These functions of protein p53 ... are coordinated so that, within a cell, it acts to integrate signals emanating from a variety of cellular stresses and allows the cell to respond to these insults by activating a set of genes whose products facilitate adaptive and protective activities, including apoptosis and growth arrest. The mutations of gene p53 are among the most prevalent molecular abnormalities in human cancers and are associated with advanced disease, poor response to therapy, and short survival. In regard to this, it is of great importance to determine the p53 status in every patient prior to the application of the treatment. Various approaches to the detection of p53 mutations have evolved in the last 21 yearsand each of them has certain advantages and certain drawbacks. The aim ofthe present study was to make a rough estimation of the frequency of p53 mutational events in the patients with NonHodgkinćs lymphoma (NHL) in total and with regard to the histological subtypes. To address this question, the immunohistochemical staining to p53 was performed using the monoclonal antibody to p53 DO-7. The study was also aimed to evaluate p53 status and define the clinical value of serological methods - i.e. ELISA determinations of protein p53 and antibodies to protein p53 in the sera of NHL patients and controls. Immunohistochemical overexpression of protein p53 was found in 21% of NHL patients, which represents an approximate assessment of the frequency of mutational events in this particular population, with the highest ocŽurrence of p53 immunoreactivity in cases of Burkittćs lymphoma, follicle center lymphoma grade III, and diffuse large B-cell lymphoma.(Abstract truncated at 2000 characters)Type of material - dissertation ; adult, seriousPublication and manufacture - Ljubljana : [B.
