Antibody-dependent cell-mediated cytotoxicity, one of the most prominent modes of action of antitumor antibodies, suffers from important limitations due to the need for optimal interactions with Fcγ ...receptors. In this work, we report the design of a new bispecific antibody format, compact and linker-free, based on the use of llama single-domain antibodies that are capable of circumventing most of these limitations. This bispecific antibody format was created by fusing single-domain antibodies directed against the carcinoembryonic antigen and the activating FcγRIIIa receptor to human Cκ and CH1 immunoglobulin G1 domains, acting as a natural dimerization motif. In vitro and in vivo characterization of these Fab-like bispecific molecules revealed favorable features for further development as a therapeutic molecule. They are easy to produce in Escherichia coli, very stable, and elicit potent lysis of tumor cells by human natural killer cells at picomolar concentrations. Unlike conventional antibodies, they do not engage inhibitory FcγRIIb receptor, do not compete with serum immunoglobulins G for receptor binding, and their cytotoxic activity is independent of Fc glycosylation and FcγRIIIa polymorphism. As opposed to anti-CD3 bispecific antitumor antibodies, they do not engage regulatory T cells as these latter cells do not express FcγRIII. Studies in nonobese diabetic/severe combined immunodeficient gamma mice xenografted with carcinoembryonic antigen-positive tumor cells showed that Fab-like bispecific molecules in the presence of human peripheral blood mononuclear cells significantly slow down tumor growth. This new compact, linker-free bispecific antibody format offers a promising approach for optimizing antibody-based therapies.

Cependant, l'un des modes d'action les plus ...importants de ces molécules, la cytotoxicité à médiation cellulaire dépendante des anticorps (ADCC), souffre de plusieurs limites en raison de la nécessité d'une interaction optimale avec le FcγRIIIA. L'équipe du Dr. Daniel Baty a conçu de nouveaux formats d'anticorps bispécifiques basés sur l'utilisation de domaine unique d'anticorps de lamas (sdAb) et capables de contourner la plupart de ces limites. Des banques de phages issus de lamas immunisés ont permis de sélectionner deux sdAbs (d'affinités différentes) dirigés contre le FcγRIIIA exprimé par des cellules NK et les macrophages, ainsi qu'un sdAb dirigé contre l'antigène tumoral carcino-embryonnaire (ACE). En utilisant les domaines CH1 et Ck d'IgG1 humaine comme motif d'hétérodimérisation et les sdabs anti-ACE et anti-FcγRIII précédemment sélectionnés, nous avons développé des formats innovants d'anticorps bispécifiques Fab-like nommés bsFabs. Ces molécules sont faciles à produire, très stables et peuvent déclencher la lyse des cellules tumorales par les cellules NK humaines à des concentrations de l'ordre du picomolaire. With fourteen antibodies used in oncology, monoclonal antibodies finally have a place in the therapeutic arsenal against cancer. However one of the modes of action of the most important of these molecules, cell-mediated cytotoxicity antibody-dependent (ADCC), suffers from several limitations due to the need for optimal interaction with the FcγRIIIA. Daniel Baty's team has developed new formats of bispecific antibodies based on the use of single domain llama antibodies (SdAb) that are capable of circumventing most of the these limitations. Phage libraries from immunized llamas were used to select two sdAbs directed against the FcγRIIIA expressed by NK cells and macrophages (with different affinities for FcγRIII), as well as one SdAb directed against the tumor antigen carcinoembryonic (CEA). Using CH1 and Ck domains of human IgG1 as a motif for heterodimerization and sdabs previously selected, we have developed innovative anti-CEA and anti-FcγRIII formats of bispecific antibodies Fab-like named bsFabs. These molecules are easy to produce, very stable and can trigger tumor cell lysis by human NK cells at picomolar concentrations. They do not bind FcγRIIB inhibitory receptor, do not compete with serum IgG and their cytotoxic activity is independent of FcγRIIIA polymorphism. In addition, they slow tumor growth in mouse model. In terms of pharmacokinetics, although bsFabs have a reasonable tumor retention, one of the limitations of these formats is size, which leads to rapid renal elimination. Such findings led us to consider the creation of new antibody formats to both increase tumor retention and secondly the half-life of antibodies in the body.
Silvoarable systems (SAS) are agroforestry systems (AFS) that combine the cultivation of trees and arable crops. Often, rows of trees delineate crop alleys in a field (alley-cropping system). SAS ...have the potential to provide many agricultural and environmental benefits, are innovative in their modern form, and are becoming more widespread. These diversified farming systems have the potential to influence biodiversity and related ecosystem services (ES). This systematic review seeks to assess the link between temperate SAS and biodiversity at all main taxonomical levels (bacteria and fungi, plants, invertebrates and vertebrates). We describe the response of biodiversity in terms of abundance, species richness and other diversity indices. This study concerns not only the potential interest of these systems for biodiversity conservation, but also the impacts of living organisms present in the system on ES and eventually ecosystem disservices. We obtained 3458 references dealing with both agroforestry and biodiversity from the Web of Sciences database. Among them, 45 articles corresponded to our specific subject and met the selection criteria. These publications were retained for further analysis. SAS have a positive impact on biodiversity conservation by increasing the abundance or diversity of the different taxonomic groups. Regarding ES, the few papers that studied the effect on pollinators or pollination also showed a positive effect. The majority of studies on natural predators and biocontrol and most of those concerning soil organisms and related supporting services showed a positive effect of SAS. However, some studies found no positive effect or even reported a negative effect in specific conditions. Data is limited for weeds, pests and damage to crop production, and results contrast according to studies and conditions. The review lists the different parameters that influence biodiversity and the subsequent services. These parameters can be divided into three main groups, namely crop management, the influence of the crop location, and agroforestry management. We also report the influence of the experimental design, for example the use of control and comparison or sampling methods. Moreover, the link with other ES is discussed (mainly in terms of provisioning but also for cultural services). Finally, we suggest future research avenues and propose advice in line with the issues discussed in this work.
•SAS are agroforestry systems where trees are closely linked to arable cropping.•SAS are beneficial for biodiversity conservation, and also usually for related ES.•The management of SAS can have a strong impact on biodiversity and associated ES.•The experimental design and protocol have a major influence on the results obtained.•It is possible to promote both biodiversity conservation and ES provision in SAS.
