Abstract
The current study aims to evaluate the effects of different gestation dietary Met/Lys (methionine, Met/lysine) ratios on the production performance of sows. Specifically, it measured the ...effect of Met on plasma urea and AA concentrations and placental vascular density of pregnant sows. A total of 325 multiparous sows (third parity, Large × White) were randomly allocated to five dietary treatments (n = 65) with five dietary Met/Lys ratios 0.27 (nutrient requirements of swine NRC 2012 level), 0.32, 0.37, 0.42, and 0.47). The litter size and weight at birth were measured and recorded. Blood samples were obtained on days 0, 40, 90, and 114 of gestation, and placenta samples were collected at parturition. The effects of different dietary Met/Lys ratios on the reproductive performance were evaluated based on the prolificacy of sows as either high (≥13 total piglets born) or low (<13 total piglets born). The results showed that dietary Met/Lys ratio had no significant effect on the reproductive performance of lower prolificacy sows (P > 0.05). However, for high-prolificacy sows, litter weight of born alive significantly increased in 0.37 Met/Lys ratios group compared with control group (P < 0.05). The gestation dietary Met/Lys ratio showed significant quadratic effects on the litter birth weight and percentage of piglets born with weight <0.9 kg (P < 0.05), and the Met/Lys ratios to achieve the best reproductive performance determined to be 0.37. Furthermore, plasma urea concentrations of sows also changed with Met/Lys ratios quadratically (P < 0.05). Increasing dietary Met/Lys ratios elevated the concentration of most plasma AA. Although the dietary Met/Lys ratio had no significant effect on the placental vascular density (P > 0.05), the gestation dietary Met/Lys ratio showed significant quadratic effects on the placental vascular density (P < 0.05). In addition, the birth weight of piglets of high-prolificacy sows was positively correlated with the placental vascular density (P < 0.01). Taken as a whole, the dietary Met/Lys ratio showed a quadratic curve relation with birth weight performance and placental angiogenesis performance, to which 0.37 ratio contributed to the best performance of high-prolificacy sows.
Abstract
The aim of this study was to investigate the effect of orexin B (OXB) on progesterone (P4) and androstenedione (A4) secretion by porcine endometrial and myometrial tissue explants and on the ...expression of key steroidogenic proteins and enzymes involved in steroid production. The hormones secretion and the expression of steroidogenic acute regulatory protein (StAR), P450 side-chain cleavage enzyme (CYP11A1), and 3β-hydroxysteroid dehydrogenase (HSD3B1) were analyzed on days 10 to 11, 12 to 13, 15 to 16, and 27 to 28 of pregnancy and during the luteal phase of the estrous cycle (days 10 to 11). Endometrial and myometrial explants were cultured in vitro in the presence of OXB (1, 10, or 100 nM) and OXB (1, 10, or 100 nM) with 1 µM of JNJ (OX2R antagonist). Gene expression was examined by real-time PCR, and steroid secretion was determined by radioimmunoassay. Orexin B modulated StAR, CYP11A1, HSD3B1 mRNA content depending on the type of uterine tissue, the applied OXB dose, and the stage of pregnancy or the estrous cycle (P < 0.05). Orexin B increased P4 secretion in all stages of early gestation (P < 0.05). Orexin B enhanced the release of A4 on days 12 to 13, 15 to 16, and 27 to 28 of gestation, whereas on days 10 to 11 of early pregnancy, A4 secretion decreased in the endometrium and increased in the myometrium (P < 0.05). These results indicate that OXB affects the expression of key steroidogenic regulators and the secretion of steroid hormones in the porcine uterus during early pregnancy.
Abstract
The objectives were to examine effects of litter size on insulin sensitivity in multiparous sows at the end of pregnancy. Twelve sows were allocated in two treatments after weaning: control ...(CTR) or ligature of the left oviduct (LIG). At 68 d of the subsequent pregnancy, catheters were implanted in a jugular vein, in a carotid artery, and in the main vein draining one uterine horn. A blood flow probe was fitted around the artery irrigating the same uterine horn. A meal test, a tolerance test, and an euglycemic hyperinsulinemic clamp test were performed at 108 ± 3 d of pregnancy. Serial blood samples were drawn simultaneously from the uterine vein and the carotid artery before and during the tests. The number of fetuses in the studied uterine horn was lower (3.7 vs. 8.0, P < 0.001), and piglets at birth were heavier (1.71 vs. 1.31 kg, P = 0.04) in the LIG sows than in the CTR sows. Treatment did not affect uterine blood flow (UBF), but UBF/fetus in the uterine horn was greater for the LIG treatment (0.67 vs. 0.34 L/min, P = 0.002). During meal test, glycemia, glucose uptake in the uterine horn and glucose uterine uptake/fetus were similar in both groups of sows, while insulin levels were higher in the LIG sows (P = 0.04). The decrease of NEFA concentrations was similar across treatments. Glucose half-life did not differ between treatments (13.4 min as a mean; P = 0.63) during tolerance test, but area under the insulin curve was greater in the LIG sows (P = 0.02). The glucose infusion rate during euglycemic hyperinsulinemic clamps was lower in the LIG sows than in CTR sows (6.1 ± 0.2 vs. 7.8 ± 0.1 mg glucose.kg−1 min−1; P = 0.01). The LIG sows are less sensitive to insulin than the CTR sows without adjustment of maternal glycemia and glucose tolerance. Insulin sensitivity adaptation to litter size in late pregnancy of sows would rather be connected to growth rate than to number of fetuses.
Abstract
Omega-3 PUFA may benefit sow reproductive performance, but effects on weaned gilts are unknown. This study evaluated the effects of supplementing omega-3 PUFA to gilts after weaning on ...growth, metabolic markers, and gene expression of steroidogenic enzymes and hormone receptors. For 52 d, gilts in the control group were fed 100 g/d of regular diets, whereas gilts in the omega-3 group were fed 75 g/d of such diets plus 25 g/d of the microalgae Schizochytium sp. (3.5 g/d of omega-3 PUFA; n = 8 gilts/group). Blood samples were collected at day 0, day 21, and day 52. Total serum cholesterol levels were lower for the omega-3 group than for the control group (P < 0.05), but high-density lipoprotein-cholesterol levels were reduced at day 52 for both groups (P < 0.05). Gilts in the omega-3 group presented lower feed intake, better feed conversion, and less-intense immunolabeling for leptin and its receptor in the cytoplasm of oocytes included in primordial/primary follicles than gilts in the control group (P < 0.05). The expression of genes coding for cholesterol side-chain cleavage and aromatase enzymes and the LH receptor in follicular cells was lower for supplemented gilts (P < 0.05). Compared with controls, supplemented gilts presented decreased serum cholesterol levels and better feed conversion, but leptin presence and gene expression for steroidogenic enzymes and for the LH receptor were lower at ovarian level.
Abstract
Accurate semen evaluation is necessary to maintain high reproductive efficiency but difficult to accomplish. The objective was to determine if the ability to bind oviduct cells or oviduct ...glycans are useful supplements to traditional semen analyses. Measuring binding to specific soluble glycans is less laborious than assessing binding to oviduct cell aggregates and more suitable for routine use. Previous work has shown that sperm binding to oviduct cells improves fertility prediction, possibly by estimating the ability of sperm to form an oviduct reservoir. The two oviduct glycan motifs, biantennary 6-sialylated N-acetyllactosamine (bi-SiaLN) and LewisX trisaccharide (LeX), that bind boar spermatozoa with high affinity and specificity were tested. Semen from 30 boars was shipped overnight for laboratory analysis and for inseminations to determine fertility (n = 3 replicates). Oviduct cell binding and traditional sperm analyses including motility and morphology were completed. Additionally, binding to soluble fluoresceinated glycans bi-SiaLN, sulfated LeX (suLeX), and the control lactosamine disaccharide (LacNAc) was measured. Inseminations were at 15 farms (>50 matings per boar) in the Midwest and farrowing data from all matings were used. Pregnancy rate (PR) and litter size (LS) were adjusted to account for different farms, number of services, number of doses inseminated, and sow parity, using the MIXED procedure in SAS 9.4. A fertility index (FI) was generated, consisting of PR × LS, to estimate boar overall fertility. Finally, the GLMSELECT procedure was used to select variables having a significant impact on PR, LS, and FI. The predictive models constructed were further analyzed using the REG procedure and accounted for 58% or more of the variation in PR, LS, and FI PR (P < 0.001, r2 = 0.60), LS (P < 0.001, r2 = 0.58), and FI (P < 0.001, r2 = 0.63). The final model for PR includes oviduct cell binding as well as boar age, % distal droplets, head morphology, tail morphology, beat/cross frequency, and curvilinear velocity. The final model for LS includes boar age, % distal droplets, tail morphology, and overall morphology. Finally, the FI model included boar age, % distal droplets, head morphology, tail morphology, curvilinear velocity, and semen volume per ejaculate. Although binding to intact oviduct cells was impactful as a means to predict PR, binding to specific soluble oviduct glycans was not a useful supplement to traditional semen analysis.
Abstract
The objective was to examine uterine artery blood flow (UBF) as well as macroscopic and microscopic placentome vascular density in nutrient-restricted Angus and Brahman heifers. Angus (n = ...6) and Brahman (n = 6) heifers were bred to a single sire and pregnancy confirmed at 30-d postbreeding. Heifers were randomly assigned to 1 of 2 dietary treatments consisting of 100% (control-fed; CON; n = 6) or 60% (total nutrient-restricted; RES; n = 6) based from net energy requirements for gestating heifers. Nutritional treatments were imposed from days 50 to 180 of gestation. On day 175 of gestation, UBF was collected ipsilateral and contralateral to the conceptus via Doppler ultrasonography. Heifers underwent Cesarean sections for collection of 2 adjacent placentomes on day 180 of gestation. The primary cotyledonary artery of 1 placentome was perfused with Alexa Fluor 647 Con A conjugate to examine macroscopic cotyledonary vascular density via an in vivo imaging system. The second placentome was fixed for microscopic immunofluorescence labeling of capillaries and separated into maternal (caruncle) and fetal (cotyledon) components for determination of angiogenic factor mRNA expression. Main effects of nutritional treatment and breed are reported in the absence of a significant nutritional treatment by breed interaction. Ipsilateral UBF was decreased (P < 0.05) by 48% in RES vs. CON, whereas breed did not influence ipsilateral UBF. Contralateral UBF was not different between nutritional treatments; however, contralateral UBF was decreased (P < 0.05) by 63% in Brahman vs. Angus cattle. Macroscopic cotyledonary vascular density was increased (P < 0.05) by 36% in RES vs. CON and 82% in Brahman vs. Angus heifers. Percent capillary area and capillary perimeter were increased (P < 0.05) in RES vs. CON and increased (P < 0.05) in Brahman vs. Angus heifers. Dietary treatments did not alter angiogenic factor expression; however, transcript abundance of caruncle and cotyledon ANGP1, FLT1, and KDR was increased (P < 0.05) in Brahman vs. Angus heifers. In summary, these data indicate compensatory responses in macroscopic and microscopic placentome blood vessel density during maternal nutrient restriction–induced reductions in UBF. Moreover, a greater macroscopic density of cotyledonary blood vessels was observed in Brahman vs. Angus heifers.

Heat shock protein 90 (Hsp90) functions as a molecular chaperone in its interaction with clients to influence multiple cellular and physiological processes. However, our current ...understanding on Hsp90’s relationship with mammalian oocyte maturation is still very limited. Here, we aimed to investigate Hsp90’s effect on pig oocyte meiotic maturation. Endogenous Hsp90α was constantly expressed at both mRNA and protein levels in porcine maturing oocytes. Addition of 2 µM 17-allylamino-17-demethoxygeldanamycin (17-AAG), the Hsp90 inhibitor, to in vitro mature cumulus–oocyte complexes (COC) significantly decreased Hsp90α protein level (P < 0.05), delayed germinal vesicle breakdown (GVBD) (P < 0.05), and impeded the first polar body (PB1) extrusion (P < 0.01) of porcine oocytes. 2 µM 17-AAG treatment during in vitro maturation also decreased the subsequent development competence as indicated by the lower cleavage (P < 0.001) and higher fragmentation (P < 0.001) rates of parthenotes, whereas no effects on the percentage and average cell number of blastocysts were found. Immunodepletion of Hsp90α by antibody microinjection into porcine oocytes at germinal vesicle and metaphase II stages induced similar defects of meiotic maturation and parthenote development, to that resulted from 2 µM inhibitor 17-AAG. For oocytes treated by 2 µM 17-AAG, the cytoplasm and membrane actin levels were weakened (P < 0.01), and the spindle assembly was disturbed (P < 0.05), due to decreased p-ERK1/2 level (P < 0.05). However, the mitochondrial function and early apoptosis were not affected, as demonstrated by rhodamine 123 staining and Annexin V assays. Our findings indicate that Hsp90α can couple with mitogen-activated protein kinase to regulate cytoskeletal structure and orchestrate meiotic maturation of porcine oocytes.
Abstract
Two experiments investigated the effects of supplementing Ca salts of soybean oil (CSSO) during early gestation on reproductive function and pregnancy rates to AI in Bos taurus beef cows. In ...Exp. 1, 771 suckled, lactating, multiparous Angus cows were divided into 22 groups of approximately 35 cows per group and timed inseminated on day 0. After AI, groups were assigned randomly to receive (as-fed basis) 100 g of ground corn + 100 g of soybean meal per cow/d, in addition to 1) 100 g/cow daily of CSSO (n = 11) or 2) 87 g of prilled saturated fat + 13 g of limestone per cow/d (CON; n = 11). Groups were maintained in individual tall fescue-dominated pastures and offered treatments from day 0 to 21. Pregnancy status was determined between days 45 and 55 via transrectal ultrasonography. Cows receiving CSSO had greater (P = 0.01) pregnancy rates to timed AI compared with CON (60.2 vs. 51.7%; SEM = 4.2). In Exp. 2, 90 suckled, lactating, multiparous Angus × Hereford cows housed in 18 drylot pens (5 cows per pen) were assigned to the same timed AI program and treatments from Exp. 1 (9 pens per treatment) and received 20 kg/d (DM basis) of grass–alfalfa hay. Transrectal ultrasonography was performed to verify ovulation and corpus luteum (CL) volume before AI (day 0), on days 7 and 15. After ultrasonography on day 15, cows diagnosed without a CL on day 0, but with a CL greater than 0.38 cm3 in volume on days 7 and 15 (2 or 3 cows per pen; CSSO, n = 20; CON, n = 24), were assigned to conceptus collection via transcervical flushing and endometrial biopsy in the uterine horn ipsilateral to the CL. Blood samples were collected for FA analysis on days 0, 7, and 15. Blood was collected from cows not assigned to conceptus collection for whole-blood RNA extraction on day 20 and for pregnancy diagnosis on day 30 by measuring concentrations of pregnancy-associated glycoproteins. Cows receiving CSSO had greater (P ≤ 0.04) mean plasma concentrations of linoleic acid and ω-6 FA compared with CON on days 7 and 15. Moreover, CSSO supplementation increased (P = 0.05) mRNA expression of interferon-tau by the conceptus and blood mRNA expression of interferon-stimulated gene 15 and 20,50-oligoadenylate synthetase on day 20 in gestating cows. Hence, post-AI CSSO supplementation to B. taurus beef cows improved pregnancy rates to timed AI, which can be associated with increased mRNA expression of interferon-tau by the conceptus when CSSO is supplemented during early gestation.
Abstract
Glycine, a component of glutathione (GSH), plays an important role in protection from reactive oxygen species (ROS) and inhibition of apoptosis. The aim of this study was to determine the ...effect of glycine on in vitro maturation (IVM) of porcine oocytes and their developmental competence after parthenogenetic activation (PA). We examined nuclear maturation, ROS levels, apoptosis, mitochondrial membrane potential (ΔΨm), and ATP concentration, as well as the expression of several genes related to oocyte maturation and development. Our studies found that treatment with glycine in IVM culture medium increased nuclear maturation rate, but varying the concentrations of glycine (0.6, 6, or 12 mM) had no significant effect. Furthermore, 6 mM glycine supported greater blastocyst formation rates and lesser apoptosis after PA than the other concentrations (P < 0.05). All the glycine treatment groups had decreased levels of ROS in both matured oocytes and at the 2-cell stage (P < 0.05). At the 2-cell stage, the 6 mM glycine group had ROS levels that were lesser than the other 2 glycine treatment groups (0.6 and 12 mM). From this, we deemed 6 mM to be the optimal condition, and we then investigated the effects of 6 mM glycine on gene expression. The expression of both FGFR2 and Hsf1 were greater than the control group in mature oocytes. The glycine treatment group had greater levels of expression of an antiapoptotic gene (Bcl2) in mature oocytes and cumulus cells and lesser levels of expression of a proapoptotic gene (Bax) in PA blastocysts (P < 0.05). In addition, mitochondrial ΔΨm and ATP concentration were increased in 6 mM glycine group compared with the control group. In conclusion, our results suggest that glycine plays an important role in oocyte maturation and later development by reducing ROS levels and increasing mitochondrial function to reduce apoptosis.
Abstract
The effects of hyperleptinemia and leptin resistance during gestation are unclear. Leptin, an important neuroendocrine regulator, has anorexic effects, but its interactions with other ...metabolic hormones during pregnancy are unclear. We examined potential roles of leptin in regulating prolactin (PRL), GH, and melatonin plasma concentrations during pregnancy in Polish Longwool ewes. Twelve estrus-synchronized ewes carrying twins after mating were randomly assigned to receive i.v. injections of saline or recombinant ovine leptin (2.5 or 5.0 µg/kg BW). Blood samples were collected (15-min intervals over 4 h) immediately before the first injection at dusk and kept under red light. Treatments were repeated at 2-wk intervals, starting before mating and continuing from days 30 to 135 of gestation. Concentrations of plasma PRL, GH, and melatonin were determined using a validated RIA. The effects of leptin on hormone plasma concentrations varied depending on pregnancy stage and leptin dose. PRL plasma concentrations were affected at most stages of pregnancy and before gestation. In non-, very early- (day 30), and late- (day 120 and 135) pregnant ewes, exogenous leptin stimulated PRL (P < 0.001) plasma concentrations, while during the second month of gestation, it decreased PRL concentrations (P < 0.01). Leptin affected GH plasma concentrations (P < 0.05) only during the first 2 mo of pregnancy, with no effects during the second part of gestation or before pregnancy. In early-pregnant ewes (day 30 and 45), leptin decreased melatonin plasma concentrations (P < 0.05), but at day 60, leptin stimulated melatonin plasma concentrations at low (P < 0.01) and high doses (P < 0.05), with no effects in ewes after 105 d of gestation. These data indicate specific pregnancy-induced endocrine adaptations to changes in energy homeostasis, supporting the hypothesis that leptin affects PRL, GH, and melatonin release during gestation.
