Cryosectioning is known as a common and well-established histological method, due to its easy accessibility, speed, and cost efficiency. However, the creation of bone cryosections is especially ...difficult. In this study, a cryosectioning protocol for trabecular bone that offers a relatively cheap and undemanding alternative to paraffin or resin embedded sectioning was developed. Sections are stainable with common histological dying methods while maintaining sufficient quality to answer a variety of scientific questions. Furthermore, this study introduces an automated protocol for analysing such sections, enabling users to rapidly access a wide range of different stainings. Therefore, an automated 'QuPath' neural network-based image analysis protocol for histochemical analysis of trabecular bone samples was established, and compared to other automated approaches as well as manual analysis regarding scattering, quality, and reliability. This highly automated protocol can handle enormous amounts of image data with no significant differences in its results when compared with a manual method. Even though this method was applied specifically for bone tissue, it works for a wide variety of different tissues and scientific questions.
FAIR handling of scientific data plays a significant role in current efforts towards a more sustainable research culture and serves as a prerequisite for the fourth scientific paradigm, that is, ...data-driven research. To enforce the FAIR principles by ensuring the reproducibility of scientific data and tracking their provenance comprehensibly, the FAIR modelling of research processes in form of automatable workflows is necessary. By providing reusable procedures containing expert knowledge, such workflows contribute decisively to the quality and the acceleration of scientific research. In this work, the requirements for a system to be capable of modelling FAIR workflows are defined and a generic concept for modelling research processes as workflows is developed. For this, research processes are iteratively divided into impartible subprocesses at different detail levels using the input-process-output model. The concrete software implementation of the identified, universally applicable concept is finally presented in form of the workflow editor KadiStudio of the Karlsruhe Data Infrastructure for Materials Science (Kadi4Mat).
Bioprinting is a method to fabricate 3D models that mimic tissue. Future fields of application might be in pharmaceutical or medical context. As the number of applicants might vary between only one ...patient to manufacturing tissue for high-throughput drug screening, designing a process will necessitate a high degree of flexibility, robustness, as well as comprehensive monitoring. To enable quality by design process optimisation for future application, establishing systematic data storage routines suitable for automated analytical tools is highly desirable as a first step. This manuscript introduces a workflow for process design, documentation within an electronic lab notebook and monitoring to supervise the product quality over time or at different locations. Lab notes, analytical data and corresponding metadata are stored in a systematic hierarchy within the research data infrastructure Kadi4Mat, which allows for continuous, flexible data structuring and access management. To support the experimental and analytical workflow, additional features were implemented to enhance and build upon the functionality provided by Kadi4Mat, including browser-based file previews and a Python tool for the combined filtering and extraction of data. The structured research data management with Kadi4Mat enables retrospective data grouping and usage by process analytical technology tools connecting individual analysis software to machine-readable data exchange formats.
Lithium-ion batteries with solid electrolytes offer safety, higher energy density and higher long-term performance, which are promising alternatives to conventional liquid electrolyte batteries. ...Lithium aluminum titanium phosphate (LATP) is one potential solid electrolyte candidate due to its high Li-ion conductivity. To evaluate its performance, influences of the experimental factors on the materials design need to be investigated systematically. In this work, a materials design strategy based on machine learning (ML) is employed to design experimental conditions for the synthesis of LATP. In the variation of parameters, we focus on the tolerance against the possible deviations in the concentration of the precursors, as well as the influence of sintering temperature and holding time. Specifically, models built with different design selection strategies are compared based on the training data assembled from previous laboratory experiments. The best one is then chosen to design new experiment parameters, followed by measuring the corresponding properties of the newly synthesized samples. A previously unknown sample with ionic conductivity of 1.09 × 10

is discovered within several iterations. In order to further understand the mechanisms governing the high ionic conductivity of these samples, the resulting phase compositions and crystal structures are studied with X-ray diffraction, while the microstructures of sintered pellets are investigated by scanning electron microscopy. Our studies demonstrate the advantages of applying machine learning in designing experimental conditions by the synthesis of desired materials, which can effectively help researchers to reduce the number of required experiments.
The ever-increasing amount of data generated from experiments and simulations in engineering sciences is relying more and more on data science applications to generate new knowledge. Comprehensive ...metadata descriptions and a suitable research data infrastructure are essential prerequisites for these tasks. Experimental tribology, in particular, presents some unique challenges in this regard due to the interdisciplinary nature of the field and the lack of existing standards. In this work, we demonstrate the versatility of the open source research data infrastructure Kadi4Mat by managing and producing FAIR tribological data. As a showcase example, a tribological experiment is conducted by an experimental group with a focus on comprehensiveness. The result is a FAIR data package containing all produced data as well as machine- and user-readable metadata. The close collaboration between tribologists and software developers shows a practical bottom-up approach and how such infrastructures are an essential part of our FAIR digital future.
Orthopaedic implants and temporary osteosynthesis devices are commonly based on Titanium (Ti). For short-term devices, cell-material contact should be restricted for easy removal after bone healing. ...This could be achieved with anti-adhesive plasma-fluorocarbon-polymer (PFP) films created by low-temperature plasma processes. Two different PFP thin film deposition techniques, microwave (MW) and radiofrequency (RF) discharge plasma, were applied to receive smooth, hydrophobic surfaces with octafluoropropane (C3F8) or hexafluorohexane (C6F6) as precursors. This study aimed at examining the immunological local tissue reactions after simultaneous intramuscular implantation of four different Ti samples, designated as MW-C3F8, MW-C6F6, RF-C3F8 and Ti-controls, in rats. A differentiated morphometric evaluation of the inflammatory reaction was conducted by immunohistochemical staining of CD68+ macrophages, CD163+ macrophages, MHC class II-positive cells, T lymphocytes, CD25+ regulatory T lymphocytes, NK cells and nestin-positive cells in cryosections of surrounding peri-implant tissue. Tissue samples were obtained on days 7, 14 and 56 for investigating the acute and chronical inflammation (n = 8 rats/group). Implants with a radiofrequency discharge plasma (RF-C3F8) coating exhibited a favorable short- and long-term immune/inflammatory response comparable to Ti-controls. This was also demonstrated by the significant decrease in pro-inflammatory CD68+ macrophages, possibly downregulated by significantly increasing regulatory T lymphocytes.
Abstract
Solutions for the generation of FAIR (Findable, Accessible, Interoperable, and Reusable) data and metadata in experimental tribology are currently lacking. Nonetheless, FAIR data production ...is a promising path for implementing scalable data science techniques in tribology, which can lead to a deeper understanding of the phenomena that govern friction and wear. Missing community-wide data standards, and the reliance on custom workflows and equipment are some of the main challenges when it comes to adopting FAIR data practices. This paper, first, outlines a sample framework for scalable generation of FAIR data, and second, delivers a showcase FAIR data package for a pin-on-disk tribological experiment. The resulting curated data, consisting of 2,008 key-value pairs and 1,696 logical axioms, is the result of (1) the close collaboration with developers of a virtual research environment, (2) crowd-sourced controlled vocabulary, (3) ontology building, and (4) numerous – seemingly – small-scale digital tools. Thereby, this paper demonstrates a collection of scalable non-intrusive techniques that extend the life, reliability, and reusability of experimental tribological data beyond typical publication practices.
The concepts and current developments of a research data infrastructure for materials science are presented, extending and combining the features of an electronic lab notebook and a repository. The ...objective of this infrastructure is to incorporate the possibility of structured data storage and data exchange with documented and reproducible data analysis and visualization, which finally leads to the publication of the data. This way, researchers can be supported throughout the entire research process. The software is being developed as a web-based and desktop-based system, offering both a graphical user interface and a programmatic interface. The focus of the development is on the integration of technologies and systems based on both established as well as new concepts. Due to the heterogeneous nature of materials science data, the current features are kept mostly generic, and the structuring of the data is largely left to the users. As a result, an extension of the research data infrastructure to other disciplines is possible in the future. The source code of the project is publicly available under a permissive Apache 2.0 license.
This article presents a detailed overview and assessment of contact-free ultraviolet light discharge systems (UVDSs) needed to control the variable electric charge level of free-flying test masses, ...which are part of high-precision inertial sensors in space. A comprehensive numerical analysis approach on the basis of experimental data is detailed. This includes ultraviolet light ray tracing, the computation of time variant electric fields inside the complex inertial sensor geometry, and the simulation of individual photoelectron trajectories. Subsequent data analysis allows determination of key parameters to set up an analytical discharge model. Such a model is an essential system engineering tool needed for requirement breakdown and subsystem specification, performance budgeting, onboard charge control software development, and instrument modeling within spacecraft end-to-end performance simulators. Different types of UVDS design concepts are presented and assessed regarding their robustness and performance. Critical hardware aspects like electron emission from air-contaminated surfaces, interfaces with other subsystems, and spacecraft operations are considered. The focus is on the modeling and performance evaluation of the existing UVDS onboard the LISA Pathfinder, a European Space Agency technology demonstrator spacecraft, which is in implementation phase. The results have motivated the design of a more robust discharge system concept for cubical test mass inertial sensors for future space missions. The analysis tools developed have been used for design optimization and performance assessment of the proposed design. A significant improvement of relevant robustness and performance figures has been achieved.
A comprehensive electrostatic finite-element (FE) analysis of the LISA Pathfinder Inertial Sensor (IS) has been carried out at Astrium GmbH. Starting with a detailed geometrical model of the IS ...housing and test mass (TM) flight units, FE results were derived from multiple analyses runs applying the Maxwell 3D field simulation software. The electrostatic forces and torques on the TM in 6DoF, as well as all non-negligible capacitances between the TM, the 18 electrodes, and the housing, have been extracted for different TM translations and rotations. The results of the FE analyses were expected to confirm the existing IS electrostatic model predictions used for performance analysis, simulations, and on-board algorithms. Major discrepancies were found, however, between the results and the model used so far. In general, FE results give considerably larger capacitance values than the equivalent infinite non-parallel plate estimates. In contrast, the FE derived forces and torques are in general significantly lower compared to the analytic IS electrostatic model predictions. In this paper, these results are discussed in detail and the reasons for the deviations are elaborated. Based on these results, an adapted analytic IS electrostatic model is proposed that reflects the electrostatic forces, torques, and stiffness values in the LISA Pathfinder IS significantly more accurate.
