Our understanding of the brown dwarf population in star-forming regions is dependent on knowing distances and proper motions and therefore will be improved through the Gaia space mission. In this ...paper, we select new samples of very low-mass objects (VLMOs) in Upper Scorpius using UKIDSS colors and optimized proper motions calculated using Gaia DR1. The scatter in proper motions from VLMOs in Upper Scorpius is now (for the first time) dominated by the kinematic spread of the region itself, not by the positional uncertainties. With age and mass estimates updated using Gaia parallaxes for early-type stars in the same region, we determine masses for all VLMOs. Our final most complete sample includes 453 VLMOs of which ∼125 are expected to be brown dwarfs. The cleanest sample is comprised of 131 VLMOs, with ∼105 brown dwarfs. We also compile a joint sample from the literature that includes 415 VLMOs, out of which 152 are likely brown dwarfs. The disk fraction among low-mass brown dwarfs ( ) is substantially higher than in more massive objects, indicating that disks around low-mass brown dwarfs survive longer than in low-mass stars overall. The mass function for is consistent with the Kroupa Initial Mass Function. We investigate the possibility that some "proper motion outliers" have undergone a dynamical ejection early in their evolution. Our analysis shows that the color-magnitude cuts used when selecting samples introduce strong bias into the population statistics due to varying levels of contamination and completeness.

We present a new algorithm for precision radial velocity (pRV) measurements, a line-by-line (LBL) approach designed to handle outlying spectral information in a simple but efficient manner. ...The effectiveness of the LBL method is demonstrated on two data sets, one obtained with SPIRou on Barnard’s star, and the other with the High Accuracy Radial velocity Planet Searcher (HARPS) on Proxima Centauri. In the near-infrared, the LBL provides a framework for meters-per-second-level accuracy in pRV measurements despite the challenges associated with telluric absorption and sky emission lines. We confirm with SPIRou measurements spanning 2.7 yr that the candidate super-Earth on a 233 day orbit around Barnard’s star is an artifact due to a combination of time sampling and activity. The LBL analysis of the Proxima Centauri HARPS post-upgrade data alone easily recovers the Proxima b signal and also provides a 2

Abstract
TRAPPIST-1 is a nearby system of seven Earth-sized, temperate, rocky exoplanets transiting a Jupiter-sized M8.5V star, ideally suited for in-depth atmospheric studies. Each TRAPPIST-1 planet ...has been observed in transmission both from space and from the ground, confidently rejecting cloud-free, hydrogen-rich atmospheres. Secondary eclipse observations of TRAPPIST-1 b with JWST/MIRI are consistent with little to no atmosphere given the lack of heat redistribution. Here we present the first transmission spectra of TRAPPIST-1 b obtained with JWST/NIRISS over two visits. The two transmission spectra show moderate to strong evidence of contamination from unocculted stellar heterogeneities, which dominates the signal in both visits. The transmission spectrum of the first visit is consistent with unocculted starspots and the second visit exhibits signatures of unocculted faculae. Fitting the stellar contamination and planetary atmosphere either sequentially or simultaneously, we confirm the absence of cloud-free, hydrogen-rich atmospheres, but cannot assess the presence of secondary atmospheres. We find that the uncertainties associated with the lack of stellar model fidelity are one order of magnitude above the observation precision of 89 ppm (combining the two visits). Without affecting the conclusion regarding the atmosphere of TRAPPIST-1 b, this highlights an important caveat for future explorations, which calls for additional observations to characterize stellar heterogeneities empirically and/or theoretical works to improve model fidelity for such cool stars. This need is all the more justified as stellar contamination can affect the search for atmospheres around the outer, cooler TRAPPIST-1 planets for which transmission spectroscopy is currently the most efficient technique.

TRPA1 is a nonselective cation channel expressed by nociceptors. Although it is widely accepted that TRPA1 serves as a broad irritancy receptor for a variety of reactive chemicals, its role in cold ...sensation remains controversial. Here, we demonstrate that mild cooling markedly increases agonist-evoked rat TRPA1 currents. In the absence of an agonist, even noxious cold only increases current amplitude slightly. These results suggest that TRPA1 is a key mediator of cold hypersensitivity in pathological conditions in which reactive oxygen species and proinflammatory activators of the channel are present, but likely plays a comparatively minor role in acute cold sensation. Supporting this, cold hypersensitivity can be induced in wild-type but not Trpa1(-/-) mice by subcutaneous administration of a TRPA1 agonist. Furthermore, the selective TRPA1 antagonist HC-030031 2-(1,3-dimethyl-2,6-dioxo-1,2,3,6-tetrahydro-7H-purin-7-yl)-N-(4-isopropylphenyl)acetamide reduces cold hypersensitivity in rodent models of inflammatory and neuropathic pain.
The present-day envelope of gaseous planets is a relic of how these giant planets originated and evolved. Measuring their elemental composition therefore presents a powerful opportunity to answer ...long-standing questions regarding planet formation. Obtaining precise observational constraints on the elemental inventory of giant exoplanets has, however, remained challenging owing to the limited simultaneous wavelength coverage of current space-based instruments. Here, we present thermal emission observations of the nontransiting hot Jupiter τ Boo b using the new wide wavelength coverage (0.95–2.50 μm) and high spectral resolution (R = 70,000) CFHT/SPIRou spectrograph. By combining a total of 20 hr of SPIRou data obtained over five nights in a full atmospheric retrieval framework designed for high-resolution data, we constrain the abundances of all the major oxygen- and carbon-bearing molecules and recover a noninverted temperature structure using a new free-shape, nonparametric temperature–pressure profile retrieval approach. We find a volume mixing ratio of log(CO) = −\({2.46}_{-0.29}^{+0.25}\) and a highly depleted water abundance of less than 0.0072 times the expected value for a solar composition envelope. Combined with upper limits on the abundances of CH4, CO2, HCN, TiO, and C2H2, this results in a gas-phase C/H ratio of \({5.85}_{-2.82}^{+4.44}\) × solar, consistent with the value of Jupiter, and an envelope C/O ratio robustly greater than 0.60, even when taking into account the oxygen that may be sequestered out of the gas phase. Combined, the inferred supersolar C/H, O/H, and C/O ratios on τ Boo b support a formation scenario beyond the water snowline in a disk enriched in CO owing to pebble drift.
Abstract Determination of fundamental parameters of stars impacts all fields of astrophysics, from galaxy evolution to constraining the internal structure of exoplanets. This paper presents a ...detailed spectroscopic analysis of Barnard’s star (otherwise known as Gl 699) that compares an exceptionally high-quality (an average signal-to-noise ratio of ∼1000 in the entire domain), high-resolution near-infrared (NIR) spectrum taken with Canada-France-Hawaii Telescope/SPIRou to PHOENIX-ACES stellar atmosphere models. The observed spectrum shows thousands of lines not identified in the models with a similarly large number of lines present in the model but not in the observed data. We also identify several other caveats, such as continuum mismatch, unresolved contamination, and spectral lines significantly shifted from their expected wavelengths; all of these can be a source of bias for the determination of abundance. Out of >10 4 observed lines in the NIR that could be used for chemical spectroscopy, we identify a short list of a few hundred lines that are reliable. We present a novel method for determining the effective temperature ( T eff ) and overall metallicity of slowly rotating M dwarfs that uses several groups of lines as opposed to bulk spectral fitting methods. With this method, we infer T eff = 3231 ± 21 K for Barnard's star, consistent with the value of 3238 ± 11 K inferred from the interferometric method. We also provide measurements of the abundance of 15 different elements for Barnard's star, including the abundances of four elements (K, O, Y, Th) never reported before for this star. This work emphasizes the need to improve current atmosphere models to fully exploit the NIR domain for chemical spectroscopy analysis.
Preconception pregnancy risk profiles-characterizing the likelihood that a pregnancy attempt results in a full-term birth, preterm birth, clinical pregnancy loss, or failure to conceive-can provide ...critical information during the early stages of a pregnancy attempt, when obstetricians are best positioned to intervene to improve the chances of successful conception and full-term live birth. Yet the task of constructing and validating risk assessment tools for this earlier intervention window is complicated by several statistical features: the final outcome of the pregnancy attempt is multinomial in nature, and it summarizes the results of two intermediate stages, conception and gestation, whose outcomes are subject to competing risks, measured on different time scales, and governed by different biological processes. In light of this complexity, existing pregnancy risk assessment tools largely focus on predicting a single adverse pregnancy outcome, and make these predictions at some later, post-conception time point.
We reframe the individual pregnancy attempt as a multistate model comprised of two nested multinomial prediction tasks: one corresponding to conception and the other to the subsequent outcome of that pregnancy. We discuss the estimation of this model in the presence of multiple stages of outcome missingness and then introduce an inverse-probability-weighted Hypervolume Under the Manifold statistic to validate the resulting multivariate risk scores. Finally, we use data from the Effects of Aspirin in Gestation and Reproduction (EAGeR) trial to illustrate how this multistate competing risks framework might be utilized in practice to construct and validate a preconception pregnancy risk assessment tool.
In the EAGeR study population, the resulting risk profiles are able to meaningfully discriminate between the four pregnancy attempt outcomes of interest and represent a significant improvement over classification by random chance.
As illustrated in our analysis of the EAGeR data, our proposed prediction framework expands the pregnancy risk assessment task in two key ways-by considering a broader array of pregnancy outcomes and by providing the predictions at an earlier, preconception intervention window-providing obstetricians and their patients with more information and opportunities to successfully guide pregnancy attempts.
BackgroundPatients with chronic viral infections including human immunodeficiency virus (HIV), hepatitis B (HBV) and hepatitis C (HCV) are at increased risk of developing malignancies. The safety and ...efficacy of ICI therapy in patients with both cancer and chronic viral infections is not well established as most clinical trials of ICIs excluded these patient populations.MethodsWe performed a retrospective analysis of patients with advanced-stage cancers and HIV, HBV, or HCV infection treated with ICI therapy at 5 MedStar Health hospitals from January 2011 to April 2018.ResultsWe identified 50 patients including 16 HIV, 29 HBV/HCV, and 5 with concurrent HIV and either HBV or HCV. In the HIV cohort (n = 21), any grade immune-related adverse events (irAEs) were 24% with grade ≥ 3 irAEs 14%. Among 5 patients with matched pre/post-treatment results, no significant changes in HIV viral load and CD4+ T-cell counts were observed. RECIST confirmed (n = 18) overall response rate (ORR) was 28% with 2 complete responses (CR) and 3 partial responses (PR). Responders included 2 patients with low baseline CD4+ T-cell counts (40 and 77 cells/ul, respectively). In the HBV/HCV cohort (n = 34), any grade irAEs were 44% with grade ≥ 3 irAEs 29%. RECIST confirmed ORR was 21% (6 PR). Among the 6 patients with known pre/post-treatment viral titers (2 HCV and 4 HBV), there was no evidence of viral reactivation.ConclusionsOur retrospective series is one of the largest case series to report clinical outcomes among HIV, HBV and HCV patients treated with ICI therapy. Toxicity and efficacy rates were similar to those observed in patients without chronic viral infections. Viral reactivation was not observed. Tumor responses occurred in HIV patients with low CD4 T-cell counts. While prospective studies are needed to validate above findings, these data support not excluding such patients from ICI–based clinical trials or treatment.

Precise measurements of chemical abundances in planetary atmospheres are necessary to constrain the formation histories of exoplanets. A recent study of WASP-127 b, a close-in puffy ...sub-Saturn orbiting its solar-type host star in 4.2 d, using Hubble Space Telescope (HST) and Spitzer revealed a feature-rich transmission spectrum with strong excess absorption at 4.5 μm. However, the limited spectral resolution and coverage of these instruments could not distinguish between CO and/or CO2 absorption causing this signal, with both low and high C/O ratio scenarios being possible. Here we present near-infrared (0.9–2.5 μm) transit observations of WASP-127 b using the high-resolution SPIRou spectrograph, with the goal to disentangle CO from CO2 through the 2.3 μm CO band. With SPIRou, we detect H2O at a t-test significance of 5.3 σ and observe a tentative (3σ) signal consistent with OH absorption. From a joint SPIRou + HST + Spitzer retrieval analysis, we rule out a CO-rich scenario by placing an upper limit on the CO abundance of log10CO <−4.0, and estimate a log10CO2 of −3.7$^{+0.8}_{-0.6}$ , which is the level needed to match the excess absorption seen at 4.5 μm. We also set abundance constraints on other major C-, O-, and N-bearing molecules, with our results favouring low C/O (0.10$^{+0.10}_{-0.06}$ ), disequilibrium chemistry scenarios. We further discuss the implications of our results in the context of planet formation. Additional observations at high and low resolution will be needed to confirm these results and better our understanding of this unusual world.
