Notre présence de 1990 à 2010 en qualité d’éducateur spécialisé sur Beaubreuil, quartier de la ville de Limoges, Haute-Vienne, nous a permis d’accompagner, d’observer et de prendre part pendant plus ...de vingt années aux processus migratoires vietnamiens. Durant tout ce temps nous avons pu constater d’un point de vue psychologique que les ruptures consécutives à l’exil, puis au choc culturel né du contact avec la société d’accueil, ont fait surgir chez les individus des problématiques complexes, notamment identitaires. Dans l’exil, pour faire face aux risques psychosociaux provoqués par l’instabilité de leur structure psychique et de leur système culturel, les vietnamiens ont investi l’espace public et l’espace privé proposés par la culture d’accueil. Ces démarches, multiples, leur ont permis dans le réaménagement de ces espaces, de retrouver, de recréer, les éléments perçus par eux comme fondamentaux de leur culture d’origine, nécessaires et indispensables au travail de rééquilibrage psychique. En France, l’injonction culturelle vietnamienne d’élaboration d’autels religieux au sein de leurs habitations a conduit les personnes à réinterpréter, au sein de dynamiques subjectives, la question des différentes composantes de leur identité, personnelle et sociale, culturelle et religieuse. Les interactions entre l’injonction de la culture d’origine et l’espace proposé par la culture d’accueil ont conduit les sujets à engager des transformations, des modifications dans l’élaboration de leurs autels religieux. En ce sens ces élaborations rendent compte et constituent des analyseurs particulièrement pertinents des processus d’interculturation.
Our presence from 1990 to 2010 as a social worker in Beaubreuil, district of the city of Limoges, Haute-Vienne, allowed us to support, observe and take part for over twenty years in the Vietnamese migration processes. All this time, we noted from a psychological perspective, that ruptures, resulting from the exile, then from the culture shock, were born by contacts with the host society, have given rise to individuals, complex problems including identity ones. In exile, to face the psychosocial risks caused by the instability of their psychic structure and their cultural system, the Vietnamese have invested public and private areas offered by the host culture. These approaches, multiple, allowed them in the redevelopment of these areas, to find, to recreate the elements perceived by them as fundamental in their native culture, necessary and essential to their work of psychic restructuring. In France, the cultural injunction of religious altars development in the private sphere has led them to reinterpret in a subjective way the question of the different components of their identity personal and social, cultural and religious The interactions between the native culture injunction and the space proposed by the host culture has engaged transformations, changes in the elaboration of religious altars. In that way, they are reflecting and are forming analyzers, particularly relevant to us, the intercultural exchange process.
We examined the mechanisms for epithelial prostanoid-dependent modulation of airway smooth muscle tone by protease-activated receptor (PARs) in mice. In ddY mice, SLIGRL-NH2, a PAR-2 agonist, and ...TFLLR-NH2, a PAR-1 agonist, caused relaxation in the tracheal and bronchial rings, which were blocked by COX-1 or COX-2 selective inhibitors. A MEK inhibitor significantly attenuated the relaxation, whereas inhibitors of either cPLA2 or iPLA2 unaffected the relaxation. SLIGRL-NH2 also induced indomethacin-sensitive bronchial and tracheal relaxation in the C57BL/6 wild-type, but not PAR-2-KO, mice. Tiypsin, a PAR-2 activator, also evoked bronchial relaxation in the wild-type mice, which was greatly reduced in the KO mice. In contrast, much higher doses of trypsin were required to induce tracheal relaxation in the wild-type mice, which was only slightly reduced in the KO mice. Our data thus suggest that COX-1, COX-2 and MEK are involved in the airway relaxation via PAR-1 and PAR-2 activation, and that the sensitivity of PAR-2 to trypsin is greatly distinct in the tracheal and bronchial epithelium.
Le travail décrit dans cette thèse porte sur l’évaluation du mode d’exposition externe et interne au xénon-133. Ce radionucléide est un des principaux produits de fission du combustible des réacteurs ...nucléaires. En cas de défaut de gaine combustible, le xénon-133 peut potentiellement exposer le personnel lors de ses interventions dans le bâtiment réacteur. En dosimétrie, les simulations Monte-Carlo sont des outils adaptés pour simuler le transport des rayonnements ionisants dans la matière. A partir des critères de radioprotection actuels, nous avons développé de nouvelles méthodes afin d’améliorer notre compréhension de l’exposition externe et interne auxénon-133 à l’intérieur d’un bâtiment réacteur. Ces nouvelles approches sont basées sur l’utilisation d’un fantôme anthropomorphe, d’une géométrie réaliste de bâtiment réacteur, de simulations Monte-Carlo GEANT4 et de modèles en compartiments. L’exposition externe dans un bâtiment réacteur a été menée en retenant un scénario d’exposition réaliste et conservatif. Nous avons quantifié le débit de dose efficace et le débit de dose équivalente au cristallin. L’exposition interne se produit lorsque le xénon-133 est inhalé. Les poumons sont les premiers organes exposés par l’inhalation du xénon-133, leur débit de dose équivalente a été quantifié. Un modèle biocinétique a été utilisé pour évaluer l’exposition interne au xénon-133. Cette thèse a permis de quantifier les grandeurs dosimétriques liées aux modes d’exposition externe et interne au xénon-133, d’étudier l’impact des changements de limites dosimétriques introduits par la Commission Internationale de Radioprotection prochainement retranscrits dans la réglementation française, et de comprendre la cinétique du xénon-133 dans le corps humain. Nous avons montré que les grandeurs dosimétriques sont nettement inférieures aux limites dosimétriques de la réglementation actuelle et future.
The work described in this thesis focuses on the external and internal dose assessment to xenon-133. During the nuclear reactor operation, fission products and radioactive inert gases, as ¹³³Xe, are generated and might be responsible for the exposure of workers incase of clad defect.Particle Monte Carlo transport code is adapted inradioprotection to quantify dosimetric quantities.The study of exposure to xenon-133 is conducted byusing Monte-Carlo simulations based on GEANT4, ananthropomorphic phantom, a realistic geometry of thereactor building, and compartmental models.The external exposure inside a reactor building isconducted with a realistic and conservative exposurescenario. The effective dose rate and the eye lensequivalent dose rate are determined by Monte-Carlosimulations. Due to the particular emission spectrum ofxenon-133, the equivalent dose rate to the lens of eyesis discussed in the light of expected new eye doselimits.The internal exposure occurs while xenon-133 isinhaled. The lungs are firstly exposed by inhalation, andtheir equivalent dose rate is obtained by Monte-Carlosimulations. A biokinetic model is used to evaluate theinternal exposure to xenon-133.This thesis gives us a better understanding to thedosimetric quantities related to external and internalexposure to xenon-133. Moreover the impacts of thedosimetric changes are studied on the current andfuture dosimetric limits. The dosimetric quantities arelower than the current and future dosimetric limits.
Most recently we have found that agonists for PAR-2 stimulate gastric mucus secretion and mucosal cytoprotection. The aim of the present study was to examine mucosal distribution of PAR-2 and to ...evaluate if PAR-2 could modulate secretion of gastric acid and pepsin/pepsinogen. The RT-PCR and immunohistochemical analyses revealed the presence of mRNAs and immunoreacitvity for PAR-2 in rat gastric mucosa. The PAR-2-activating peptide SLIGRL-NH_2 , administered i.v. in combination with amastatin, an aminopeptidase inhibitor, suppressed carbachol-induced acid output for 30 min, although it had no effect on basal acid output in a resting state. In contrast, the PAR-2 agonist affected neither basal nor carbachol-triggered pepsin/pepsinogen secretion for 30 min. Our results demonstrate the presence of PAR-2 in the gastric mucosa and suggest that the decreased acid output, in addition to increased mucus secretion, might contribute to the PAR-2-mediated gastric mucosal cytoprotection.
We investigated the characteristics of 3-mercaptopropionyl-Phe-Cha-Cha-Arg-Lys-Pro-Asn-Asp-Lys-NH_2 (Mp-NH_2 ), a presumed PAR_1 /thrombin receptor antagonist, using a Ca signaling assay in human ...embryonic kidney (HEK) 293 cells that naturally express both PAR_1 and PAR_2 , and in sarcoma virus transformed rat kidney epithelial (KNRK) cells, which had been transfected with rat PAR_2 . In HEK 293 cells where no heterologous desensitization develops between PAR_1 and PAR_2 , Mp-NH_2 itself increased Ca^2+ _i , while it reduced the increase in Ca^2+ _i by subsequently applied TFLLR-NH_2 , a specific PAR_1 agonist (possibly, due to antagonism of PAR_1 ), and abolished the Ca response to subsequent SLIGRL-NH_2 , a specific PAR_2 agonist (presumably, due to homologous desensitization). Conversely, TFLLR-NH_2 failed to alter the Ca response to subsequent Mp-NH_2 , whereas SLIGRL-NH_2 completely abolished it (presumably, due to homologous desensitization). Furthermore, Mp-NH_2 activated the PAR_2 -transfected KNRK cells, but not the background KNRK cells. These findings strongly suggest that Mp-NH_2 has an agonistic activity on PAR_2 , in addition to its antagonistic activity on PAR_1 .
L’imagerie 3γ est une nouvelle technique d’imagerie médicale nucléaire qui a été proposée au laboratoire Subatech. Cette technique consiste à localiser tridimensionellement la position de la ...désintégration d’un radioisotope innovant émetteur (β+, γ), le 44Sc. Il s’agit pour cela d’associer la détection des deux photons gamma de 511 keV issus de la désintégration d’un positon, assurée par une couronne de détecteurs de tomogaphie à émission de positon, à la détection dutroisième photon par un télescope Compton au xénon liquide. La position de l’interaction entre le photon et le xénon, ainsi que l’énergie déposée, sont relevées grâce à la mesure du signal d’ionisation à l’aide d’un chambre MICROMEGAS (MICROMEsh GAseousStructure), tandis que le déclenchement de l’acquisition et la mesure du temps de l’interaction sont assurés parla détection du signal de scintillation. Le principe de la TPC est ainsi utilisé pour l’imagerie Compton.Afin de faire la preuve expérimentale de faisabilité de l’imagerie 3γ, un prototype de petite dimension, XEMIS (XEnon Medical Imaging System), a été développé.Cette thèse constitue une étape importante vers cette preuve de faisabilité. Les travaux qui y sont exposés portent sur la caractérisation de la réponse du détecteur pour un faisceau de gammas de 511 keV et sur l’analyse des données qui en sont issues. La mesure des résolutions en énergie et temporelle seront exposés, ainsi que celle de la pureté du xénon liquide.
3γ imaging is a new nuclear medical imaging technique which has been suggested by Subatech laboratory. This technique involves locating three-dimensional position of the decay of an innovative radioisotope (β+, γ) emitter the 44Sc. The principle consist in the detection of two photons of 511 keV gamma rays from the decay of the positron, provided by a PET ring detector, associated to the detection of the third photon by a Liquid xenon Compton telescope. The energy deposited in the interaction between the photon and xenon and its position are identified by measuring the ionization signal with a MICROMEGAS chamber (MicroMesh Gaseous Structure), while the trigger and time measurement of the interaction are provided by the detection of thescintillation signal. The principle of the TPC is thus usedto Compton imaging.In order to demonstrate experimentally the feasibility of imaging 3γ, a small prototype, XEMIS (Xenon MedicalImaging System) was developed. This thesis is an important step towards the proof of feasibility. In this work are exposed the characterization of the detector response for a beam of 511 keV gamma rays and the analysis of data derived from it. The measurement of energy and time resolutions will be presented, as well as the purity of the liquid xenon.
