The long-term consequences of childhood abuse on adult mental health have been a major focus of research. Much less attention has been directed to its effects on physical health outcomes. By use of ...data from the Ontario Health Survey (n = 9,953), the association between retrospective reports of childhood physical and sexual abuse and adult health and health care utilization was examined in men and women. The population health survey was conducted from November 1990 to March 1991 in the Canadian province of Ontario. An association of moderate strength was found between childhood abuse and multiple health problems, poor or fair self-rated health, pain that interferes with activities, disability due to physical health problems, and frequent emergency room and health professional visits but not frequent general practitioner visits. These effects were more pronounced in females and younger respondents. The strength of the associations reported here with odds ratios of 1.3–2.2 was lower than that found between childhood abuse and adult mental health, with odds ratios of 1.9–3.4. Given the growing evidence of the long-term effects of childhood abuse, greater efforts are clearly needed in developing more effective strategies for the prevention and treatment of child abuse.
Protein-protein interactions (PPIs) between modular binding domains and their target peptide motifs are thought to largely depend on the intrinsic binding specificities of the domains. The large ...family of SRC Homology 3 (SH3) domains contribute to cellular processes via their ability to support such PPIs. While the intrinsic binding specificities of SH3 domains have been studied in vitro, whether each domain is necessary and sufficient to define PPI specificity in vivo is largely unknown. Here, by combining deletion, mutation, swapping and shuffling of SH3 domains and measurements of their impact on protein interactions in yeast, we find that most SH3s do not dictate PPI specificity independently from their host protein in vivo. We show that the identity of the host protein and the position of the SH3 domains within their host are critical for PPI specificity, for cellular functions and for key biophysical processes such as phase separation. Our work demonstrates the importance of the interplay between a modular PPI domain such as SH3 and its host protein in establishing specificity to wire PPI networks. These findings will aid understanding how protein networks are rewired during evolution and in the context of mutation-driven diseases such as cancer.
Most lactobacilli produce extracellular polysaccharides that are considered to contribute to the probiotic effect of many strains. Lacticaseibacillus rhamnosus CNCM I-3690 is an anti-inflammatory ...strain able to counterbalance gut barrier dysfunction. In this study ten spontaneous variants of CNCM I-3690 with different EPS-production were generated and characterized by their ropy phenotype, the quantification of the secreted EPS and genetic analysis. Amongst them, two were further analysed in vitro and in vivo: an EPS over-producer (7292) and a low-producer derivative of 7292 (7358, with similar EPS levels than the wild type (WT) strain). Our results showed that 7292 does not have anti-inflammatory profile in vitro, and lost the capacity to adhere to the colonic epithelial cells as well as the protective effect on the permeability. Finally, 7292 lost the protective effects of the WT strain in a murine model of gut dysfunction. Notably, strain 7292 was unable to stimulate goblet cell mucus production and colonic IL-10 production, all key features for the beneficial effect of the WT strain. Furthermore, transcriptome analysis of colonic samples from 7292-treated mice showed a down-regulation of anti-inflammatory genes. Altogether, our results point out that the increase of EPS production in CNCM I-3690 impairs its protective effects and highlight the importance of the correct EPS synthesis for the beneficial effects of this strain.
Lysis of dairy starters is a prerequisite for optimum cheese maturation, since intracellular starter enzymes, particularly peptidases, can then play their role. Here we describe the different methods ...used to detect starter lysis in situ and current knowledge concerning the impact of lysis on cheese ripening, particularly the increase of free amino acids due to early lysis and the reduction of bitterness by hydrolysis of large hydrophobic peptides. Recent results obtained on the impact of lysis on lipolysis and amino acid catabolism are also described. Then, we present current knowledge regarding the mechanisms involved, focussing mainly on the model most investigated:
Lactococcus lactis. Recent advances concerning the molecular characterization of peptidoglycan hydrolases are summarized (sequence, structure, regulation) together with current knowledge of the relationship between lysogeny and lysis. Lastly, we review the different approaches proposed to control or induce lysis in situ. In conclusion, we point out unaddressed questions.

Abstract
The flower is a bisexual reproductive unit where both genders compete for resources. Counting pollen and ovules in flowers is essential to understand how much is invested in each gender. Classical methods to count very numerous pollen grains and ovules are inefficient when pollen grains are tightly aggregated, and when fertilization rates of ovules are unknown. In this study we have therefore developed novel counting techniques based on computed tomography. In order to demonstrate the potential of our methods in very difficult cases, we counted pollen and ovules across inflorescences of deceptive and rewarding species of European orchids, which possess both very large numbers of pollen grains (tightly aggregated) and ovules. Pollen counts did not significantly vary across inflorescences and pollination strategies, whereas deceptive flowers had significantly more ovules than rewarding flowers. The within-inflorescence variance of pollen-to-ovule ratios in rewarding flowers was four times higher than in deceptive flowers, possibly demonstrating differences in the constraints acting on both pollination strategies. We demonstrate the inaccuracies and limitations of previously established methods, and the broad applicability of our new techniques: they allow measurement of reproductive investment without restriction on object number or aggregation, and without specimen destruction.
In arid and semiarid rangelands, soil erosion has been widely considered an important soil degradation process and one of the main factors responsible for declining soil fertility. In this study, we ...determined the sediment production and the enrichment ratios of clay, organic C, and total N by using rainfall simulations on runoff plots (0.60 × 1.67 m) in three plant communities of northeastern Patagonia: grass (GS), degraded grass with scattered shrubs (DGS), and degraded shrub steppes (DSS). Our results clearly indicate that spatial variability in soil loss rate and enrichment process exists as a result of the local differences in both plant composition and soil surface characteristics. Sediment production was significantly lower in the GS (14.2 g m−2) compared with the DGS and DSS (38.2 and 51.5 g m−2, respectively). In the GS, the enrichment ratio of clay was significantly greater (3.9) and enrichment ratio of organic C was lower (3.1) than in the DGS and the DSS, though differences in enrichment ratios of total N were not significant. The high rate of soil loss and nutrients through overland-flow may limit the opportunities that promote the pathway from DGS back to GS community, favoring the dominance of shrubs.

In grazed semiarid ecosystems, considerable spatial variability in soil infiltration exists as a result of vegetation and soil patchiness. Despite widespread recognition that important interactions ...and feedbacks occur between vegetation, runoff and erosion, currently there is only limited quantitative information on the control mechanisms that lead to differences in infiltration from different vegetation types. In this paper, we determine (i) the relationship between vegetation and soil surface characteristics and (ii) the soil infiltration rate by using rainfall simulations on runoff plots (0.60 × 1.67 m) in three plant communities of northeastern Patagonia: grass (GS), degraded grass with scattered shrubs (DGS), and degraded shrub steppes (DSS). Our results clearly indicate that vegetation and soil infiltration are closely coupled. Total infiltration was significantly higher in the GS (69.6 mm) compared with the DGS and DSS (42.9 and 28.5 mm, respectively). In the GS, soil infiltration rate declined more slowly than the others communities, reaching a terminal infiltration rate significantly greater (57.7 mm) than those of DGS and DSS (25.7 and 12.9 mm, respectively). The high rate of water losses via overland-flow may limit the possibilities for grass seedling emergence and establishment and favor the persistent dominance of shrubs.

Quasi-free scattering reactions of the type (p,2p) were measured for the first time exclusively in complete and inverse kinematics, using a 12C beam at an energy of ∼400 MeV/u as a benchmark. This ...new technique has been developed to study the single-particle structure of exotic nuclei in experiments with radioactive-ion beams. The outgoing pair of protons and the fragments were measured simultaneously, enabling an unambiguous identification of the reaction channels and a redundant measurement of the kinematic observables. Both valence and deeply-bound nucleon orbits are probed, including those leading to unbound states of the daughter nucleus. Exclusive (p,2p) cross sections of 15.8(18) mb, 1.9(2) mb and 1.5(2) mb to the low-lying 0p-hole states overlapping with the ground state (3/2−) and with the bound excited states of 11B at 2.125 MeV (1/2−) and 5.02 MeV (3/2−), respectively, were determined via γ-ray spectroscopy. Particle-unstable deep-hole states, corresponding to proton removal from the 0s-orbital, were studied via the invariant-mass technique. Cross sections and momentum distributions were extracted and compared to theoretical calculations employing the eikonal formalism. The obtained results are in a good agreement with this theory and with direct-kinematics experiments. The dependence of the proton–proton scattering kinematics on the internal momentum of the struck proton and on its separation energy was investigated for the first time in inverse kinematics employing a large-acceptance measurement.
Drosophila Crumbs (Crb) and its mammalian ortholog CRB3 control epithelial polarity through poorly understood molecular mechanisms. Elucidating these mechanisms is crucial, because the physiology of ...epithelia largely depends on the polarized architecture of individual epithelial cells. In addition, loss of CRB3 favors tumor cell growth, metastasis and epithelial to mesenchymal transition (EMT). Using Drosophila embryos, we report that Rac1 sustains PI3K signaling, which is required for Rac1 activation. Crb represses this positive-feedback loop. Notably, this property confers to Crb its ability to promote epithelial integrity in vivo, because attenuation of either Rac1 or PI3K activity rescues the crb mutant phenotype. Moreover, inhibition of Rac1 or PI3K results in Crb-dependent apical membrane growth, whereas Rac1 activation restricts membrane localization of Crb and interferes with apical domain formation. This illustrates that Crb and the Rac1-PI3K module are antagonists, and that the fine balance between the activities of these proteins is crucial to maintain epithelial organization and an appropriate apical to basolateral ratio. Together, our results elucidate a mechanism that mediates Crb function and further define the role of PI3K and Rac1 in epithelial morphogenesis, allowing for a better understanding of how distinct membrane domains are regulated in polarized epithelial cells.
