    Nicotiana tabacurn L. cv. Samsun tobacco plants were infected with two strainsof Agrobacterium rhizogenes, ATCC 15834 and ATCC 11325, to transform plant cells and establish hairy root cultures. ... Strain ATCC 15834 was more efficient, and transformation succeeded on 100% of tobacco explants. The growth of the main root axis was also faster and more secondary roots formed compared with the culture established by strain ATCC 11325. Plant growth regulators are important endogenous and exogenous factors in the process of establishing of hairy root cultures. lAA and JA are known to stimulate the formation of secondary rools on non-transformed plants. Their effect on the growth of tobacco hairy root cultures was investigated using two strains of Agrobacterium rhizogenes. Added to the culture medium, JA and IAA affect both cultures similarly. JA in concentrations of 1-10 microM inhibited the growth of the main root axis, while IAA inhibited growth at concentrations of 0.1-10 microM. Lower concentrations (0.01 microM) of both hormones had no effect on the length of the main root axis. 10 microM JA and 10 microM IAA inhibited thenumber of secondary and tertiary roots. The inhibitory effects of high JA and IAA concentrations were also reflected in the fresh and dry weights of thecultures.
