PurposeSupplier Selection (SS) is one of the vital decisions frequently executed by numerous industries. In recent times, the number of suppliers has increased enormously depending on a wide range of ...criteria. A selection of suppliers is a sensitive process that may impact various supply chain activities. The purpose of this research is to explore an underutilized technique called PROMETHEE II method for SS.Design/methodology/approachVarious tools and techniques are available under multi-criteria decision-making tools, which sometimes creates confusion in researchers' minds regarding reliability. PROMETHEE II was the most prominent method for ranking all available alternatives that ultimately avoid decision-making errors. To execute this equal and unequal weights approach has been used with three case studies.FindingsIn this research, three case studies have been used and soved with the help of the PROMETHEE II approach. The study also provides fundamental insights into the supplier's ranking on different criteria using sensitivity analysis. Further, criteria were divided as per benefits and non-beneficial to get a robust result. The pros and cons of PROMETHEE II approaches are also highlighted compared to other MCDM tools in this study.Originality/valueMost of the SS research uses either AHP or TOPSIS as per existing literature. There are very few attempts highlighted in the literature that use PROMETHEE II for the SS problem with sensitivity analysis. The proposed method is probable to motivate decision-makers to consider using a more sophisticated method like PROMETHEE II in supplier evaluation processes. This study opens a new direction for the ranking of suppliers in the field of the supply chain. The study also bears significant practical as well as managerial implications.
PurposeThe study of supply chain disruption (SCD) and supply chain resilience (SCR) remains to be studied deeply in the field of business management. The purpose of this paper is to showcase a ...framework of SCR strategies to reduce the adverse effects of SCD using systematic literature review and data visualization.Design/methodology/approachUsing a systematic literature review approach, the paper explores the concepts of supply chain disruption and supply chain resilience. Through rigorous systematic screening, authors studied papers on both the concepts and have proposed a framework for the same. The authors also have used data visualization and network diagram approaches for better understanding of the topic.FindingsThe systematic literature review of both the concepts brings out some exciting results which give a new direction to supply chain field. The outcome of this research also outlines numerous future research direction, which will be useful for the research community.Practical implicationsThe numerous strategies of SCR should be implemented by manufacturing as well as a service organization. The framework reported in this research help academician and practitioners to understand SCR and to easily overcome any level of disruption. Supply chain managers must also formulate strategies accordingly and make plans to continually expand the system.Originality/valueThis research is the first such attempt to showcase a formal systematic framework and co-occurrence networks as well as overlay networks of SCR and SCD.
BACKGROUNDAdenoid cystic carcinoma (ACC) is a rare malignancy arising in salivary glands and other sites, characterized by high rates of relapse and distant spread. Recurrent/metastatic (R/M) ACCs ...are generally incurable, due to a lack of active systemic therapies. To improve outcomes, deeper understanding of genetic alterations and vulnerabilities in R/M tumors is needed.METHODSAn integrated genomic analysis of 1,045 ACCs (177 primary, 868 R/M) was performed to identify alterations associated with advanced and metastatic tumors. Intratumoral genetic heterogeneity, germline mutations, and therapeutic actionability were assessed.RESULTSCompared with primary tumors, R/M tumors were enriched for alterations in key Notch (NOTCH1, 26.3% vs. 8.5%; NOTCH2, 4.6% vs. 2.3%; NOTCH3, 5.7% vs. 2.3%; NOTCH4, 3.6% vs. 0.6%) and chromatin-remodeling (KDM6A, 15.2% vs. 3.4%; KMT2C/MLL3, 14.3% vs. 4.0%; ARID1B, 14.1% vs. 4.0%) genes. TERT promoter mutations (13.1% of R/M cases) were mutually exclusive with both NOTCH1 mutations (q = 3.3 × 10-4) and MYB/MYBL1 fusions (q = 5.6 × 10-3), suggesting discrete, alternative mechanisms of tumorigenesis. This network of alterations defined 4 distinct ACC subgroups: MYB+NOTCH1+, MYB+/other, MYBWTNOTCH1+, and MYBWTTERT+. Built upon the resource‐based view, the study examines the critical human and behavioral factors (HBFs) on adopting sustainable supply chain practices in the automobile sector (AS). The Interpretive ...Structural Modeling (ISM) and Decision‐Making Trial and Evaluation Laboratory (DEMATEL) method is applied to understand the causeeffect relationships between (HBFs), evidenced by a case study in the automobile industry of India. The causeeffect relationships are informed by an expert team consisting of 18 members working in academia, the automobile industry, and professional consulting companies. The experts provided their inputs based on their direct experience and expertise in this subject matter. The research findings highlight that sustainable supply chain cost (end to end) is a critical success factor. Mangers in the AS should prioritize and focus on sustainable supply chain costs (end to end) to pursue sustainable practices because such critical success factors can foster information sharing behavior. The firms could develop both hard and soft critical HBFs such as Information technology (IT) infrastructure and employee training to build up HBFs to integrate a supply chain. The study provides insight to academicians, policymakers, and practitioners about critical HBFs and develops sustainability goals and policies accordingly.

We address the problem of automatically learning the main steps to complete a certain task, such as changing a car tire, from a set of narrated instruction videos. The contributions of this paper are ...three-fold. First, we develop a new unsupervised learning approach that takes advantage of the complementary nature of the input video and the associated narration. The method solves two clustering problems, one in text and one in video, applied one after each other and linked by joint constraints to obtain a single coherent sequence of steps in both modalities. Second, we collect and annotate a new challenging dataset of real-world instruction videos from the Internet. The dataset contains about 800,000 frames for five different tasks 1 that include complex interactions between people and objects, and are captured in a variety of indoor and outdoor settings. Third, we experimentally demonstrate that the proposed method can automatically discover, in an unsupervised manner, the main steps to achieve the task and locate the steps in the input videos.
Lungs resected for adenocarcinomas often harbour minute discrete foci of cytologically atypical pneumocyte proliferations designated as atypical adenomatous hyperplasia (AAH). Evidence suggests that ...AAH represents an initial step in the progression to adenocarcinoma in situ (AIS), minimally invasive adenocarcinoma (MIA) and fully invasive adenocarcinoma. Despite efforts to identify predictive markers of malignant transformation, alterations driving this progression are poorly understood. Here we perform targeted next-generation sequencing on multifocal AAHs and different zones of histologic progression within AISs and MIAs. Multiregion sequencing demonstrated different genetic drivers within the same tumour and reveal that clonal expansion is an early event of tumorigenesis. We find that KRAS, TP53 and EGFR mutations are indicators of malignant transition. Utilizing droplet digital PCR, we find alterations associated with early neoplasms in paired circulating DNA. This study provides insight into the heterogeneity of clonal events in the progression of early lung neoplasia and demonstrates that these events can be detected even before neoplasms have invaded and acquired malignant potential.
Cell-free DNA shed by cancer cells has been shown to be a rich source of putative tumor-specific biomarkers. Because cell-free DNA from brain and spinal cord tumors cannot usually be detected in the ...blood, we studied whether the cerebrospinal fluid (CSF) that bathes the CNS is enriched for tumor DNA, here termed CSF-tDNA. We analyzed 35 primary CNS malignancies and found at least one mutation in each tumor using targeted or genome-wide sequencing. Using these patient-specific mutations as biomarkers, we identified detectable levels of CSF-tDNA in 74% 95% confidence interval (95% CI) = 57−88% of cases. All medulloblastomas, ependymomas, and high-grade gliomas that abutted a CSF space were detectable (100% of 21 cases; 95% CI = 88−100%), whereas no CSF-tDNA was detected in patients whose tumors were not directly adjacent to a CSF reservoir (P< 0.0001, Fisher’s exact test). These results suggest that CSF-tDNA could be useful for the management of patients with primary tumors of the brain or spinal cord.
The measurement of circulating nucleic acids has transformed the management of chronic viral infections such as HIV. The development of analogous markers for individuals with cancer could similarly ...enhance the management of their disease. DNA containing somatic mutations is highly tumor specific and thus, in theory, can provide optimum markers. However, the number of circulating mutant gene fragments is small compared to the number of normal circulating DNA fragments, making it difficult to detect and quantify them with the sensitivity required for meaningful clinical use. In this study, we applied a highly sensitive approach to quantify circulating tumor DNA (ctDNA) in 162 plasma samples from 18 subjects undergoing multimodality therapy for colorectal cancer. We found that ctDNA measurements could be used to reliably monitor tumor dynamics in subjects with cancer who were undergoing surgery or chemotherapy. We suggest that this personalized genetic approach could be generally applied to individuals with other types of cancer.
Saliva has significant advantages as a test medium for detection of SARS-CoV-2 infection in patients, such as ease of collection, minimal requirement of supplies and trained personnel, and safety. ...Comprehensive validation in a large cohort of prospectively collected specimens with unknown SARS-CoV-2 status should be performed to evaluate the potential and limitations of saliva-based testing. We developed a saliva-based testing pipeline for detection of SARS-CoV-2 nucleic acids using real-time reverse transcription PCR (RT-PCR) and droplet digital PCR (ddPCR) readouts, and measured samples from 137 outpatients tested at a curbside testing facility and 29 inpatients hospitalized for COVID-19. These measurements were compared to the nasal swab results for each patient performed by a certified microbiology laboratory. We found that our saliva testing positively detects 100% (RT-PCR) and 93.75% (ddPCR) of curbside patients that were identified as SARS-CoV-2 positive by the Emergency Use Authorization (EUA) certified nasal swab testing assay. Quantification of viral loads by ddPCR revealed an extremely wide range, with 1 million-fold difference between individual patients. Our results demonstrate for both community screening and hospital settings that saliva testing reliability is on par with that of the nasal swabs in detecting infected cases, and has potential for higher sensitivity when combined with ddPCR in detecting low-abundance viral loads that evade traditional testing methods.
