5-Aminolevulinic acid (ALA) is the universal precursor for tetrapyrrole biosynthesis and is synthesized in plants in three enzymatic steps: ligation of glutamate (Glu) to tRNA GlU by glutamyl-tRNA ...synthetase, reduction of activated Glu to Glu-1-semialdehyde by glutamyl-tRNA reductase (GluTR), and transamination to ALA by Glu 1-semialdehyde aminotransferase. ALA formation controls the metabolic flow into the tetrapyrrole biosynthetic pathway. GluTR is proposed to be the key regulatory enzyme that is tightly controlled at transcriptional and posttranslational levels. We identified a GluTR binding protein (GluTRBP; previously called PROTON GRADIENT REGULATION7) that is localized in chloroplasts and part of a 300-kD protein complex in the thylakoid membrane. Although the protein does not modulate activity of ALA synthesis, the knockout of GluTRBP is lethal in Arabidopsis thaliana, whereas mutants expressing reduced levels of GluTRBP contain less heme. GluTRBP expression correlates with a function in heme biosynthesis. It is postulated that GluTRBP contributes to subcompartmentalized ALA biosynthesis by maintaining a portion of GluTR at the plastid membrane that funnels ALA into the heme biosynthetic pathway. These results regarding GluTRBP support a model of plant ALA synthesis that is organized in two separate ALA pools in the chloroplast to provide appropriate substrate amounts for balanced synthesis of heme and chlorophyll.
Receiver-operating characteristic curves from confidence ratings and remember/know (R/K) judgments are often used to estimate the contribution of familiarity and recollection to recognition memory. ...Both coming with specific advantages and disadvantages, which could be reduced by their combination. Little is known how the combination of both methods impacts response behaviour. This could be particularly important for emotional memory research, which is susceptible to variation in meta-mnemonic processes. We obtained reference performance indices from the two methods, instructing individuals to give confidence ratings or R/K judgments in one step. Against these, we contrasted R/K judgments in a two-step format and two combined formats, confidence ratings followed by R/K judgments and vice versa. Regarding reference formats, confidence ratings resulted in more liberal response criteria and false alarm rates than R/K judgments. Two-step R/K judgments and confidence ratings followed by R/K judgments resulted in patterns similar to one-step R/K judgments. Reversing the order resulted in more liberal response biases, higher hit and false alarms rates. Recollection and familiarity were unaffected by response formats. Valence effects did not vary with response formats. The present results suggest that confidence ratings followed by R/K judgments provide the advantages of both without biasing response behaviour.
Introduction
The influence of limb malrotation on long-leg radiographs (LLR) is frequently discussed in literature. This systematic review aimed to describe the influence of limb rotation on ...alignment measurements alone and in combination with knee flexion, and determine its clinical impact.
Materials and methods
A literature search was conducted in June 2021 using the databases MEDLINE, Cochrane, Web of Science (Clarivate Analytics), and Embase. The search term ((radiograph OR X-ray) AND (position OR rotation) AND limb alignment) was used. Database query, record screening, and study inclusion and exclusion were performed by two reviewers independently. Experimental studies (using either specimens or synthetic bones) or clinical studies (prospective or retrospective using radiographs of patients) analyzing the influence of limb rotation on anatomic and mechanical limb alignment measurements were included. Characteristics and results of the included studies were summarized, simplified, and grouped for comparison to answer the research question. Studies were compared descriptively, and no meta-analysis was performed.
Results
A total of 22 studies were included showing large heterogeneity, comprising studies with cadavers, patients, and synthetic bones. Most studies (7 out of 8) reported that external rotation (ER) causes less apparent valgus and leads to more varus and internal rotation (IR) causes more valgus and leads to less varus. However, there is no consensus on the extent of rotation influencing alignment measures. Studies reported about an average change of > 2° (

= 4) hip-knee-ankle angle (HKA) between 15°IR and 15°ER. There is a consensus that the impact of rotation on mechanical alignment is higher if additional sagittal knee angulation, such as knee flexion, is present. All five studies analyzing the influence of rotation combined with knee flexion (5°–15°) showed an HKA change of > 2° between 15°IR and 15°ER.
Conclusion
Malrotation is frequently present on LLR, possibly influencing the measured alignment especially in knees with extension deficit. Surgeons must consider this when measuring and treating deformities (high tibial osteotomy or total knee arthroplasties), and analyzing surgical outcomes. Especially in patients with osteoarthritis with knee extension deficits or postoperative swelling, the effect of malrotation is significantly greater.
This introduction to the special issue aims to bridge the gap between two fields - policing on the one hand and public management on the other - which have seen only sporadic cross-fertilization so ...far. We begin by outlining the benefits of engaging with policing as a distinctive subject of public management study but one with generalizable and challenging insights for public management. We then provide a brief overview of the papers in the special issue and draw out key themes, which hopefully will inspire academics to engage with policing research as a contribution to public management.

Payments for ecosystem services (PES) have played an increasingly important role in environmental conservation. In line with international trends, a growing number of governmental programs in China are embracing these policy innovations, which were labelled under the broad notion of “eco-compensation”. However, recent research indicates that the understanding of state–civil society and central–local relationships are knowledge gaps in the institutional arrangement of such policies. As one of the most important eco-compensation schemes, the Sloping Land Conversion Program (SLCP) is confronting many challenges to incentivise the local actors. In this study we examine the network of actors of the SLCP by focusing on the collaboration of state actor and civil-society actors and develop a typology of local governance models. To find out how different stakeholders perceive the implementation of the program and how they influence each other, we rely on Social Network Analysis (SNA) and a household survey. The results of our case studies in three townships show that local governance models derive and shape the program implementation. A new actor, which never appeared in the official document and previous research, was discovered. We found that local actors and civil-society actors could potentially bring together government and local farmers for up-front negotiations. In order to mediate conflicts, local government, village committees, forest field stations and farmers may be forced to coordinate and ideally deliberate the terms and conditions of implementation and management options. Therefore, for developing further such eco-compensation schemes in rural China, particular attention should be paid to the local actors and their cooperation for the participatory governance.
Signal transduction networks generate characteristic dynamic activities to process extracellular signals and guide cell fate decisions such as to divide or differentiate. The differentiation of ...pluripotent cells is controlled by FGF/ERK signaling. However, only a few studies have addressed the dynamic activity of the FGF/ERK signaling network in pluripotent cells at high time resolution. Here, we use live cell sensors in wild-type and Fgf4-mutant mouse embryonic stem cells to measure dynamic ERK activity in single cells, for defined ligand concentrations and differentiation states. These sensors reveal pulses of ERK activity. Pulsing patterns are heterogeneous between individual cells. Consecutive pulse sequences occur more frequently than expected from simple stochastic models. Sequences become more prevalent with higher ligand concentration, but are rarer in more differentiated cells. Our results suggest that FGF/ERK signaling operates in the vicinity of a transition point between oscillatory and non-oscillatory dynamics in embryonic stem cells. The resulting heterogeneous dynamic signaling activities add a new dimension to cellular heterogeneity that may be linked to divergent fate decisions in stem cell cultures.
Abstract
In recent years, several ensemble-based Kalman filter algorithms have been developed that have been classified as ensemble square root Kalman filters. Parallel to this development, the ...singular “evolutive” interpolated Kalman (SEIK) filter has been introduced and applied in several studies. Some publications note that the SEIK filter is an ensemble Kalman filter or even an ensemble square root Kalman filter. This study examines the relation of the SEIK filter to ensemble square root filters in detail. It shows that the SEIK filter is indeed an ensemble square root Kalman filter. Furthermore, a variant of the SEIK filter, the error subspace transform Kalman filter (ESTKF), is presented that results in identical ensemble transformations to those of the ensemble transform Kalman filter (ETKF), while having a slightly lower computational cost. Numerical experiments are conducted to compare the performance of three filters (SEIK, ETKF, and ESTKF) using deterministic and random ensemble transformations. The results show better performance for the ETKF and ESTKF methods over the SEIK filter as long as this filter is not applied with a symmetric square root. The findings unify the separate developments that have been performed for the SEIK filter and the other ensemble square root Kalman filters.
In particulate soft matter systems the average number of contacts Z of a particle is an important predictor of the mechanical properties of the system. Using x-ray tomography, we analyze packings of ...frictional, oblate ellipsoids of various aspect ratios α, prepared at different global volume fractions ϕg. We find that Z is a monotonically increasing function of ϕg for all α. We demonstrate that this functional dependence can be explained by a local analysis where each particle is described by its local volume fraction ϕl computed from a Voronoi tessellation. Z can be expressed as an integral over all values of ϕl: Z(ϕg,α,X)=∫Zl(ϕl,α,X)P(ϕl|ϕg)dϕl. The local contact number function Zl(ϕl,α,X) describes the relevant physics in term of locally defined variables only, including possible higher order terms X. The conditional probability P(ϕl|ϕg) to find a specific value of ϕl given a global packing fraction ϕg is found to be independent of α and X. Our results demonstrate that for frictional particles a local approach is not only a theoretical requirement but also feasible.
