This article proposes neural network (NN) based observer schemes and a sensorless robust optimal control scheme for partially unknown permanent magnet synchronous motors with disturbances and ...saturating voltages. First, an NN-observer scheme is designed to estimate back-electromotive force (EMF), for which the mathematical model in rotary or stationary reference frames is relaxed. The NN weight tuning law is designed via Lyapunov theory to guarantee that EMF is ultimately uniformly bounded. Second, to compensate the inexact extraction of the estimated back-EMF at any speed conditions, disturbances, and NN approximation errors, another NN-observer scheme is designed to estimate the tracking errors of rotor position and speed, for which low-pass filters and/or phase-locked loops are not needed. Third, a sensorless saturated robust optimal control scheme dealing with general disturbances and saturating voltages is designed. The scheme includes the augmented feedforward controller to transform the speed and current tracking problem into an optimal control problem. Finally, the feedback control law and worst disturbance law are obtained without estimating unknown internal dynamics. The effectiveness of the proposed schemes is tested through simulations and comparative experiments on a load drive application with a DSP board TMS320F28379D.
Despite recent advances in typhoid fever control, asymptomatic carriage of Salmonella Typhi in the gallbladder remains poorly understood. Aiming to understand if S. Typhi becomes genetically adapted ...for long-term colonisation in the gallbladder, we performed whole genome sequencing on a collection of S. Typhi isolated from the gallbladders of typhoid carriers. These sequences were compared to contemporaneously sampled sequences from organisms isolated from the blood of acute patients within the same population. We found that S. Typhi carriage was not restricted to any particular genotype or conformation of antimicrobial resistance genes, but was largely reflective of S. Typhi circulating in the general population. However, gallbladder isolates showed a higher genetic variability than acute isolates, with median pairwise SNP distances of 21 and 13 SNPs (p = 2.8x10-9), respectively. Within gallbladder isolates of the predominant H58 genotype, variation was associated with a higher prevalence of nonsense mutations. Notably, gallbladder isolates displayed a higher frequency of non-synonymous mutations in genes encoding hypothetical proteins, membrane lipoproteins, transport/binding proteins, surface antigens, and carbohydrate degradation. Specifically, we identified several gallbladder-specific non-synonymous mutations involved in LPS synthesis and modification, with some isolates lacking the Vi capsular polysaccharide vaccine target due to the 134Kb deletion of SPI-7. S. Typhi is under strong selective pressure in the human gallbladder, which may be reflected phylogenetically by long terminal branches that may distinguish organisms from chronic and acute infections. Our work shows that selective pressures asserted by the hostile environment of the human gallbladder generate new antigenic variants and raises questions regarding the role of carriage in the epidemiology of typhoid fever.
Shigellosis is the major global cause of dysentery. Shigella sonnei, which has historically been more commonly isolated in developed countries, is undergoing an unprecedented expansion across ...industrializing regions in Asia, Latin America, and the Middle East. The precise reasons underpinning the epidemiological distribution of the various Shigella species and this global surge in S. sonnei are unclear but may be due to three major environmental pressures. First, natural passive immunization with the bacterium Plesiomonas shigelloides is hypothesized to protect populations with poor water supplies against S. sonnei. Improving the quality of drinking water supplies would, therefore, result in a reduction in P. shigelloides exposure and a subsequent reduction in environmental immunization against S. sonnei. Secondly, the ubiquitous amoeba species Acanthamoeba castellanii has been shown to phagocytize S. sonnei efficiently and symbiotically, thus allowing the bacteria access to a protected niche in which to withstand chlorination and other harsh environmental conditions in temperate countries. Finally, S. sonnei has emerged from Europe and begun to spread globally only relatively recently. A strong selective pressure from localized antimicrobial use additionally appears to have had a dramatic impact on the evolution of the S. sonnei population. We hypothesize that S. sonnei, which exhibits an exceptional ability to acquire antimicrobial resistance genes from commensal and pathogenic bacteria, has a competitive advantage over S. flexneri, particularly in areas with poorly regulated antimicrobial use. Continuing improvement in the quality of global drinking water supplies alongside the rapid development of antimicrobial resistance predicts the burden and international distribution of S. sonnei will only continue to grow. An effective vaccine against S. sonnei is overdue and may become one of our only weapons against this increasingly dominant and problematic gastrointestinal pathogen.
Physical layer wireless communications are more and more essential, and are vulnerable to malicious users owing to the nature of broadcast channels. Herein, we consider a centralized multi-channel ...cognitive radio network in the presence of eavesdroppers (EVEs). In the network, the secondary base station (SBS) shares currently free primary channels to simultaneously communicate with secondary users (SUs), while passive eavesdroppers attempt to overhear data in the secondary communications. Each limited-battery SU is equipped with two antennas (one for transmitting signals, and other for receiving signals) and is powered by a solar energy harvester. Meanwhile, the SBS equipped with multiple antennas can operate in full-duplex (FD) transmission mode (simultaneously transmit and receive signals) or in half-duplex (HD) transmission mode (transmit and receive signals in turn during each half of a time slot) with the SUs. In this paper, we propose a novel scheme to maximize the secondary system security of the multi-channel cognitive system in the presence of multiple passive EVEs, in which the EVEs are able to overhear the data of the SBS-SU transmissions on all the primary channels. The problem of decision making is formulated as the framework of a partially observable Markov decision process (POMDP), and an optimal solution is achieved by adopting value iteration-based dynamic programming. Specifically, in each time slot, the SBS allocates optimal channel and optimal action (i.e. either stay silent or employ HD/FD transmission modes with optimal transmission power) for each SU in order to obtain maximum long-term secrecy rate for the secondary system.

Shigella spp. are some of the key pathogens responsible for the global burden of diarrhoeal disease. These facultative intracellular bacteria belong to the family Enterobacteriaceae, together with ...other intestinal pathogens, such as Escherichia coli and Salmonella spp. The genus Shigella comprises four different species, each consisting of several serogroups, all of which show phenotypic similarity, including invasive pathogenicity. DNA sequencing suggests that this similarity results from the convergent evolution of different Shigella spp. founders. Here, we review the evolutionary relationships between Shigella spp. and E . coli, and we highlight how the genomic plasticity of these bacteria and their acquisition of a distinctive virulence plasmid have enabled the development of such highly specialized pathogens. Furthermore, we discuss the insights that genotyping and whole-genome sequencing have provided into the phylogenetics and intercontinental spread of Shigella spp.
The interplay between bacterial antimicrobial susceptibility, phylogenetics and patient outcome is poorly understood. During a typhoid clinical treatment trial in Nepal, we observed several treatment ...failures and isolated highly fluoroquinolone-resistant Salmonella Typhi (S. Typhi). Seventy-eight S. Typhi isolates were genome sequenced and clinical observations, treatment failures and fever clearance times (FCTs) were stratified by lineage. Most fluoroquinolone-resistant S. Typhi belonged to a specific H58 subclade. Treatment failure with S. Typhi-H58 was significantly less frequent with ceftriaxone (3/31; 9.7%) than gatifloxacin (15/34; 44.1%)(Hazard Ratio 0.19, p=0.002). Further, for gatifloxacin-treated patients, those infected with fluoroquinolone-resistant organisms had significantly higher median FCTs (8.2 days) than those infected with susceptible (2.96) or intermediately resistant organisms (4.01)(pS. Typhi clade internationally, but there are no data regarding disease outcome with this organism. We report an emergent new subclade of S. Typhi-H58 that is associated with fluoroquinolone treatment failure.
Maternal disorders are the third leading cause of sepsis globally, accounting for 5.7 million (12%) cases in 2017. There are increasing concerns about the emergence of antimicrobial resistance (AMR) ...in bacteria commonly causing maternal sepsis. Our aim is to describe the protocol for a clinical and microbiology laboratory study to understand risk factors for and the bacterial etiology of maternal sepsis in a tertiary Obstetrics and Gynaecology Hospital. This case-control study aims to recruit 100 cases and 200 controls at Tu Du Hospital in Ho Chi Minh City, Vietnam, which had approximately 55,000 births in 2022. Women aged greater than or equal to 18 years and greater than or equal to 28 weeks gestation having a singleton birth will be eligible for inclusion as cases or controls, unless they have an uncomplicated localised or chronic infection, or an infection with SARS-CoV-2. Cases will include pregnant or recently pregnant women with sepsis recognised between the onset of labour and/or time of delivery/cessation of pregnancy for up to 42 days post-partum. Sepsis will be defined as suspected or confirmed infection with an obstetrically modified Sequential Organ Failure Assessment score of greater than or equal to 2, treatment with intravenous antimicrobials and requested cultures of any bodily fluid. Controls will be matched by age, location, parity, mode of delivery and gestational age. Primary and secondary outcomes are risk factors associated with the development of maternal sepsis, the frequency of adverse outcomes due to maternal sepsis, bacterial etiology and AMR profiles of cases and controls. This study will improve understanding of the epidemiology and clinical implications of maternal sepsis management including the presence of AMR in women giving birth in Vietnam. It will help us to determine whether women in this setting are receiving optimal care and to identify opportunities for improvement.
Striped catfish are one of the main aquaculture species in Asia. However, the low level of omega‐3 fatty acids in its body makes this species less attractive compared with marine fish. This study ...aimed to investigate the effect of different oils on growth performance, body composition, and fatty acids in striped catfish juveniles. A basal diet (T0) was formulated to contain no oil, while four other diets (T1, T2, T3, and T4) were supplemented at 4% with different oils, including salmon oil (SALO), soybean oil (SBO), linseed oil (LSO), and rapeseed oil (RSO). Catfish juveniles with initial body weight (23–24 g/fish) were randomly stocked in 500‐L fiberglass tanks, divided into five treatments, three replicates for each, and fed the experimental diets for 60 days. Results from this study revealed that fish fed the LSO‐supplemented diet had significantly (p < 0.05) improved whole body omega‐3 (alpha‐linoleic acid, 18:3n3) fatty acid levels (7.86%), total n‐3 fatty acids (9.12%), and higher growth than those fed the other diets. In conclusion, the findings suggest that linseed oil can be used to improve growth, omega‐3 fatty acid, C18:3–3 (ALA), and total n‐3 PUFA deposition in the whole body of striped catfish juveniles.
