Anaerobic digestates from sewage sludge (SSADs) are a by-product of the wastewater treatment process that still preserves a certain agronomic interest for its richness in plant nutrients and organic ...matter. Fertilizing properties of two liquid and two dewatered SSADs were tested on tomato plants (Solanum lycopersicum L.). Pot experiments were performed on sandy soil and peat substrate under greenhouse conditions with a SSADs application rate of 170 kg N/ha over a period of three months. Beneficial effects of SSADs were reported on different growth parameters, revealing an increase in biomass and height up to 37.5 and 6-folds over untreated control. No phytotoxic effect occurred on SSAD-exposed plants. Chemical analysis of soils treated with SSADs showed enrichment of macro- and micro-nutrients as well as organic matter. In some cases, the chemical characterization of leaves revealed an enhancement of uptaken macronutrients. This study contributed in general to deepen the knowledge on the short-term growing season fertilizing effects of SSAD. Despite the treatment dosage was calculated only on nitrogen requirements, the study highlighted the importance of the other nutrients and organic matter on plant growth.

Sewage sludge production in European countries has widely raised in the last decade and its fate is currently landfilling, incinerators, composting or land application. To explore its agronomic potential, the main target of this work is to understand the effects of anaerobic digestates from sewage sludge (SSAD). To this aim, four different SSADs (two liquids and two dewatered) were characterized. On the liquid ones, Germination Index was evaluated through a plate bioassay with Lepidium sativum L. seeds; low concentrations of SSAD (2.5%) improved GI in one case, while at higher concentrations phytotoxic effects occurred in both. Then, pot experiments were set in climate chamber with Cucumis sativus L. grown for 30 days on two different substrates: a sandy, alkaline and poor soil, and peat substrate. All SSADs and a mineral fertilizer were used at three increasing dosages: 85, 170, 255 kg of nitrogen per hectare (kg N/ha). Results in terms of germination, dry biomass, chlorophyll content, net photosynthesis, stomatal conductance, CO2 concentration in substomatal cavity and root development were compared to a not treated control.
All treatments gave results significantly higher or similar to control on all the parameters evaluated. Moreover, the intermediate nitrogen dosage (170 kg N/ha) generally showed the highest results compared to other dosages, especially for dewatered SSADs. All these results were much more evident for cucumber plants grown on an the alkaline, sandy and poor soil than on peat substrate, such demonstrating that SSADs have a fertilizing effect for plants growing on this kind of soil.
Wastewater production is rising all over the world and one of the most difficult problems is the disposal of sewage sludge (SS). It is known that SS contains certain quantities of added-value ...compounds, such as humic acids (HA) which in turn have beneficial effects on soil quality and plant growth. On the other hand, SS can retain many pollutants, such as heavy metals. The present work aimed to implement an HA alkaline extraction protocol from anaerobic sewage sludge (ASS). Subsequently, the HA were quantified in ASS, in HA extract and in commercial HA, used as a benchmark, which gave results of 12.53%, 26.87% and 77.87% (on dry matter basis), respectively. FESEM and EDX analyses on lyophilized HA extract confirmed that no heavy metals had passed into the extract. Afterwards, in order to allow controlled release of the HA in soils, alginate beads containing the HA extract were created. Finally, a pot experiment in a greenhouse was performed using Chilean lettuce plants (Lactuca sativa L.) treated with alginate-HA extract beads. At the end of the greenhouse experiments, the hypogean dry biomass of the treated plants was significantly higher than for non-treated plants. The relevance of this study relies not only on the exploitation of green chemistry principles, by converting a waste stream into a high-value product, but also on the application of an approach following a circular economy model.

Reported here is the correlation between the transfection efficiency of cationic liposome/DNA complexes (lipoplexes) and the structural evolution that they undergo when interacting with anionic ...membrane lipids. Multicomponent lipoplexes, incorporating from three to six lipid species simultaneously, presented a much higher transfection efficiency than binary lipoplexes, which are more commonly used for gene-delivery purposes. The discovery that a high transfection efficiency can be achieved by employing multicomponent complexes at a lower-than-ever-before membrane charge density of lipoplexes was of primary significance. Synchrotron small-angle X-ray diffraction (SAXD) experiments showed that anionic liposomes made of dioleoylphosphatidylglycerol (DOPG) disintegrated the lamellar phase of lipoplexes. DNA unbinding was measured by electrophoresis on agarose gels. Most importantly, structural changes induced by anionic lipids strictly depended on the lipid composition of lipoplexes. We found evidence of the existence of three different regimes of stability related to the interaction between complexes and anionic membranes. Both unstable (with low membrane charge density, σM) and highly stable lipoplexes (with high σM) exhibited low transfection efficiency whereas highly efficient multicomponent lipoplexes exhibited an “optimal stability”. This intermediate regime reflects a compromise between two opposing constraints: protection of DNA in the cytosol and endosomal escape. Here we advance the concept that structural stability, upon interaction with cellular anionic lipids, is a key factor governing the transfection efficiency of lipoplexes. Possible molecular mechanisms underlying experimental observations are also discussed.
The lifelong education of coaches is one of the priorities of the European Union. Therefore, the purpose of this study was to investigate Italian elite coaches' motivation to engage in a ...sport-related academic education, and its demands, barriers, support, and relocation issues in relation to their dual career (DC) path. Sixteen Italian elite coaches (e.g., certified fourth-level national team coaches, senior team coaches) enrolled in a specifically tailored Bachelor's degree in sports sciences at the University of Rome Foro Italico (Italy) volunteered for this study. A qualitative approach integrating inductive and deductive reasoning, and thematic analysis was applied to participants' responses to an open-ended item survey. Independently from relocation, student-coaches' DC perceptions resulted in 15 lower-order themes further organized in 5 high-er-order themes (e.g., Benefit, Challenge, Expectation, Organization, and Support), each related to the contexts (e.g., Personal, Academic, Sport), the DC dimension (e.g., micro, meso, macro, and policy), and the DC push (e.g., facilitating) /pull (e.g., hindering) factors. The elite coaches' insights emphasize the complexity of the coach lifelong education at university level, and provide valuable information for promoting European and National (e.g., Italian) DC recommendations for elite sportspersons through a cooperation between sport bodies and higher educational institutions.
The swimming larvae of most solitary ascidians belonging to the Ascidiidae family bear three anterior, simple conic adhesive papillae. They secrete adhesive substances that are used to effect ...transitory settlement at the beginning of the metamorphosis. The adhesive papillae of newly hatched Phallusia mamillata larvae examined by the SEM are covered by the tunic. When the larvae are about to settle, the tunic becomes fenestrated over the central part of the papilla and bulb-ended microvilli protrude through the holes. These papillae have two types of elongated cells: many peripheral cells and few larger central cells with microvilli and bundles of microtubules oriented along the major axis of the cells. We have done immunofluorescence experiments with an anti-β-tubulin monoclonal antibody (clone 2-28-33) reacting with axonal microtubules. Only the central cells of the papillae were stained and the axons appeared to arise from the proximal ends of these cells. These axons form a long nerve that reaches the brain vesicle. Branches of the same nerve appear to connect to the basal ends of the peripheral cells. By confocal laser microscopy we were able to follow the course of the papillary nerve. The two nerves connecting the dorsal papillae fuse together into a single nerve that runs posteriorly. The nerve connecting the ventral papilla runs posteriorly for a long tract before fusing with the nerve of the dorsal papillae just near the brain. The reported observations raise the hypothesis that the central cells of the adhesive papillae might be primary sensory neurons and that they may have chemosensory function.
In semiarid regions, vegetated ecosystems can display abrupt and unexpected changes, i.e., transitions to different states, due to drifting or time-varying parameters, with severe consequences for ...the ecosystem and the communities depending on it. Despite intensive research, the early identification of an approaching critical point from observations is still an open challenge. Many data analysis techniques have been proposed, but their performance depends on the system and on the characteristics of the observed data (the resolution, the level of noise, the existence of unobserved variables, etc.). Here, we propose an entropy-based approach to identify an upcoming transition in spatiotemporal data. We apply this approach to observational vegetation data and simulations from two models of vegetation dynamics to infer the arrival of an abrupt shift to an arid state. We show that the permutation entropy (PE) computed from the probabilities of two-dimensional ordinal patterns may provide an early warning indicator of an approaching tipping point, as it may display a maximum (or minimum) before decreasing (or increasing) as the transition approaches. Like other spatial early warning indicators, the spatial permutation entropy does not need a time series of the system dynamics, and it is suited for spatially extended systems evolving on long time scales, like vegetation plots. We quantify its performance and show that, depending on the system and data, the performance can be better, similar or worse than the spatial correlation. Hence, we propose the spatial PE as an additional indicator to try to anticipate regime shifts in vegetated ecosystems.

Identifying approaching bifurcations and regime transitions from observations is an important challenge in time series analysis with practical applications in many fields of science. Well-known indicators are the increase in spatial and temporal correlations. However, the performance of these indicators depends on the system under study and on the type of approaching bifurcation, and no indicator provides a reliable warning for any system and bifurcation. Here we propose an indicator that simultaneously takes into account information about spatial and temporal correlations. By performing a bivariate correlation analysis of signals recorded in pairs of adjacent spatial points, and analyzing the distribution of lag times that maximize the cross-correlation, we find that the variance of the lag distribution displays an extreme value that is a consistent early warning indicator of the approaching bifurcation. We demonstrate the reliability of this indicator using different types of models that present different types of bifurcations, including local bifurcations (transcritical, saddle-node, supercritical and subcritical Hopf), and global bifurcations.
Cirrhosis is a diffuse pathophysiological state of the liver considered to be the final stage of various liver injuries, characterized by chronic necroinflammatory and fibrogenetic processes, with ...subsequent conversion of normal liver architecture into structurally abnormal nodules, dense fibrotic septa, concomitant parenchymal exaustment and collapse of the liver tissue. Alcoholic liver disease and chronic infections due to HBV and/or HCV constitute the main causes of liver cirrhosis worldwide. During a lag time of 15 to 30 years, chronic liver diseases can lead to liver cirrhosis and its complications. Active hepatic inflammation plays a pivotal role in the inflammation- necrosis-regeneration process, which eventually leads to liver cirrhosis and hepatocellular carcinoma. Prognosis of liver cirrhosis is highly variable and influenced by several variables, such as etiology, severity of liver disease, presence of complications and comorbidities. In advanced cirrhosis, survival decreases to one or two years. Correct advanced diagnosis and selected treatment with different molecules may help in understanding mechanisms of fibrogenesis, the driving forces of cirrhosis's pathogenesis, and the scrupulous approach to more effective therapeutic procedures. Prevention of fibrosis with further deterioration of liver function through specific treatments is always required, through the removal of the underlying causes of liver disease. Advanced liver disease, with subsequent complications, requires targeted treatment. Therefore, the aim of this review is to assess the diagnosis and treatment of liver cirrhosis on the pathophysiological bases, searching for relevant studies published in English using the PubMed database from 2011 to the present.

Barro et al. show that serum ...neurofilament light chain (NfL) levels are increased in patients versus healthy controls and reflect radiological and clinical disease activity and progression. High serum NfL is associated with greater subsequent brain and spinal cord atrophy.
Abstract
Neuro-axonal injury is a key factor in the development of permanent disability in multiple sclerosis. Neurofilament light chain in peripheral blood has recently emerged as a biofluid marker reflecting neuro-axonal damage in this disease. We aimed at comparing serum neurofilament light chain levels in multiple sclerosis and healthy controls, to determine their association with measures of disease activity and their ability to predict future clinical worsening as well as brain and spinal cord volume loss. Neurofilament light chain was measured by single molecule array assay in 2183 serum samples collected as part of an ongoing cohort study from 259 patients with multiple sclerosis (189 relapsing and 70 progressive) and 259 healthy control subjects. Clinical assessment, serum sampling and MRI were done annually; median follow-up time was 6.5 years. Brain volumes were quantified by structural image evaluation using normalization of atrophy, and structural image evaluation using normalization of atrophy, cross-sectional, cervical spinal cord volumes using spinal cord image analyser (cordial). Results were analysed using ordinary linear regression models and generalized estimating equation modelling. Serum neurofilament light chain was higher in patients with a clinically isolated syndrome or relapsing remitting multiple sclerosis as well as in patients with secondary or primary progressive multiple sclerosis than in healthy controls (age adjusted P < 0.001 for both). Serum neurofilament light chain above the 90th percentile of healthy controls values was an independent predictor of Expanded Disability Status Scale worsening in the subsequent year (P < 0.001). The probability of Expanded Disability Status Scale worsening gradually increased by higher serum neurofilament light chain percentile category. Contrast enhancing and new/enlarging lesions were independently associated with increased serum neurofilament light chain (17.8% and 4.9% increase per lesion respectively; P < 0.001). The higher the serum neurofilament light chain percentile level, the more pronounced was future brain and cervical spinal volume loss: serum neurofilament light chain above the 97.5th percentile was associated with an additional average loss in brain volume of 1.5% (P < 0.001) and spinal cord volume of 2.5% over 5 years (P = 0.009). Serum neurofilament light chain correlated with concurrent and future clinical and MRI measures of disease activity and severity. High serum neurofilament light chain levels were associated with both brain and spinal cord volume loss. Neurofilament light chain levels are a real-time, easy to measure marker of neuro-axonal injury that is conceptually more comprehensive than brain MRI.
