Intermittent fasting (IF) may limit metabolic adaptations that reduce energy expenditure, potentially by stimulating white adipose tissue (WAT) browning. The aim of this study was to examine the effects of 8 wk of IF on energy metabolism and markers of WAT browning in lean and diet-induced obese mice and in women who were overweight or obese.
Male C57 BL/6 J mice were fed chow or a high-fat diet (HFD; 43%) for 8 wk before undergoing IF (3 non-consecutive d/wk) for an additional 8 wk. Food intake, energy expenditure, and inguinal and gonadal fat pads were collected in fed or fasted conditions (22 h, IF mice only). Subcutaneous adipose tissue (SAT) was also collected at baseline, and after 8 wk of IF (in the fed state, and after a 24-h fast), in women with overweight or obesity. Uncoupling protein 1 (UCP1) was assessed by quantitative real-time polymerase chain reaction (mice and humans) and immunohistochemistry (mice).
IF reduced body weight and energy intake in HFD fed mice and reduced gonadal and inguinal fat pad weights in both diet groups. IF increased energy expenditure, meal number, Ucp1 mRNA levels in inguinal and gonadal fat depots, and UCP1 protein in inguinal fat in both diet groups on fed days. In women, IF reduced body weight and fat mass, but did not alter UCP1mRNA levels.

There is a growing body of literature using functional MRI to study the acute and long-term effects of concussion on functional brain networks. To date, studies have largely focused on changes in ...pairwise connectivity strength between brain regions. Less is known about how concussion affects whole-brain network topology, particularly the "small-world" organization which facilitates efficient communication at both local and global scales. The present study addressed this knowledge gap by measuring local and global efficiency of 26 concussed athletes at acute injury, return to play (RTP) and one year post-RTP, along with a cohort of 167 athletic controls. On average, concussed athletes showed no alterations in local efficiency but had elevated global efficiency at acute injury, which had resolved by RTP. Athletes with atypically long recovery, however, had reduced global efficiency at 1 year post-RTP, suggesting long-term functional abnormalities for this subgroup. Analyses of nodal efficiency further indicated that global network changes were driven by high-efficiency visual and sensorimotor regions and low-efficiency frontal and subcortical regions. This study provides evidence that concussion causes subtle acute and long-term changes in the small-world organization of the brain, with effects that are related to the clinical profile of recovery.
A variety of organisms have evolved mechanisms to detect and respond to light, in which the response is mediated by protein structural changes after photon absorption. The initial step is often the ...photoisomerization of a conjugated chromophore. Isomerization occurs on ultrafast time scales and is substantially influenced by the chromophore environment. Here we identify structural changes associated with the earliest steps in the trans-to-cis isomerization of the chromophore in photoactive yellow protein. Femtosecond hard x-ray pulses emitted by the Linac Coherent Light Source were used to conduct time-resolved serial femtosecond crystallography on photoactive yellow protein microcrystals over a time range from 100 femtoseconds to 3 picoseconds to determine the structural dynamics of the photoisomerization reaction.
Genomic evaluations of dairy cattle in the United States have been available for Brown Swiss, Holsteins, and Jerseys since 2009. As of January 2013, 1,023 Ayrshires had genotypes in the North ...American database. Evaluation accuracy was assessed using genomic evaluations based on 646 bulls with 2008 traditional evaluations to predict daughter performance of up to 180 bulls in 2012. Mean gain in reliability over parent average for all traits was 8.2 percentage points. The highest gains were for protein yield (16.9 percentage points), milk yield (16.6 percentage points), and stature (16.2 percentage points). Twelve single nucleotide polymorphisms were useful for Ayrshire breed determination. Fewer breed-determining SNP were available for Ayrshires than for Holsteins, Jerseys, and Brown Swiss because of the similarity of Ayrshires and Holsteins. A haplotype that affects fertility was identified on chromosome 17 and traces back in the genotyped population to the bull Selwood Betty’s Commander (born in 1953). The haplotype carrier frequency for genotyped Ayrshires was 26.1%. Sire conception rate was decreased by 4.3±2.5 percentage points for carriers of the haplotype as determined by 618 matings of carrier sire by carrier maternal grandsire. Genomic evaluations for Ayrshires were officially implemented in the United States in April 2013.
Abstract
Mid-infrared (mid-IR) observations are powerful in identifying heavily obscured active galactic nuclei (AGN) that have weak emission in other wavelengths. Data from the Mid-Infrared ...Instrument (MIRI) on board the James Webb Space Telescope provides an excellent opportunity to perform such studies. We take advantage of the MIRI imaging data from the Cosmic Evolution Early Release Science Survey to investigate the AGN population in the distant universe. We estimate the source properties of MIRI-selected objects by utilizing spectral energy distribution (SED) modeling, and classify them into star-forming galaxies (SFs), SF-AGN mixed objects, and AGN. The source numbers of these types are 433, 102, and 25, respectively, from four MIRI pointings covering ∼9 arcmin
2
. The sample spans a redshift range of ≈0–5. We derive the median SEDs for all three source types, respectively, and publicly release them. The median MIRI SED of AGN is similar to the typical SEDs of hot dust-obscured galaxies and Seyfert 2s, for which the mid-IR SEDs are dominated by emission from AGN-heated hot dust. Based on our SED-fit results, we estimate the black hole accretion density (BHAD; i.e., total BH growth rate per comoving volume) as a function of redshift. At
z
< 3, the resulting BHAD agrees with the X-ray measurements in general. At
z
> 3, we identify a total of 27 AGN and SF-AGN mixed objects, leading to that our high-
z
BHAD is substantially higher than the X-ray results (∼0.5 dex at
z
≈ 3–5). This difference indicates MIRI can identify a large population of heavily obscured AGN missed by X-ray surveys at high redshifts.
To facilitate routine genomic evaluation, a database was constructed to store genotypes for 50,972 single nucleotide polymorphisms (SNP) from the Illumina BovineSNP50 BeadChip (Illumina Inc., San ...Diego, CA). Multiple samples per animal are allowed. All SNP genotypes for a sample are stored in a single row. An indicator specifies whether the genotype for a sample was selected for use in genomic evaluation. Samples with low call rates or pedigree conflicts are designated as unusable. Among multiple samples that qualify for use in genomic evaluation, the one with the highest call rate is designated as usable. When multiple samples are stored for an animal, a composite is formed during extraction by using SNP genotypes from other samples to replace missing genotypes. To increase the number of SNP available, scanner output for approximately 19,000 samples was reprocessed. Any SNP with a minor allele frequency of ≥1% for Holsteins, Jerseys, or Brown Swiss was selected, which was the primary reason that the number of SNP used for USDA genomic evaluations increased. Few parent–progeny conflicts (≤1%) and a high call rate (≥90%) were additional requirements that eliminated 2,378 SNP. Because monomorphic SNP did not degrade convergence during estimation of SNP effects, a single set of 43,385 SNP was adopted for all breeds. The use of a database for genotypes, detection of conflicts as genotypes are stored, online access for problem resolution, and use of a single set of SNP for genomic evaluations have simplified tracking of genotypes and genomic evaluation as a routine and official process.
Previous biogeographic studies of late Cretaceous (late Campanian) vertebrate faunas in the Western Interior Basin (WIB) of North America have suggested the presence of faunal and floral ...provincialism, characterized by distinct northern and southern ‘biomes.’ However, the “provincialism hypothesis” has been questioned based largely on the contention that the investigated faunas were recovered from a series of diachronous, time-transgressive deposits, and are therefore non-correlative.
Extensive work in several fossiliferous units of late Campanian age, including the Dinosaur Park, Judith River, Two Medicine, Kaiparowits, Fruitland/Kirtland, and Aguja formations, has greatly increased understanding of WIB vertebrate faunas and their chronostratigraphic relationships. Here updated and greatly expanded faunal and chronostratigraphic datasets are utilized to undertake an extensive biogeographic analysis of these six terrestrial fossiliferous formations within the WIB of North America. Quantitative biogeographic comparisons of the formations and their constituent faunas are conducted using four statistical methods: Analysis of Similarity, Q-mode cluster analysis, Parsimony Analysis of Endemicity, and Correspondence Analysis.
The results of this study provide strong support for highly divergent faunas in northern and southern regions of the WIB, with a latitudinal faunal gradient as an interface. Yet the nature of the interface between these faunas remains unclear, with possibilities including: 1) two or more discrete provinces separated by a zone (or zones) of faunal mixing; and 2) a continuous latitudinal gradient or cline, with no discrete zones of endemism. Lacking evidence of any physiographic barrier to north–south dispersal, climatic variation within the WIB is regarded as the most likely explanation for the overarching biogeographic patterns observed for late Campanian vertebrate taxa.

This study compared the effects of daily calorie restriction (DR) versus intermittent fasting (IF) on markers of inflammation and extracellular matrix deposition in adipose tissue and skeletal muscle in a controlled feeding trial in women with overweight or obesity.
Women (N = 76) were randomised to one of three diets and provided with all foods at 100% (IF100) or 70% (IF70 and DR70) of calculated energy requirements for 8 weeks. IF groups ate breakfast prior to fasting for 24-h on 3 non-consecutive days/week. Weight, body composition, serum non-esterified fatty acids (NEFA), tumour necrosis factor-alpha (TNFα), interleukin-6 (IL-6), interleukin-10 (IL-10), M1- and M2-macrophage markers by qPCR and immunohistochemistry in adipose tissue and skeletal muscle were measured following a 12-h overnight fast (fed day, all groups) and a 24-h fast (IF groups only).
IF70 resulted in greater weight and fat losses and reductions in serum NEFA versus DR70 and IF100 (P < 0.05) after fed days. Markers of inflammation in serum (TNFα, IL6 and IL10), subcutaneous adipose tissue and skeletal muscle (CD68, CD40 and CD163) were unchanged by DR or IF after fed days. After fasting, NEFA, M1-macrophages (CD40+) in adipose tissue, and M2-macrophages (CD163+) in muscle were increased in IF70 and IF100 (all P < 0.05) and the changes in NEFA and mRNA of pan-macrophage marker CD68 in adipose tissue were positively correlated (r = 0.56, P = 0.002).
Unlike caloric restriction, IF transiently elevated markers of macrophage infiltration in adipose tissue and skeletal muscle, possibly in response to marked increases in adipose tissue lipolysis.
Background Pulmonary arteriovenous malformations (PAVMs) lead to stroke, brain abscess, and hemorrhage in hereditary hemorrhagic telangiectasia (HHT). The current screening approach for PAVMs in HHT ...patients with chest radiograph (CXR) and oxygen shunt study has not been validated and is thought to be insensitive. We hypothesized that agitated saline contrast echocardiography (ECHO) would be a useful screening test for PAVMs. Methods and Results A total of 106 sequential HHT patients underwent screening for PAVMs with ECHO in a prospective study. If the test was positive, or if the CXR or shunt study suggested PAVMs, pulmonary angiography was performed. A positive ECHO was defined as appearance of bubbles in the left atrium after injection of agitated saline solution. A positive shunt study was defined as a partial pressure of oxygen in arterial blood <500 mm Hg while breathing 100% oxygen. The mean age was 41 years (range 15-80 years); 66% were female. Forty-four patients had positive ECHO. Forty-one of the 44 patients underwent angiography. Three patients declined further testing. Thirty-three of the 41 patients who underwent angiography were diagnosed with PAVMs. Of the 62 patients with a negative ECHO, 18 underwent angiography because of either a shunt study or CXR that was suggestive of PAVMs. Of these 18 patients, 2 had PAVMs. In the total population of 106 patients, 35 (33%) had PAVMs. ECHO was the only positive screening test in 11 of 35 (31%) patients. The diagnosis of PAVMs in these 11 patients would have otherwise been missed. Conclusions ECHO is a useful screening tool for PAVMs in HHT. (Am Heart J 2001;141:243-6.)
