The southeast region of Brazil experienced in austral summer 2014 a major drought event leading to a number of impacts in water availability for human consumption, agricultural irrigation and ...hydropower production. This study aims to perform a diagnostic analysis of the observed climate conditions during this event, including an inspection of the occurred precipitation anomalies in the context of previous years, and an investigation of possible relationships with sea surface temperatures and atmospheric circulation patterns. The sea surface temperature analysis revealed that the southwestern South Atlantic Ocean region near the coast of southeast Brazil showed strong negative association with precipitation over southeast Brazil, indicating that increased sea temperatures in this ocean region are consistent with reduced precipitation as observed in summer 2014. The circulation analysis revealed prevailing anti-cyclonic anomalies at lower levels (850 hPa) with northerly anomalies to the west of southeast Brazil, channeling moisture from the Amazon towards Paraguay, northern Argentina and southern Brazil, and drier than normal air from the South Atlantic Ocean towards the southeast region of Brazil. This circulation pattern was found to be part of a large-scale teleconnection wave train linked with the subsidence branch of the Walker circulation in the tropical east Pacific, which in turn was generated by an anomalous tropical heat source in north/northeastern Australia. A regional Hadley circulation with an ascending branch to the south of the subsidence branch of the Walker circulation in the tropical east Pacific was identified as an important component connecting the tropical and extratropical circulation. The ascending branch of this Hadley circulation in the south Pacific coincided with an identified Rossby wave source region, which contributed to establishing the extratropical component of the large-scale wave train connecting the south Pacific and the Atlantic region surrounding southeast Brazil. This connection between the Pacific and the Atlantic was confirmed with Rossby ray tracing analyses. The local circulation response was associated to downward air motion (subsidence) over Southeast Brazil, contributing to the expressive negative precipitation anomalies observed during summer 2014, and leading to a major drought event in the historical context. The analysis of atmospheric and oceanic patterns of this event helped defining a schematic framework leading to the observed drought conditions in southeast Brazil, including the involved teleconnections, blocking high pressure, radiative and humidity transport effects.
This pilot study evaluated the clinical efficacy of a mouthwash containing 1% Matricaria chamomilla L. (MTC) extract in reducing gingival inflammation and plaque formation in patients undergoing ...orthodontic treatment with fixed appliances. This randomized, double-blind, placebo-controlled study enrolled a total of 30 males and females (age, 10-40 years) with fixed orthodontic appliances and a minimum of 20 natural teeth. The participants were allocated to three groups (n = 10 each) and asked to rinse with 15 mL of a placebo, 0.12% chlorhexidine (CHX), or 1% MTC mouthwash, immediately after brushing for 1 min, in the morning and evening, for 15 days. Data (mean ± SD) on visible plaque index (VPI) and gingival bleeding index (GBI) were recorded on days 1 and 15. The placebo group exhibited increases in VPI and GBI (10.2% and 23.1%, respectively) from day 1 to day 15. As compared with placebo, VPI and GBI significantly decreased in the MTC group (−25.6% and −29.9%, respectively) and the CHX group (−39.9% and −32.0%, respectively). In summary, MTC reduced biofilm accumulation and gingival bleeding in patients with gingivitis, probably because of its antimicrobial and anti-inflammatory activities.(J Oral Sci 58, 569-574, 2016)
Abstract Objective The study aims to evaluate the effect of alendronate (ALD) on bone-specific alkaline phosphatase (BALP) serum levels on periodontal bone loss in Wistar rats. Design Periodontitis ...was induced by ligature around the upper second molar in 36 male Wistar rats (±200 g). Groups of six animals received 0.9% saline (SAL) or ALD (0.01; 0.05; 0.25 mg kg−1 , s.c.), over 11 days; then they were sacrificed and their maxillae were removed to be defleshed and stained for macroscopic or histopathological analysis. Blood samples were collected for BALP, transaminases and total alkaline phosphatase (TALP) serum dosage, and haematologic study. Rats were weighed daily. Results Periodontitis induction caused reduction of BALP, intense alveolar bone loss (ABL), cementum and periodontal ligament destructions and intense leucocyte infiltration seen microscopically. Systemically, periodontitis induced leucocytosis, weight loss and TALP reduction. ALD (0.25 mg kg−1 ) prevented BALP reduction (19.17 ± 1.36 U l−1 ) when compared to SAL (13.6 ± 1.5), as well as prevented ABL, by 57.2%, when compared to SAL (4.80 + 0.18 mm2 ), which was corroborated by histological findings (ALD 0.25 mg kg−1 = 1.5 (1–2) and SAL = 3 (2–3)) ( p < 0.05). ALD did not alter transaminases but reduced TALP levels ( p < 0.05). ALD 0.25 mg kg−1 reduced 6th-h neutrophilia (2.50 ± 0.22 cell × 103 mm−3 ) and 7th- (12.29 ± 0.66) and 11th-day lymphomonocytosis (15.74 ± 0.52) when compared to SAL (5.20 ± 0.28; 18.24 ± 1.05; and 23.21 ± 1.48, respectively). ALD did not alter the weight loss. Conclusion ALD prevented BALP reduction and ABL and reduced inflammatory infiltrate, without causing systemic alterations.

The essential oil at 100, 300 and 500 mg/kg exhibited significant protection against ulcers induced by ethanol, indometacin and HCl/ethanol (P < 0.001). The essential oil caused a marked reduction in the exudate volume and inhibited leucocyte and neutrophil influx (P < 0.05) in carrageenan-induced pleurisy. Moreover, the essential oil significantly decreased nitric oxide (NO) and interleukin-1 (IL-1) levels, without affecting tumour necrosis factor-alpha production.

This study describes a methodology to prepare and characterize PUF poly(urea formaldehyde) microcapsules containing 5-ethylidene-2-norbornene (ENB) as self-healing agent, as well as the compatibility ...evaluation of ENB in the presence of Grubb's catalyst with an epoxy matrix. First, the results of an adhesion assay by lap shear proved chemical compatibility between the epoxy matrix and self-healing agent in the presence of Grubb's catalyst. After evaluating the chemical compatibility, microcapsules with ENB were synthesized in an oil-in-water emulsion system. Thereafter, the microcapsules were morphologically, chemically and thermally characterized in which a granulometric dispersion between 30-140 μm and an average size of 69.8 ± 4.9 μm were observed. The SEM (scanning electron microscope) results showed that the average thickness of microcapsules shell was 3.6 ± 0.4 μm. The thermogravimetric analysis (TGA) showed that microcapsules are susceptible to rupture and consequent ENB release in temperatures greater than 230 °C, demonstrating that microcapsules are suitable for applications in materials with self-healing capacity.
Genomic studies may generate massive amounts of data, bringing interpretation challenges. Efforts for the differentiation of benign and pathogenic variants gain importance. In this article, we used ...segregation analysis and other molecular data to reclassify to benign or likely benign several rare clinically curated variants of autosomal dominant inheritance from a cohort of 500 Brazilian patients with rare diseases. This study included only symptomatic patients who had undergone molecular investigation with exome sequencing for suspected diseases of genetic etiology. Variants clinically suspected as the causative etiology and harbored by genes associated with highly-penetrant conditions of autosomal dominant inheritance underwent Sanger confirmation in the proband and inheritance pattern determination because a "de novo" event was expected. Among all 327 variants studied, 321 variants were inherited from asymptomatic parents. Considering segregation analysis, we have reclassified 51 rare variants as benign and 211 as likely benign. In our study, the inheritance of a highly penetrant variant expected to be de novo for pathogenicity assumption was considered as a non-segregation and, therefore, a key step for benign or likely benign classification. Studies like ours may help to identify rare benign variants and improve the correct interpretation of genetic findings.
Type 1 diabetes mellitus (T1DM) is associated with low bone mass and a higher risk for fractures. Here, we investigated the functional role of Dkk1 in T1DM-induced bone loss in mice. T1DM was induced in 10-week-old male mice with Dkk1-deficiency in late osteoblasts/osteocytes (Dkk1
;Dmp1-Cre, cKO) and littermate control mice by 5 subsequent injections of streptozotocin (40 mg/kg). Age-matched, non-diabetic control groups received citrate buffer instead. At week 12, calvarial defects were created in subgroups of each cohort. After a total of 16 weeks, weight, fat, the femoral bone phenotype and the area of the bone defect were analyzed using µCT and dynamic histomorphometry. During the experiment, diabetic WT and cKO mice did not gain body weight compared to control mice. Further they lost their perigonadal and subcutaneous fat pads. Diabetic mice had highly elevated serum glucose levels and impaired glucose tolerance, regardless of their Dkk1 levels. T1DM led to a 36% decrease in trabecular bone volume in Cre- negative control animals, whereas Dkk1 cKO mice only lost 16%. Of note, Dkk1 cKO mice were completely protected from T1DM-induced cortical bone loss. T1DM suppressed the bone formation rate, the number of osteoblasts at trabecular bone, serum levels of P1NP and bone defect healing in both, Dkk1-deficient and sufficient, mice. This may be explained by increased serum sclerostin levels in both genotypes and the strict dependence on bone formation for bone defect healing. In contrast, the number of osteoclasts and TRACP 5b serum levels only increased in diabetic control mice, but not in Dkk1 cKO mice. In summary, Dkk1 derived from osteogenic cells does not influence the development of T1DM but plays a crucial role in T1DM-induced bone loss in male mice by regulating osteoclast numbers.
Pulmonary tuberculosis (PTB) can lead to lung tissue damage (LTD) and compromise the pulmonary capacity of TB patients that evolve to severe PTB. The molecular mechanisms involved in LTD during ...anti-tuberculous treatment (ATT) remain poorly understood.
We evaluated the role of neutrophil extracellular trap (NET) and the occurrence of LTD through chest radiographic images, the microbial load in sputum, and inflammatory serum profile (IL-12p40/p70, IL-8, IL-17A, IL-23, VEGF-A, MMP-1, and -8, galectin-3, citrunillated histone H3-cit-H3, alpha-1-antitrypsin-α1AT, C-reactive protein-CRP and albumin) in a cohort of 82 PTB patients before and after 60 days of ATT. Using univariate analysis, LTD was associated with neutrophilia and increase of several inflammatory proteins involved in the neutrophil-mediated response, being cit-H3 the more related to the event. In the multivariate analysis, neutrophilia and cit-H3 appear as directly related to LTD. The analysis of the ROC curve at day 60 presented AUC of 0.97 (95.0% CI 0.95-1). Interestingly, at day 0 of ATT, these biomarkers demonstrated fine relation with LTD showing an AUC 0.92 (95.0% CI 0.86-0.99). Despite of that, the same molecules have no impact in culture conversion during ATT.
Our data revealed that NETs may play a key role in the pathway responsible for non-specific inflammation and tissue destruction in PTB. High level of cit-H3 and low level of α1AT was observed in the serum of severe TB patients, suggesting a breakdown in the intrinsic control of NET-driven tissue damage. These data show a new insight to knowledge TB immunopathogenesis, the role of neutrophil and NET pathway. Likewise, we identified possible biomarkers to screening of PTB patients eligible to adjuvants therapies, as anti-inflammatories and alpha-1-antitrypsin.
The taxonomy of Polyporales is complicated by the variability in key morphological characters across families and genera, now being gradually resolved through molecular phylogenetic analyses. Here a ...new resupinate species, Crystallicutis damiettensis sp. nov. found on the decayed trunks of date palm (Phoenix dactylifera) trees in the fruit orchards of the Nile Delta region of Egypt is reported. Multigene phylogenetic analyses based on ITS, LSU, EF1α, RPB1 and RPB2 loci place this species in Irpicaceae, and forming a distinct clade with Ceraceomyces serpens and several other hitherto unnamed taxa, which we also incorporate into a new genus Crystallicutis. We name two of these species, Crystallicutis huangshanensis sp. nov. and Crystallicutis rajchenbergii sp. nov. The distinctive feature of Crystallicutis gen. nov. is the presence of crystal-encrusted hyphae in the hymenium and subiculum. Basidiomes are usually honey-yellow with white margins but there is variability in the presence of clamp connections and cystidia, as noted for other genera within Irpicacae. C. damiettensis is hitherto consistently associated with date palms killed by the red palm weevil Rhynchophorus ferrugineus, a highly damaging and invasive pest, recently spread to the Mediterranean region. C. damiettensis causes rapid wood decay by a potentially unusual white-rot mechanism and may play a role in the damage caused by R. ferrugineus.
