To investigate the postconditioning protective effect of penehyclidine hydrochloride (PHC) against anoxia/reoxygenation (A/R) injury in H9c2 cells along with the involved mechanism and timing effect.
...We divided H9c2 cells into 7 groups: control group, A/R group and PHC+A/R groups at 0 min, 5 mins, 10 mins, 20 mins, 30 mins, respectively (treated with 0.1 μm/L PHC at 0 min, 5 mins, 10 mins, 20 mins, 30 mins after the reoxygenation procedure began). Cell apoptosis, oxidative stress, intracellular Ca
concentration, mitochondrial membrane potential and mitochondrial permeability transition pore (MPTP) opening were explored. Bcl-2, Bax, Cyt C, caspase-3 and caspase-9 levels were measured.
A/R significantly increased both cell injury and cell apoptosis. PHC showed postconditioning protective effect by attenuating superoxide production, decreasing Ca
overload, restraining MPTP activities, restoring mitochondrial membrane potential, regulating cell apoptosis proteins and modulation of mitochondrial pathway. Earlier administration of PHC offered greater postconditioning protective effect.
H9c2 cells were protected by PHC from A/R injury regardless of timing of PHC administration (0 min, 5 mins, 10 mins, 20 mins, 30 mins). However, earlier administration of PHC resulted in better PHC postconditioning protection.
Phase-modulated metasurfaces (PMMs) have attracted considerable attention because of their widespread application in many fields. However, the most existing PMMs are only available in a relatively ...narrow band. In this paper, we propose a design of a broadband PMM based on vanadium dioxide (VO2) patches. The unit cell of the proposed PMM is composed of a surface layer supported by a 0.5 mm F4B with a dielectric constant of εr = 2.65, and a dielectric layer with a metal ground on its back. The surface layer consists of two opposite T-shaped metal patches connected by a VO2 patch. The VO2 patch can be used as an active element due to the insulator-to-metal transition of VO2. The process of phase transition is achieved by direct heating with a heating plate, without additional feeders affecting the performance of the PMM. The designed PMM can achieve a broadband phase difference within 180° ± 37° from 6.94 to 20.84 GHz, with a fractional bandwidth of 100%. Both full wave simulation and measurement results show that the proposed metasurface presents a good phase reconstruction performance over a wideband frequency range.
Abstract Objective This study investigates the relationship between hukou conversion and the psychological integration of rural older migrants, exploring the mediating role of accessibility to health ...resources. Methods The 3,963 valid samples of rural older migrants included in the study were sourced from the 2017 China Migrants Dynamic Survey (CMDS). The study established a multiple linear regression model for estimation and utilized inverse probability-weighted regression adjustment (IPWRA) method to correct for the selection bias of hukou conversion. Results Compared to older migrants with rural hukou , merit-based (β = 0.384, 95% CI: 0.265 to 0.504), family-based (β = 0.371, 95% CI: 0.178 to 0.565) and policy-based (β = 0.306, 95% CI: 0.124 to 0.487) converters have significantly higher psychological integration. These findings remain robust even after addressing the potential issue of endogenous selection bias using the IPWRA method. Bootstrap mediating effect tests indicate that hukou conversion can indirectly affect psychological integration through the mediator role of health resources accessibility. Conclusion Accessibility of health resources mediates the association between hukou conversion and psychological integration. Policymakers should enhance the implementation of hukou conversion, strengthen the health resource guarantee system, and achieve a deeper psychological integration among rural older migrants.
This Letter reports the first extraction of individual antineutrino spectra from ^{235}U and ^{239}Pu fission and an improved measurement of the prompt energy spectrum of reactor antineutrinos at ...Daya Bay. The analysis uses 3.5×10^{6} inverse beta-decay candidates in four near antineutrino detectors in 1958 days. The individual antineutrino spectra of the two dominant isotopes, ^{235}U and ^{239}Pu, are extracted using the evolution of the prompt spectrum as a function of the isotope fission fractions. In the energy window of 4-6 MeV, a 7% (9%) excess of events is observed for the ^{235}U (^{239}Pu) spectrum compared with the normalized Huber-Mueller model prediction. The significance of discrepancy is 4.0σ for ^{235}U spectral shape compared with the Huber-Mueller model prediction. The shape of the measured inverse beta-decay prompt energy spectrum disagrees with the prediction of the Huber-Mueller model at 5.3σ. In the energy range of 4-6 MeV, a maximal local discrepancy of 6.3σ is observed.
Sensitive detection of off-target effects is important for translating CRISPR-Cas9 nucleases into human therapeutics. In vitro biochemical methods for finding off-targets offer the potential ...advantages of greater reproducibility and scalability while avoiding limitations associated with strategies that require the culture and manipulation of living cells. Here we describe circularization for in vitro reporting of cleavage effects by sequencing (CIRCLE-seq), a highly sensitive, sequencing-efficient in vitro screening strategy that outperforms existing cell-based or biochemical approaches for identifying CRISPR-Cas9 genome-wide off-target mutations. In contrast to previously described in vitro methods, we show that CIRCLE-seq can be practiced using widely accessible next-generation sequencing technology and does not require reference genome sequences. Importantly, CIRCLE-seq can be used to identify off-target mutations associated with cell-type-specific single-nucleotide polymorphisms, demonstrating the feasibility and importance of generating personalized specificity profiles. CIRCLE-seq provides an accessible, rapid, and comprehensive method for identifying genome-wide off-target mutations of CRISPR-Cas9.

) nanoparticles-modified potassium titanate whiskers (AlNPs-MPTW), which were used as ...reinforcements to produce MPTW MC nylon and AlNPs-MPTW MC nylon, respectively, through in-situ polymerization. Then the thermodynamic stability and mechanical properties of these two kinds of nylon prepared and unmodified MC nylon were studied, proving that AlNPs-MPTW MC nylon possesses better performance. It was found that the prepared AlNPs-MPTW MC nylon with 0.1 wt% AlNP content and 1 wt% MPTW content delivered the optimal properties, specifically tensile strength 97±6 MPa, tensile modulus 3140±45 MPa, flexural strength 116±8 MPa, flexural modulus 3640±53 MPa, and notched impact strength 11.61 kJ/m

. The results showed that multiscale synergistic modified MC nylon possesses remarkable reinforcing and toughening effects, thereby enjoying very broad application prospects.
Using data samples with a total integrated luminosity of 20.1 fb−1 collected by the BESIII detector operating at the BEPCII collider, the cross section of the process e+ e− → π+ π− ψ ( 3686 ) is ...measured at center-of-mass energies between 4.0076 and 4.6984 GeV. The measured cross section is consistent with previous results, and with much improved precision. A fit to the measured energy-dependent cross section, which includes three Breit-Wigner functions and a nonresonant contribution, confirms the existence of the charmonium-like states Y (4220), Y (4390), and Y (4660). This is the first observation of the Y (4660) at the BESIII experiment.

By analyzing (448.1±2.9)×106 ψ(3686) events collected with the BESIII detector operating at the BEPCII collider, the decays of χcJ→4KS0 (J=0, 1, 2) are observed for the first time with statistical ...significances of 26.5σ, 5.9σ and 11.4σ, respectively. The product branching fractions of ψ(3686)→γχcJ, χcJ→4KS0 are presented, and the branching fractions of χcJ→4KS0 decays are determined to be Bχc0→4KS0=(5.76±0.34±0.38)×10−4, Bχc1→4KS0=(0.35±0.09±0.03)×10−4 and Bχc2→4KS0=(1.14±0.15±0.08)×10−4, where the first uncertainties are statistical and the second are systematic, respectively.
Induced pluripotent stem (iPS) cells are derived by epigenetic reprogramming, but their DNA methylation patterns have not yet been analyzed on a genome-wide scale. Here, we find substantial ...hypermethylation and hypomethylation of cytosine-phosphate-guanine (CpG) island shores in nine human iPS cell lines as compared to their parental fibroblasts. The differentially methylated regions (DMRs) in the reprogrammed cells (denoted R-DMRs) were significantly enriched in tissue-specific (T-DMRs; 2.6-fold, P < 10(-4)) and cancer-specific DMRs (C-DMRs; 3.6-fold, P < 10(-4)). Notably, even though the iPS cells are derived from fibroblasts, their R-DMRs can distinguish between normal brain, liver and spleen cells and between colon cancer and normal colon cells. Thus, many DMRs are broadly involved in tissue differentiation, epigenetic reprogramming and cancer. We observed colocalization of hypomethylated R-DMRs with hypermethylated C-DMRs and bivalent chromatin marks, and colocalization of hypermethylated R-DMRs with hypomethylated C-DMRs and the absence of bivalent marks, suggesting two mechanisms for epigenetic reprogramming in iPS cells and cancer.
