Proteolytic enzymes, which catalyze the breakdown of peptide bonds in proteins, are regulated at ... various levels. The most important for regulating the activity itself are peptide inhibitors, which can reversibly or irreversibly inhibit enzymatic activity. The action of endogenous inhibitors is insufficient in various diseases and uncontrolled proteolytic activity can lead to tissue damage, increased cell proliferation and invasiveness, or accumulation of certain proteins that cells cannot degrade quickly enough. The application of endogenous or other natural inhibitors for the treatment of diseases associated with uncontrolled proteolytic activity has not been successful, primarily due to their non-specificity, poor biological availability, and instability. Small synthetic inhibitors have proven to be more useful, specifically binding to a particular target and not causing side effects.
