Peppers (Capsicum spp.) are one of the most important vegetables and their double use (vegetable or spice) and two commercial stages (unripe and fully ripe) contributed to their use in many recipes ...and fast diffusion from America. Nowadays, Spain is a center of diversity for C. annuum, with many landraces, offering a great opportunity for adaptation to organic cultivation. Furthermore, Capsicum peppers contain many bioactive compounds, essential to provide high added-value to these cultivars, especially for organic markets, although knowledge about the effect of organic cultivation on Capsicum fruit quality is still scarce. Here, 37 accessions of Spanish landraces and foreign materials from C. annuum and other species were grown under organic and conventional conditions and evaluated for ascorbic acid (AAC), total phenolics (TP) and total red and yellow/orange carotenoids, considering both ripening stages. A large genotypic variation was found within each ripening stage and growing condition for the studied traits. Also, both stages showed high levels, although fully ripe fruits were the richest. Organic conditions enabled higher levels in fully ripe fruits of AAC and TP on average (135 vs 117 mg·100 g-1 and 232 vs 206 mg·100 g-1) and in most accessions, although the genotype×growing conditions interaction also contributed, but at lower extent, to the observed variation. Significant genotype×ripening stage and growing conditions×ripening stage interactions were also found, suggesting that the magnitude of the increase with ripening depends on the accession and growing conditions. By contrast, there were no differences between growing conditions for carotenoids and differences were mainly due to the genotype factor. Finally, the large genotypic variation and favourable organic conditions allowed identifying several materials from different types and uses with very high levels of bioactive compounds for organic cultivation, in both ripening stages but particularly at fully ripe stage (>500 mg·100 g-1).
Projected changes in precipitation extremes and their uncertainties are evaluated using an ensemble of global climate models from phase 6 of the Coupled Model Intercomparison Project (CMIP). They are ...scaled by corresponding changes either in global mean surface temperature (ΔGSAT) or in local surface temperature (ΔT) and are expressed in terms of 20-yr return values (RV20) of annual maximum one-day precipitation. Our main objective is to quantify the model response uncertainty and to highlight the regions where changes may not be consistent with the widely used assumption of a Clausius–Clapeyron (CC) rate of ≈7%/K. When using a single realization for each model, as in the latest report from the Intergovernmental Panel on Climate Change (IPCC), the assessed inter-model spread includes both model uncertainty and internal variability, which can be however assessed separately using a large ensemble. Despite the overestimated inter-model spread, our results show a robust enhancement of extreme precipitation with more than 90% of models simulating an increase of RV20. Moreover, this increase is consistent with the CC rate of ≈7%/K over about 83% of the global land domain when scaled by (ΔGSAT). Our results also advocate for producing multiple single model initial condition ensembles in the next CMIP projections, to better filter internal variability out in estimating the response of extreme events.
Changes in the global water cycle are expected as a result of anthropogenic climate change, but large uncertainties exist in how these changes will be manifest regionally. This is especially the case ...over the tropical oceans, where observed estimates of precipitation and evaporation disagree considerably. An alternative approach is to examine changes in near-surface salinity. Datasets of observed tropical Pacific and Atlantic near-surface salinity combined with climate model simulations are used to assess the possible causes and significance of salinity changes over the late twentieth century. Two different detection methodologies are then applied to evaluate the extent to which observed large-scale changes in near-surface salinity can be attributed to anthropogenic climate change.
Basin-averaged observed changes are shown to enhance salinity geographical contrasts between the two basins: the Pacific is getting fresher and the Atlantic saltier. While the observed Pacific and interbasin-averaged salinity changes exceed the range of internal variability provided from control climate simulations, Atlantic changes are within the model estimates. Spatial patterns of salinity change, including a fresher western Pacific warm pool and a saltier subtropical North Atlantic, are not consistent with internal climate variability. They are similar to anthropogenic response patterns obtained from transient twentieth- and twenty-first-century integrations, therefore suggesting a discernible human influence on the late twentieth-century evolution of the tropical marine water cycle. Changes in the tropical and midlatitudes Atlantic salinity levels are not found to be significant compared to internal variability. Implications of the results for understanding of the recent and future marine tropical water cycle changes are discussed.
Purpose
This paper aims to contrast the expectations placed on the smart destination as a theoretical management approach with the reality of emergent tech-savvy tourism demand by examining the ...response of this segment to three critical dimensions of technology use in the context of smart destinations. Tech-savvy tourists are here represented by highly educated Spanish millennial tourists.
Design/methodology/approach
Data were obtained through an online survey and analysed through descriptive techniques and exploratory factor analysis using SPSS. Literature review was critical for setting the foundations of the research in this novel context.
Findings
Results suggest the existence of a gap between the smart destination theoretical expectations and created hype and the real response of the examined demand in the three main scopes for this tourist-smart destination technology-based relationship, namely, mobile technology use, data sharing for personalised experiences and smart technologies for enhanced experiences.
Research limitations/implications
Convenience sampling was used, and the results of the study cannot be generalised to all millennial tourists. The research is a first approximation to the interrelationship between tourists and smart destinations.
Practical/implications
Tourists’ role and experiences mediated by information and communication technologies (ICTs) are decisive in smart destinations. Destination Management Organisations (DMOs), through their policies and actions, ought to take into account the limited predisposition of tourists in using mobile devices, sharing data and using smart technologies for their experiences. Privacy concerns appear to have special relevance for tourists and, therefore, for the future of smart destinations.
Originality/value
The findings offer relevant insights for smart destinations from a neglected angle hitherto, as they introduce several interesting nuances which do not match entirely the fast track taken by institutions, media and academia. This is critical for better understanding tourists in the current panorama, for DMOs and for the theoretical foundations of smart tourism. Besides, the exploratory data analysis reveals potential dimensions of millennials’ behaviour, which can be useful for further investigations.
Purpose: The present study assessed the role of an amino acid-based formula (AAF) in the growth of infants with cow's milk protein allergy (CMPA). Methods: Non-breastfed, term infants aged 0-6 months ...with symptoms suggestive of CMPA were recruited from 10 pediatric centers in China. After enrollment, infants were started on AAF for two weeks, followed by an open food challenge (OFC) with cow's milk-based formula (CMF). Infants with confirmed CMPA remained on AAF until 9 months of age, in conjunction with a cow's milk protein-free complementary diet. Body weight, length, and head circumference were measured at enrollment and 9 months of age. Measurements were converted to weight-for-age, length-for-age, and head circumference-for-age Z scores (WAZ, LAZ, HCAZ), based on the World Health Organization growth reference. Results: Of 254 infants (median age 16.1 weeks, 50.9% male), 218 (85.8%) were diagnosed with non-IgE-mediated CMPA, 33 (13.0%) tolerated CMF, and 3 (1.2%) did not complete the OFC. The mean WAZ decreased from 0.119 to -0.029 between birth and enrollment (p=0.067), with significant catch-up growth to 0.178 at 9 months of age (p=0.012) while being fed the AAF. There were no significant changes in LAZ (0.400 vs. 0.552; p=0.214) or HCAZ (-0.356 vs. -0.284; p=0.705) from the time of enrollment to age 9 months, suggesting normal linear and head growth velocity. Conclusion: The amino acid-based study formula, in conjunction with a cow's milk proteinfree complementary diet, supported normal growth till 9 months of age in a cohort of Chinese infants with challenge-confirmed non-IgE-mediated CMPA.
Cerebral organoids are 3D stem cell-derived models that can be utilized to study the human brain. The current consensus is that cerebral organoids consist of cells derived from the neuroectodermal ...lineage. This limits their value and applicability, as mesodermal-derived microglia are important players in neural development and disease. Remarkably, here we show that microglia can innately develop within a cerebral organoid model and display their characteristic ramified morphology. The transcriptome and response to inflammatory stimulation of these organoid-grown microglia closely mimic the transcriptome and response of adult microglia acutely isolated from post mortem human brain tissue. In addition, organoid-grown microglia mediate phagocytosis and synaptic material is detected inside them. In all, our study characterizes a microglia-containing organoid model that represents a valuable tool for studying the interplay between microglia, macroglia, and neurons in human brain development and disease.
Multiple angiogenesis inhibitors have been therapeutically validated in preclinical cancer models, and several in clinical trials. Here we report that angiogenesis inhibitors targeting the VEGF ...pathway demonstrate antitumor effects in mouse models of pancreatic neuroendocrine carcinoma and glioblastoma but concomitantly elicit tumor adaptation and progression to stages of greater malignancy, with heightened invasiveness and in some cases increased lymphatic and distant metastasis. Increased invasiveness is also seen by genetic ablation of the
Vegf-A gene in both models, substantiating the results of the pharmacological inhibitors. The realization that potent angiogenesis inhibition can alter the natural history of tumors by increasing invasion and metastasis warrants clinical investigation, as the prospect has important implications for the development of enduring antiangiogenic therapies.
Describing the relationship between a weather event and climate change—a science usually termed event attribution—involves quantifying the extent to which human influence has affected the frequency ...or the strength of an observed event. In this study we show how event attribution can be implemented through the application of nonstationary statistics to transient simulations, typically covering the 1850–2100 period. The use of existing CMIP-style simulations has many advantages, including their availability for a large range of coupled models and the fact that they are not conditional to a given oceanic state. We develop a technique for providing a multimodel synthesis, consistent with the uncertainty analysis of long-term changes. Last, we describe how model estimates can be combined with historical observations to provide a single diagnosis accounting for both sources of information. The potential of this new method is illustrated using the 2003 European heat wave and under a Gaussian assumption. Results suggest that (i) it is feasible to perform event attribution using transient simulations and nonstationary statistics, even for a single model; (ii) the use of multimodel synthesis in event attribution is highly desirable given the spread in single-model estimates; and (iii) merging models and observations substantially reduces uncertainties in human-induced changes. Investigating transient simulations also enables us to derive insightful diagnostics of how the targeted event will be affected by climate change in the future.
Therapy-induced resistance remains a significant hurdle to achieve long-lasting responses and cures in cancer patients. We investigated the long-term consequences of genetically impaired angiogenesis ...by engineering multiple tumor models deprived of endoglin, a co-receptor for TGF-β in endothelial cells actively engaged in angiogenesis. Tumors from endoglin-deficient mice adapted to the weakened angiogenic response, and refractoriness to diminished endoglin signaling was accompanied by increased metastatic capability. Mechanistic studies in multiple mouse models of cancer revealed that deficiency for endoglin resulted in a tumor vasculature that displayed hallmarks of endothelial-to-mesenchymal transition, a process of previously unknown significance in cancer biology, but shown by us to be associated with a reduced capacity of the vasculature to avert tumor cell intra- and extravasation. Nevertheless, tumors deprived of endoglin exhibited a delayed onset of resistance to anti-VEGF (vascular endothelial growth factor) agents, illustrating the therapeutic utility of combinatorial targeting of multiple angiogenic pathways for the treatment of cancer.
