The broad distribution and often high densities of the cyanobacterium Trichodesmium spp. in oligotrophic waters imply a substantial role for this one taxon in the oceanic N cycle of the marine ...tropics and subtropics. New results from 154 stations on six research cruises in the North Atlantic Ocean show depth‐integrated N2 fixation by Trichodesmium spp. at many stations that equalled or exceeded the estimated vertical flux of NO3− into the euphotic zone by diapycnal mixing. Areal rates are consistent with those derived from several indirect geochemical analyses. Direct measurements of N2 fixation rates by Trichodesmium are also congruent with upper water column N budgets derived from parallel determinations of stable isotope distributions, clearly showing that N2 fixation by Trichodesmium is a major source of new nitrogen in the tropical North Atlantic. We project a conservative estimate of the annual input of new N into the tropical North Atlantic of at least 1.6 × 1012 mol N by Trichodesmium N2 fixation alone. This input can account for a substantial fraction of the N2 fixation in the North Atlantic inferred by several of the geochemical approaches.
Abstract Hot Jupiters were many of the first exoplanets discovered in the 1990s, but in the decades since their discovery the mysteries surrounding their origins have remained. Here we present nine ...new hot Jupiters (TOI-1855 b, TOI-2107 b, TOI-2368 b, TOI-3321 b, TOI-3894 b, TOI-3919 b, TOI-4153 b, TOI-5232 b, and TOI-5301 b) discovered by NASA’s TESS mission and confirmed using ground-based imaging and spectroscopy. These discoveries are the first in a series of papers named the Migration and Evolution of giant ExoPlanets survey and are part of an ongoing effort to build a complete sample of hot Jupiters orbiting FGK stars, with a limiting Gaia G -band magnitude of 12.5. This effort aims to use homogeneous detection and analysis techniques to generate a set of precisely measured stellar and planetary properties that is ripe for statistical analysis. The nine planets presented in this work occupy a range of masses (0.55 M J < M P < 3.88 M J ) and sizes (0.967 R J < R P < 1.438 R J ) and orbit stars that have an effective temperature in the range of 5360 K < T eff < 6860 K with Gaia G -band magnitudes ranging from 11.1 to 12.7. Two of the planets in our sample have detectable orbital eccentricity: TOI-3919 b ( e = 0.259 − 0.036 + 0.033 ) and TOI-5301 b ( e = 0.33 − 0.10 + 0.11 ). These eccentric planets join a growing sample of eccentric hot Jupiters that are consistent with high-eccentricity tidal migration, one of the three most prominent theories explaining hot Jupiter formation and evolution.
Fidaxomicin is an approved therapy for Clostridium difficile-associated diarrhea (CDAD) in adults. The safety of fidaxomicin in children has not been reported.
In this study (ClinicalTrials.gov ...identifier NCT01591863), pediatric patients with CDAD received twice-daily oral fidaxomicin at a dose of 16 mg/kg per day (up to 200 mg) for 10 days in an open-label study. Plasma and fecal samples were collected for pharmacokinetic assessments. The primary outcome measure was safety, which was assessed by adverse-event (AE), laboratory, and physical examination/vital-sign monitoring. Efficacy was determined through early and sustained clinical response rates (clinical response without recurrence of CDAD).
The study enrolled 40 patients (11 months to 17 years of age), many with underlying comorbidity, including neoplasm (23.7%), gastrointestinal disorder (78.9%), and history of CDAD (60.5%). Plasma fidaxomicin and OP-1118 (the major fidaxomicin metabolite) 3- to 5-hour postdose concentrations were 0.6 to 87.4 and 2.4 to 882.0 ng/mL, respectively, and no age-related trends were seen. Fecal fidaxomicin concentrations within 24 hours of the last dose averaged 3228 µg/g, and higher concentrations and greater variability in the youngest age group were found. AEs were reported in 73.7% of the patients; most of them were mild (44.7%) to moderate (21.1%) and were considered treatment-related in 15.8% of the patients. Overall, the early clinical response rate was 92.1%. The rate of sustained clinical response (clinical response without recurrence through 28 days after treatment) was 65.8% overall.
Fidaxomicin was well tolerated in children with CDAD and has a pharmacokinetic profile in children similar to that in adults. The clinical response rate was high.
Pharmacologic inhibition of tumor necrosis factor-alpha (TNF-α) has been used in the management of a variety of inflammatory conditions. Recently, reports on the development of sarcoid-like ...granulomatous disease at multiple systemic sites after treatment with TNF-α inhibitors have emerged, although, to the authors' knowledge, orbital manifestations of this problem have not been previously described. A 48-year-old woman who received injections of adalimumab for the treatment of psoriatic arthritis developed right-sided orbital pain and inflammation. Orbital biopsy of a focal lesion demonstrated sarcoid-like granulomatosis, and a workup for other causes of this problem was noncontributory. This report represents the first documented case of this phenomenon in the orbit, and possible mechanisms are discussed in this presentation. Given the expanding role of TNF-α inhibitors and the increased frequency of their use, clinicians should be aware of this possible side effect.
Describing the distribution and clearance of HIV surrogates within the gastrointestinal tract to inform rectal microbicide development.
Radiolabeled simulated HIV-infected semen was administered, ...imaged, and biopsied to simulate and measure colonic HIV distribution after anal intercourse.
Healthy male subjects with a history of receptive anal intercourse and experience with the use of anal sex toys were recruited to this study. Apheresis isolated leukocytes were collected before simulated intercourse. These autologous leukocytes, radiolabeled with 9.25 MBq (111)Indium-oxine (cell-associated HIV surrogate), and sulfur colloid particles, labeled with 37 MBq (99m)Technectium (cell-free HIV surrogate), were mixed in 3 mL autologous seminal plasma. This simulated HIV-infected semen was administered to subjects via an artificial phallus with urethra after 5 minutes of simulated intercourse. Postdosing dual isotope Single photon emission computed tomography coupled with traditional computed tomography (SPECT/CT) images were acquired at 1, 4, 8, and 24 hours. At 5 hours postdosing, colon biopsies were collected, CD4 cells were extracted, and samples analyzed for radioactivity.
SPECT/CT images showed similar luminal distribution for both surrogates, with migration limited to the rectosigmoid colon in all subjects. SPECT showed at least 75% overlap in distribution of both surrogates up to 4 hours after dosing. Biopsies indicate that 2.4% of CD4 cells extracted from rectosigmoid colon tissue were exogenously administered.
Our HIV surrogates stayed within the rectosigmoid colon for 24 hours. Exogenously dosed autologous lymphocytes and HIV-sized particles migrate to similar locations and associate with the colonic tissue in the lumen.
Abstract JWST has ushered in an era of unprecedented ability to characterize exoplanetary atmospheres. While there are over 5000 confirmed planets, more than 4000 Transiting Exoplanet Survey ...Satellite (TESS) planet candidates are still unconfirmed and many of the best planets for atmospheric characterization may remain to be identified. We present a sample of TESS planets and planet candidates that we identify as “best-in-class” for transmission and emission spectroscopy with JWST. These targets are sorted into bins across equilibrium temperature T eq and planetary radius R p and are ranked by a transmission and an emission spectroscopy metric (TSM and ESM, respectively) within each bin. We perform cuts for expected signal size and stellar brightness to remove suboptimal targets for JWST. Of the 194 targets in the resulting sample, 103 are unconfirmed TESS planet candidates, also known as TESS Objects of Interest (TOIs). We perform vetting and statistical validation analyses on these 103 targets to determine which are likely planets and which are likely false positives, incorporating ground-based follow-up from the TESS Follow-up Observation Program to aid the vetting and validation process. We statistically validate 18 TOIs, marginally validate 31 TOIs to varying levels of confidence, deem 29 TOIs likely false positives, and leave the dispositions for four TOIs as inconclusive. Twenty-one of the 103 TOIs were confirmed independently over the course of our analysis. We intend for this work to serve as a community resource and motivate formal confirmation and mass measurements of each validated planet. We encourage more detailed analysis of individual targets by the community.
ABSTRACT
We present the discovery and characterization of six short-period, transiting giant planets from NASA’s Transiting Exoplanet Survey Satellite (TESS) -- TOI-1811 (TIC 376524552), TOI-2025 ...(TIC 394050135), TOI-2145 (TIC 88992642), TOI-2152 (TIC 395393265), TOI-2154 (TIC 428787891), and TOI-2497 (TIC 97568467). All six planets orbit bright host stars (8.9 <G < 11.8, 7.7 <K < 10.1). Using a combination of time-series photometric and spectroscopic follow-up observations from the TESS Follow-up Observing Program Working Group, we have determined that the planets are Jovian-sized (RP = 0.99--1.45 RJ), have masses ranging from 0.92 to 5.26 MJ, and orbit F, G, and K stars (4766 ≤ Teff ≤ 7360 K). We detect a significant orbital eccentricity for the three longest-period systems in our sample: TOI-2025 b (P = 8.872 d, 0.394$^{+0.035}_{-0.038}$), TOI-2145 b (P = 10.261 d, e = $0.208^{+0.034}_{-0.047}$), and TOI-2497 b (P = 10.656 d, e = $0.195^{+0.043}_{-0.040}$). TOI-2145 b and TOI-2497 b both orbit subgiant host stars (3.8 < log g <4.0), but these planets show no sign of inflation despite very high levels of irradiation. The lack of inflation may be explained by the high mass of the planets; $5.26^{+0.38}_{-0.37}$ MJ (TOI-2145 b) and 4.82 ± 0.41 MJ (TOI-2497 b). These six new discoveries contribute to the larger community effort to use TESS to create a magnitude-complete, self-consistent sample of giant planets with well-determined parameters for future detailed studies.
