High fat or fructose induces non-alcoholic fatty liver disease (NAFLD) accompanied of mitochondrial dysfunction and oxidative stress. Controversy remains about whether fructose or fat is more ...deleterious for NAFLD development. To get more insights about this issue and to determine if the severity of liver disease induced by fructose or fat is related to degree of mitochondrial dysfunction, we compared the effects of diets containing high fat (HF), fructose (Fr) or high fat plus fructose (HF + Fr) on NAFLD development, mitochondrial function, ROS production and lipid peroxidation.
Wistar rats were assigned to four groups: Control, fed with standard rodent chow; High fat (HF), supplemented with lard and hydrogenated vegetable oil; Fructose (Fr), supplemented with 25% fructose in the drinking water; High fat plus fructose group (HF + Fr), fed with both HF and Fr diets. Rats were sacrificed after 6 weeks of diets consumption and the liver was excised for histopathological analysis by hematoxylin and eosin staining and for mitochondria isolation. Mitochondrial function was evaluated by measuring both mitochondrial respiration and complex I activity. Lipid peroxidation and ROS production were evaluated in mitochondria by the thiobarbituric acid method and with the fluorescent ROS probe 2,4-H
DCFDA, respectively.
Fr group underwent the lower degree of both liver damage and mitochondrial dysfunction that manifested like less than 20% of hepatocytes with microvesicular steatosis and partial decrease in state 3 respiration, respectively. HF group displayed an intermediate degree of damage as it showed 40% of hepatocytes with microvesicular steatosis and diminution of both state 3 respiration and complex I activity. HF + Fr group displayed more severe damage as showed microvesicular steatosis in 60% of hepatocytes and inflammation, while mitochondria exhibited fully inhibited state 3 respiration, impaired complex I activity and increased ROS generation. Exacerbation of mitochondrial lipid peroxidation was observed in both the Fr and HF + Fr groups.
Severity of liver injury induced by fructose or fat was related to the degree of dysfunction and oxidative damage in mitochondria. Attention should be paid on the serious effects observed in the HF + Fr group as the typical Western diet is rich in both fat and carbohydrates.
The performance of vehicle safety systems depends very much on the accuracy of the signals coming from vehicle sensors. Among them, the wheel speed is of vital importance. This paper describes a new ...method to obtain the wheel speed by using Sin-Cos encoders. The methodology is based on the use of the Savitzky-Golay filters to optimally determine the coefficients of the polynomials that best fit the measured signals and their time derivatives. The whole process requires a low computational cost, which makes it suitable for real-time applications. This way it is possible to provide the safety system with an accurate measurement of both the angular speed and acceleration of the wheels. The proposed method has been compared to other conventional approaches. The results obtained in simulations and real tests show the superior performance of the proposed method, particularly for medium and low wheel angular speeds.
Non-alcoholic fatty liver disease (NAFLD) is characterized by lipid accumulation in hepatocytes, and in advanced stages, by inflammation and fibrosis. Excessive ROS production due to mitochondrial ...dysfunction contributes to NAFLD development, making the decrease in mitochondrial ROS production an emerging target to alleviate NAFLD. Previously, we have shown that avocado oil, a source of several bioactive compounds with antioxidant effects, decreases oxidative stress by improving the function of the mitochondrial electron transport chain (ETC) and decreasing ROS levels in mitochondria of diabetic and hypertensive rats. Therefore, we tested in this work whether avocado oil alleviates NAFLD by attenuating mitochondrial dysfunction, oxidative stress and inflammation. NAFLD was induced in rats by a high fat-high fructose (HF) diet administered for six (HF6) or twelve (HF12) weeks. Hepatic steatosis, hypertrophy and inflammation were detected in both the HF6 and HF12 groups. Hyperglycemia was observed only in the HF12 group. The HF6 and HF12 groups displayed dyslipidemia, impairments in mitochondrial respiration, complex III activity, and electron transfer in cytochromes in the complex III. This led to an increase in the levels of ROS and lipid peroxidation. The substitution of the HF6 diet by standard chow and avocado oil for 6 weeks (HF6+AVO + D), or supplementation of the HF12 diet with avocado oil (HF12 + AVO), ameliorated NAFLD, hyperglycemia, dyslipidemia, and counteracted mitochondrial dysfunctions and oxidative stress. The substitution of the HF6 diet by standard chow without avocado oil did not correct many of these abnormalities, confirming that the removal of the HF diet is not enough to counteract NAFLD and mitochondrial dysfunction. In summary, avocado oil decreases NAFLD by improving mitochondrial function, oxidative stress, and inflammation.
Background
Visceral adiposity index (VAI) has been identified as a cardiometabolic risk marker in children and adolescents which reflects abdominal fat distribution. The aim of the present study was ...to evaluated the predictive capacity of VAI, a body shape index (ABSI), atherogenic index of plasma (AIP), and triglycerides and glucose index (TyG index) compared with classical anthropometric measurements to discriminate metabolic syndrome (MetS).
Methods
This retrospective study included 1372 individuals. Anthropometric, clinical, and biochemical measurements were used to screen the prevalence of MetS components and to calculate VAI, ABSI, TyG index, and AIP.
Results
The discriminatory capacity among the variables was assessed by the area under the receiver operating characteristic (ROC) curve (AUC). VAI was the variable with the highest AUC with 0.932 CI 95% (0.917–0.948), followed by AIP with 0.914 CI 95% (0.897–0.931), and TyG index with 0.889 CI 95% (0.871–0.908).
Conclusion
VAI is a promising tool to identify MetS in the late adolescence setting. Among the novel adiposity indexes VAI, AIP, TyG index are able to determine MetS presence, while ABSI is not capable.
PARP inhibition can induce anti-neoplastic effects when used as monotherapy or in combination with chemo- or radiotherapy in various tumor settings; however, the basis for the anti-metastasic ...activities resulting from PARP inhibition remains unknown. PARP inhibitors may also act as modulators of tumor angiogenesis. Proteomic analysis of endothelial cells revealed that vimentin, an intermediary filament involved in angiogenesis and a specific hallmark of EndoMT (endothelial to mesenchymal transition) transformation, was down-regulated following loss of PARP-1 function in endothelial cells. VE-cadherin, an endothelial marker of vascular normalization, was up-regulated in HUVEC treated with PARP inhibitors or following PARP-1 silencing; vimentin over-expression was sufficient to drive to an EndoMT phenotype. In melanoma cells, PARP inhibition reduced pro-metastatic markers, including vasculogenic mimicry. We also demonstrated that vimentin expression was sufficient to induce increased mesenchymal/pro-metastasic phenotypic changes in melanoma cells, including ILK/GSK3-β-dependent E-cadherin down-regulation, Snail1 activation and increased cell motility and migration. In a murine model of metastatic melanoma, PARP inhibition counteracted the ability of melanoma cells to metastasize to the lung. These results suggest that inhibition of PARP interferes with key metastasis-promoting processes, leading to suppression of invasion and colonization of distal organs by aggressive metastatic cells.

The influence of different support materials (alumina, ceria, β-silicon carbide and yttria-stabilized zirconia) on the catalytic behaviour of Ni catalysts for the dry reforming reaction and the tri-reforming process has been studied. The influence on the catalytic performance of the composition of the atmosphere surrounding the Ni/YSZ catalyst during the calcination step was also analysed. Temperature-programmed reduction experiments showed remarkable differences in the reduction profile and the degree of reduction of the catalysts as a function of both the support used and the calcination conditions. Ni/YSZ-O2, the catalyst calcined under an oxygen-poor atmosphere, presented a higher reducibility as a consequence of the higher number of oxygen vacancies in the surface of the support. The Ni/Al2O3 catalyst gave the lowest CH4 and CO2 reaction rates as a consequence of its low reducibility due to the formation of Ni aluminate. The Ni/CeO2 catalyst showed the lowest H2/CO molar ratio for the tri-reforming process. This result can be explained on considering the higher basicity of this catalyst, as shown by CO2-TPD experiments. The CeO2 and β-SiC catalysts had the best characteristics as catalytic supports for the tri-reforming process.
Time series of European sardine (Sardina pilchardus) landings from 1962 and environmental variables from 1978 in the northern Alboran Sea are analysed. European sardine spawns in the northern Alboran ...Sea from mid‐autumn to late winter at a temperature range slightly higher than the one observed in the nearby Eastern North Atlantic and the North Western Mediterranean. Individuals hatched during autumn and winter are incorporated to the fishery during the following summer and autumn producing the maximum annual landings. These landings show both a decreasing long‐term trend and a strong inter‐annual variability. Although further research is needed, the warming trend of sea surface temperature and the decrease in upwelling intensity inferred from empirical orthogonal function (EOF) analyses could have some influence on the negative trends of sardine landings. The inter‐annual variability of sardine abundance seems to be related to the wind intensity at a local scale, the second principal component of the chlorophyll concentration and the sardine abundance during the preceding year. If the inter‐annual variability is considered, a linear model including these three variables with a one‐year time lag allows to explain 79% of the sardine landings variance. If the negative linear trend is also considered, the model explains 86% of the variance. These results indicate that the body condition of spawners, linked to the food availability during the preceding year, is the main factor controlling the recruitment success. The possibility of predicting sardine landings 1 year in advance could have important implications for fishery management.
The development of new control algorithms in vehicles requires high economic resources, mainly due to the use of generic real-time instrumentation and control systems. In this work, we proposed a ...low-cost electronic control unit (ECU) that could be used for both development and implementation. The proposed electronic system used a hybrid system on chip (SoC) between a field-programmable gate array (FPGA) and an Advanced RISC (reduced instruction set computer) Machine (ARM) processor that allowed the execution of parallel tasks, fulfilling the real-time requirements that vehicle controls demand. Another feature of the proposed electronic system was the recording of measured data, allowing the performance of the implemented algorithm to be evaluated. All this was achieved by using modular programming that, without the need for a real-time operating system, executed the different tasks to be performed, exploiting the parallelism offered by the FPGA as well as the dual core of the ARM processor. This methodology facilitates the transition between the designing, testing, and implementation stages in the vehicle. In addition, our system is programmed with a single binary file that integrates the code of all processors as well as the hardware description of the FPGA, which speeds up the updating process. In order to validate and demonstrate the performance of the proposed electronic system as a tool for the development and implementation of control algorithms in vehicles, a series of tests was carried out on a test bench. Different traction control system (TCS) algorithms were implemented and the results were compared.
Biopolymers such as chitosan (CHT) or alginate (ALG) are among the most prominent for health-related applications due to their broad bioactivity. Their combination for the preparation of membranes is ...hereby proposed as an application for wound healing with the incorporation of lavender essential oil (LEO), widely known for its antioxidant and antimicrobial properties. The preparation of CHT, CHT + LEO, ALG, ALG + LEO, and CHT/ALG + LEO membranes was accomplished, and its composition was analyzed using Fourier Transform Infrared Spectroscopy (FTIR). The water absorption capacity and oil release profile of the membranes revealed higher water uptake capacity when a lower LEO release was obtained. The combined CHT/ALG + LEO film showed a water uptake percentage of 638% after 48 h and a maximum LEO release concentration of 42 mg/L. Cytotoxicity and biocompatibility of the prepared membranes were studied using a HaCaT cell line, with an assessment of cell viability regarding film leachables, DNA quantification, and DAPI-phalloidin staining. The results revealed that the indirect contact of the prepared membranes via its leachables does not compromise cell viability, and upon direct contact, cells do not adhere or proliferate on the surface of the membranes. Moreover, the CHT/ALG + LEO membrane increases cell proliferation, making it suitable for applications in wound healing.
