  • Vloga sinaptotagmina I pri sekrecijski aktivnosti podganjih hipofiznih celic : magistrsko delo

Resolving the mechanisms of exocytosis and endocytosis is important for the understanding of numerous physiological and pathophysiological processes. It is already known, that an increase in calcium ... concentration inside the cell acts as a trigger for regulated exocytosis. Molecular events, which transduce a rise of calcium to vesicle fusion are poorly understood. In neurons, the release of neurotransmitter appears to involve synaptotagmin I, a calcium binding protein of synaptic vesicles, that appears to serve as a calcium sensor. It is likely, but not studied yet, that synaptotagmin I plays a role in regulated exocytosis of neuroendocrine cells. More than 11 synaptotagmin isoforms have been reported in mammals. Firstly, we have investigated which ofthe synaptotagmin isoforms are expressed in rat melanotrophs. Using RT-PCR analysis we looked at the expression of rat synaptotagmin isoforms I to VII. In controls, the expression of synaptotagmin isoforms was compared to the expression in hippocampus, spinal cord and cerebellum. We found that synaptotagmins I, IV, V in VII are aboundantly expressed, whereas the signal of expression of sinaptotagmin III is weak in the pars intermedia cells. Synaptotagmins II and VI were not detected in our study. (Abstract truncated at 2000 characters)
