ABSTRACT We present X-ray source catalogs for the 7 Ms exposure of the Chandra Deep Field-South (CDF-S), which covers a total area of 484.2 arcmin2. Utilizing wavdetect for initial source detection ...and ACIS Extract for photometric extraction and significance assessment, we create a main source catalog containing 1008 sources that are detected in up to three X-ray bands: 0.5-7.0 keV, 0.5-2.0 keV, and 2-7 keV. A supplementary source catalog is also provided, including 47 lower-significance sources that have bright ( ) near-infrared counterparts. We identify multiwavelength counterparts for 992 (98.4%) of the main-catalog sources, and we collect redshifts for 986 of these sources, including 653 spectroscopic redshifts and 333 photometric redshifts. Based on the X-ray and multiwavelength properties, we identify 711 active galactic nuclei (AGNs) from the main-catalog sources. Compared to the previous 4 Ms CDF-S catalogs, 291 of the main-catalog sources are new detections. We have achieved unprecedented X-ray sensitivity with average flux limits over the central 1 arcmin2 region of 1.9 × 10−17, 6.4 × 10−18, and 2.7 × 10−17 erg cm−2 s−1 in the three X-ray bands, respectively. We provide cumulative number-count measurements observing, for the first time, that normal galaxies start to dominate the X-ray source population at the faintest 0.5-2.0 keV flux levels. The highest X-ray source density reaches 50,500 deg−2, and 47% 4% of these sources are AGNs ( 23,900 deg−2).
Cancer cells are known to execute reprogramed metabolism of glucose, amino acids and lipids. Here, we report a significant role of cholesterol metabolism in cancer metastasis. By using label-free ...Raman spectromicroscopy, we found an aberrant accumulation of cholesteryl ester in human pancreatic cancer specimens and cell lines, mediated by acyl-CoA cholesterol acyltransferase-1 (ACAT-1) enzyme. Expression of ACAT-1 showed a correlation with poor patient survival. Abrogation of cholesterol esterification, either by an ACAT-1 inhibitor or by shRNA knockdown, significantly suppressed tumor growth and metastasis in an orthotopic mouse model of pancreatic cancer. Mechanically, ACAT-1 inhibition increased intracellular free cholesterol level, which was associated with elevated endoplasmic reticulum stress and caused apoptosis. Collectively, our results demonstrate a new strategy for treating metastatic pancreatic cancer by inhibiting cholesterol esterification.

Although the skeleton is one of the predominant sites for breast cancer metastasis, why breast cancer cells often become dormant after homing to bone is not well understood. Here, we reported an ...intrinsic self-defense mechanism of bone cells against breast cancer cells: a critical role of connexin (Cx) 43 hemichannels in osteocytes in the suppression of breast cancer bone metastasis. Cx43 hemichannels allow passage of small molecules between the intracellular and extracellular environments. The treatment of bisphosphonate drugs, either alendronate (ALN) or zoledronic acid (ZOL), opened Cx43 hemichannels in osteocytes. Conditioned media (CM) collected from MLO-Y4 osteocyte cells treated with bisphosphonates inhibited the anchorage-independent growth, migration and invasion of MDA-MB-231 human breast cancer cells and Py8119 mouse mammary carcinoma cells, and this inhibitory effect was attenuated with Cx43(E2), a specific hemichannel-blocking antibody. The opening of osteocytic Cx43 hemichannels by mechanical stimulation had similar inhibitory effects on breast cancer cells and this inhibition was attenuated by Cx43(E2) antibody as well. These inhibitory effects on cancer cells were mediated by ATP released from osteocyte Cx43 hemichannels. Furthermore, both Cx43 osteocyte-specific knockout mice and osteocyte-specific Δ130-136 transgenic mice with impaired Cx43 gap junctions and hemichannels showed significantly increased tumor growth and attenuated the inhibitory effect of ZOL. However, R76W transgenic mice with functional hemichannels but not gap junctions in osteocytes did not display a significant difference. Together, our studies establish the specific inhibitory role of osteocytic Cx43 hemichannels, and exploiting the activity of this channel could serve as a de novo therapeutic strategy.
Large scale epitaxial growth and transfer of monolayer MoS2 has attracted great attention in recent years. Here, we report the wafer-scale epitaxial growth of highly oriented continuous and uniform ...monolayer MoS2 films on single-crystalline sapphire wafers by chemical vapor deposition (CVD) method. The epitaxial film is of high quality and stitched by many 0°, 60° domains and 60°-domain boundaries. Moreover, such wafer-scale monolayer MoS2 films can be transferred and stacked by a simple stamp-transfer process, and the substrate is reusable for subsequent growth. Our progress would facilitate the scalable fabrication of various electronic, valleytronic, and optoelectronic devices for practical applications.
Abstract
The interaction between the solar wind and the magnetosphere is one of the most important research subjects in the fields of astrophysics and space physics. For more than half a century, ...based on the pressure balance assumption between the solar wind and the magnetosphere and considering other important factors, such as the interplanetary magnetic field and magnetic reconnection process, the dynamic processes at the magnetopause have been extensively analyzed. However, the responses of magnetopause to the solar wind dynamic pressure variations are still complicated to understand. Here, we show that the interaction between the solar wind and the magnetosphere can be regarded as a quasi-elastodynamic process. The driving frequency of the solar wind is determined as a crucial reason for the phase difference between solar wind dynamic pressure variations and magnetopause standoff distance. The low-pass filter effect and oscillation properties of the magnetopause can also be well explained by the forced damped vibrations. Moreover, the quasi-elastodynamic processes predict deformations at the magnetopause, which resemble the magnetopause surface wave. Finally, a three-dimensional time-dependent magnetopause model is constructed and verified by observation. Based on 12,242 magnetopause crossing events, it is found that the new model reveals ∼9.7% better prediction accuracy than the widely used time-independent model. These results can also shed light on our understanding of the solar-wind–magnetopause interaction for other planets.
Purpose
Non-alcoholic fatty liver disease (NAFLD) is considered as both a vital risk factor and a consequence of type 2 diabetes mellitus (T2DM). Low total testosterone (TT) is common in men with ...T2DM, contributing to increased risks of metabolic diseases. This study aimed to investigate the association between TT levels and the prevalence of NAFLD in men with T2DM.
Methods
In this cross-sectional study, 1005 men with T2DM were enrolled in National Metabolic Management Center (MMC) of First Affiliated Hospital of Wenzhou Medical University between January 2017 and August 2021. NAFLD was diagnosed using ultrasound as described by the Chinese Liver Disease Association. Overweight/obesity was defined as body mass index (BMI) ≥ 25 kg/m
2
according to WHO BMI classifications.
Results
Individuals without NAFLD had higher serum TT levels than those with NAFLD. After adjustments for potential confounding factors, the top tertile was significantly associated with lower prevalence of NAFLD compared with the bottom tertile of TT level odds ratio (OR) 0.303, 95% confidence interval (CI) 0.281–0.713;
P
< 0.001. The association between TT with NAFLD in individuals with normal weight (OR 0.175, 95% CI 0.098–0.315;
P
< 0.001) was stronger than in individuals with overweight/obesity (OR 0.509, 95% CI 0.267–0.971;
P
= 0.040). There was a significant interaction of TT with overweight/obesity (
P
for interaction = 0.018 for NAFLD).
Conclusion
Higher serum TT was significantly associated with a lower prevalence of NAFLD in men with T2DM. We found that the relationship of TT and NAFLD was stronger in individuals with non-overweight/obesity.
The debris flow is one of the geological hazards; its occurrence is complex, fuzzy, and random. And it is affected by many indices; a new multi-index assessment method is proposed to analyze the risk ...level of debris flow based on the entropy weight-normal cloud model in Banshanmen gully. The index weight is calculated by using the entropy weight method. Then, the certainty degree of each index belonging to the corresponding cloud is obtained by using the cloud model. The final risk level of debris flow is determined according to the synthetic certainty degree. The conclusions are drawn that the method is feasible and accurate rate of risk estimation for debris flow is very high, so a new method and thoughts for the risk assessment of debris flow can be provided in the future.
The primary resonance and nonlinear vibrations of the functionally graded graphene platelet (FGGP)-reinforced rotating pretwisted composite blade under combined the external and multiple parametric ...excitations are investigated with three different distribution patterns. The FGGP-reinforced rotating pretwisted composite blade is simplified to the rotating pretwisted composite cantilever plate reinforced by the functionally graded graphene platelet. It is novel to simplify the leakage of the airflow in the tip clearance to the non-uniform axial excitation. The rotating speed of the steady state adding a small periodic perturbation is considered. The aerodynamic load subjecting to the surface of the plate is simulated as the transverse excitation. Utilizing the first-order shear deformation theory, von Karman nonlinear geometric relationship, Lagrange equation and mode functions satisfying the boundary conditions, three-degree-of-freedom nonlinear ordinary differential equations of motion are derived for the FGGP-reinforced rotating pretwisted composite cantilever plate under combined the external and multiple parametric excitations. The primary resonance and nonlinear dynamic behaviors of the FGGP-reinforced rotating pretwisted composite cantilever plate are analyzed by Runge–Kutta method. The amplitude–frequency response curves, force–frequency response curves, bifurcation diagrams, maximum Lyapunov exponent, phase portraits, waveforms and Poincare map are obtained to investigate the nonlinear dynamic responses of the FGGP-reinforced rotating pretwisted composite cantilever plate under combined the external and multiple parametric excitations.
Aims
The aims of this study were to investigate the effects of probiotics and antibiotics on microbial composition, short chain fatty acids (SCFAs) concentration and free fatty acid receptor 2/3 ...(FFAR2/3) expression in boiler chickens.
Methods and Results
A total of 150 1‐day‐old male broilers were randomly allocated into three groups, control (CON) group, probiotics (PB) group and antibiotics (ATB) group. Results indicated that PB improved the average body weight from 1 to 21 days and feed intake from 21 to 42 days (P < 0·05), while ATB improved the feed efficiency from 1 to 42 days (P < 0·05). Based on 16s rRNA sequencing, PB treatment increased the amount of kingdom bacteria, and the relative abundance of the main bacteria including acetate and butyrate producing bacteria of phylum Firmicutes, family Ruminococcaceae and genus Faecalibacterium. ATB treatment also increased the relative abundance of phylum Firmicutes, family Ruminococcaceae and Lachnospiraceae, however, it introduced some pathogenic bacteria, such as bacteria of family Rikenellaceae and Enterobacteriaceae. Gas chromatography and mass spectrometry (GC‐MS) assay revealed that PB increased acetate and butyrate concentrations at both 21 and 42 days, and propionate at 42 days in the colorectum. Moreover qRT‐PCR analysis showed PB treatment significantly activated the FFAR2/3 mRNA expressions. On the contrast, ATB treatment lowered the colorectal propionate at 21 days, and decreased acetate, propionate and butyrate concentrations at 42 days, accompanied with decreased FFAR2/3 mRNA expressions.
Conclusions
Compared to the CON birds, an enriched SCFAs producing bacteria with higher SCFAs contents and activated FFAR2/3 expressions are prominent features of PB birds. However, antibiotics treatment plays the reverse effect compared to PB treatment.
Significance and Impact of the Study
This study brings a significant idea that less SCFAs concentration may be another reason why the antibiotics inhibit the immune system development and immunity of the body.
