In light microscopy, refractive index mismatches between media and sample cause spherical aberrations that often limit penetration depth and resolution. Optical clearing techniques can alleviate ...these mismatches, but they are so far limited to fixed samples. We present Iodixanol as a non-toxic medium supplement that allows refractive index matching in live specimens and thus substantially improves image quality in live-imaged primary cell cultures, planarians, zebrafish and human cerebral organoids.
Recent decades have seen two important developments that animate the authors in this forum: the reemergence of Jewish studies scholarship in Russia and Poland - stimulated by often heroic individuals ...who emerged from the shadows of Communism - and growing postCommunist contestation and dispute over the sites of memory on which Jews lived and created. Ellie Schainker excavates the fascinating story of master archivist Benyamin Lukin, who played a central role in unearthing critical sources related to Russian Jewish history already in pre-1989 Leningrad, later in post-Soviet St. Petersburg, and finally in Jerusalem as head of the Eastern European section of the Central Archives of the History of the Jewish People. ChaeRan Freeze calls attention to the "affective revolution" in the work of scholars today who have recovered love from its erstwhile status as a flattened platonic ideal usually reserved for revered rabbis or books.
Part of the interpretive intrigue in "The Stranger," as in "Before the Law," is to ponder how equivalent Simmel's notion is to the Jew, who is mentioned in passing on but two occasions (in the latter ...case, in a discussion of the irony that the most impermanent residents of medieval Frankfurt, the Jews, were uniquely levied a fixed tax). The import of revisiting this figure is, at least, threefold: first, it allows us to piece together a genealogy of the stranger in modern society in order to remind ourselves that while the scale and pace of human alienation may be different today, its presence and form are not; second, it offers us an opportunity to explore, as Paul Mendes-Flohr did forty years ago, the method for studying those whom Isaac Deutscher memorably called "non-Jewish Jews";2 and third, it is an invitation to revisit important concepts and apparent antinomies that have stimulated, challenged, and provoked modern sociological, philosophical, and historical discourse since the early twentieth century. In attempting to apply his insight to the present, Liska offers an extended gloss on Simmel as she proposes an "alternative Jewish exemplarity . . . that would encompass the paradigmatic unity of detachment and fixity underlying Simmel's idea of the stranger."
Epigenetic regulation is based on a network of complexes that modulate the chromatin character and structure of the genome to impact gene expression, cell fate, and development. Thus, epigenetic ...modulators represent novel therapeutic targets used to treat a range of diseases, including malignancies. Infectious pathogens such as herpesviruses are also regulated by cellular epigenetic machinery, and epigenetic therapeutics represent a novel approach used to control infection, persistence, and the resulting recurrent disease. The histone H3K27 methyltransferases EZH2 and EZH1 (EZH2/1) are epigenetic repressors that suppress gene transcription via propagation of repressive H3K27me3-enriched chromatin domains. However, while EZH2/1 are implicated in the repression of herpesviral gene expression, inhibitors of these enzymes suppressed primary herpes simplex virus (HSV) infection
and
Furthermore, these compounds blocked lytic viral replication following induction of HSV reactivation in latently infected sensory ganglia. Suppression correlated with the induction of multiple inflammatory, stress, and antipathogen pathways, as well as enhanced recruitment of immune cells to
infection sites. Importantly, EZH2/1 inhibitors induced a cellular antiviral state that also suppressed infection with DNA (human cytomegalovirus, adenovirus) and RNA (Zika virus) viruses. Thus, EZH2/1 inhibitors have considerable potential as general antivirals through the activation of cellular antiviral and immune responses.
A significant proportion of the world's population is infected with herpes simplex virus. Primary infection and subsequent recurrent reactivation can result in diseases ranging from mild lesions to severe ocular or neurological damage. Herpesviruses are subject to epigenetic regulation that modulates viral gene expression, lytic replication, and latency-reactivation cycles. Thus, epigenetic pharmaceuticals have the potential to alter the course of infection and disease. Here, while the histone methyltransferases EZH2/1 are implicated in the suppression of herpesviruses, inhibitors of these repressors unexpectedly suppress viral infection
and
by induction of key components of cellular innate defense pathways. These inhibitors suppress infection by multiple viral pathogens, indicating their potential as broad-spectrum antivirals.
The Eternal Dissident offers rare insight into one of the most inspiring and thought-provoking Reform rabbis of the twentieth century, Leonard Beerman, who was renowned both for his eloquent and ...challenging sermons and for his unrelenting commitment to social action. Beerman was a man of powerful word and action—a probing intellectual and stirring orator, as well as a nationally known opponent of McCarthyism, racial injustice, and Israeli policy in the occupied territories. The shared source of Beerman’s thought and activism was the moral imperative of the Hebrew prophets, which he believed bestowed upon the Jewish people their role as the “eternal dissident.” This volume brings Beerman to life through a selection of his most powerful writings, followed by commentaries from notable scholars, rabbis, and public personalities that speak to the quality and ongoing relevance of Beerman’s work.
Antisemitism, which has been called the "longest hatred," appeared to be on a sharp decline in the United States in the early years of the new century. Over the past five years, antisemitism has ...surged to life, registering a 100% increase in reports of antisemitic incidents in the United States between 20016-18. That period coincides, and not by accident, with the presidency of Donald Trump, who declared himself to be a friend of Jews and a strong supporter of the state of Israel, has lapsed into stereotypical representations of Jews as beholden to money and loyal only to their own. In this way, the boundary between philosemitism and antisemitism became hard to trace. It is especially noteworthy that Trump arrogated to himself the right to define Jews, a move that calls to mind the infamous declaration of the mayor of Vienna in the late nineteenth century, Karl Lueger: "Who is a Jew-that I determine." This paper explores the naming of Jews not only in the context of Trump's declarations, but also policy formulations such as his Executive Order on antisemitism and the IHRA definition.
Three-dimensional (3D) mesostructured semiconductors show promising properties and applications; however, to date, few methods exist to synthesize or fabricate such materials. Metal can diffuse along ...semiconductor surfaces, and even trace amounts can change the surface behavior. We exploited the phenomena for 3D mesoscale lithography, by showing one example where iterated deposition-diffusion-incorporation of gold over silicon nanowires forms etchant-resistant patterns. This process is facet-selective, producing mesostructured silicon spicules with skeletonlike morphology, 3D tectonic motifs, and reduced symmetries. Atom-probe tomography, coupled with other quantitative measurements, indicates the existence and the role of individual gold atoms in forming 3D lithographic resists. Compared to other more uniform silicon structures, the anisotropic spicule requires greater force for detachment from collagen hydrogels, suggesting enhanced interfacial interactions at the mesoscale.
A leading scholar of Jewish history's bracing and challenging case for the role of the historian todayWhy do we study history? What is the role of the historian in the contemporary world? These ...questions prompted David N. Myers's illuminating and poignant call for the relevance of historical research and writing. His inquiry identifies a number of key themes around which modern Jewish historians have wrapped their labors: liberation, consolation, and witnessing. Through these portraits, Myers revisits the chasm between history and memory, revealing the middle space occupied by modern Jewish historians as they work between the poles of empathic storytelling and the critical sifting of sources.History, properly applied, can both destroy ideologically rooted myths that breed group hatred and create new memories that are sustaining of life. Alive in these investigations is Myers's belief that the historian today can and should attend to questions of political and moral urgency. Historical knowledge is not a luxury to society but an essential requirement for informed civic engagement, as well as a vital tool in policy making, conflict resolution, and restorative justice.
Abstract The etiology of community-acquired pneumonia (CAP) is determined in less than half of the patients based on cultures of sputum and blood plus testing urine for the antigens of Streptococcus ...pneumoniae and Legionella pneumophila . This study added nasal polymerase chain reaction (PCR) probes for S. pneumoniae , Staphylococcus aureus , and respiratory viruses. Serum procalcitonin (PCT) levels were measured. Pathogens were identified in 78% of the patients. For detection of viruses, patients were randomized to either a 5-virus laboratory-generated PCR bundle or the 17-virus FilmArray PCR platform. The FilmArray PCR platform detected more viruses than the laboratory-generated bundle and did so in less than 2 hours. There were fewer days of antibiotic therapy, P = 0.003, in CAP patients with viral infections and a low serum PCT levels.
The unstable political conditions of Zunz's day, punctuated by a rising tide of anti-Jewish expression and violence, and the absence of any enduring institutional framework to support Jewish studies, ...make his foresight all the more impressive. Nor could he have predicted the sheer number of scholars or range of topics represented at a meeting of the Association for Jewish Studies or World Congress of Jewish Studies. ...he sought to approach the Bible with the critical tools of Wissenschaft des Judentums rather than consign its study to Protestant scholars who, he believed, were intent on chronicling Judaism's decline. ...he attempted to restore pride of place to the Bible in modern Jewish scholarship, thereby rebalancing Leopold Zunz's focus on the wide body of postbiblical.
