Exocytosis involves fusion of neurotransmitter- and hormone- containing vesicles with the plasma membrane. In neurones and neuroendocrine cells this process can be very fast and not easilly studied ...by standard biochemical methods. Here we describe an approach to monitor fast secretory activity in single cells by measuring membrane capacitance and intracellular Ca2+ dynamicsusing whole-cell patch-clamp technique and furaptra based Ca2+ photometry. As an example, the biochemical nature of regulated exocytosis in rat melanotrophs and cortical astrocytes is examined.
