The introduction of oxygen vacancies (Ov) has been regarded as an effective method to enhance the catalytic performance of photoanodes in oxygen evolution reaction (OER). However, their stability ...under highly oxidizing environment is questionable but was rarely studied. Herein, NiFe‐metal–organic framework (NiFe‐MOFs) was conformally coated on oxygen‐vacancy‐rich BiVO4 (Ov‐BiVO4) as the protective layer and cocatalyst, forming a core–shell structure with caffeic acid as bridging agent. The as‐synthesized Ov‐BiVO4@NiFe‐MOFs exhibits enhanced stability and a remarkable photocurrent density of 5.3±0.15 mA cm−2 at 1.23 V (vs. RHE). The reduced coordination number of Ni(Fe)‐O and elevated valence state of Ni(Fe) in NiFe‐MOFs layer greatly bolster OER, and the shifting of oxygen evolution sites from Ov‐BiVO4 to NiFe‐MOFs promotes Ov stabilization. Ovs can be effectively preserved by the coating of a thin NiFe‐MOFs layer, leading to a photoanode of enhanced photocurrent and stability.
A core–shell Ov‐BiVO4@NiFe‐MOFs photoanode was constructed via a coordination‐assisted self‐assembly method. A NiFe‐MOFs thin layer acts as protective layer and cocatalyst to shift active sites from oxygen vacancies to NiFe‐MOFs, leading to improved stability and activity for OER. This molecular‐based approach tailors the coordination and electronic structure of active sites and provides mechanistic insights for rational design of photocatalysts.
The low electrocatalytic activity of pristine graphite felt (GF) electrodes toward V(II)/V(III) and V(IV)/V(V) redox couples is a major concern in vanadium redox flow batteries (VRFBs). For ...overcoming this challenge, herein a novel composite electrode is proposed comprising of two components: multidimensional frame carbon (MFC) derived from edge‐rich carbon and GF that serves as the frame for the in situ growth of MFC. The high electrocatalytic activity, rapid charge migration, and reduced local current emanating from the 0D, 2D, and 3D coexistent structures of the MFC material, respectively, enhance the performance of the GF. Consequently, the battery assembled using the MFC GF electrode achieves a maximum current density of 500 mA cm−2, along with high stability and preeminent energy efficiency at a current density of 200 mA cm−2 for over 400 cycles. For the first time via density functional theory analysis on VRFBs, this study reveals that the edge‐rich carbon atoms possess higher electrocatalytic activity in both positive and negative electrolytes than the plane carbon atoms and heteroatoms. Therefore, this study is of immense significance in guiding and promoting the application of edge‐rich carbon in the battery‐based energy storage industry.

Using recycled concrete and crumb rubber as aggregates to produce green concrete is a promising technology toward sustainability in the construction industry. In this study, the compressive and flexural behaviours of a new type of concrete material, rubber crumb and steel-fibre-reinforced recycled aggregate concrete (RSRAC), are investigated. To popularise the application of this new type of green building material, an experimental study was conducted to investigate the effect of the rubber content on the compressive and flexural behaviours of RSRAC. A total of 18 cubes (150mm) and 18 cylinders (150mm×200mm) were tested under axial compressive loading, and 18 prisms of 150×150×550mm were tested subjected to three-point bending. The crumb rubber content was varied in the investigation at levels of 0%, 4%, 8%, 12% and 16% by volume substitution of sand. Recycled concrete aggregate (RCA) was introduced into the concrete mixture by 100% volume substitution of natural coarse aggregate (NCA), and 1% volumetric quantity of steel fibre was added to the concrete mixture. The effect of the rubber content on the compressive and flexural strength, failure mode, modulus of elasticity and toughness of RSRAC was analysed. The results indicate that RSRAC with an optimal rubber content displays good compressive behaviour compared with normal NCA concrete. RSRAC is also a more environmentally friendly alternative to normal rubber concrete for use in theflexural members of concrete structures.

Many plant viruses with monopartite or bipartite genomes have been developed as efficient expression vectors of foreign recombinant proteins. Nonetheless, due to lack of multiple insertion ...sites in these plant viruses, it is still a big challenge to simultaneously express multiple foreign proteins in single cells. The genome of Beet necrotic yellow vein virus (BNYVV) offers an attractive system for expression of multiple foreign proteins owning to a multipartite genome composed of five positive‐stranded RNAs. Here, we have established a BNYVV full‐length infectious cDNA clone under the control of the Cauliflower mosaic virus 35S promoter. We further developed a set of BNYVV‐based vectors that permit efficient expression of four recombinant proteins, including some large proteins with lengths up to 880 amino acids in the model plant Nicotiana benthamiana and native host sugar beet plants. These vectors can be used to investigate the subcellular co‐localization of multiple proteins in leaf, root and stem tissues of systemically infected plants. Moreover, the BNYVV‐based vectors were used to deliver NbPDS guide RNAs for genome editing in transgenic plants expressing Cas9, which induced a photobleached phenotype in systemically infected leaves. Collectively, the BNYVV‐based vectors will facilitate genomic research and expression of multiple proteins, in sugar beet and related crop plants.
Direct myocardial and vascular injuries due to severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection-driven inflammation is the leading cause of acute cardiac injury associated with ...coronavirus disease 2019 (COVID-19). However, in-depth knowledge of the injury characteristics of the heart affected by inflammation is lacking. In this study, using a quantitative spatial proteomics strategy that combines comparative anatomy, laser-capture microdissection, and histological examination, we establish a region-resolved proteome map of the myocardia and microvessels with obvious inflammatory cells from hearts of patients with COVID-19. A series of molecular dysfunctions of myocardia and microvessels is observed in different cardiac regions. The myocardia and microvessels of the left atrial are the most susceptible to virus infection and inflammatory storm, suggesting more attention should be paid to the lesion and treatment of these two parts. These results can guide in improving clinical treatments for cardiovascular diseases associated with COVID-19.

Hornworts, liverworts and mosses are three early diverging clades of land plants, and together comprise the bryophytes. Here, we report the draft genome sequence of the hornwort Anthoceros angustus. ...Phylogenomic inferences confirm the monophyly of bryophytes, with hornworts sister to liverworts and mosses. The simple morphology of hornworts correlates with low genetic redundancy in plant body plan, while the basic transcriptional regulation toolkit for plant development has already been established in this early land plant lineage. Although the Anthoceros genome is small and characterized by minimal redundancy, expansions are observed in gene families related to RNA editing, UV protection and desiccation tolerance. The genome of A. angustus bears the signatures of horizontally transferred genes from bacteria and fungi, in particular of genes operating in stress-response and metabolic pathways. Our study provides insight into the unique features of hornworts and their molecular adaptations to live on land.
Tripartite motif-containing 44 (TRIM44) was reported to be involved in the tumorigenesis of several tumors, but its function in laryngeal squamous cell carcinoma has not been investigated yet. In the ...present study, we aimed to elucidate the function of TRIM44 in laryngeal squamous cell carcinoma, and identify the compounds which could inhibit TRIM44 expression. Our results showed that TRIM44 was upregulated in tumor tissues and cell lines of laryngeal squamous cell carcinoma. Knockdown of TRIM44 significantly inhibited cell growth of laryngeal squamous cell carcinoma by suppressing TLR4, phosphorylated AKT and phosphorylated NF-κB p65 expression in vitro. Moreover, TRIM44 knockdown inhibited tumor growth in nude mice, which further suggested that TRIM44 exerted oncogenic activity in laryngeal squamous cell carcinoma. Interestingly, it was found that nuciferine significantly inhibited the mRNA levels of TRIM44 after screening a small natural compound library. Our further studies showed nuciferine markedly downregulated the protein levels of TRIM44 and its substrate TLR4 in a concentration-dependent manner in laryngeal squamous cell carcinoma cells. Moreover, the activation of downstream kinases of TLR4 such as AKT signaling pathway was also inhibited by nuciferine. Additionally, nuciferine markedly inhibited cell survival of laryngeal squamous cell carcinoma in a concentration-dependent manner. In contrast, TRIM44 overexpression significantly reduced the cytotoxicity of nuciferine in laryngeal squamous cell carcinoma cells. In conclusion, this study indicated that inhibiting TRIM44 would be a useful strategy for the treatment of laryngeal squamous cell carcinoma, and nuciferine could be a potential chemical applicated in the therapy of laryngeal squamous cell carcinoma.
Hepatitis B virus (HBV) can integrate into the human genome, contributing to genomic instability and hepatocarcinogenesis. Here by conducting high-throughput viral integration detection and RNA ...sequencing, we identify 4,225 HBV integration events in tumour and adjacent non-tumour samples from 426 patients with HCC. We show that HBV is prone to integrate into rare fragile sites and functional genomic regions including CpG islands. We observe a distinct pattern in the preferential sites of HBV integration between tumour and non-tumour tissues. HBV insertional sites are significantly enriched in the proximity of telomeres in tumours. Recurrent HBV target genes are identified with few that overlap. The overall HBV integration frequency is much higher in tumour genomes of males than in females, with a significant enrichment of integration into chromosome 17. Furthermore, a cirrhosis-dependent HBV integration pattern is observed, affecting distinct targeted genes. Our data suggest that HBV integration has a high potential to drive oncogenic transformation.
Background
To reveal detailed histopathological changes, virus distributions, immunologic properties and multi‐omic features caused by SARS‐CoV‐2 in the explanted lungs from the world's first ...successful lung transplantation of a COVID‐19 patient.
Materials and methods
A total of 36 samples were collected from the lungs. Histopathological features and virus distribution were observed by optical microscope and transmission electron microscope (TEM). Immune cells were detected by flow cytometry and immunohistochemistry. Transcriptome and proteome approaches were used to investigate main biological processes involved in COVID‐19‐associated pulmonary fibrosis.
Results
The histopathological changes of the lung tissues were characterized by extensive pulmonary interstitial fibrosis and haemorrhage. Viral particles were observed in the cytoplasm of macrophages. CD3+CD4− T cells, neutrophils, NK cells, γ/δ T cells and monocytes, but not B cells, were abundant in the lungs. Higher levels of proinflammatory cytokines iNOS, IL‐1β and IL‐6 were in the area of mild fibrosis. Multi‐omics analyses revealed a total of 126 out of 20,356 significant different transcription and 114 out of 8,493 protein expression in lung samples with mild and severe fibrosis, most of which were related to fibrosis and inflammation.
Conclusions
Our results provide novel insight that the significant neutrophil/ CD3+CD4− T cell/ macrophage activation leads to cytokine storm and severe fibrosis in the lungs of COVID‐19 patient and may contribute to a better understanding of COVID‐19 pathogenesis.
N-linked glycans on immunoglobulin G (IgG) have been associated with pathogenesis of diseases and the therapeutic functions of antibody-based drugs; however, low-abundance species are difficult to ...detect. Here we show a glycomic approach to detect these species on human IgGs using a specialized microfluidic chip. We discover 20 sulfated and 4 acetylated N-glycans on IgGs. Using multiple reaction monitoring method, we precisely quantify these previously undetected low-abundance, trace and even ultra-trace N-glycans. From 277 patients with rheumatoid arthritis (RA) and 141 healthy individuals, we also identify N-glycan biomarkers for the classification of both rheumatoid factor (RF)-positive and negative RA patients, as well as anti-citrullinated protein antibodies (ACPA)-positive and negative RA patients. This approach may identify N-glycosylation-associated biomarkers for other autoimmune and infectious diseases and lead to the exploration of promising glycoforms for antibody therapeutics.Post-translational modifications can affect antibody function in health and disease, but identification of all variants is difficult using existing technologies. Here the authors develop a microfluidic method to identify and quantify low-abundance IgG N-glycans and show some of these IgGs can be used as biomarkers for rheumatoid arthritis.
