Serious infections owing to vancomycin-resistant enterococci have historically proven to be difficult clinical cases, requiring combination therapy and management of treatment-related toxicity. ...Despite the introduction of new antibiotics with activity against vancomycin-resistant enterococci to the therapeutic armamentarium, significant challenges remain. An understanding of the factors driving the emergence of resistance in vancomycin-resistant enterococci, the dynamics of gastrointestinal colonization and microbiota-mediated colonization resistance, and the mechanisms of resistance to the currently available therapeutics will permit clinicians to be better prepared to tackle these challenging hospital-associated pathogens.

The separation of global horizontal irradiance (GHI) into its direct and diffuse components is necessary in a variety of applications, most specially in solar energy utilization, where knowledge of direct normal irradiance (DNI) is of paramount importance. Here a novel and efficient model, referred to as GISPLIT, is presented to perform this task accurately, using time series of measured data at 1-min resolution. To better describe the radiative effects of different cloud situations, the model takes advantage of a preliminary classification of the sky conditions into six sky types. An empirical submodel is assigned to each sky class to split GHI into its components, using a limited number of predictors that are related to GHI’s magnitude and variability, and to coincident estimates of the clear-sky irradiance components. Those submodels are trained and validated using rigorously quality-assessed measurements from 120 radiometric stations over all continents and all five major Köppen-Geiger (KG) climate classes, totaling ≈64 million valid data points. Four model versions are evaluated using training data for either all KG climate regions combined or conditioned by KG climate, and either with or without additional support from machine learning. The validation of the four versions suggests that the conditioning by KG climate does not add any significant benefit over the “all-climates” training approach and that, overall, the model version trained with data from all KG climates combined and supported by machine learning generally predicts DNI with the best RMSE results at unseen sites, although with little difference over the other versions.
Using hourly records from 51 rain gauges, spanning between 22 and 28 yr, the authors study the diurnal cycle of precipitation over the tropical Andes of Colombia. Analyses are developed for the ...seasonal march of the diurnal cycle and its interannual variability during the two phases of El Nino-Southern Oscillation (ENSO). Also, the diurnal cycle is analyzed at intra-annual time scales, associated with the westerly and easterly phases of the Madden-Julian oscillation, as well as higher-frequency variability ( < 10 days), mainly associated with tropical easterly wave activity during ENSO contrasting years. Five major general patterns are identified: (i) precipitation exhibits clear-cut diurnal (24 h) and semidiurnal (12 h) cycles; (ii) the minimum of daily precipitation is found during the morning hours (0900-1100 LST) regardless of season or location; (iii) a predominant afternoon peak is found over northeastern and western Colombia; (iv) over the western flank of the central Andes, precipitation maxima occur either near midnight, or during the afternoon, or both; and (v) a maximum of precipitation prevails near midnight amongst stations located on the eastern flank of the central Cordillera. The timing of diurnal maxima is highly variable in space for a fixed time, although a few coherent regions are found in small groups of rain gauges within the Cauca and Magdalena River valleys. Overall, the identified strong seasonal variability in the timing of rainfall maxima appears to exhibit no relationship with elevation on the Andes. The effects of both phases of ENSO are highly consistent spatially, as the amplitude of hourly and daily precipitation diminishes (increases) during El Nino (La Nina), but the phase remains unaltered for the entire dataset. We also found a generalized increase (decrease) in hourly and daily rainfall rates during the westerly (easterly) phase of the Madden-Julian oscillation, and a diminished (increased) high-frequency activity in July-October and February-April during El Nino (La Nina) years, associated, among others, with lower (higher) tropical easterly wave (4-6 day) activity over the Caribbean.
Pancreatic cancer is one of the most aggressive and lethal types of cancer, and more effective therapeutic agents are urgently needed. Overexpressed cell surface antigens are ideal targets for ...therapy with monoclonal antibody (mAb)-based drugs, but none have been approved for the treatment of pancreatic cancer. Here, we report development of two novel mouse mAbs, KU42.33C and KU43.13A, against the human pancreatic cancer cell line BxPC-3. Using ELISA, flow cytometry, competitive assay and immunoprecipitation followed by mass spectrometry, we discovered that these two mAbs target two distinct epitopes on the external domain of CD109 that are overexpressed by varying amounts in human pancreatic cancer cell lines. Treatment with these two naked antibodies alone did not affect tumour cell growth or migration
. Of the two mAbs, only KU42.33C was useful in determining the expression of CD109 in tumour cells by Western blot and immunohistochemistry. Interestingly, immunohistochemistry of human pancreatic carcinoma tissue arrays with mAb KU42.33C showed that 94% of the 65 human pancreatic adenocarcinoma cases were CD109 positive, with no expression in normal pancreatic tissues. Our results suggest that these two novel mAbs are excellent tools for determining the expression level of CD109 in the tumour specimens and sera of patients with a wide range of cancers, in particular pancreatic cancer, and for investigating its diagnostic, prognostic and predictive value. Further research is warranted and should aim to unravel the therapeutic potential of the humanised forms or conjugated versions of such antibodies in patients whose tumours overexpress CD109 antigen.
Despite step-down inhibitory avoidance procedures that have been widely implemented in rats and mice to study learning and emotion phenomena, performance of other species in these tasks has received ...less attention. The case of the Mongolian gerbil is of relevance considering the discrepancies in the parameters of the step-down protocols implemented, especially the wide range of foot-shock intensities (i.e., 0.4-4.0 mA), and the lack of information on long-term performance, extinction effects, and behavioral patterning during these tasks. Experiment 1 aimed to (a) characterize gerbils' acquisition, extinction, and steady-state performance during a multisession (i.e., extended) step-down protocol adapted for implementation in a commercially-available behavioral package (Video Fear Conditioning System-MED Associates Fairfax, VT, USA), and (b) compare gerbils' performance in this task with two shock intensities - 0.5 vs. 1.0 mA-considered in the low-to-mid range. Results indicated that the 1.0 mA protocol produced more reliable and clear evidence of avoidance learning, extinction, and reacquisition in terms of increments in freezing and on-platform time as well as suppression of platform descent. Experiment 2 aimed to (a) assess whether an alternate protocol consisting of a random delivery of foot shocks could replicate the effects of Experiment 1 and (b) characterize gerbils' exploratory behavior during the step-down task (jumping, digging, rearing, and probing). Random shocks did not reproduce the effects observed with the first protocol. The data also indicated that a change from random to response-dependent shocks affects (a) the length of each visit to the platform, but not the frequency of platform descends or freezing time, and (b) the patterns of exploratory behavior, namely, suppression of digging and rearing, as well as increments in probing and jumping. Overall, the study demonstrated the feasibility of the extended step-down protocol for studying steady performance, extinction, and reacquisition of avoidance behavior in gerbils, which could be easily implemented in a commercially available system. The observation that 1.0 mA shocks produced a clear and consistent avoidance behavior suggests that implementation of higher intensities is unnecessary for reproducing aversive-conditioning effects in this species. The observed patterning of freezing, platform descents, and exploratory responses produced by the change from random to periodic shocks may relate to the active defensive system of the gerbil. Of special interest is the probing behavior, which could be interpreted as risk assessment and has not been reported in other rodent species exposed to step-down and similar tasks.
Nanosized CeO2 and CuO/CeO2 samples, active for CO-PROX or related processes were comparatively examined by O2 probe electron paramagnetic resonance and in situ Raman and X-ray diffraction ...techniques. Their behavior toward CO reduction, as well as the oxygen-handling properties of the CO-reduced samples, was explored. An appreciable reduction of the ceria bulk was detected on treatment under CO at 473 K. On the basis of the analysis of the evolution of different oxygen-derived species (superoxide, peroxide, O - ) on low-temperature (77-300 K) oxygen chemisorption on the CO-reduced samples, a general picture of the redox properties of the samples is presented. Results demonstrate that the presence of copper promotes completion of the redox cycle under CO/O2 by favoring both ceria reduction and oxidation. This can be relevant to explaining the remarkable oxidation activity and synergetic effects observed for catalysts combining CuO and CeO2 .
The Acatlán Complex of southern Mexico is a vestige of a Paleozoic Ocean inferred to be either the Cambro-Ordovician Iapetus and/or the Ordovician-Carboniferous Rheic oceans. Ordovician granitoids in ...the complex have been interpreted as either the products of dehydration melting, arc or rift magmatism, however, the geochemistry of felsic rocks is inconclusive. The geochemistry of a recently discovered, major, Ordovician–earliest Silurian mafic igneous suite associated with these granitoids is critical to defining the tectonic setting of the igneous event, and to paleogeographic reconstructions. Such data from three areas in the Acatlán Complex (Xayacatlán, Patlanoaya, and Cuaulote) document tholeiitic, within-plate characteristics with a source in either primitive mantle or mantle previously modified by subduction-related magmatism possibly combined with crustal contamination. This, combined with their occurrence as a dike swarm intruding rift-passive margin clastic sedimentary rocks, indicates rifting of a continental margin. Mafic dikes at Xayacatlán yielded a concordant U–Pb TIMS zircon age of 442
±
1 Ma and a
40Ar/
39Ar hornblende plateau age of 434
±
3 Ma. The age of mafic magmatism at other localities is defined by the ages of associated granitoids intruded at ca. 461 Ma and by the age of the youngest detrital zircon in the host rocks: 496
±
25 Ma at Patlanoaya. Previously published age data suggest that this igneous event may have started earlier at 478
±
5 Ma (Early Ordovician). Although the life spans of the Iapetus and Rheic oceans overlap during the Ordovician, subduction and collision characterize the former, whereas the latter was in a rift–drift mode. Thus, this Ordovician–earliest Silurian magmatism is more consistent with rifting along the southern margin of the Rheic Ocean. Initiation of rifting at ca. 480 Ma is contemporaneous with separation of peri-Gondwanan terranes, such as Avalonia and Carolinia, from Amazonia–Oaxaquia. Subsequent rifting may be analogous to Mesozoic–Cenozoic western North America where transtensional deformation is related to dispersal of terranes (e.g. Baja California and Cretaceous Baja British Columbia) leading to a gradual collapse of the inboard continental margin.
Calanchi (plural of calanco) are typical badland landforms of the Italian landscape. They consist of dense networks of V-shaped valleys, with a sparse or absent vegetation cover, which frequently ...develop on unconsolidated or poorly consolidated clayey deposits. In this paper, the dimensional analysis and the incomplete self-similarity theory were used to deduce a model relating the volume of sediments eroded from the calanchi area to a set of geometric attributes of their tributary areas. The morphometric characteristics of 209 calanchi basins were used to calibrate and validate the model. The predictive skill of the model was assessed by calculating the mean square error and the Nash–Sutcliffe model efficiency coefficient. The model was found to provide a reliable prediction of eroded volume from basins entirely covered by calanchi landforms as well as from basins only partially affected by badland erosion processes. Furthermore, as the residuals between the calculated and the measured volume of the eroded material are normally distributed, the deterministic model may be considered complete, meaning that no other variables are necessary to explain the studied physical process. In other words, the model is able to predict erosion volume in calanchi landforms from a deterministic point of view.

The objective of this study was to compare the microbial community metabolic function from both unsaturated and saturated constructed wetland mesocosms (CWs) when treating the pesticide tebuconazole. ...The comparison was performed for both interstitial water and substrate biofilm by community level physiological profiling (CLPP) via BIOLOG™ EcoPlates. For each CW design (saturated or unsaturated), six mesocosms were established including one unplanted and five planted individually with either Juncus effusus, Typha latifolia, Berula erecta, Phragmites australis or Iris pseudacorus. Microbial activity and metabolic richness of interstitial water from unsaturated CWs were significantly lower than that from saturated CWs. However, in general, the opposite result was observed for biofilm samples. Wetland plants promoted significantly higher biofilm microbial activity and metabolic richness than unplanted CWs in both CW designs. Differences in the microbial community functional profiles between plant species were only found for saturated CWs. Biofilm microbial metabolic richness was generally statistically higher than that of interstitial water in both unsaturated (1.4–24 times higher) and saturated (1.2–1.7 times higher) CWs. Carbon source (guild) utilization patterns were generally different between interstitial water and biofilm samples. Functionality of the biofilm microbial community was positively correlated to the removal of all pollutants (TN, NH4+-N, TP, TOC and tebuconazole) for both unsaturated and saturated CWs, suggesting the biofilm plays a more important role in pollutant removal than the interstitial water microbial community. Thus, merely observing the interstitial water microbial communities may underestimate the role of the microbial community in CW performance. Interestingly, the ability for the biofilm microbial community to utilize amino acids and amines/amides was positively correlated with tebuconazole removal in all system types.
