We present a new common stratigraphic timescale for the North Greenland Ice Core Project (NGRIP) and GRIP ice cores. The timescale covers the period 7.9–14.8 kyr before present and includes the ...Bølling, Allerød, Younger Dryas, and early Holocene periods. We use a combination of new and previously published data, the most prominent being new high‐resolution Continuous Flow Analysis (CFA) impurity records from the NGRIP ice core. Several investigators have identified and counted annual layers using a multiparameter approach, and the maximum counting error is estimated to be up to 2% in the Holocene part and about 3% for the older parts. These counting error estimates reflect the number of annual layers that were hard to interpret, but not a possible bias in the set of rules used for annual layer identification. As the GRIP and NGRIP ice cores are not optimal for annual layer counting in the middle and late Holocene, the timescale is tied to a prominent volcanic event inside the 8.2 kyr cold event, recently dated in the DYE‐3 ice core to 8236 years before A. D. 2000 (b2k) with a maximum counting error of 47 years. The new timescale dates the Younger Dryas‐Preboreal transition to 11,703 b2k, which is 100–150 years older than according to the present GRIP and NGRIP timescales. The age of the transition matches the GISP2 timescale within a few years, but viewed over the entire 7.9–14.8 kyr section, there are significant differences between the new timescale and the GISP2 timescale. The transition from the glacial into the Bølling interstadial is dated to 14,692 b2k. The presented timescale is a part of a new Greenland ice core chronology common to the DYE‐3, GRIP, and NGRIP ice cores, named the Greenland Ice Core Chronology 2005 (GICC05). The annual layer thicknesses are observed to be log‐normally distributed with good approximation, and compared to the early Holocene, the mean accumulation rates in the Younger Dryas and Bølling periods are found to be 47 ± 2% and 88 ± 2%, respectively.
Volcanic eruptions contribute to climate variability, but quantifying these contributions has been limited by inconsistencies in the timing of atmospheric volcanic aerosol loading determined from ice ...cores and subsequent cooling from climate proxies such as tree rings. Here we resolve these inconsistencies and show that large eruptions in the tropics and high latitudes were primary drivers of interannual-to-decadal temperature variability in the Northern Hemisphere during the past 2,500 years. Our results are based on new records of atmospheric aerosol loading developed from high-resolution, multi-parameter measurements from an array of Greenland and Antarctic ice cores as well as distinctive age markers to constrain chronologies. Overall, cooling was proportional to the magnitude of volcanic forcing and persisted for up to ten years after some of the largest eruptive episodes. Our revised timescale more firmly implicates volcanic eruptions as catalysts in the major sixth-century pandemics, famines, and socioeconomic disruptions in Eurasia and Mesoamerica while allowing multi-millennium quantification of climate response to volcanic forcing.
Citrullination of proteins, a post-translational conversion of arginine residues to citrulline, is recognized in rheumatoid arthritis, but largely undocumented in cancer. Here we show that ...citrullination of the extracellular matrix by cancer cell derived peptidylarginine deiminase 4 (PAD4) is essential for the growth of liver metastases from colorectal cancer (CRC). Using proteomics, we demonstrate that liver metastases exhibit higher levels of citrullination and PAD4 than unaffected liver, primary CRC or adjacent colonic mucosa. Functional significance for citrullination in metastatic growth is evident in murine models where inhibition of citrullination substantially reduces liver metastatic burden. Additionally, citrullination of a key matrix component collagen type I promotes greater adhesion and decreased migration of CRC cells along with increased expression of characteristic epithelial markers, suggesting a role for citrullination in promoting mesenchymal-to-epithelial transition and liver metastasis. Overall, our study reveals the potential for PAD4-dependant citrullination to drive the progression of CRC liver metastasis.
The far-infrared dielectric function of a wide range of organic molecules is dominated by vibrations involving a substantial fraction of the atoms forming the molecule and motion associated with ...intermolecular hydrogen bond vibrations. Due to their collective nature such modes are highly sensitive to the intra- and intermolecular structure and thus provide a unique fingerprint of the conformational state of the molecule and effects of its environment. We demonstrate the use of terahertz time-domain spectroscopy (THz-TDS) for recording the far-infrared (0.5-4.0 THz) dielectric function of the four nucleobases and corresponding nucleosides forming the building blocks of deoxyribose nucleic acid (DNA). We observe numerous distinct spectral features with large differences between the molecules in both frequency-dependent absorption coefficient and index of refraction. Assisted by results from density-functional calculations we interpret the origin of the observed resonances as vibrations of hydrogen bonds between the molecules.
In this review we summarize our knowledge of using stable isotopes (
15N/
14N,
13C/
12C) to better understand the trophic ecology of oribatid mites. Our aims are (a) to recapitulate the history of ...stable isotope research in soil animals with a focus on oribatid mites, (b) to present new stable isotope data for oribatid mites and overview the current state of knowledge of oribatid mite trophic niche differentiation, (c) to compile problems and limitations of stable isotope based analyses of trophic relationships and (d) to suggest future challenges, questions and problems that may be solved using stable isotope analyses and other novel techniques for improving our understanding on the trophic ecology of soil invertebrates. We conclude that (1) in addition to
15N/
14N ratios,
13C/
12C ratios contribute to our understanding of the trophic ecology of oribatid mites, allowing, e.g. separation of lichen- and moss-feeding species, (2) there likely are many lichen but few moss feeding oribatid mite species, (3) oribatid mite species that are endophagous as juveniles are separated by their stable isotope signatures from all other oribatid mite species, (4) fungivorous oribatid mite species cannot be separated further, e.g. the fungal taxa they feed on cannot be delineated. A particular problem in using stable isotope data is the difficulty in determining signatures for basal food resources, since decomposing material, fungi and lichens comprise various components differing in stable isotope signatures;
13C/
12C ratios and potentially other isotopes may help in identifying the role of these resources for decomposer animal nutrition.

Cytotoxic T lymphocytes (CTLs) kill infected and cancerous cells. We detected transfer of cytotoxic multiprotein complexes, called supramolecular attack particles (SMAPs), from CTLs to target cells. ...SMAPs were rapidly released from CTLs and were autonomously cytotoxic. Mass spectrometry, immunochemical analysis, and CRISPR editing identified a carboxyl-terminal fragment of thrombospondin-1 as an unexpected SMAP component that contributed to target killing. Direct stochastic optical reconstruction microscopy resolved a cytotoxic core surrounded by a thrombospondin-1 shell of ~120 nanometer diameter. Cryo-soft x-ray tomography analysis revealed that SMAPs had a carbon-dense shell and were stored in multicore granules. We propose that SMAPs are autonomous extracellular killing entities that deliver cytotoxic cargo targeted by the specificity of shell components.

cell population directly associated with IL-6 levels in plasma, and a long-lasting CD11b

cell population. Trafficking of monocytes is mediated by TARC (CCL17) and associates with viral load measured in bronchial brushes. We also describe associations between disease outcomes and high levels of cell infiltration in lungs including CD11b
CD16
macrophages and CD11b
neutrophils. Accumulation of macrophages is long-lasting and detectable even in animals with mild or no signs of disease. Interestingly, animals with anti-inflammatory responses including high IL-10:IL-6 and kynurenine to tryptophan ratios show less severe illness. Our results unravel cellular mechanisms of COVID-19 and suggest that NHP may be appropriate models to test immune therapies.

The success of the Magnetospheric Multiscale mission depends on the accurate measurement of the magnetic field on all four spacecraft. To ensure this success, two independently designed and built ...fluxgate magnetometers were developed, avoiding single-point failures. The magnetometers were dubbed the digital fluxgate (DFG), which uses an ASIC implementation and was supplied by the Space Research Institute of the Austrian Academy of Sciences and the analogue magnetometer (AFG) with a more traditional circuit board design supplied by the University of California, Los Angeles. A stringent magnetic cleanliness program was executed under the supervision of the Johns Hopkins University’s Applied Physics Laboratory. To achieve mission objectives, the calibration determined on the ground will be refined in space to ensure all eight magnetometers are precisely inter-calibrated. Near real-time data plays a key role in the transmission of high-resolution observations stored on board so rapid processing of the low-resolution data is required. This article describes these instruments, the magnetic cleanliness program, and the instrument pre-launch calibrations, the planned in-flight calibration program, and the information flow that provides the data on the rapid time scale needed for mission success.

Polymers typically show only low absorption in the far-infrared or terahertz (THz) frequency range and are thus often used as window or lens materials for THz spectroscopy and imaging systems. A comprehensive study of the far-infrared properties of polymers yet reveals that THz spectroscopy is a very convenient technology for the investigation of various material properties including the morphology. Here, we report the dielectric parameters refraction index
n and absorption coefficient
κ of various polymers determined by THz time-domain spectroscopy with a very high precision. The observed features are attributed to macromolecular origin. Furthermore, we show that the temperature-dependent lattice mode shifts of highly-crystalline polymers reveal information about thermo-morphological interactions between the amorphous and the crystalline phase. A temperature-dependent refractometric data base of various standard polymers completes the paper. This data base enables the determination of the glass transition temperature

of these polymers and allows the extrapolation of the index of refraction across a broad range of temperatures above and below

g
for a given material.
Data availability and temporal resolution make it challenging to unravel the anatomy (duration and temporal phasing) of the Last Glacial abrupt climate changes. Here, we address these limitations by ...investigating the anatomy of abrupt changes using sub-decadal-scale records from Greenland ice cores. We highlight the absence of a systematic pattern in the anatomy of abrupt changes as recorded in different ice parameters. This diversity in the sequence of changes seen in ice-core data is also observed in climate parameters derived from numerical simulations which exhibit self-sustained abrupt variability arising from internal atmosphere-ice-ocean interactions. Our analysis of two ice cores shows that the diversity of abrupt warming transitions represents variability inherent to the climate system and not archive-specific noise. Our results hint that during these abrupt events, it may not be possible to infer statistically-robust leads and lags between the different components of the climate system because of their tight coupling.
