In the current study, we determined the functional significance of sodium-dependent/-independent glucose transporters at the neurovasculature during oxygen glucose deprivation (OGD). Confluent brain ...endothelial cells cocultured with astrocytes were exposed to varying degrees of in vitro stroke conditions. Glucose transporter (GLUT) 1 and sodium glucose cotransporter (SGLT) activity were investigated by luminal membrane uptake and transport studies using (3)HD-glucose and also by (14)Calpha-methyl D-glucopyranoside (AMG), a specific, nonmetabolized substrate of SGLT. In vivo middle cerebral artery occlusion experiments were tested to determine whether blood-brain barrier (BBB) SGLT activity was induced during ischemia. Increases in luminal D-glucose and AMG uptake and transport were observed with in vitro stroke conditions. Specific inhibitor experiments suggest a combined role for both SGLT and GLUT1 at the BBB during OGD. A time-dependent increase in the uptake of AMG was also seen in mice exposed to permanent focal ischemia, and this increase was sensitive to the SGLT inhibitor, phlorizin. Infarct and edema ratio during ischemia were significantly decreased by the inhibition of this transporter. These results show that both GLUT1 and SGLT play a role at the BBB in the blood-to-brain transport of glucose during ischemic conditions, and inhibition of SGLT during stroke has the potential to improve stroke outcome. Pharmacological modulation of this novel BBB transporter could prove to be a brain vascular target in stroke.
Bambusicola, gen. nov., is introduced for four new saprobic taxa from culms of bamboos in northern Thailand. One species, chosen as the generic type, has both sexual and asexual morphs, one species ...has the sexual morph only and two species have the asexual morph only. The four new species, Bambusicola massarinia, B. bambusae, B. irregulispora and B. splendida are illustrated and described. Isolates of each species were sequenced using LSU, SSU and ITS genes and phylogenetic analysis shows the genus to be a distinct clade in the suborder “Massarineae”. Morphological differences between Bambusicola and some related genera are noted. Bambusicola is characterized by small, cone-shaped ascomata, slightly broad and fusiform ascospores and a coelomycete asexual morph with light brown conidia. Bambusicola clusters in Trematosphaeriaceae, but the closeness of this relationship cannot be resolved.

During an ongoing investigation of Ascomycetes from plant substrates, three saprobic species were found from plant substrates. Two new species, Leptosphaeria regiae and Neomicrosphaeropsis juglandis ...were isolated from dead branches of Juglans regia from Turkey. Another species is introduced herein as Subplenodomus urticae sp. nov within the family Leptosphaeriaceae found on Urtica dioica in Italy. Multigene phylogenies based on combined LSU, ITS, SSU, and β-tubulin DNA sequence data generated from maximum likelihood and MrBayes analyses indicate that Leptosphaeria regiae is related to L. slovacica and forms an independent lineage within the genus Leptosphaeria. Subplenodomus urticae is basal to S. iridicola and its establishment as a new species is strongly supported. Neomicrosphaeropsis juglandis forms a moderately supported lineage in between N. italica and N. elaeagni in the Didymellaceae. Full morphological details are provided herein and phylogenetic relationships of the three new species are also discussed.
Coelomycete is a general term used for asexual fungi which produce conidia in fruiting bodies: pycnidial, acervular, cupulate, pycnothyria or stromatic conidiomata. The group contains numerous plant ...pathogenic, saprobic and endophytic species associated with a wide range of hosts. Traditionally, morphological characters and host associations have been used as criteria to identify and classify coelomycetes, and this has resulted in a poor understanding of their generic and species boundaries. DNA based taxonomic studies have provided a better outlook of the phylogenetic and evolutionary trends in coelomycetes. However, the present outcomes represent only a preliminary step towards the understanding of coelomycetes. Many genera have not been revisited since they were first described. The present study revises the classification of the hyaline-spored coelomycetes and provides a modern taxonomic framework based on both morphology and phylogeny. In total, 248 genera were investigated, of which less than 100 are known to have sequence data. Multi-locus sequence data analyses of 28S nrDNA, 18S nrDNA, ITS, RNA polymerase II second largest subunit (
rpb2
), and part of the translation elongation factor 1-alpha gene (
tef1
) and β-tubulin (
tub2
) gene regions were analysed. As a result, three new genera and 23 new species are introduced. In addition, three new links between sexual and asexual genera are provided. There are 138 genera that lack sequence data, and these are treated as Ascomycota, genera

constitute a strongly supported independent lineage and are related to
P.griseo-olivaceum
and
P.africanum
. The number of nucleotide differences, across the genes analysed, also supports establishment of
P.ovale
as a new species.
Epidemiological and clinical observations provide consistent evidence that regular intake of aspirin may effectively inhibit the occurrence of epithelial tumors; however, the molecular mechanisms are ...not completely understood. In the present study, we determined the ability of aspirin to acetylate and post-translationally modify cellular proteins in HCT-116 human colon cancer cells to understand the potential mechanisms by which it may exerts anti-cancer effects. Using anti-acetyl lysine antibodies, here we demonstrate that aspirin causes the acetylation of multiple proteins whose molecular weight ranged from 20 to 200 kDa. The identity of these proteins was determined, using immuno-affinity purification, mass spectrometry and immuno-blotting. A total of 33 cellular proteins were potential targets of aspirin-mediated acetylation, while 16 were identified as common to both the control and aspirin-treated samples. These include enzymes of glycolytic pathway, cytoskeleton proteins, histones, ribosomal and mitochondrial proteins. The glycolytic enzymes which were identified include aldolase, glyceraldehyde-3-phosphate dehydrogenase, enolase, pyruvate kinase M2, and lactate dehydrogenase A and B chains. Immunoblotting experiment showed that aspirin also acetylated glucose-6-phosphate dehydrogenase and transketolase, both enzymes of pentose phosphate pathway involved in ribonucleotide biosynthesis. In vitro assays of these enzymes revealed that aspirin did not affect pyruvate kinase and lactate dehydrogenase activity; however, it decreased glucose 6 phosphate dehydrogenase activity. Similar results were also observed in HT-29 human colon cancer cells. Selective inhibition of glucose-6-phosphate dehydrogenase may represent an important mechanism by which aspirin may exert its anti-cancer effects through inhibition of ribonucleotide synthesis.

L. (Rosaceae) in the Zhaotong region of Yunnan Province, China. The isolates were characterized using multi-locus phylogenetic analyses and were found to represent a new monophyletic lineage in Melanommataceae (Pleosporales, Dothideomycetes). This new clade was named as
Dematiomelanomma yunnanense
gen. et sp. nov. which consists of both sexual and asexual morphs. The sexual morph is characterized by globose to subglobose ascomata with a central ostiole, cylindrical asci with a pedicel and ocular chamber, and muriform, ellipsoidal to fusiform ascospores. The asexual morph has synanamorphs including both brown, muriform macroconidia and hyaline, round to oblong or ellipsoidal microconidia. These findings contribute to the understanding of fungal diversity in grasslands and related vegetation types in Yunnan Province, China.
