Aspirin, compared with placebo, reduces the risk of cardiovascular events by 25% in populations of patients with and without known arterial vascular disease. However, the phenomena of 'aspirin ...resistance' that has been described in 5'--50% of this population may critically reduce aspirin's efficacy. One study has suggested that aspirin-resistant patients have a 3.5 times higher risk of cardiovascular death. Presently, there are no established, simple, broadly used methods determining antiplatelet properties of aspirin, while conventional aggregometry requires special equipment and trained personnel. We sought to determine the validity of an Ultegra analyzer with the novel aspirin-sensitive cartridge before and after one pill of non-enteric coated aspirin (325 mg) in subjects with multiple risk factors for coronary artery disease. One hundred and fifty-four volunteers were enrolled into the multicenter study, but six of them were excluded. Data from 148 participants were analyzed. Platelets were assessed twice at baseline (pre-aspirin), and after 2-30 h (post-aspirin). We employed 5 micromol/l epinephrine-induced conventional aggregometry, and aspirin response units stimulated by propyl gallate with the point-of-care Ultegra analyzer. A single pill of aspirin reduced platelet-rich plasma aggregation from 72 +/- 21% to 25 +/- 10%, and diminished aspirin reduction units from 647 +/- 95 to 436 +/- 69. The overall agreement between the two methods was 87% with 85% sensitivity for Ultegra and 88% for platelet aggregation, respectively. The correlation between the two methods was 0.902. Timely determination of aspirin resistance represents an indispensable application in current medicine. The Ultegra RPFA-ASA analyzer is a novel, fast method that could be used in clinical practice for monitoring efficacy of aspirin, and for triaging the aspirin-resistant population. The clinical implications of these data need to be proven in randomized trials.
Changes in dielectric properties and thermal stability of the alpha-aluminum hydride polymorph (α-AlH
3
) after irradiation with electrons with an energy of 7 MeV were studied by dielectric ...spectroscopy, differential scanning calorimetry, thermogravimetry, and powder X-ray diffraction. Dielectric relaxation was observed in α-AlH
3
samples irradiated using an electron beam with a frequency of the electric field of more than 5 MHz. The dielectric properties of α-AlH
3
samples depend on the absorbed dose and the duration of time elapsed after the cessation of irradiation, while the temperature of the onset of decomposition and the kinetics of thermal decomposition of α-AlH
3
samples vary little for absorbed doses up to 160 kGy. Radiation-chemical yield of the formation of the metallic phase during α-AlH
3
radiolysis was determined by potentiometric titration.
Depression after acute coronary syndromes (ACSs) has been identified as an independent risk factor for subsequent cardiac death. Enhanced platelet activation has been hypothesized to represent 1 of ...the mechanisms underlying this association. Selective serotonin reuptake inhibitors (SSRIs) are known to inhibit platelet activity. Whether treatment of depressed post-ACS patients with SSRIs alters platelet function was not known. Accordingly, we serially assessed the release of established platelet/endothelial biomarkers in patients treated with sertraline vs placebo in the Sertraline AntiDepressant Heart Attack Randomized Trial (SADHART).
Plasma samples (baseline, week 6, and week 16) were collected from patients randomized to sertraline (n=28) or placebo (n=36). Anticoagulants, aspirin, and ADP-receptor inhibitors were permitted in this study. Platelet factor 4, beta-thromboglobulin (betaTG), platelet/endothelial cell adhesion molecule-1, P-selectin, thromboxane B2, 6-ketoprostaglandin F1a, vascular cell adhesion molecule-1, and E-selectin were measured by ELISA. Treatment with sertraline was associated with substantially less release of platelet/endothelial biomarkers than was treatment with placebo. These differences attained statistical significance for betaTG (P=0.03) at weeks 6 and 16 and for P-selectin (P=0.04) at week 16. Repeated-measures ANOVA revealed a significant advantage for sertraline vs placebo for diminishing E-selectin and betaTG concentrations across the entire treatment period.
Treatment with sertraline in depressed post-ACS patients is associated with reductions in platelet/endothelial activation despite coadministration of widespread antiplatelet regimens including aspirin and clopidogrel. The antiplatelet and endothelium-protective properties of SSRIs might represent an attractive additional advantage in patients with depression and comorbid coronary artery and/or cerebrovascular disease.
The immobilization of α-chymotrypsin on the surface of boron silicate glass microspheres is conducted via the technique of multilayer adsorption of polyelectrolytes. It is shown that the enzyme is ...adsorbed on both positively and negatively charged surfaces and its activity is partially preserved relative to that in solution. The activity of the enzyme depends on the number of polyectrolyte layers preliminarily adsorbed on glass microspheres and on the charge of the surface. The activity of α-chymotrypsin adsorbed on the negatively charged surface is four times higher than the activity of this enzyme adsorbed on a positively charged surface.
Introduction: unfavorable phenomena in society make new demands for the upbringing and development of the child's personality, for transforming his views on the surrounding reality, for changing his ...attitude towards people, himself and his own activity. However, some theorists and practitioners of education defend the position of denying the need for a school of educational work and shift their entire attention only to the problems of teaching, focusing on the technological development of education. However, it is quite obvious that the school has always been, is and will be a teaching and educational institution responsible not only for educating children, but also for the development of the child's personality. In modern conditions, the need to focus the content of educational programs on Russian culture, traditions of people's life, local history is actualized. The purpose of the article is to present the results of the long-term experience of one of the leading schools of Nizhny Novgorod, MAOU "School No. 187 with in-depth study of individual subjects"..Materials and methods: the diversity of the material studied required the use of different approaches and a set of complementary methods that ensured the objectivity and scientific validity of the research results: theoretical methods: comparative-comparative analysis of philosophical, psychological, pedagogical, methodological literature; method of scientific synthesis of factual materials; inductive and deductive methods in their unity in the generalization of materials; pedagogical monitoring: evaluation and analysis of products of activity. Methodological basis is system, personality-activity and culturological approaches. Results: in the scientific research, the conceptual foundations of the spiritual and moral system of the upbringing of the school are revealed; the modern approach to development and realization of educational process on the basis of orthodox values and cultural traditions is proved. The expediency of introduction of administrative-organizational, pedagogical, psychological, technological conditions of introduction of the system of education in school is proved. The development of the personality of the teacher in the spirit of traditional Orthodox values is determined by the fundamental condition for the realization of the tasks of spiritual and moral education of students in school. Features of introduction of innovative forms of the organization of educational process are considered. The criteria for the effectiveness of spiritual and moral education in the educational organization, diagnostic tools are presented.Discussion and Conclusions: the long-term experience of the school proves the readiness of the school to effectively ensure the formation of spiritual, moral, and civil security of the individual in pupils, successful socialization and professional self-determination of students, which ultimately contributes to increasing civil responsibility for the fate of the country, security and sustainable development of the country.
Some methodological approaches to solving several key problems of dynamic loading and structural strength analysis of Permanent Space Station (PSS)modules developed on the basis of the working ...experience of Soviet and Russian PSS and the International Space station (ISS) are presented. The solutions of the direct and semi-inverse problems of PSS structure dynamics are mathematically stated. Special attention is paid to the use of the results of ground structural strength tests of space station modules and the data on the actual flight actions on the station and its dynamic responses in the orbital operation regime. The procedure of determining the dynamics and operation life parameters of elements of the PSS modules is described.
Results are presented for experimental investigations of the dependence of the diameter of soil-cement columns on production parameters of jet grouting and various soil conditions.
Cell cultures of higher organisms, especially human cells, are increasingly used in medical, pharmaceutical, and scientific research. The main difficulty for cell cultures is hidden nonlethal ...contamination by mycoplasmas, viruses, and cross contamination with other cells. We suggest using PCR kits designed and officially employed in clinical diagnostics as an accessible and reliable method for monitoring the purity of cell cultures. We tested 50 human cell lines using commercial diagnostic systems for detection of papillomaviruses, herpes viruses, adenoviruses,
Mycoplasma hominis
, and total bacterial mass. No contamination was revealed in the cell lines tested. For the cell lines that contained integrated parts of viral genomes, the presence of the corresponding DNA sequences was confirmed. These diagnostic systems can be effectively used to test the purity of cell lines and for qualitative detection of possible contamination, as well as for quantitative evaluations using calculation of viral load, just as is practiced in clinical diagnostics.
A search for the rare decays $B^0_s$→$μ$+$μ$- and B0→$μ$+$μ$- is performed at the LHCb experiment using data collected in pp collisions corresponding to a total integrated luminosity of 4.4 fb-1. An ...excess of $B^0_s$→$μ$+$μ$- decays is observed with a significance of 7.8 standard deviations, representing the first observation of this decay in a single experiment. The branching fraction is measured to be $B$($μ$+$μ$-)=(3.0 ± $0.6^{+0.3}_{-0.2}$) × 10-9, where the first uncertainty is statistical and the second systematic. The first measurement of the $B^0_s$→$μ$+$μ$- effective lifetime, τ($B^0_s$→($μ$+$μ$-) = 2.04 ± 0.44 ± 0.05 ps, is reported. No significant excess of B0→($μ$+$μ$- decays is found, and a 95% confidence level upper limit, $B$(B0→($μ$+$μ$-) < 3.4 × 10-10, is determined. All results are in agreement with the standard model expectations.
