During the last decade Ebola virus has caused several outbreaks in Africa. The recombinant vesicular stomatitis virus-vectored Zaire Ebola (rVSVΔG-ZEBOV-GP) vaccine has proved safe and immunogenic ...but is reactogenic. We previously identified the first innate plasma signature response after vaccination in Geneva as composed of five monocyte-related biomarkers peaking at day 1 post-immunization that correlates with adverse events, biological outcomes (haematological changes and viremia) and antibody titers. In this follow-up study, we sought to identify additional biomarkers in the same Geneva cohort and validate those identified markers in a US cohort.
Additional biomarkers were identified using multiplexed protein biomarker platform O-link and confirmed by Luminex. Principal component analysis (PCA) evaluated if these markers could explain a higher variability of the vaccine response (and thereby refined the initial signature). Multivariable and linear regression models evaluated the correlations of the main components with adverse events, biological outcomes, and antibody titers. External validation of the refined signature was conducted in a second cohort of US vaccinees (n=142).
Eleven additional biomarkers peaked at day 1 post-immunization: MCP2, MCP3, MCP4, CXCL10, OSM, CX3CL1, MCSF, CXCL11, TRAIL, RANKL and IL15. PCA analysis retained three principal components (PC) that accounted for 79% of the vaccine response variability. PC1 and PC2 were very robust and had different biomarkers that contributed to their variability. PC1 better discriminated different doses, better defined the risk of fever and myalgia, while PC2 better defined the risk of headache. We also found new biomarkers that correlated with reactogenicity, including transient arthritis (MCP-2, CXCL10, CXCL11, CX3CL1, MCSF, IL-15, OSM). Several innate biomarkers are associated with antibody levels one and six months after vaccination. Refined PC1 correlated strongly in both data sets (Geneva: r = 0.97, P < 0.001; US: r = 0.99, P< 0.001).
Eleven additional biomarkers refined the previously found 5-biomarker Geneva signature. The refined signature better discriminated between different doses, was strongly associated with the risk of adverse events and with antibody responses and was validated in a separate cohort.
The prototype of the NIKA2 camera, NIKA, is a dual-band instrument operating at the IRAM 30-m telescope, which can observe the sky simultaneously at 150 and 260 GHz. One of the main goals of NIKA ...(and NIKA2) is to measure the pressure distribution in galaxy clusters at high angular resolution using the thermal Sunyaev-Zel’dovich (tSZ) effect. Such observations have already proved to be an excellent probe of cluster pressure distributions even at intermediate and high redshifts. However, an important fraction of clusters host sub-millimeter and/or radio point sources, which can significantly affect the reconstructed signal. Here we report on <20 arcsec angular resolution observations at 150 and 260 GHz of the cluster MACS J1423.8+2404, which hosts both radio and sub-millimeter point sources. We examine the morphological distribution of the tSZ signal and compare it to other datasets. The NIKA data are combined with Herschel satellite data to study the spectral energy distribution (SED) of the sub-millimeter point source contaminants. We then perform a joint reconstruction of the intracluster medium (ICM) electronic pressure and density by combining NIKA, Planck, XMM-Newton, and Chandra data, focusing on the impact of the radio and sub-millimeter sources on the reconstructed pressureprofile. We find that large-scale pressure distribution is unaffected by the point sources because of the resolved nature of the NIKA observations. The reconstructed pressure in the inner region is slightly higher when the contribution of point sources are removed. We show that it is not possible to set strong constraints on the central pressure distribution without accurately removing these contaminants. The comparison with X-ray only data shows good agreement for the pressure, temperature, and entropy profiles, which all indicate that MACS J1423.8+2404 is a dynamically relaxed cool core system. The present observations illustrate the possibility of measuring these quantities with a relatively small integration time, even at high redshift and without X-ray spectroscopy. This work is part of a pilot study aiming at optimizing tSZ observations with the future NIKA2 camera.

The Adjuvant System AS01 contains monophosphoryl lipid A (MPL) and the saponin QS-21 in a liposomal formulation. AS01 is included in recently developed vaccines against malaria and varicella zoster ...virus. Like for many other adjuvants, induction of adaptive immunity by AS01 is highly dependent on the ability to recruit and activate dendritic cells (DCs) that migrate to the draining lymph node for T and B cell stimulation. T cell responses. Vaccinated mice lacking Flt3 or the chemokine receptor CCR2 showed a marked deficit in inf-cDC2 recruitment and failed to raise proper antibody and T cell responses. Thus, the adjuvant activity of AS01 is associated with the potent activation of subsets of cDC2s, including the newly described inf-cDC2s.
The cosmetics industry is searching for efficient and sustainable substances capable of stabilizing emulsions or colloidal dispersions that are thermodynamically unstable because of their high ...surface energy. Therefore, surfactants are commonly used to stabilize the water/oil interface. However, the presence of a surfactant is not always sufficient to obtain stable emulsions on the one hand, and conventional surfactants are often subject to such controversies as their petroleum origin and environmental concerns on the other hand. As a consequence, among other challenges, it is obvious that research related to new-natural, biodegradable, biocompatible, available, competitive-surfactants are nowadays more intensive. This study aims to valorize a natural gum from

) as a sustainable emulsifier and stabilizer for oil-in-water (O/W) emulsions, and to evaluate how the nature of the fatty phase could affect this potential. To this end, O/W emulsions were prepared at room temperature using three different oils varying in composition, using a rotor-stator mixer. Resulting mixtures were characterized using optical microscopy, laser granulometry, rheology, pH and stability monitoring over time. The results demonstrated good potential for the gum as an emulsifying agent.

) for the preparation and stabilization of the different O/W emulsions. The best results were observed for cocoglyceride oil due to its stronger effect of lowering interfacial tension (IFT) thus acting as a co-emulsifier. Therefore, overall results showed that
gum is undoubtedly a highly promising new bio-sourced and environmentally friendly emulsifier/stabilizer for many applications including cosmetics.

Water–Soluble Extracts (WSE) were obtained from Banana Pseudo–stems (BPS) using a simple and robust deconstruction procedure. WSE were characterized by FTIR, WAXS, TGA–DTG analysis. With the objective of developing fully bio–based polymer formulations, WSE fillers were blended to Poly Lactic Acid (PLA) using a solution blending, casting and thermocompression process. Thermal and mechanical properties were investigated by Differential Scanning Calorimetry (DSC), Dynamic Mechanical Analysis (DMA) and uniaxial tensile tests on neat–PLA (n–PLA) and PLA/WSE blends at various filler contents. Thermal DSC investigations established that WSE act as a plasticizer on PLA. This was further confirmed by DMA by the shift of the loss factor (tan δ) peak maximum towards lower temperatures. WSE also significantly impacted the thermally induced crystallization of PLA and the nucleating power of WSE was assessed by isothermal and non–isothermal studies. WSE had a detrimental effect on PLA brittleness and a slightly positive influence on its stiffness in the glassy state, whereas the drawability remained fairly acceptable when PLA–based materials where drawn at 75 °C above Tg. Moreover, WSE induced an earlier strain–hardening of the composite films as compared to neat PLA. These results underline the valorization potential of such agricultural by–products for sustainable polymer applications in the packaging industry.
Soil function may be affected by cropping practices impacting the soil microbial community. The effect of different phosphorus (P) fertilization rates (0, 20, or 40 kg P₂O₅ ha⁻¹) on soil microbial ...diversity was studied in 8-year-old alfalfa monocultures. The hypothesis that P fertilization modifies soil microbial community was tested using denaturing gradient gel electrophoresis and phospholipids fatty acid (PLFA) profiling to describe soil bacteria, fungi, and arbuscular mycorrhizal (AM) fungi diversity. Soil parameters related to fertility (soil phosphate flux, soluble P, moisture, phosphatase and dehydrogenase assays, and carbon and nitrogen content of the light fraction of soil organic matter) were also monitored and related to soil microbial ribotype profiles. Change in soil P fertility with the application of fertilizer had no effect on crop yield in 8 years, but on the year of this study was associated with shifts in the composition of fungal and bacterial communities without affecting their richness, as evidenced by the absence of effect on the average number of ribotypes detected. However, variation in soil P level created by a history of differential fertilization did not significantly influence AM fungi ribotype assemblages nor AM fungi biomass measured with the PLFA 16:1ω5. Fertilization increased P flux and soil soluble P level but reduced soil moisture and soil microbial activity, as revealed by dehydrogenase assay. Results suggest that soil P fertility management could influence soil processes involving soil microorganisms. Seasonal variations were also recorded in microbial activity, soil soluble P level as well as in the abundance of specific bacterial and fungal PLFA indicators of soil microbial biomass.

/AS03 > AS04 > Alum) at peak response, at D360, and following their increases post-antigen recall (D390). Proportions of high-avidity antibodies increased post-dose 2 across all groups and persisted at D360, but avidity maturation appeared to be more strongly promoted by AS vs. Alum. Post-antigen recall, frequencies of subjects with high-avidity antibodies increased only markedly in the AS groups. Among the AS, total antibody responses were lowest for AS04. However, proportions of high-avidity antibodies were similar between groups, suggesting that MPL in AS04 contributes to avidity maturation. Specific combinations of immunoenhancers in the AS, regardless of their individual nature, increase antibody persistence and avidity maturation.
