    There is a big gap between the many discoveries of genes abnormal in monogenicdiseases and the very few gene therapies showing at least transitory clinical effectiveness. Primary immunodeficiency ... diseases are among the few diseases with cell biologies favourable to today's gene therapy techniques; haemopoietic stem cells are easily accessible, and either a significant selection (growth) advantage or a low chimerism of gene transduced cells may already achieve a healthy phenotype. This review focuses on the progress in gene therapy for primary immunodeficiency diseases and addresses safety aspects encountered with to-day's retroviral vectors.
