RNA interference (RNAi) targeting lethal genes in insects has great potential for sustainable crop protection. Compared with traditional double‐stranded (ds)RNA delivery systems, nanoparticles such ...as chitosan, liposomes, and cationic dendrimers offer advantages in delivering dsRNA/small interfering (si)RNA to improve RNAi efficiency, thus promoting the development and practice of RNAi‐based pest management strategies. Here, we illustrate the limitations of traditional dsRNA delivery systems, reveal the mechanism of nanoparticle‐mediated RNAi, summarize the recent progress and successful applications of nanoparticle‐mediated RNAi in pest management, and finally address the prospects of nanoparticle‐based RNA pesticides.
The detection and characterization of extended structures is a crucial goal in high-contrast imaging. However, these structures face challenges in data reduction, leading to over-subtraction from ...speckles and self-subtraction with most existing methods. Iterative post-processing methods offer promising results, but their integration into existing pipelines is hindered by selective algorithms, the high computational cost, and algorithmic regularization. To address this for reference differential imaging (RDI), here we propose a data imputation concept for the Karhunen–Loève transform (DIKL) by modifying two steps in the standard Karhunen–Loève image projection (KLIP) method. Specifically, we partition an image to two matrices: an anchor matrix that focuses only on the speckles to obtain the DIKL coefficients, and a boat matrix that focuses on the regions of astrophysical interest for speckle removal using DIKL components. As an analytical approach, DIKL achieves high-quality results with significantly reduced computational cost (~3 orders of magnitude less than iterative methods). Being a derivative method of KLIP, DIKL is seamlessly integrable into high-contrast imaging pipelines for RDI observations.
Metallic nanoparticles (MNPs) with their diverse physical and chemical properties have been applied in various biomedical domains. The increasing demand for MNPs has attracted researchers to develop ...straightforward, inexpensive, simple, and eco-friendly processes for the enhanced production of MNPs. To discover new biomedical applications first requires knowledge of the interactions of MNPs with target cells. This review focuses on plant and microbial synthesis of biological MNPs, their cellular uptake, biocompatibility, any biological consequences such as cytotoxicity, and biomedical applications. We highlighted the involvement of biomolecules in capping and stabilization of MNPs and the effect of physicochemical parameters particularly the pH on the synthesis of MNPs. Recently achieved milestones to understand the role of synthetic biology (SynBiol) in the synthesis of tailored MNPs are also discussed.
Diabetes is a disorder of glucose metabolism, and over 90% are type 2 diabetes. Diabetic cardiomyopathy (DCM) is one of the type 2 diabetes complications, usually accompanied by changes in myocardial ...structure and function, together with cardiomyocyte apoptosis. Our study investigated the effect of curcumin on regulating oxidative stress (OS) and apoptosis in DCM. In vivo, diabetes was induced in an experimental rat model by streptozoticin (STZ) together with high‐glucose and high‐fat (HG/HF) diet feeding. In vitro, H9c2 cardiomyocytes were cultured with high‐glucose and saturated free fatty acid palmitate. Curcumin was orally or directly administered to rats or cells, respectively. Streptozoticin ‐induced diabetic rats showed metabolism abnormalities and elevated markers of OS (superoxide dismutase SOD, malondialdehyde MDA, gp91phox, Cyt‐Cyto C), enhanced cell apoptosis (Bax/Bcl‐2, Cleaved caspase‐3, TUNEL‐positive cells), together with reduced Akt phosphorylation and increased Foxo1 acetylation. Curcumin attenuated the myocardial dysfunction, OS and apoptosis in the heart of diabetic rats. Curcumin treatment also enhanced phosphorylation of Akt and inhibited acetylation of Foxo1. These results strongly suggest that apoptosis was increased in the heart of diabetic rats, and curcumin played a role in diabetic cardiomyopathy treatment by modulating the Sirt1‐Foxo1 and PI3K‐Akt pathways.
Surface-enhanced Raman spectroscopy (SERS) inherits the rich chemical fingerprint information on Raman spectroscopy and gains sensitivity by plasmon-enhanced excitation and scattering. In particular, ...most Raman peaks have a narrow width suitable for multiplex analysis, and the measurements can be conveniently made under ambient and aqueous conditions. These merits make SERS a very promising technique for studying complex biological systems, and SERS has attracted increasing interest in biorelated analysis. However, there are still great challenges that need to be addressed until it can be widely accepted by the biorelated communities, answer interesting biological questions, and solve fatal clinical problems. SERS applications in bioanalysis involve the complex interactions of plasmonic nanomaterials with biological systems and their environments. The reliability becomes the key issue of bioanalytical SERS in order to extract meaningful information from SERS data. This review provides a comprehensive overview of bioanalytical SERS with the main focus on the reliability issue. We first introduce the mechanism of SERS to guide the design of reliable SERS experiments with high detection sensitivity. We then introduce the current understanding of the interaction of nanomaterials with biological systems, mainly living cells, to guide the design of functionalized SERS nanoparticles for target detection. We further introduce the current status of label-free (direct) and labeled (indirect) SERS detections, for systems from biomolecules, to pathogens, to living cells, and we discuss the potential interferences from experimental design, measurement conditions, and data analysis. In the end, we give an outlook of the key challenges in bioanalytical SERS, including reproducibility, sensitivity, and spatial and time resolution.
Abstract
Background
The triglyceride-glucose index (TyG index) has been regarded as a reliable alternative marker of insulin resistance and an independent predictor of cardiovascular outcomes. ...Whether the TyG index predicts adverse cardiovascular events in patients with diabetes and acute coronary syndrome (ACS) remains uncertain. The aim of this study was to investigate the prognostic value of the TyG index in patients with diabetes and ACS.
Methods
A total of 2531 consecutive patients with diabetes who underwent coronary angiography for ACS were enrolled in this study. Patients were divided into tertiles according to their TyG index. The primary outcomes included the occurrence of major adverse cardiovascular events (MACEs), defined as all-cause death, non-fatal myocardial infarction and non-fatal stroke. The TyG index was calculated as the ln (fasting triglyceride level mg/dL × fasting glucose level mg/dL/2).
Results
The incidence of MACE increased with TyG index tertiles at a 3-year follow-up. The Kaplan–Meier curves showed significant differences in event-free survival rates among TyG index tertiles (P = 0.005). Multivariate Cox hazards regression analysis revealed that the TyG index was an independent predictor of MACE (95% CI 1.201–1.746; P < 0.001). The optimal TyG index cut-off for predicting MACE was 9.323 (sensitivity 46.0%; specificity 63.6%; area under the curve 0.560; P = 0.001). Furthermore, adding the TyG index to the prognostic model for MACE improved the C-statistic value (P = 0.010), the integrated discrimination improvement value (P = 0.001) and the net reclassification improvement value (P = 0.019).
Conclusions
The TyG index predicts future MACE in patients with diabetes and ACS independently of known cardiovascular risk factors, suggesting that the TyG index may be a useful marker for risk stratification and prognosis in patients with diabetes and ACS.

We apply the vectorized non-negative matrix factorization (NMF) method to the post-processing of the direct imaging data of exoplanetary systems such as circumstellar disks. NMF is an iterative ...approach, which first creates a nonorthogonal and non-negative basis of components using the given reference images and then models a target with the components. The constructed model is then rescaled with a factor to compensate for the contribution from the disks. We compare NMF with existing methods (classical reference differential imaging method, and the Karhunen-Loève image projection algorithm) using synthetic circumstellar disks and demonstrate the superiority of NMF: with no need of prior selection of references, NMF not only can detect fainter circumstellar disks but also better preserves their morphology and does not require forward modeling. As an application to a well-known disk example, we process the archival Hubble Space Telescope STIS coronagraphic observations of HD 181327 with different methods and compare them, and NMF is able to extract some circumstellar materials inside the primary ring for the first time. In an appendix, we mathematically investigate the stability of NMF components during the iteration and the linearity of NMF modeling.
In this paper, pattern division multiple access (PDMA), which is a novel nonorthogonal multiple access scheme, is proposed for fifth-generation (5G) radio networks. The PDMA pattern defines the ...mapping of transmitted data to a resource group that can consist of time, frequency, and spatial resources or any combination of these resources. The pattern is introduced to differentiate signals of users sharing the same resources, and the pattern is designed with disparate diversity order and sparsity so that PDMA can take the advantage of the joint design of transmitter and receiver to improve system performance while maintaining detection complexity to a reasonable level. System level simulation results show that PDMA can support six times simultaneous connections than that of conventional and at least 30% improvement in spectrum efficiency over orthogonal frequency division multiple access.
Endoplasmic reticulum stress (ERS) and unfolded protein response are the critical processes of tumour biology. However, the roles of ERS regulatory genes in pancreatic adenocarcinoma (PAAD) remain ...elusive. A novel ERS‐related risk signature was constructed using the Lasso regression analysis. Its prognostic value, immune effect, metabolic influence, mutational feature and therapeutic correlation were comprehensively analysed through multiple bioinformatic approaches. The biofunctions of KDELR3 and YWHAZ in pancreatic cancer (PC) cells were also investigated through colony formation, Transwell assays, flow cytometric detection and a xenograft model. The upstream miRNA regulatory mechanism of KDELR3 was predicted and validated. ERS risk score was identified as an independent prognostic factor and could improve traditional prognostic model. Meanwhile, it was closely associated with metabolic reprogramming and tumour immune. High ERS risk enhanced glycolysis process and nucleotide metabolism, but was unfavourable for anti‐tumour immune response. Moreover, ERS risk score could act as a potential biomarker for predicting the efficacy of ICBs. Overexpression of KDELR3 and YWHAZ stimulated the proliferation, migration and invasion of SW1990 and BxPC‐3 cells. Silencing KDELR3 suppressed tumour growth in a xenograft model. miR‐137 could weaken the malignant potentials of PC cells through inhibiting KDELR3 (5′‐AGCAAUAA‐3′). ERS risk score greatly contributed to PAAD clinical assessment. KDELR3 and YWHAZ possessed cancer‐promoting capacities, showing promise as a novel treatment target.
