Objectives
The chemical composition, antimicrobial and synergistic effect, and cytotoxic activity of Citrus limon (lemon), Piper nigrum (green pepper) and Melaleuca alternifoila (tea tree) essential ...oils (EOs) were investigated.
Methods
Chemical analyses of essential oils were tested by GC‐FID and GC‐MS spectroscopy. The antimicrobial activity assay was conducted using microdilution method against several oral bacteria and Candida spp. originating from the humans with oral disorders. The synergistic antimicrobial activity was evaluated using checkerboard method. The cytotoxicity evaluation of EOs was assessed using MTT test.
Key findings
Limonene (37.5%) and β‐pinene (17.9%) were the major compounds in C. limon oil, β‐pinene (34.4%), δ‐3‐carene (19.7%), limonene (18.7%) and α‐pinene (10.4%) in P. nigrum oil and terpinen‐4‐ol (38.6%) and γ‐terpinene (21.7%) in M. alternifolia oil. The broad‐spectrum antimicrobial activity was achieved by tested three EOs, with C. limon oil being the strongest against bacteria and M. alternifolia oil strongest against fungi. The EOs demonstrated synergism; their combined application revealed an increase in antimicrobial activity. All tested essential oils showed lower cytotoxic activity in comparison with the positive control, and the obtained results confirmed a dose‐dependent activity.
Conclusions
The results of this study encourage use of tested EOs in development of a novel agent intended for prevention or therapy of corresponding oral disorders.

In this study, the petals collected from peony hybrid Paeonia lactiflora "Bowl of Beauty" and Calendula officinalis L. were extracted using an ethanol-water mixture assisted with microwave and ...ultrasonic treatment. The isolation of Calendula officinalis L. essential oil was done by hydrodistillation as well. The total phenolic and flavonoids content in the extracts and oil were determined and their antioxidant activity was evaluated. The highest total phenolic content was found for the extracts of hybrid P. lactiflora and C. officinalis L. obtained by ultrasound extraction (83.16 and 114.47 mg GA/g, respectively), while the flavonoid content obtained by microwave-assisted extraction was relatively high (123.48 and 65.29 mg QE/g, respectively). The highest antioxidant activity was obtained in DPPH• and ABTS•+ assay for the microwave-assisted extraction of hybrid P. lactiflora (79% and 83%) and ultrasound-assisted extraction of C. officinalis L. (45% and 49%), respectively. To improve antioxidant activity of both types of examined analytes (extracts and essential oil), the pectin biopolymer film (as a carrier) was prepared in the process of enzymatically assisted catalysis. Optical microscopy and FTIR spectroscopy were used for the characterization of obtained materials. The films, with essential oil of C. officinalis L. and gallic acid, showed significantly increased percentage inhibition in DPPH• and ABTS•+ test (91% and 95%, respectively) after 10 minutes. The results, also, showed that all formulations of pectin biopolymer film, modified with gallic acid, can be successfully applied as a carrier for both types of ingredients.
In this study the content of chloroplast pigments (chlorophyll a, b, a+b, and carotenoids) in the leaves of Geranium macrorrhizum L., Doronicum columnae Ten., Aegopodium podagraria L. and Tussilago ...farfara L. from a beech forest that had undergone fire on Vidlič Mountain was determined. The same species of plants from a place that had not been exposed to fire were taken as controls. Chloroplast pigments were determined from acetone extracts of these plants spectrophotometrically. In the first year after the fire the content of chlorophyll a, b and a+b in Geranium macrorrhizum L. and Doronicum columnae Ten. was greater than in the plants not exposed to fire. The results were the opposite for Aegopodium podagraria L. and Tussilago farfara L. These differences can be attributed to the different physiology of the plants and consequently their different adaptation patterns. The carotenoid content was higher in the plant species at the fire site than in the area not exposed to fire.

Svrha je ovoga rada bila ispitati citotoksični, proapoptotski, antimigratorni i pro-antioksidacijski učinak metanolnih, acetonskih i etil-acetatnih ekstrakata dvaju vrsta lišaja, i to: Pseudevernia ...furfuracea i Platismatia glauca na tumorske stanične linije debelog crijeva (HCT-116 i SW-480). Citotoksični efekti na tumorskim staničnim linijama debelog crijeva uspoređeni su s onima na staničnoj liniji normalnih humanih fibroblasta (MRC-5). Citotoksičnost je ispitana MTT testom, stanična migracija Transwell esejom, dok je apoptoza praćena fluorescentnom metodom pomoću akridin narančastog bojila i etidijevog bromida. Prooksidacijski/antioksidacijski učinak praćen je spektrofotometrijskim mjerenjem koncentracije redoks parametara. Ispitani ekstrakti imali su značajan citotoksični učinak na tumorske stanične linije, bez zapaženog utjecaja na normalnu staničnu liniju. Najjača citotoksičnost postignuta je nakon 72 h obrade staničnih linija ekstraktima vrste P. furfuracea (IC50= (21,2±1.3) µg/mL pri obradi HCT-116 stanica etil-acetatnim ekstraktom i IC50=(51.3±0.8) µg/mL pri obradi stanica SW-480 acetonskim ekstraktom). Ekstrakti P. furfuracea imali su značajan proapoptotski i prooksidacijski učinak, dok je metanolni ekstrakt te vrste imao najjači antimigratorni učinak na ispitane tumorske stanične linije. Svi ispitani ekstrakti vrste P. glauca imali su znatan citotoksični učinak na stanice HCT-116 nakon 72 h tretmana (IC50<40 μg/mL), dok su metanolni i acetonski ekstrakti imali citotoksični učinak na stanice SW-480 nakon 24 h, s izraženom proapoptotskom/nekrotskom aktivnošću. Svi navedeni efekti posljedica su induciranog oksidacijskog stresa u tretiranim stanicama. Može se zaključiti da ekstrakti lišaja bitno mijenjaju vijabilnost i migratorni potencijal ispitanih tumorskih staničnih linija karcinoma debelog crijeva. Dobiveni rezultati pokazuju da su stanice HCT-116 osetljivije na ispitane ekstrakte, pri čemu ekstrakti vrste P. furfuracea pokazuju bolji proapototski i antimigratorni učinak. Ispitane vrste lišaja mogu se smatrati izvorima antikancerogenih aktivnih tvari.
Glutathione-S transferases (GSTs) are xenobiotic-conjugation enzymes involved in the detoxification process of heterocyclic aromatic amines and polycyclic aromatic hydrocarbons, widely recognized ...risk factors of colorectal cancer (CRC) development. Polymorphism in GSTs often leads to alteration or complete lack of enzyme activity, which might have an effect on CRC carcinogenesis. Aim of this study was to investigate GST gene variants as risk factors in patients with CRC. A total of 523 CRC patients administered for surgical resection and 400 matched controls were included. Deletion polymorphism of GSTs M1 and T1 was investigated by polymerase chain reaction. Single nucleotide polymorphism of GST A1 and P1 was investigated by restriction fragment length polymorphism method. The association between GST genotype and risk of CRC development was found in carriers of GSTT1-null and GSTP1-variant genotypes individually (

= 0.016, respectively). Furthermore, statistically significant association was found when combination of GSTP1-variant genotype with any of other three common GST genotypes was analyzed with respect to CRC susceptibility. Additionally, patients with combined GSTM1-null/GSTT1-null/GSTA1 low-activity/GSTP1-variant genotype showed 2.71-fold increased risk of developing CRC (
p
= 0.037). This study supports hypothesis that GST polymorphisms might have an important role in the process of the CRC development. Additionally, GSTM1-null/ GSTT1-null/ GSTA1 low-activity/ GSTP1-variant genotype could be combination of GST genotypes whose carriers are more prone to CRC development.
