Nigella sativa is known for the safety profile, containing a wealth of useful antiviral compounds. The main protease (Mpro, 3CLpro) of severe acute respiratory syndrome 2 (SARS-CoV-2) is being ...considered as one of the most attractive viral target, processing the polyproteins during viral pathogenesis and replication. In the current investigation we analyzed the potency of active component, thymoquinone (TQ) of Nigella sativa against SARS-CoV-2 Mpro. The structures of TQ and Mpro was retrieved from PubChem (CID10281) and Protein Data Bank (PDB ID 6MO3) respectively. The Mpro and TQ were docked and the complex was subjected to molecular dynamic (MD) simulations for a period 50ns. Protein folding effect was analyzed using radius of gyration (Rg) while stability and flexibility was measured, using root means square deviations (RMSD) and root means square fluctuation (RMSF) respectively. The simulation results shows that TQ is exhibiting good binding activity against SARS-CoV-2 Mpro, interacting many residues, present in the active site (His41, Cys145) and also the Glu166, facilitating the pocket shape. Further, experimental approaches are needed to validate the role of TQ against virus infection. The TQ is interfering with pocket maintaining residues as well as active site of virus Mpro which may be used as a potential inhibitor against SARS-CoV-2 for better management of COVID-19.
Diabetes mellitus (DM), an endocrine syndrome characterized by high blood glucose levels due to abrogated insulin activity. The existing treatments for DM have side effects and varying degrees of ...efficacy. Therefore, it is paramount that novel approaches be developed to enhance the management of DM. Therapeutic plants have been accredited as having comparatively high efficacy with fewer adverse effects. The current study aims to elucidate the phytochemical profile, anti-hyperlipidemic, and anti-diabetic effects of methanolic extract D. salicifolia (leaves) in Alloxan-induced diabetic mice. Alloxan was injected intraperitoneally (150 mg kg-1, b.w), to induced diabetes in mice. The mice were divided into three groups (n=10). Group 1 (normal control) received normal food and purified water, Group II (diabetic control) received regular feed and clean water and group III (diabetic treated) received a methanolic extract of the plant (300 mg kg-1) for 28 days with a typical diet and clean water throughout the experiment. Blood samples were collected to checked serum glucose and concentration of LDL, TC, TG. The extract demonstrated significant antihyperglycemic activity (P<0.05), whereas improvements in mice's body weight and lipid profiles were observed after treatment with the extract. This study establishes that the extract has high efficacy with comparatively less toxicity that can be used for DM management.
Abstract The supplementation of Selenium-enriched probiotics is effective in reducing oxidative stress and maintaining meat quality stability in broiler chicken especially under heat stress. An ...experimental study was conducted to perform Comparative analysis of Selenium yeast with inorganic Se in broilers under heat stress. A total of 120 broilers chicks of one day were assigned to 4 groups each consisting 30 chicks fed on same basal diet but different selenium sources. The basal diet of group D1 was not supplemented with Se source (Negative control), group D2 basal diet was supplemented with inorganic selenium (Sodium selenite 0.22mg/Kg starter phase and 0.15mg/Kg finisher phase), group D3 basal diet was supplemented with commercially available organic selenium (Seleno-methionine 0.22mg/Kg starter phase and 0.15mg/Kg finisher phase) and group D4 basal diet was supplemented with self-developed organic selenium (Se-enriched yeast 0.22mg/Kg starter phase and 0.15mg/Kg finisher phase). The performance parameters i.e. feed intake (FI), live body weight (BW) and FCR were not significantly (p>0.05) effected by selenium supplementation in the starter phase but were significantly (p<0.05) effected in the finisher phase. Selenium supplementation significantly (p<0.05) effected serum Se level in different supplemented groups. Higher serum Se value (58.20±0.06) was recorded in D4 group. Similarly significantly lower selenium value was recorded for D4 and higher was recorded for D1 (11.36±0.08). However lower serum Paraoxonase (PON) value was recorded for D4 (13.24±0.01) and higher for D1 (13.33±0.03). Comparatively self-developed Se enriched yeast increased the Se accumulation and improved antioxidant system. Glutathione peroxidase (GPx) was found higher in D4 (12.333±0.03) followed by D3, D2 and D1 respectively. Whereas superoxide dismutase (SOD) was significantly lower (p<0.05) in D4 (0.1437±0.003) followed by D3 (0.1457±0.002). Selenium supplementation increased the bird’s survival rate. Birds fed on Se enriched yeast showed higher Se deposition and better antioxidant capacity as compared to other sources of selenium. Se-enriched yeast displayed an improved result on Se deposition in tissues, and oxidative capacity, meat tenderness and immune response level as compared to other sources of selenium.
Resumo A suplementação de probióticos enriquecidos com selênio é eficaz na redução do estresse oxidativo e na manutenção da estabilidade da qualidade da carne em frangos de corte, especialmente sob estresse por calor. Um estudo experimental foi conduzido para realizar uma análise comparativa da levedura selênio com o Se inorgânico em frangos de corte sob estresse térmico. Um total de 120 pintos de um dia foi dividido em 4 grupos, cada um consistindo de 30 pintos alimentados com a mesma dieta basal, mas com diferentes fontes de selênio. A dieta basal do grupo D1 não foi suplementada com fonte de Se (controle negativo), a dieta basal do grupo D2 foi suplementada com selênio inorgânico (selenito de sódio 0,22 mg / kg fase inicial e 0,15 mg / kg fase finalizadora), a dieta basal do grupo D3 foi suplementada com selênio orgânico disponível comercialmente (fase inicial de seleno-metionina 0,22 mg / kg e fase finalizadora de 0,15 mg / kg) e a dieta basal do grupo D4 foi suplementada com selênio orgânico autodesenvolvido (fermento enriquecido com Se 0,22 mg / kg fase inicial e 0,15 mg / kg fase finalizadora). Os parâmetros de desempenho, ou seja, consumo de ração (FI), peso corporal vivo (PC) e FCR não foram significativamente (p > 0,05) afetados pela suplementação de selênio na fase inicial, mas foram significativamente (p < 0,05) afetados na fase final. A suplementação de selênio afetou significativamente (p < 0,05) o nível de Se sérico em diferentes grupos suplementados. O maior valor de Se sérico (58,20 ± 0,06) foi registrado no grupo D4. Da mesma forma, valor de selênio significativamente menor foi registrado para D4 e maior foi registrado para D1 (11,36 ± 0,08). No entanto, um valor mais baixo de Paraoxonase (PON) sérica foi registrado para D4 (13,24 ± 0,01) e mais alto para D1 (13,33 ± 0,03). A levedura enriquecida com Se comparativamente autodesenvolvida aumentou o acúmulo de Se e melhorou o sistema antioxidante. A glutationa peroxidase (GPx) foi encontrada maior em D4 (12,333 ± 0,03) seguido por D3, D2 e D1 respectivamente. Enquanto a superóxido dismutase (SOD) foi significativamente menor (p < 0,05) em D4 (0,1437 ± 0,003) seguido por D3 (0,1457 ± 0,002). A suplementação de selênio aumentou a taxa de sobrevivência da ave. Aves alimentadas com levedura enriquecida com Se apresentaram maior deposição de Se e melhor capacidade antioxidante em comparação com outras fontes de selênio. A levedura enriquecida com Se apresentou um resultado melhorado na deposição de Se nos tecidos, capacidade oxidativa, maciez da carne e nível de resposta imune em comparação com outras fontes de selênio.
Adding organic matter to soil proved an efficient strategy for restoring soil fertility and improving crop dry matter – an indicator of yield potential. In this regard, evaluating the sources of ...organic matter (OM) (control, poultry manure PM, farmyard manure FYM, compost, and mungbean residues MR) to provide 120 kg N ha-1 and effective microbes (EM) (0, 100, 200, and 300 L ton-1 of OM) as 2% solution proceeded in field conditions during 2017–2019. The sowing of wheat seeds (cv. Pirsabak 2015) at 120 kg ha-1 took place in the field using RCB design with four replications. Results showed that PM/FYM had delayed the phenology and improved the biomass-related parameters, dry matter (DM) accumulation, and crop growth rate (CGR) more than compost and MR. However, the results were more pronounced when applied with 300 L EM ton-1 of OM from the PM. The maximum DM (55%) accumulation in plant parts occurred beyond 100 days after sowing (DAS). A marked increase in DM and CGR beyond 60 DAS and a reduction in CGR beyond 120 DAS appeared irrespective of treatments. In the case of EM, the 300 L ton-1 revealed superior in terms of growth, DM accumulation, CGR, and delayed phenology. Structural equation modeling suggested that DM production gained a direct effect from crop phenology (46.1%) and crop stand (30.4%) but no indirect effect from crop growth (24.8%). In conclusion, the 300 L EM ton-1 of OM applied to PM or FYM had improved the crop stand, development, and DM production in wheat.
Early maturity and genotype by environment interaction (GEI) have always been challenging concerns for breeders in selecting appropriate parents for breeding programs. The presented study aimed to ...investigate early maturity and the dimension of gene action, as well as, assess the performance of half-diallel populations using eight advanced sweet corn inbred lines and their 28 F1 hybrids with two commercial checks for maturity and yield-related traits in the spring of 2018 at the Nowshera (plain) and Swat (hilly) areas, Khyber Pakhtunkhwa, Pakistan. Analysis revealed significant differences among the genotypes for the studied traits over both locations. General combining ability (GCA) effects were significant for all the traits at both the agro-climatic conditions, except 100-kernel weight, with the specific combining ability (SCA) effects relevant for grain yield at both locations. The GCA-SCA ratio for studied traits indicated dominance gene action, which also gained support by higher values of SCA than GCA variances. Based on the results, the identified inbred lines SWTS-1-8 and SODS-1 serve as good general combiners for traits like earliness and grain yield attributes, making them better parents to improve the stated characteristics in sweet corn. However, the F1 hybrids, i.e., NARCCCRI-19 × CCRI-34 at Nowshera and CCRI-34 × SODS-1 at Swat, showed the best specific combiners for maturity. Likewise, F1 hybrids, i.e., SWTS-1-4 × SWTS-1-8 and SWTS-1-8 × CCRIS-34, emerged as desirable for grain yield at Nowshera and Swat, respectively. The inbred lines for the mentioned hybrids can be a source of germplasm improvement, breaking through undesirable linkages in future sweet corn breeding programs.
Recent advances in wireless inter-vehicle communication systems enable the establishment of vehicular ad-hoc networks (VANET) and create significant opportunities for the deployment of a wide variety ...of applications and services to vehicles. In this work, we investigate the problem of developing services that can provide car drivers with time-sensitive information about traffic conditions and roadside facilities. We introduce the vehicular information transfer protocol (VITP), a location- aware, application-layer, communication protocol designed to support a distributed service infrastructure over vehicular ad- hoc networks. We describe the key design concepts of the VITP protocol and infrastructure. We provide an extensive simulation study of VITP performance on large-scale vehicular networks under realistic highway and city traffic conditions. Our results demonstrate the viability and effectiveness of VITP in providing location-aware services over VANETs.
The Veterans Health Administration system is one of the largest integrated health care providers in the United States, delivering medical care to > 9 million veterans. Barriers to delivering ...efficient health care include geographical limitations as well as long wait times. Telehealth has been used as a solution by many different health care services. However, it has not been as widely used in cancer care. In 2018, the US Department of Veterans Affairs (VA) Pittsburgh Healthcare System expanded the use of telehealth to provide antineoplastic therapies to rural patients by creating a clinical video telehealth clinic of the Virtual Cancer Care Network. This allows oncologists located at the tertiary center to virtually deliver care to remote sites. The recent COVID-19 pandemic forced oncologists across the VA system to adopt telehealth to provide continuity of care. On the basis of our review and personal experience, we have outlined opportunities for telehealth to play a role in every step of the cancer care journey from diagnosis to therapy to surveillance to clinical trials for medical, surgical, and radiation oncology. There are many advantages, such as decreased travel time and potential cost savings; however, there continues to be challenges with veterans having access to devices and the Internet as well as understanding how to use telehealth equipment. The lessons learned from this assessment of the VA telehealth system for cancer care can be adopted and integrated into other health systems. In the future, there needs to be evaluation of how telehealth can be further incorporated into oncology, satisfaction of veterans using telehealth services, overcoming telehealth barriers, and defining metrics of success.
Speech enhancement plays an important role in Automatic Speech Recognition (ASR) even though this task remains challenging in real-world scenarios of human-level performance. To cope with this ...challenge, an explicit denoising framework called Deep Denoising Autoencoder (DDAE) is introduced in this paper. The parameters of DDAE encoder and decoder are optimized based on the backpropagation criterion, where all denoising autoencoders are stacked up instead of recurrent connections. For better speech estimation in real and noisy environments, we include matched and mismatched noisy and clean pairs of speech data to train the DDAE. The DDAE has the ability to achieve optimal results even for a limited amount of training data. Our experimental results show that the proposed DDAE outperformed the baseline algorithms. The DDAE shows superior performances based on three-evaluation metrics in noisy and clean pairs of speech data compared to three baseline algorithms.
Pyrazinamide (PZA) is a key first-line antibiotic used for the short-course treatment of drug-sensitive and multidrug-resistant (MDR) isolates of tuberculosis. PZA exhibits potent action against ...semi-dormant bacilli in acidic environments. However, mutations that occur in target genes may cause technical difficulties in the diagnosis of PZA resistance during drug susceptibility testing. The objective of the current study is to identify mutations in pncAWT rpsA and rpsAWT panD genes among PZA-resistant isolates of Mycobacterium tuberculosis (MTB) circulating in the Pashtun dominant region, Khyber Pakhtunkhwa, Pakistan. We selected 18 PZA-resistant pncAWT strains from the Provincial Tuberculosis Reference Laboratory (PTRL) Khyber Pakhtunkhwa to investigate mutations in the coding region of rpsA and panD genes. The experiments were repeated for drug susceptibility testing using MGIT 960 automated system. In addition, eighteen PZA-resistant rpsA genes along with 5 susceptible strains and one H37Rv strain were sequenced. All 18 isolates were PZA-resistant. The majority of these isolates exhibited multidrug resistance (MDR) (13/18). We identified 14 non-synonymous and one synonymous mutation in the coding region of rpsA in 11 strains. All mutations were scattered throughout the gene and not reported previously. Further, we did not identify any mutation in 7 rpsAWT panD genes. Mutations in rpsA but not in panD occur in PZA-resistant pncAWT MTB isolates circulating in Khyber Pakhtunkhwa, Pakistan.
