In this paper we report a stable, label-free, bacteriophage-based detection of Escherichia coli (E. coli) using ultra sensitive long-period fiber gratings (LPFGs). Bacteriophage T4 was covalently immobilized on optical fiber surface and the E. coli binding was investigated using the highly accurate spectral interrogation mechanism. In contrast to the widely used surface plasmon resonance (SPR) based sensors, no moving part or metal deposition is required in our sensor, making the present sensor extremely accurate, very compact and cost effective. We demonstrated that our detection mechanism is capable of reliable detection of E. coli concentrations as low as 103cfu/ml with an experimental accuracy greater than 99%.
Mechanical strain is a powerful tuning knob for excitons, Coulomb-bound electron-hole complexes dominating optical properties of two-dimensional semiconductors. While the strain response of bright ...free excitons is broadly understood, the behaviour of dark free excitons (long-lived excitations that generally do not couple to light due to spin and momentum conservation) or localized excitons related to defects remains mostly unexplored. Here, we study the strain behaviour of these fragile many-body states on pristine suspended WSe
kept at cryogenic temperatures. We find that under the application of strain, dark and localized excitons in monolayer WSe
-a prototypical 2D semiconductor-are brought into energetic resonance, forming a new hybrid state that inherits the properties of the constituent species. The characteristics of the hybridized state, including an order-of-magnitude enhanced light/matter coupling, avoided-crossing energy shifts, and strain tunability of many-body interactions, are all supported by first-principles calculations. The hybridized excitons reported here may play a critical role in the operation of single quantum emitters based on WSe
. Furthermore, the techniques we developed may be used to fingerprint unidentified excitonic states.
We quantify the fraction of the cosmic infrared background (CIB) that originates from galaxies identified in the UV/optical/near-infrared by stacking 81,250 (~35.7 arcmin super(-2)) K-selected ...sources (K sub(AB) < 24.0) split according to their rest-frame U - V versus V - J colors into 72,216 star-forming and 9034 quiescent galaxies, on maps from Spitzer/MIPS (24 mu m), Herschel/PACS (100, 160 mu m), Herschel/SPIRE (250, 350, 500 mu m), and AzTEC (1100 mu m). Of that total, about 95% originates from star-forming galaxies, while the remaining 5% is from apparently quiescent galaxies. The CIB at lambda lap 200 mu m appears to be sourced predominantly from galaxies at z lap 1, while at lambda gap 200 mu the bulk originates from 1 lap z lap 2. Stacking analyses were performed using simstack, a novel algorithm designed to account for possible biases in the stacked flux density due to clustering. Damaged or superfluous cells are typically eliminated by apoptosis. Although apoptosis is a cell-autonomous process, apoptotic cells communicate with their environment in different ways. Here we ...describe a mechanism whereby cells under apoptotic stress can promote survival of neighbouring cells. We find that upon apoptotic stress, cells release the growth factor FGF2, leading to MEK-ERK-dependent transcriptional upregulation of pro-survival BCL-2 proteins in a non-cell autonomous manner. This transient upregulation of pro-survival BCL-2 proteins protects neighbouring cells from apoptosis. Accordingly, we find in certain cancer types a correlation between FGF-signalling, BCL-2 expression and worse prognosis. In vivo, upregulation of MCL-1 occurs in an FGF-dependent manner during skin repair, which regulates healing dynamics. Importantly, either co-treatment with FGF-receptor inhibitors or removal of apoptotic stress restores apoptotic sensitivity to cytotoxic therapy and delays wound healing. These data reveal a pathway by which cells under apoptotic stress can increase resistance to cell death in surrounding cells. Beyond mediating cytotoxic drug resistance, this process also provides a potential link between tissue damage and repair.
In this work we discussed a label-free biosensing application of long-period gratings (LPGs) optimized in refractive index (RI) sensitivity by deposition of thin tantalum oxide (TaOx) overlays. ...Comparing to other thin film and materials already applied for maximizing the RI sensitivity, TaOx offers good chemical and mechanical stability during its surface functionalization and other biosensing experiments. It was shown theoretically and experimentally that when RI of the overlay is as high as 2 in IR spectral range, for obtaining LPGs ultrasensitive to RI, the overlay's thickness must be determined with subnanometer precision. In this experiment the TaOx overlays were deposited using Atomic Layer Deposition method that allowed for achieving overlays with exceptionally well-defined thickness and optical properties. The TaOx nano-coated LPGs show RI sensitivity determined for a single resonance exceeding 11,500 nm/RIU in RI range nD= 1.335–1.345 RIU, as expected for label-free biosensing applications. Capability for detection of various in size biological targets, i.e., proteins (avidin) and bacteria (Escherichia coli), with TaOx-coated LPGs was verified using biotin and bacteriophage adhesin as recognition elements, respectively. It has been shown that functionalization process, as well as type of recognition elements and target analyte must be taken into consideration when the LPG sensitivity is optimized. In this work optimized approach made possible detection of small in size biological targets such as proteins with sensitivity reaching 10.21 nm/log(ng/ml).

Background Imaging of Cosmic Extragalactic Polarization (BICEP) is a bolometric polarimeter designed to measure the inflationary B-mode polarization of the cosmic microwave background (CMB) at degree ...angular scales. During three seasons of observing at the South Pole (2006 through 2008), BICEP mapped ~2% of the sky chosen to be uniquely clean of polarized foreground emission. Here, we present initial results derived from a subset of the data acquired during the first two years. We present maps of temperature, Stokes Q and U, E and B modes, and associated angular power spectra. We demonstrate that the polarization data are self-consistent by performing a series of jackknife tests. We study potential systematic errors in detail and show that they are sub-dominant to the statistical errors. We measure the E-mode angular power spectrum with high precision at 21 <= ell <= 335, detecting for the first time the peak expected at ell ~ 140. The measured E-mode spectrum is consistent with expectations from a ΛCDM model, and the B-mode spectrum is consistent with zero. The tensor-to-scalar ratio derived from the B-mode spectrum is r = 0.02+0.31 -0.26, or r < 0.72 at 95% confidence, the first meaningful constraint on the inflationary gravitational wave background to come directly from CMB B-mode polarization.
We study the evolution of the radio spectral index and far-infrared/radio correlation (FRC) across the star-formation rate – stellar masse (i.e. SFR–M∗) plane up to z ~ 2. We start from a ...stellar-mass-selected sample of galaxies with reliable SFR and redshift estimates. We then grid the SFR–M∗ plane in several redshift ranges and measure the infrared luminosity, radio luminosity, radio spectral index, and ultimately the FRC index (i.e. qFIR) of each SFR–M∗–z bin. The infrared luminosities of our SFR–M∗–z bins are estimated using their stacked far-infrared flux densities inferred from observations obtained with the Herschel Space Observatory. Their radio luminosities and radio spectral indices (i.e. α, where Sν ∝ ν−α) are estimated using their stacked 1.4 GHz and 610 MHz flux densities from the Very Large Array and Giant Metre-wave Radio Telescope, respectively. Our far-infrared and radio observations include the most widely studied blank extragalactic fields – GOODS-N, GOODS-S, ECDFS, and COSMOS – covering a total sky area of ~2.0 deg2. Using this methodology, we constrain the radio spectral index and FRC index of star-forming galaxies with M∗ > 1010 M⊙ and 0 <z< 2.3. We find that α1.4 GHz610 MHz does not evolve significantly with redshift or with the distance of a galaxy with respect to the main sequence (MS) of the SFR–M∗ plane (i.e. Δlog (SSFR)MS = log SSFR(galaxy) /SSFRMS(M∗,z) ). Instead, star-forming galaxies have a radio spectral index consistent with a canonical value of 0.8, which suggests that their radio spectra are dominated by non-thermal optically thin synchrotron emission. We find that the FRC index, qFIR,displays a moderate but statistically significant redshift evolution as qFIR(z) = (2.35 ± 0.08) × (1 + z)−0.12 ± 0.04, consistent with some previous literature. Finally, we find no significant correlation between qFIR and Δlog (SSFR)MS, though a weak positive trend, as observed in one of our redshift bins (i.e. Δ qFIR /Δ Δlog (SSFR)MS = 0.22 ± 0.07 at 0.5 <z< 0.8), cannot be firmly ruled out using our dataset.
The potential for Planck to detect clusters of dusty, star-forming galaxies at z > 1 is tested by examining the Herschel-SPIRE images of Planck Early Release Compact Source Catalog sources lying in ...fields observed by the Herschel Multitiered Extragalactic Survey. Of the 16 Planck sources that lie in the ∼90 sq. deg. examined, we find that 12 are associated with single bright Herschel sources. The remaining four are associated with overdensities of Herschel sources, making them candidate clusters of dusty, star-forming galaxies. We use complementary optical/near-IR data for these 'clumps' to test this idea, and find evidence for the presence of galaxy clusters in all four cases. We use photometric redshifts and red sequence galaxies to estimate the redshifts of these clusters, finding that they range from 0.8 to 2.3. These redshifts imply that the Herschel sources in these clusters, which contribute to the detected Planck flux, are forming stars very rapidly, with typical total cluster star formation rates >1000 M yr−1. The high-redshift clusters discovered in these observations are used to constrain the epoch of cluster galaxy formation, finding that the galaxies in our clusters are 1-1.5 Gyr old at z ∼ 1-2. Prospects for the discovery of further clusters of dusty galaxies are discussed, using not only all sky Planck surveys, but also deeper, smaller area, Herschel surveys.
We report on the design, simulation and experimental demonstration of a new type of waveguide crossing based on subwavelength gratings in silicon waveguides. We used 3D finite-difference time-domain ...simulations to minimize loss, crosstalk and polarization dependence. Measurement of fabricated devices show that our waveguide crossings have a loss as low as -0.023 dB/crossing, polarization dependent loss of < 0.02 dB and crosstalk <-40 dB.
