Chronic inflammation is an established risk factor for the onset of cancer, and the inflammatory cytokine IL-6 has a role in tumorigenesis by enhancing proliferation and hindering apoptosis. As ...factors stimulating proliferation also downregulate p53 expression by enhancing ribosome biogenesis, we hypothesized that IL-6 may cause similar changes in inflamed tissues, thus activating a mechanism that favors neoplastic transformation. Here, we showed that IL-6 downregulated the expression and activity of p53 in transformed and untransformed human cell lines. This was the consequence of IL-6-dependent stimulation of c-MYC mRNA translation, which was responsible for the upregulation of rRNA transcription. The enhanced rRNA transcription stimulated the MDM2-mediated proteasomal degradation of p53, by reducing the availability of ribosome proteins for MDM2 binding. The p53 downregulation induced the acquisition of cellular phenotypic changes characteristic of epithelial-mesenchymal transition, such as a reduced level of E-cadherin expression, increased cell invasiveness and a decreased response to cytotoxic stresses. We found that these changes also occurred in colon epithelial cells of patients with ulcerative colitis, a very representative example of chronic inflammation at high risk for tumor development. Histochemical and immunohistochemical analysis of colon biopsy samples showed an upregulation of ribosome biogenesis, a reduced expression of p53, together with a focal reduction or absence of E-cadherin expression in chronic colitis in comparison with normal mucosa samples. These changes disappeared after treatment with anti-inflammatory drugs. Taken together, the present results highlight a new mechanism that may link chronic inflammation to cancer, based on p53 downregulation, which is activated by the enhancement of rRNA transcription upon IL-6 exposure.

04′W, altitude 1100 m) in the vintage 2018. The date of occurrence of main phenological stages, productiveindexes, clusters characteristics and grape qualitative indexes was evaluated. There was no difference for budbreak date, but flowering, veraison an maturity time varied between regions. The development of plants is slower when they are cultivated at a higher altitude. The number of clusters per plant and yield were higher at 830 m for all varieties. For productive indexes Felicia and Calardis Blanc varieties stood out in relation to Aromera in all parameters. Among the evaluated varieties, Felicia and Calardis Blanc were better adapted to the lower altitude region and had higher productivity and the same grape quality. On the other hand, Aromera presented higher productivity at 830 m but higher soluble solids content at 1100 m.
Despite the increasing understanding about differences in carbon cycling between temperate and tropical freshwater systems, our knowledge on the importance of organic matter (OM) pools on light ...absorption properties in tropical lakes is very scarce. We performed a factorial mesocosm experiment in a tropical lake (Minas Gerais, Brazil) to evaluate the effects of increased concentrations of allochthonous and autochthonous OM, and differences in light availability on the light absorption characteristics of chromophoric dissolved organic matter (CDOM). Autochthonous OM deriving from phytoplankton ( ∼ Chl a) was stimulated by addition of nutrients, while OM from degradation of terrestrial leaves increased allochthonous OM, and neutral shading was used to manipulate light availability. Effects of the additions and shading on DOC, Chl a, nutrients, total suspended solid concentrations (TSM) and spectral CDOM absorption were monitored every 3 days. CDOM quality was characterized by spectral indices (S250–450, S275–295, S350–450, SR and SUVA254). Effects of carbon sources and shading on the spectral CDOM absorption was investigated through principal component (PCA) and redundancy (RDA) analyses. The two different OM sources affected CDOM quality very differently and shading had minor effects on OM levels, but significant effects on OM quality, especially in combination with nutrient additions. Spectral indices (S250–450 and SR) were mostly affected by allochthonous OM addition. The PCA showed that enrichment by allochthonous carbon had a strong effect on the CDOM spectra in the range between 300 and 400 nm, while the increase in autochthonous carbon increased absorption at wavelengths below 350 nm. Our study shows that small inputs of allochthonous OM can have large effects on the spectral light absorption compared to large production of autochthonous OM, with important implications for carbon cycling in tropical lakes.
The objective of this work was to evaluate Brazilian wine competitiveness in domestic market in relation to imported wines marketed in Brazil. The work is based on quantitative sources provided by ...different institutions: International Organization of Vine and Wine (OIV), Brazilian Union of Viticulture (UVIBRA), Ministry of Agriculture, Livestock, and Food Supply (MAPA) and Brazilian Agricultural Research Corporation (EMBRAPA). The data referring to marketing of Brazilian and imported wines refer to the period from 1993 to 2016; and for sparkling wines data refer to the period from 1998 to 2016. During the periods evaluated, there was a 73.86% increase in wine sales and a 446% increase in sparkling wines sale; however, when we compared the volume of Brazilian wines and Brazilian sparkling wines sold, there was a reduction of 38% in the marketing of these wines, while sparkling wines commercialization recorded an increase of 518% in the volume sold. Regarding the volume of imported products, there was an increase of 737% in imported wines marketing, and an increase of 275% in imported sparkling wines marketing. In 1993, only 19.3% of imported wines were sold in relation to the total volume marketed. As of 2016, imported wines accounted for 82.1% of the volume of wines sold in Brazil. For sparkling wines, there was an inverse situation, with a reduction in the marketing of imported sparkling wines in relation to the total sold, since in 1998 imported sparkling wine accounted for 29% of Brazilian sparkling wine market and in 2016 this percentage dropped to 18.2%. It is concluded that sparkling wines are the products with greatest potential of Brazilian wine market. While still wines presents low competitiveness in relation to imported wines, being necessary to create market strategies that propitiate a greater acceptance of Brazilian wines, besides fiscal incentives to the grape producers and winemakers, reducing production costs, resulting in a lower price paid by final consumer.

Novel 3D ...high-resolution simulations show the soft X-ray, keV hot plasma cools rapidly via radiative emission at the high-density interface of the turbulent eddies, stimulating a top-down condensation cascade of warm 104 K filaments. The kpc-scale ionized (optical/ultraviolet) filaments form a skin enveloping the neutral filaments (optical/infrared/21 cm). The peaks of the warm filaments further condense into cold molecular clouds (<50 K; radio) with total mass of several 107 M⊙ and inheriting the turbulent kinematics. In the core, the clouds collide inelastically, mixing angular momentum and leading to Chaotic Cold Accretion (CCA). CCA plays important role in AGN feedback and unification, the evolution of BHs, galaxies and clusters.
The fueling of black holes is one key problem in the evolution of baryons in the universe. Chaotic cold accretion (CCA) profoundly differs from classic accretion models, as Bondi and thin disc ...theories. Using 3D high-resolution hydrodynamic simulations, we now probe the impact of rotation on the hot and cold accretion flow in a typical massive galaxy. In the hot mode, with or without turbulence, the pressure-dominated flow forms a geometrically thick rotational barrier, suppressing the black hole accretion rate to ~1/3 of the spherical case value. When radiative cooling is dominant, the gas loses pressure support and quickly circularizes in a cold thin disk; the accretion rate is decoupled from the cooling rate, although it is higher than that of the hot mode. In the more common state of a turbulent and heated atmosphere, CCA drives the dynamics if the gas velocity dispersion exceeds the rotational velocity, i.e., turbulent Taylor number Tat< 1. Extended multiphase filaments condense out of the hot phase via thermal instability (TI) and rain toward the black hole, boosting the accretion rate up to 100 times the Bondi rate (Ṁ• ~ Ṁcool). Initially, turbulence broadens the angular momentum distribution of the hot gas, allowing the cold phase to condense with prograde or retrograde motion. Subsequent chaotic collisions between the cold filaments, clouds, and a clumpy variable torus promote the cancellation of angular momentum, leading to high accretion rates. As turbulence weakens (Tat > 1), the broadening of the distribution and the efficiency of collisions diminish, damping the accretion rate ∝ Tat-1, until the cold disk drives the dynamics. This is exacerbated by the increased difficulty to grow TI in a rotating halo. The simulated sub-Eddington accretion rates cover the range inferred from AGN cavity observations. CCA predicts inner flat X-ray temperature and r-1 density profiles, as recently discovered in M 87 and NGC 3115. The synthetic Hα images reproduce the main features of cold gas observations in massive ellipticals, as the line fluxes and the filaments versus disk morphology. Such dichotomy is key for the long-term AGN feedback cycle. As gas cools, filamentary CCA develops and boosts AGN heating; the cold mode is thus reduced and the rotating disk remains the sole cold structure. Its consumption leaves the atmosphere in hot mode with suppressed accretion and feedback, reloading the cycle.
Treating patients with systemic lupus erythematosus (SLE) with steroids and immunosuppressive drugs may interfere in the presence of potentially opportunistic microorganisms in the oral cavity. The ...aim of this study was to evaluate the presence of Candida spp., Staphylococcus spp., Enterobacteria and Pseudomonas spp. in the oral cavity of SLE patients, compared with healthy controls. A group of 40 patients who had received therapy for at least 60 days was selected (19–53 years). For the control group, 40 healthy individuals matched for age, gender and use of partial prosthesis were selected. Oral rinse samples were collected and plated on specific culture media. After incubation, the number of colony forming units (CFU) was obtained and the isolates were identified at species level. Microbial counts were compared between SLE and control by analysis of variance (ANOVA) and Mann–Whitney (p < 0.05 significant). Microorganism counts in patients with and without immunosuppressive drugs, as well with active and inactive disease (according to SLEDAI score) were also compared. No significant differences in CFU/mL between SLE and control patients were observed (yeasts, p = 0.55; Staphylococci, p = 0.24; Enterobacteria/Pseudomonas spp., p = 0.26). No differences in microbial counts were observed regarding clinical parameters tested. The most frequent species isolated in the SLE group were Candida albicans, Staphylococcus epidermidis and Klebsiella oxytoca. In conclusion, no differences in frequency and microorganism levels were found between SLE patients and healthy individuals.
A fundamental gap in the current understanding of galaxies concerns the thermodynamical evolution of ordinary, baryonic matter. On the one hand, radiative emission drastically decreases the thermal ...energy content of the interstellar plasma (ISM), inducing a slow cooling flow towards the centre. On the other hand, the active galactic nucleus (AGN) struggles to prevent the runaway cooling catastrophe, injecting huge amount of energy into the ISM. The present study intends to investigate thoroughly the role of mechanical AGN feedback in (isolated or massive) elliptical galaxies, extending and completing the mass range of tested cosmic environments. Our previously successful feedback models in galaxy clusters and groups demonstrated that AGN outflows, self-regulated by cold gas accretion, are able to quench the cooling flow properly without destroying the cool core. Via three-dimensional hydrodynamic simulations (flash 3.3), also including stellar evolution, we show that massive mechanical AGN outflows can indeed solve the cooling-flow problem for the entire life of the galaxy, at the same time reproducing typical observational features and constraints such as buoyant underdense bubbles, elliptical shock cocoons, sonic ripples, dredge-up of metals, subsonic turbulence and extended filamentary or nuclear cold gas. In order to avoid overheating and totally emptying the isolated galaxy, the frequent mechanical AGN feedback should be less powerful and efficient (ε∼ 10−4) compared with the heating required for more massive and bound ellipticals surrounded by the intragroup medium (ε∼ 10−3).
To compare the oral prevalence and antimicrobial susceptibility of candida spp., staphylococci, enterobacteriaceae, and pseudomonas spp. from ankylosing spondylitis (AS) patients receiving ...conventional and anti-TNF-α therapy.
The study included 70 AS patients, diagnosed according to the modified New York criteria (1984). The volunteers were divided into 2 groups: a biological group (AS BioG) (n=35) (on anti-TNF-α therapy) and a conventional group (AS ConvG) (n=35). The control group (ContG) (n=70) was made up of healthy individuals matched for age, gender, and oral conditions. After clinical examination, oral rinse samples were collected and plated in specific culture media. The number of colony-forming units per milliliter (cfu/ml) was obtained, and isolates were identified using the API system. Antimicrobial susceptibility tests were performed according to the NCCLS guidelines. Prevalence and counts of microorganisms were statistically compared between the 3 groups, using the Mann-Whitney and Chi-square tests. Significance level was set at 5%.
In both the AS BioG and the AS ConvG, staphylococci counts were higher than that in the ContG (p<0.0001). Candida albicans and staphylococcus epidermidis were the most commonly found species in all the groups. Serratia marcescens and klebsiella oxytoca were more prevalent in the AS BioG and the AS ConvG, respectively. Two candida isolates (2.8%) from the AS BioG and 5 (10.8%) from the AS ConvG were resistant to amphotericin B and 5-fluorocytosine. A low percentage of staphylococci isolates was resistant to amoxicillin, ciprofloxacin, and doxycycline.
Higher counts of staphylococci were observed in both AS groups, regardless of the current therapy, age, sex, and oral conditions. Anti-TNF-α therapy could not be correlated with increased counts of microorganisms.
