We established a relationship among the immune-related genes, tumor-infiltrating immune cells (TIICs), and immune checkpoints in patients with osteosarcoma. The gene expression data for osteosarcoma ...were downloaded from UCSC Xena and GEO database. Immune-related differentially expressed genes (DEGs) were detected to calculate the risk score. "Estimate" was used for immune infiltrating estimation and "xCell" was used to obtain 64 immune cell subtypes. Furthermore, the relationship among the risk scores, immune cell subtypes, and immune checkpoints was evaluated. The three immune-related genes (TYROBP, TLR4, and ITGAM) were selected to establish a risk scoring system based on their integrated prognostic relevance. The GSEA results for the Hallmark and KEGG pathways revealed that the low-risk score group exhibited the most gene sets that were related to immune-related pathways. The risk score significantly correlated with the xCell score of macrophages, M1 macrophages, and M2 macrophages, which significantly affected the prognosis of osteosarcoma. Thus, patients with low-risk scores showed better results with the immune checkpoints inhibitor therapy. A three immune-related, gene-based risk model can regulate macrophage activation and predict the treatment outcomes the survival rate in osteosarcoma.
Until recently, a Chinese rural-to-urban migrant worker's citizenship was primarily seen as a matter of household registration (hukou), since these workers were excluded from China's urban hukou ...system and therefore did not share in the benefits of urban citizenship. This conception of Chinese migrant worker's citizenship is no longer entirely accurate. Using the case of the 'points system' implemented in Dongguan city, this article examines in detail the citizenship demands of migrant workers, the government's responses, and the gaps between the demands and the response. Based on Dongguan's experience, a 'differentiated citizenship' structure is becoming universal in Chinese cities. The development of urban citizenship for these migrant workers is along the path of 'greater inclusion but differentiated exclusion.'
We study the possibly existing anisotropy in the accelerating expansion Universe with the Union2 type la supernovae data and gamma-ray burst data. We construct a direction-dependent dark energy model ...and constrain the anisotropy direction and strength of modulation. We find that the maximum anisotropic deviation direction is (l, b) = (126degrees, 13degrees) or, equivalently, (l, b) = (306degrees, -13degrees), and the current anisotropy level is (ProQuest: Formulae and/or non-USASCII text omitted) (1sigma confidence level with Union2 data). Our results do not show strong evidence for the anisotropic dark energy model. We also discuss potential methods that may distinguish the peculiar velocity field from the anisotropic dark energy model.
The intent of this meta-analysis was to examine the efficacy of thoracolumbar interfascial plane block (TLIP) for pain control after lumbar spinal surgery.
Randomized controlled trials (RCTs) ...published on PubMed, CENTRAL, Scopus, Embase, and Web of Science databases up to February 10, 2023, comparing TLIP with no or sham block or wound infiltration for lumbar spinal surgeries were included. Pain scores, total analgesic consumption, and postoperative nausea and vomiting (PONV) were analyzed.
Seventeen RCTs were eligible. Comparing TLIP with no block or sham block, the meta-analysis showed a significant decrease of pain scores at rest and movement at 2 h, 8 h, 12 h, and 24 h. Pooled analysis of four studies showed a significant difference in pain scores at rest between TLIP and wound infiltration group at 8 h but not at 2 h, 12 h, and 24 h. Total analgesic consumption was significantly reduced with TLIP block as compared to no block/sham block and wound infiltration. TLIP block also significantly reduced PONV. GRADE assessment of the evidence was moderate.
Moderate quality evidence indicates that TLIP blocks are effective in pain control after lumbar spinal surgeries. TLIP reduces pain scores at rest and movement for up to 24 h, reduces total analgesic consumption, and the incidence of PONV. However, evidence of its efficacy as compared to wound infiltration of local anesthetics is scarce. Results should be interpreted with caution owing low to moderate quality of the primary studies and marked heterogeneity.
Ewing's sarcoma has a poor prognosis and high metastasis rate; thus, it is critical to explore prognostic biomarkers of m6A-related genes. Two datasets were downloaded from the Gene Expression ...Omnibus database, m6A-related genes were extracted, and prognostic models were constructed using the least absolute shrinkage and selection operator and multivariate COX regression analyses. Immune cell composition and drug sensitivity analyses were performed, and our analysis was validated using laboratory methods of immunohistochemical specific staining and qRT-PCR. Ewing's sarcoma prognostic model demonstrated that the survival rate of cases in the high-risk group was much lower than that of the low-risk group. Naïve B cells, macrophages M0, macrophages M1, and resting mast cells are closely associated with Ewing's sarcoma. METTL14 and YTHDF2 are strongly associated with multiple drug sensitivity. Immunohistochemical specific staining revealed higher expression of both METTL14 and YTHDF2 in Ewing's sarcoma than in the paraneoplastic tissues. The results of qRT-PCR showed that METTL14 expression was significantly higher in both ES cell lines than in the control cell line. The prognostic model constructed using m6A-related genes METTL14 and TYHDF2, can be a potential prognostic biomarker for Ewing's sarcoma, with the survival rate of cases in the high-risk group being much lower than that of the low-risk group.
Myosin heavy chain 9 (MYH9) plays an essential role in human diseases, including multiple cancers; however, little is known about its role in gliomas. In the present study, we revealed that HMGA1 and ...MYH9 were upregulated in gliomas and their expression correlated with WHO grade, and HMGA1 promoted the acquisition of malignant phenotypes and chemoresistance of glioma cells by regulating the expression of MYH9 through c-Jun-mediated transcription. Moreover, MYH9 interacted with GSK-3β to inhibit the expression of GSK-3β protein by promoting its ubiquitination; the downregulation of GSK-3β subsequently promoted the nuclear translocation of β-catenin, enhancing growth, invasion, migration, and temozolomide resistance in glioma cells. Expression levels of HMGA1 and MYH9 were significantly correlated with patient survival and should be considered as independent prognostic factors. Our findings provide new insights into the role of HMGA1 and MYH9 in gliomagenesis and suggest the potential application of HMGA1 and MYH9 in cancer therapy in the future.
Nanofluid offers more opportunities and challenges over the traditional surfactant and polymer solutions during enhanced oil recovery (commonly referred to as tertiary oil recovery) due to its ...remarkable properties. This review mainly discusses the preparation methods of amphiphilic nanoparticles due to their higher interface activity than sole hydrophilic or hydrophobic nanoparticles (SHNPs). The nanofluids' stability is reviewed in this work. Moreover, the mechanisms of nanofluids in enhancing oil recovery (N-EOR) in terms of interfacial tension reduction, wettability alteration, foam stabilization, emulsion stabilization, structural disjoining pressure, and depressurization-increasing injection are mainly summarized and reviewed. Also, the synergistic effects of nanofluids and traditional surfactants and polymers are discussed. Finally, nanofluids’ challenges and prospects are also outlined. The nanofluids can still be regarded as an outstanding candidate for enhancing oil recovery significantly in the future although there are limitations on their applications from laboratory scale to field scale.
Aristolochic acid analogues (AAAs), naturally existing in herbal
Aristolochia
and
Asarum
genera, were once widely used in traditional pharmacopeias because of their anti-inflammatory properties, but ...lately they were identified as potential nephrotoxins and mutagens. A method for rapid characterization of AAAs in serum was developed using ion mobility spectrometry coupled with mass spectrometry (IMS-MS). Five AAAs, containing four aristolochic acids and one aristolactam, were separated and identified within milliseconds. AAAs were separated in gas phase based on the difference of their ion mobility (
K

), and then identified based on their

values,

, and product ions from MS/MS. Quantitative analysis of AAAs was performed using an internal standard with a satisfactory sensitivity. Limits of detection (signal-to-noise = 3) and quantification (signal-to-noise = 10) were 1–5 ng/mL and 3–8 ng/mL, respectively. The method was validated and successfully applied to the pharmacokinetics study of AAAs in rats, offering a promising way for fast screening and evaluation of AAAs in biological samples.

This paper examines multi-field behavior analysis of piezomagnetic microchips panels in a multi-field environment. The displacement field is modeled using the third-order theory for more accurate ...analysis and satisfying the shear stress condition. The principle of virtual work is employed to obtain the electro-magneto-elastic governing equations of the microchips. The numerical results are obtained to investigate the effect of significant parameters such as micro-scale parameters, electro-magnetic loads, and length to radius ratio. The radial and axial displacements are increased with the increase of micro-scale parameters, while the circumferential displacement is decreased significantly.
Lower-phase microemulsions with core-shell structure were prepared by microemulsion dilution method. The high temperature resistant systems were screened and the performance evaluation experiments ...were conducted to clarify the spontaneous imbibition mechanisms in ultra-low permeability and tight oil reservoirs, and to direct the field microfracture huff and puff test of oil well. The microemulsion system (O-ME) with cationic-nonionic surfactant as hydrophilic shell, No.3 white oil as oil phase core has the highest imbibition recovery; its spontaneous imbibition mechanisms include: the ultra-low interfacial tension and wettability reversal significantly reduce oil adhesion work to improve oil displacement efficiency, the nanoscale “core-shell structure” formed can easily enter micro-nano pores and throats to expand the swept volume, in addition, the remarkable effect of dispersing and solubilizing crude oil can improve the mobility of crude oil. Based on the experimental results, a microfracture huff and puff test of O-ME was carried out in Well YBD43-X506 of Shengli Oilfield. After being treated, the well had a significant increase of daily fluid production to 5 tons from 1.4 tons, and an increase of daily oil production to 2.7 tons from 1.0 ton before treatment.
