Mesenchymal stem cells (MSCs) have been isolated from a variety of tissues, such as bone marrow, skeletal muscle, dental pulp, bone, umbilical cord and adipose tissue. MSCs are used in regenerative ...medicine mainly based on their capacity to differentiate into specific cell types and also as bioreactors of soluble factors that will promote tissue regeneration from the damaged tissue cellular progenitors. In addition to these regenerative properties, MSCs hold an immunoregulatory capacity, and elicit immunosuppressive effects in a number of situations. Not only are they immunoprivileged cells, due to the low expression of class II Major Histocompatibilty Complex (MHC-II) and costimulatory molecules in their cell surface, but they also interfere with different pathways of the immune response by means of direct cell-to-cell interactions and soluble factor secretion. In vitro, MSCs inhibit cell proliferation of T cells, B-cells, natural killer cells (NK) and dendritic cells (DC), producing what is known as division arrest anergy. Moreover, MSCs can stop a variety of immune cell functions: cytokine secretion and cytotoxicity of T and NK cells; B cell maturation and antibody secretion; DC maturation and activation; as well as antigen presentation. It is thought that MSCs need to be activated to exert their immunomodulation skills. In this scenario, an inflammatory environment seems to be necessary to promote their effect and some inflammation-related molecules such as tumor necrosis factor-α and interferon-γ might be implicated. It has been observed that MSCs recruit T-regulatory lymphocytes (Tregs) to both lymphoid organs and graft. There is great controversy concerning the mechanisms and molecules involved in the immunosuppressive effect of MSCs. Prostaglandin E2, transforming growth factor-β, interleukins- 6 and 10, human leukocyte antigen-G5, matrix metalloproteinases, indoleamine-2,3-dioxygenase and nitric oxide are all candidates under investigation. In vivo studies have shown many discrepancies regarding the immunomodulatory properties of MSCs. These studies have been designed to test the efficacy of MSC therapy in two different immune settings: the prevention or treatment of allograft rejection episodes, and the ability to suppress abnormal immune response in autoimmune and inflammatory diseases. Preclinical studies have been conducted in rodents, rabbits and baboon monkeys among others for bone marrow, skin, heart, and corneal transplantation, graft versus host disease, hepatic and renal failure, lung injury, multiple sclerosis, rheumatoid arthritis, diabetes and lupus diseases. Preliminary results from some of these studies have led to human clinical trials that are currently being carried out. These include treatment of autoimmune diseases such as Crohn's disease, ulcerative colitis, multiple sclerosis and type 1 diabetes mellitus; prevention of allograft rejection and enhancement of the survival of bone marrow and kidney grafts; and treatment of resistant graft versus host disease. We will try to shed light on all these studies, and analyze why the results are so contradictory.
Context.
It has become clear in recent years that binarity plays a crucial role in many aspects of planetary nebulae (PNe), particularly with regard to the striking morphologies they exhibit. To ...date, there are nearly 60 known binary central stars of PNe (bCSPNe). However, both theory and observation indicate that this figure represents only the tip of the iceberg, with the Galactic PN population hosting orders of magnitude more stars.
Aims.
We are involved in a search for new bCSPNe with the aim of enhancing the statistical validation of the key role of binarity in the formation and shaping of PNe. New discoveries of bCSPNe and their characterization carry important implications not only for understanding PN evolution, but also for studying binary evolution and the common-envelope phase, which is still poorly understood.
Methods.
We used data from the TESS satellite to search for variability in the eight CSPNe that belong to the two-minute cadence of preselected targets in Cycle 1, with their available pipeline-extracted light curves. We identified strong periodicities and analysed them in the context of the binary scenario.
Results.
All the CSPNe but one (Abell 15) show clear signs of periodic variability in TESS. The cause of this variability can be attributed to different effects, some of them requiring the presence of a companion star. We find simple sinusoidal modulations in several of the systems, compatible with irradiation effects. In addition, two of the central stars (PG 1034+001 and NGC 5189) also show photometric variations due to ellipsoidal variations and other signs of variability that are probably caused by star spots or relativistic Doppler-beaming. The case of the well-studied Helix Nebula is of particular interest; here we constructed a series of binary models to explain the modulations we see in the light curve. We find that the variability constrains the possible companion to be very low-mass main-sequence star or sub-stellar object. We also identify, in substantial detail, the individual pulsation frequencies of NGC 246.
The importance of long-period binaries for the formation and evolution of planetary nebulae is still rather poorly understood, which in part is due to the lack of central star systems that are known ...to comprise such long-period binaries. Here, we report on the latest results from the on-going Mercator-HERMES survey for variability in the central stars of planetary nebulae. We present a study of the central stars of NGC 1514, BD+30°623, the spectrum of which shows features associated with a hot nebular progenitor as well as a possible A-type companion. Cross-correlation of high-resolution HERMES spectra against synthetic spectra shows the system to be a highly eccentric (e ~ 0.5) double-lined binary with a period of ~3300 days. Previous studies indicated that the cool component might be a horizontal branch star of mass ~0.55 M⊙, but the observed radial velocity amplitudes rule out such a low mass. If we assume that the nebular symmetry axis and binary orbital plane are perpendicular, then the data are more consistent with a post-main-sequence star ascending towards the giant branch. We also present the continued monitoring of the central star of LoTr 5, HD 112313, which has now completed one full cycle, allowing the orbital period (P~2700 days) and eccentricity (e ~ 0.3) to be derived. To date, the orbital periods of BD+30°623 and HD 112313 are the longest to have been measured spectroscopically in the central stars of planetary nebulae. Furthermore, these systems, along with BD+33°2642, comprise the only spectroscopic wide-binary central stars currently known.
Paracetamol (PCT), or acetaminophen, is an important drug used worldwide for various clinical purposes. However, the excessive or indiscriminate use of PCT can provoke liver and kidney dysfunction; ...hence, it is essential to determine the amount of this target in biological samples. In this work, we develop a quick, simple, and sensitive voltammetric method using chemically modified electrodes to determine PCT in complex matrices, including human serum and commercial solid formulations. We modify the carbon paste electrode with stevensite monoclinic clay mineral (Stv-CPE), using cyclic voltammetry, differential pulse voltammetry, and electrochemical impedance spectroscopy to characterise and detect PCT. The kinetics study provides a better electrochemical characterisation of the electrode behaviour, finding the detection and quantitation limits of 0.2 μM and 0.5 μM under favourable conditions. Further, the best linear working concentration range is 0.6-100 μM for PCT, applying the proposed method to the quantitative determination of PCT content in reference tablet formulations and biological samples for validation.
The liver centralizes the systemic metabolism and thus controls and modulates the functions of the central and peripheral nervous systems, the immune system, and the endocrine system. In addition, ...the liver intervenes between the splanchnic and systemic venous circulation, determining an abdominal portal circulatory system. The liver displays a powerful regenerative potential that rebuilds the parenchyma after an injury. This regenerative mission is mainly carried out by resident liver cells. However, in many cases this regenerative capacity is insufficient and organ failure occurs. In normal livers, if the size of the liver is at least 30% of the original volume, hepatectomy can be performed safely. In cirrhotic livers, the threshold is 50% based on current practice and available data. Typically, portal vein embolization of the part of the liver that is going to be resected is employed to allow liver regeneration in two-stage liver resection after portal vein occlusion (PVO). However, hepatic resection often cannot be performed due to advanced disease progression or because it is not indicated in patients with cirrhosis. In such cases, liver transplantation is the only treatment possibility, and the need for transplantation is the common outcome of progressive liver disease. It is the only effective treatment and has high survival rates of 83% after the first year. However, donated organs are becoming less available, and mortality and the waiting lists have increased, leading to the initiation of living donor liver transplantations. This type of transplant has overall complications of 38%. In order to improve the treatment of hepatic injury, much research has been devoted to stem cells, in particular mesenchymal stem cells (MSCs), to promote liver regeneration. In this review, we will focus on the advances made using MSCs in animal models, human patients, ongoing clinical trials, and new strategies using 3D organoids.
ABSTRACT
We present high-resolution, long-slit optical spectra and images of the planetary nebula NGC 1514. The position velocity maps of the O iii emission line reveal complex kinematics with ...multiple structures. A morpho-kinematical analysis suggests an inner shell, originally spherical and now distorted by several bubbles, and an attached outer shell. The two well-defined, mid-infrared rings of NGC 1514 are not detected in our high-resolution, long-slit spectra, which prevented us from doing a kinematical analysis of them. Based exclusively on their morphology, we propose a barrel-like structure to explain the rings. Several ejection processes have been possibly involved in the formation of the nebula, although a time sequence is difficult to establish with the current data. We also analyse intermediate-resolution, long-slit spectra with the goal of studying the physical parameters and chemical abundances of NGC 1514. The nebular spectra reveal a moderate-excitation nebula with weak emission lines of Ar iii, Ne iii, He i, and He ii. Neither N ii nor other low-excitation emission lines are detected. We found an electron temperature around 14 000 K in the gas and an electron density in the range of 2000–4000 cm−3.
Paracetamol (acetaminophen) (PAR), caffeine (CAF) and tramadol hydrochloride (TRA) are important drugs widely used for many clinical purposes. Determination of their contents is of the paramount ...interest. In this respect, a quick, simple and sensitive isocratic RP-HPLC method with photodiode array detection was developed for the determination of paracetamol, caffeine and tramadol in pharmaceutical formulations. An improved sensitive procedure was also evolved for tramadol using a fluorescence detector system. A C18 column and a mobile phase constituted by methanol/phosphate were used. LODs were found to be 0.2 μg/mL, 0.1 μg/mL and 0.3 μg/mL for paracetamol, caffeine and tramadol hydrochloride, respectively, using photodiode-array detection. Alternatively, LOD for tramadol decreased to 0.1 μg/mL with the fluorescence detector. Other notable analytical figures of merit include the linear concentration ranges, 0.8–270 μg/mL, 0.4–250 μg/mL and 1.0–300 (0.2–40) μg/mL, for the same ordered analytes (including the fluorescence detector). The proposed method was successfully applied for the quantitative determination of the three drugs in tablet dosage forms.
Abstract
LoTr 5 is a planetary nebula with an unusual long-period binary central star. As far as we know, the pair consists of a rapidly rotating G-type star and a hot star, which is responsible for ...the ionization of the nebula. The rotation period of the G-type star is 5.95 d and the orbital period of the binary is now known to be ∼2700 d, one of the longest in central star of planetary nebulae. The spectrum of the G central star shows a complex H α double-peaked profile which varies with very short time-scales, also reported in other central stars of planetary nebulae and whose origin is still unknown. We present new radial velocity observations of the central star which allow us to confirm the orbital period for the long-period binary and discuss the possibility of a third component in the system at ∼129 d to the G star. This is complemented with the analysis of archival light curves from Super Wide Angle Search for Planets, All Sky Automated Survey, and Optical Monitoring Camera. From the spectral fitting of the G-type star, we obtain an effective temperature of Teff = 5410 ± 250 K and surface gravity of log g = 2.7 ± 0.5, consistent with both giant and subgiant stars. We also present a detailed analysis of the H α double-peaked profile and conclude that it does not present correlation with the rotation period and that the presence of an accretion disc via Roche lobe overflow is unlikely.
ABSTRACT
The estimation of the binary fraction of hot subdwarfs is key to shed light on the different evolution scenarios proposed to explain the loss of the hydrogen envelope during the red giant ...branch phase. In this paper, we analyse the spectral energy distribution of the hot subdwarfs included in a recent and comprehensive catalogue with the aim of identifying companions. Our methodology shows a performance superior to the photometric criteria used in that study, identifying 202 objects wrongly classified as binaries according to their spectral energy distributions, and finding 269 new binaries. Out of an initial sample of 3186 objects, we classified 2469 as single and 615 as binary hot subdwarfs. The rest of the objects (102) were not classified because of their inadequate spectral energy distribution fitting due, in turn, to poor quality photometry. Effective temperatures, luminosities, and radii were computed for 192 singles and 42 binaries. They, in particular the binary sample, constitute an excellent data set to further perform a more careful spectroscopic analysis that could provide detailed values for the chemical composition, masses, ages, rotation properties, or reflection effects for the shortest period systems. The results obtained in this paper will be used as a reference for a forthcoming work where we aim to generalize binary and single hot subdwarf classification using Artificial Intelligence-based techniques.
