Despite the ratio of incoming discharges being recognized as a key parameter in open-channel confluence hydrodynamics, little is known about the flow patterns when the tributary provides more than ...90% of the total discharge. This paper offers a systematic study of flow features when the tributary becomes increasingly dominant in a 90° confluence with a fixed concordant bed. Large-eddy simulations are used to investigate the three-dimensional complex flow patterns for three different discharge ratios. It is found that the tributary flow impinges on the opposing bank when the tributary flow becomes sufficiently dominant, causing a recirculating eddy in the upstream channel of the confluence, which induces significant changes in the incoming velocity distribution. Moreover, it results in stronger helicoidal cells in the downstream channel, along with zones of upwelling flow. In turn, the changed flow patterns also influence the mixing layer and the flow recovery. Finally, intermittent events of stronger upwelling flow are discerned. Improved understanding of flow patterns at confluences where the tributary is dominant is applicable to both engineering and earth sciences.
Cancer immunotherapies have induced long-lasting responses in cancer patients including those with melanoma and head and neck squamous cell carcinoma (HNSCC). However, the majority of treated ...patients does not achieve clinical benefit from immunotherapy because of systemic tumor-induced immunosuppression. Monocytic myeloid-derived suppressor cells (M-MDSCs) are implicated as key players in inhibiting anti-tumor immune responses and their frequencies are closely associated with tumor progression. Tumor-derived signals, including signaling via STAT3-COX-2, induce the transformation of monocytic precursors into suppressive M-MDSCs. In a retrospective assessment, we observed that survival of melanoma patients undergoing dendritic cell vaccination was negatively associated with blood M-MDSC levels. Previously, it was shown that platinum-based chemotherapeutics inhibit STAT signaling. Here, we show that cisplatin and oxaliplatin treatment interfere with the development of M-MDSCs, potentially synergizing with cancer immunotherapy. +
M-MDSCs induced by tumor cells from melanoma patients. This was confirmed in HNSCC patients where intravenous cisplatin treatment drastically lowered M-MDSC frequency while monocyte levels remained stable. In treated patients, expression of COX-2 and arginase-1 in M-MDSCs was significantly decreased after two rounds of cisplatin, indicating inhibition of STAT3 signaling. In line, the capacity of M-MDSCs to inhibit activated T cell responses ex vivo was significantly decreased after patients received cisplatin. These results show that platinum-based chemotherapeutics inhibit the expansion and suppressive activity of M-MDSCs in vitro and in cancer patients. Therefore, platinum-based drugs have the potential to enhance response rates of immunotherapy by overcoming M-MDSC-mediated immunosuppression.
To investigate the historical development of the tidally averaged transport of sandy sediments in the main branch of the Scheldt estuary over the last decades (1950–2013), a 2D exploratory model has ...been developed. This model comprises the depth-averaged (2DH) shallow water equations, driven by an asymmetric tidal forcing at the seaward side, as well as an advection-diffusion equation to describe the depth-integrated dynamics of the suspended sediment concentration. The tidally averaged sand transport results from a subtle balance between the various contributions; advective contributions due to internally generated and externally prescribed overtides and the diffusive and topographically induced contributions. A seaward tidally averaged sand transport is found near the open boundary, whose magnitude has increased since ca. 1950. Moving upstream, the magnitude of the seaward transport decreases and changes into a smaller landward transport with a local maximum near the landward boundary. This maximum has increased over the years. Varying parameters that capture changes in the environment, e.g. historical changes in the bathymetry, future mean sea-level rise or changes in tidal forcing, results in changes in the tidally averaged sand transport that are systematically analysed and related to changes in the various contributions. Our model shows that there is a competition, in terms of determining the magnitude and the direction of the tidally averaged sand transport, between the effects of historical bathymetric changes, changes in tidal forcing and (projected) SLR. Even small changes in the tidal forcing at the seaward boundary can have a large impact on the magnitude and the direction of the tidally averaged sand transport. This hampers accurate predictions of sediment transport and morphodynamic changes in tidal systems, due to the uncertainty in the response of the tidal dynamics to the projected sea-level rise.
The presented model allows for fast simulations of the near-field behaviour of overflow dredging plumes. Overflow dredging plumes occur when dredging vessels employ a dropshaft release system to ...discharge the excess sea water, which is pumped into the trailing suction hopper dredger (TSHD) along with the dredged sediments. The fine sediment fraction in the loaded water-sediment mixture does not fully settle before it reaches the overflow shaft. By consequence, the released water contains a fine sediment fraction of time-varying concentration. The sediment grain size is in the range of clays, silt and fine sand; the sediment concentration varies roughly between 10 and 200 g/l in most cases, peaking at even higher value with short duration. In order to assess the environmental impact of the increased turbidity caused by this release, plume dispersion predictions are often carried out. These predictions are usually executed with a large-scale model covering a complete coastal zone, bay, or estuary. A source term of fine sediments is implemented in the hydrodynamic model to simulate the fine sediment dispersion. The large-scale model mesh resolution and governing equations, however, do not allow to simulate the near-field plume behaviour in the vicinity of the ship hull and propellers. Moreover, in the near-field, these plumes are under influence of buoyancy forces and air bubbles. The initial distribution of sediments is therefore unknown and has to be based on crude assumptions at present. The initial (vertical) distribution of the sediment source is indeed of great influence on the final far-field plume dispersion results. In order to study this near-field behaviour, a highly-detailed computationally fluid dynamics (CFD) model was developed. This model contains a realistic geometry of a dredging vessel, buoyancy effects, air bubbles and propeller action, and was validated earlier by comparing with field measurements. A CFD model requires significant simulation times, which is not available in all situations. For example, to allow correct representation of overflow plume dispersion in a real-time forecasting model, a fast assessment of the near-field behaviour is needed. For this reason, a semi-analytical parameter model has been developed that reproduces the near-field sediment dispersion obtained with the CFD model in a relatively accurate way. In this paper, this so-called grey-box model is presented.
Confluences with dominant tributary inflow are found to exhibit long-periodic alternations of the flow patterns. They are shown to exist both in laboratory experiments and in numerical simulations. ...By means of a modal decomposition, insight is given into these long-periodic oscillations. The origin of these oscillations is investigated and their significant influence on the secondary flow patterns in the downstream channel is revealed.
Confluences play a major role in the dynamics of networks of natural and man-made open channels, and field measurements on river confluences reveal that discordance in bed elevation is common. ...Studies of schematized confluences with a step at the interface between the tributary and the main channel bed reveal that bed elevation discordance is an important additional control for the confluence hydrodynamics. This study aimed to improve understanding of the influence of bed elevation discordance on the flow patterns and head losses in a right-angled confluence of an open channel with rectangular cross-sections. A large eddy simulation (LES)-based numerical model was set up and validated with experiments by others. Four configurations with different bed discordance ratios were investigated. The results confirm that, with increasing bed elevation discordance, the tributary streamlines at the confluence interface deviate less from the geometrical confluence angle, the extent of the recirculation zone (RZ) gets smaller, the ratio of the water depth upstream to that downstream of the confluence decreases, and the water level depression reduces. The bed elevation discordance also leads to the development of a large-scale structure in the lee of the step. Despite the appearance of the large-scale structure, the reduced extent of the RZ and associated changes in flow deflection/contraction reduce total head losses experienced by the main channel with an increase of the bed discordance ratio. It turns out that bed elevation discordance converts the lateral momentum from the tributary to streamwise momentum in the main channel more efficiently.

Background
Acoustic positioning telemetry is nowadays widely used in behavioural ecology of aquatic animals. Data on the animal’s geographical location and its changes through time are used ...to study for instance movement patterns, habitat use and migration. The acoustic signals are detected by stand-alone receivers, allowing to collect huge amounts of data over long periods of time. However, large volumes of data might contain large errors. The traditional Time Difference of Arrival (TDOA) method used to calculate underwater positions, is a point-by-point approach: every position is calculated independently of all other positions. This method assumes that the acoustic transmissions follow a linear path. In many environments, this assumption is violated, for instance by reflections of the acoustic signal against hard surfaces, such as rock formations and concrete walls, or by diffractions around obstacles. Hence, acoustic positioning datasets usually require additional filtering. Unfortunately, the performance of the available filtering techniques is often unclear or ambiguous, especially when reflections occur. An alternative to the point-by-point approach, is a track-oriented approach, as used by YAPS (Yet Another Positioning System). This novel algorithm uses the information that is present in previous and subsequent positions, by combining a model of fish behaviour with Time of Arrival (TOA) of the signals on the receivers. In this study, we investigated the performance of two filtering techniques applied to positions provided by the Vemco Positioning System (VPS) in a highly reflective environment. We compared the unfiltered VPS positions with a standard filtering technique, making use of the Horizontal Positioning Error (HPE), and developed a new filter based on receiver cluster classification. Finally, we recalculated the positions with YAPS and compared the performance of this system to the two filtering techniques.
Results
The performance of the VPS system was strongly impeded by the multiple reflections occurring in this study area, but lowering the power output of transmitters can slightly attenuate this issue. None of the filtering techniques was able to compensate for reflections and to improve the positioning accuracy significantly. Only the YAPS algorithm could cope with the high level of reflectivity in this study site.
Conclusions
Point-by-point algorithms might fail to provide accurate fine-scale tracks in a highly reflective acoustic environment. As this study has shown, the YAPS algorithm can provide a successful alternative, even in these difficult conditions.

Background
Acoustic positioning telemetry allows to collect large amounts of data on the movement of aquatic animals by use of autonomous receiver stations. Essential in this process is the ...conversion from raw signal detections to reliable positions. A new advancement in the domain is Yet Another Positioning Solver (YAPS), which combines the detection data on the receivers with a model of animal movement. This transparent, flexible and on-line available positioning algorithm overcomes problems related to traditional point-by-point positioning and filtering techniques. However, its performance has only been tested on data from one telemetry system, providing transmitters with stable burst interval. To investigate the performance of YAPS on different system parameters and settings, we conducted a simulation study.
Results
This paper discusses the effect of varying burst types, burst intervals, number of observations, reflectivity levels of the environment, levels of out-of-array positioning and temporal receiver resolution on positioning accuracy. We found that a receiver resolution better than 1 ms is required for accurate fine-scale positioning. The positioning accuracy of YAPS increases with decreasing burst intervals, especially when the number of observations is low, when reflectivity is high or when information out-of-array is used. However, when the burst interval is stable, large burst intervals (in the order of 1 to 2 min) can be chosen without strongly hampering the accuracy (although this results in information loss). With random burst intervals, the accuracy can be much improved if the random sequence is known.
Conclusions
As it turns out, the key to accurate positioning is the burst type. If a stable burst interval is not possible, the availability of the random sequence improves the positioning of random burst interval data significantly.
