97% of hepatic flow to 1 lung in 2 cases. Sensitivity studies show that changes in pulmonary flow split can affect the hepatic flow distribution dramatically but that some Y-graft and T-junction designs are relatively less sensitive than offset designs. The Y-graft design offers moderate improvements over the traditional designs in power loss and superior vena cava pressure in all patients. Conclusions The Y-graft Fontan design achieves overall superior hemodynamic performance compared with traditional designs. However, the results emphasize that no one-size-fits-all solution is available that will universally benefit all patients and that designs should be customized for individual patients before clinical application.
To test the hypothesis that delayed brain development in fetuses with d-transposition of the great arteries or hypoplastic left heart syndrome heightens their postnatal susceptibility to acquired ...white matter injury.
This is a cohort study across 3 sites. Subjects underwent fetal (third trimester) and neonatal preoperative magnetic resonance imaging of the brain to measure total brain volume as a measure of brain maturity and the presence of acquired white matter injury after birth. White matter injury was categorized as no-mild or moderate-severe based on validated grading criteria. Comparisons were made between the injury groups.
A total of 63 subjects were enrolled (d-transposition of the great arteries: 37; hypoplastic left heart syndrome: 26). White matter injury was present in 32.4% (n = 12) of d-transposition of the great arteries and 34.6% (n = 8) of those with hypoplastic left heart syndrome. Overall total brain volume (taking into account fetal and neonatal scan) was significantly lower in those with postnatal moderate-severe white matter injury compared with no-mild white matter injury after adjusting for age at scan and site in d-transposition of the great arteries (coefficient: 14.8 mL, 95% confidence interval, −28.8 to −0.73, P = .04). The rate of change in total brain volume from fetal to postnatal life did not differ by injury group. In hypoplastic left heart syndrome, no association was noted between overall total brain volume and change in total brain volume with postnatal white matter injury.
Lower total brain volume beginning in late gestation is associated with increased risk of postnatal moderate-severe white matter injury in d-transposition of the great arteries but not hypoplastic left heart syndrome. Rate of brain growth was not a risk factor for white matter injury. The underlying fetal and perinatal physiology has different implications for postnatal risk of white matter injury.
