The paper is theoretical and empirical. Its basic purpose is to examine the resilience of organizational startups during the post-COVID-19 pandemic. The basic research method used in the study was ...CAWI. The group of surveyed companies consisted of startups operating in Zachodniopomorskie, Wielkopolskie, and Lubuskie voivodeships. The survey covered 62 enterprises in 2022. The research revealed that owing to their organizational resilience the startups were able to adapt to the changes in the market. Thus, on the one hand, they were able to quickly tailor their offer to the current needs of customers, and on the other hand, they could modify the processes taking place inside the company. As a result of these measures, the startups increased their revenues, which may directly determine the future competitiveness of the surveyed companies.
Here we report the development of a proteolysis targeting chimera (PROTAC) based on the combination of the unique features of the sirtuin rearranging ligands (SirReals) as highly potent and ...isotype-selective Sirt2 inhibitors with thalidomide, a bona fide cereblon ligand. For the first time, we report the formation of a PROTAC by Cu(I)-catalyzed cycloaddition of a thalidomide-derived azide to an alkynylated inhibitor. This thalidomide-derived azide as well as the highly versatile linking strategy can be readily adapted to alkynylated ligands of other targets. In HeLa cells, our SirReal-based PROTAC induced isotype-selective Sirt2 degradation that results in the hyperacetylation of the microtubule network coupled with enhanced process elongation. Thus, our SirReal-based PROTAC is the first example of a probe that is able to chemically induce the degradation of an epigenetic eraser protein.
The smart home is a field that uses smart devices and gadgets to automate tasks and operations in the house, and it is expected that this study subject will become a focal point in future ...civilizations. The adoption of smart home services faces significant obstacles in terms of reliability and stability. The Software Defined Networking (SDN) paradigm offers up novel approaches and trends to embrace smart technologies to tackle the issues of smart homes. To avoid accidents, we developed a vertical SDN-based multi-level structure consisting of two controllers with a parent-child connection. It enables smart homes to make optimal use of current services while also providing new services to smart homes and buildings, opening the path for future smart services. In addition, we created an Application Programming Interface (API) for the smart home to make it easier to use current services and pave the path for future smart services. We designed numerous jobs depending on their categorization to give smart services, which is beneficial for smart homes. Local topologies, cloud topologies, and cloud-local topologies have all been compared in the evaluation section. The performance of POX, NOX, and Flood Light controllers is measured using three performance metrics: mean throughput, Round-Trip Time (RTT), and packet loss across all three topologies. Our findings revealed that the cloud-local topology had the lowest packet loss ratio of 1.4%, while the local and cloud topologies had 1.9% and 2.3%, respectively.
In the era of industrial digitalization, companies are increasingly investing in tools and solutions that allow their processes, machines, employees, and even the products themselves, to be ...integrated into a single integrated network for data collection, data analysis, the evaluation of company development, and performance improvement. To study the impact of Industry 4.0 on the company we used Porter’s (1985) value chain model, which is particularly useful when paying particular attention to corporate areas which have a primary role in customer value creation. Since the primary impact of Industry 4.0 is perceived in value-creating processes, and has so far had the greatest transformative effect in this area, the model can be considered to be appropriate. The objective of our research is to discover how companies operating in Hungary interpret the phenomenon of Industry 4.0, what Internet of Things (IoT) tools they use to support their processes, and what critical issues they face during adaptation. We applied a dual methodology in our investigation: We sent an online questionnaire to manufacturing and logistical service companies to investigate the IoT tools they use, and the problems they face, and received 43 answers we could evaluate. We also conducted four expert interviews with manufacturing firms to get deeper insights into the application, critical issues and development phases of IoT tools. During our research, we found that the spread of real-time data across companies—given the availability of appropriate analytical tools and methods—can have a significant impact on the entire company. In the case of CPS (Cyber Physical System), CPPS and Big Data Technologies, companies using them have been evaluated as having a higher level of logistic service, more efficient processes with their partners, improved cooperation between certain logistic functions, and higher market and financial performance and competitiveness. Applying more efficient production processes, and achieving better productivity and economies of scale, might also result in increased economic sustainability. Furthermore, we have found that companies have started on the path to digital evolution, and investments of this type have already begun.
The concept of circular economy-based performance has gained significant interest within the highly competitive business arena and environment-concerned stakeholders. This study assumes that ...environment oriented HRM practices have significant effects on circular economy-based performance. This study aims to investigate the contributory effect of five selected components of green HRM on the circular economy-based performance of environment-concerned banking organizations. A total of 418 managers of commercial banks are the unit of analysis, and data were derived using a structured questionnaire from the respondents who are working in managerial positions in banking organizations in Bangladesh. The gathered data has been encoded by SPSS and analyzed through PLS-SEM 4. The results of this study reveal that all components of green HRM, i.e., green employment design, planning, staffing, training, and benefits significantly contribute to the circular economy-based performance of the banking sector. Thus, green HRM effectively promotes the circular performance of banks to provide a distinctive perspective for adding value to the environmental concerns in an emerging economy. Big data is one of the most recent business and technical issues in the age of technology. Hundreds of millions of events occur every day. The financial field is deeply involved in the calculation of ...big data events. As a result, hundreds of millions of financial transactions occur in the financial world each day. Therefore, financial practitioners and analysts consider it an emerging issue of the data management and analytics of different financial products and services. Also, big data has significant impacts on financial products and services. Therefore, identifying the financial issues where big data has a significant influence is also an important issue to explore with the influences. Based on these concepts, the objective of this paper was to show the current landscape of finance dealing with big data, and also to show how big data influences different financial sectors, more specifically, its impact on financial markets, financial institutions, and the relationship with internet finance, financial management, internet credit service companies, fraud detection, risk analysis, financial application management, and so on. The connection between big data and financial-related components will be revealed in an exploratory literature review of secondary data sources. Since big data in the financial field is an extremely new concept, future research directions will be pointed out at the end of this study.
DJ-1, a multi-functional protein with antioxidant properties, protects dopaminergic neurons against Parkinson's disease (PD). The oligomerization/assembly of alpha-synuclein (SYN), promoted by ...Tubulin Polymerization Promoting Protein (TPPP/p25), is fatal in the early stage of PD. The pathological assembly of SYN with TPPP/p25 inhibits their proteolytic degradation. In this work, we identified DJ-1 as a new interactive partner of TPPP/p25, and revealed its influence on the association of TPPP/p25 with SYN. DJ-1 did not affect the TPPP/p25-derived tubulin polymerization; however, it did impede the toxic assembly of TPPP/p25 with SYN. The interaction of DJ-1 with TPPP/p25 was visualized in living human cells by fluorescence confocal microscopy coupled with Bifunctional Fluorescence Complementation (BiFC). While the transfected DJ-1 displayed homogeneous intracellular distribution, the TPPP/p25-DJ-1 complex was aligned along the microtubule network. The anti-aggregative effect of DJ-1 on the pathological TPPP/p25-SYN assemblies was established by the decrease in the intensity of their intracellular fluorescence (BiFC signal) and the increase in the proteolytic degradation of SYN complexed with TPPP/p25 due to the DJ-1-derived disassembly of SYN with TPPP/p25. These data obtained with HeLa and SH-SY5Y cells revealed the protective effect of DJ-1 against toxic SYN assemblies, which assigns a new function to the antioxidant sensor DJ-1.

Dysregulation of human ...Sirt2 (hSirt2) activity has been associated with the pathogenesis of cancer, inflammation, and neurodegeneration, which makes the modulation of hSirt2 activity a promising strategy for pharmaceutical intervention. The sirtuin rearranging ligands (SirReals) have recently been discovered by us as highly potent and isotype-selective hSirt2 inhibitors. Here, we present a well-defined structure–activity relationship study, which rationalizes the unique features of the SirReals and probes the limits of modifications on this scaffold regarding inhibitor potency. Moreover, we present a crystal structure of hSirt2 in complex with an optimized SirReal derivative that exhibits an improved in vitro activity. Lastly, we show cellular hyperacetylation of the hSirt2 targeted tubulin caused by our improved lead structure.

This paper addresses the challenges of the transition from a fossil fuel-dependent to a bio-based economy and implications related to the production of food, feed, bioenergy and other bio-based materials. The objective is to provide a comprehensive review of global biomass and biomass-based energy supplies and demand, with particular attention to the EU. Furthermore, factors related to setting priorities in the use of non-food biomass are discussed, as food security will remain the top priority. Finally, the changes in the bioenergy balance indicators in the Member States of the EU and new plant breeding technologies are analyzed. Overall, this study describes the complexity of the bio-based value chains in making decisions on how best to use biomass. The article presents a comprehensive review on global biomass and biomass based energy supplies and demand, discusses the European chemical industry perspective, analyzes the changes in the biomass based energy balance indicators in the Member States of the EU, and considers the challenges of the new plant breeding technologies.

The bioeconomy is a highly complex, cross-sectorial concept covering all sectors and systems that rely on biomass. Based on the analysis of the size of bioeconomy in the V4 countries it can be concluded that the sustainable transition is a great challenge and can be achieved by developing national circular bioeconomy strategies. It is extremely difficult to analyse the contribution of different sectors to the performance of the circular bioeconomy. Currently, the contribution of different sectors to the biomass-based economy is very complex, therefore a strategic socio-economic planning and optimal resource allocation is necessary to achieve the goals of the bioeconomy. This paper aims to determine the place and role of the bioeconomy in the structural change of the linear economy, based on detailed matrices of in- and output relations between different sectors. Based on the input-output matrices, as well as centrality and flow metrics of network analysis, the role of the bioeconomy in the linear economy, its multiplicative effects and future research implications are analysed.
