Regional declines in polar bear (
) populations have been attributed to changing sea ice conditions, but with limited information on the causative mechanisms. By simultaneously measuring field ...metabolic rates, daily activity patterns, body condition, and foraging success of polar bears moving on the spring sea ice, we found that high metabolic rates (1.6 times greater than previously assumed) coupled with low intake of fat-rich marine mammal prey resulted in an energy deficit for more than half of the bears examined. Activity and movement on the sea ice strongly influenced metabolic demands. Consequently, increases in mobility resulting from ongoing and forecasted declines in and fragmentation of sea ice are likely to increase energy demands and may be an important factor explaining observed declines in body condition and survival.
The diets of the eight species of ursids range from carnivory (e.g., polar bears, Ursus maritimus) to insectivory (e.g., sloth bears, Melursus ursinus), omnivory (e.g., brown bears, U. arctos), and ...herbivory (e.g., giant pandas, Ailuropoda melanoleuca). Dietary energy availability ranges from the high-fat, highly digestible, calorically dense diet of polar bears (~ 6.4 kcal digestible energy/g fresh weight) to the high-fiber, poorly digestible, calorically restricted diet (~ 0.7) of giant pandas. Thus, ursids provide the opportunity to examine the extent to which dietary energy drives evolution of energy metabolism in a closely related group of animals. We measured the daily energy expenditure (DEE) of captive brown bears in a relatively large, zoo-type enclosure and compared those values to previously published results on captive brown bears, captive and free-ranging polar bears, and captive and free-ranging giant pandas. We found that all three species have similar mass-specific DEE when travel distances and energy intake are normalized even though their diets differ dramatically and phylogenetic lineages are separated by millions of years. For giant pandas, the ability to engage in low-cost stationary foraging relative to more wide-ranging bears likely provided the necessary energy savings to become bamboo specialists without greatly altering their metabolic rate.
Blastic plasmacytoid dendritic cell neoplasm (BPDCN) is a rare disease of controversial origin recently recognized as a neoplasm deriving from plasmacytoid dendritic cells (pDCs). Nevertheless, it ...remains an orphan tumor with obscure biology and dismal prognosis. To better understand the pathobiology of BPDCN and discover new targets for effective therapies, the gene expression profile (GEP) of 25 BPDCN samples was analyzed and compared with that of pDCs, their postulated normal counterpart. Validation was performed by immunohistochemistry (IHC), whereas functional experiments were carried out ex vivo. For the first time at the molecular level, we definitely recognized the cellular derivation of BPDCN that proved to originate from the myeloid lineage and in particular, from resting pDCs. Furthermore, thanks to an integrated bioinformatic approach we discovered aberrant activation of the NF-kB pathway and suggested it as a novel therapeutic target. We tested the efficacy of anti-NF-kB-treatment on the BPDCN cell line CAL-1, and successfully demonstrated by GEP and IHC the molecular shutoff of the NF-kB pathway. In conclusion, we identified a molecular signature representative of the transcriptional abnormalities of BPDCN and developed a cellular model proposing a novel therapeutic approach in the setting of this otherwise incurable disease.
These European Society for Clinical Microbiology and Infectious Diseases and European Confederation of Medical Mycology Joint Clinical Guidelines focus on the diagnosis and management of ...mucormycosis. Only a few of the numerous recommendations can be summarized here. To diagnose mucormycosis, direct microscopy preferably using optical brighteners, histopathology and culture are strongly recommended. Pathogen identification to species level by molecular methods and susceptibility testing are strongly recommended to establish epidemiological knowledge. The recommendation for guiding treatment based on MICs is supported only marginally. Imaging is strongly recommended to determine the extent of disease. To differentiate mucormycosis from aspergillosis in haematological malignancy and stem cell transplantation recipients, identification of the reverse halo sign on computed tomography is advised with moderate strength. For adults and children we strongly recommend surgical debridement in addition to immediate first-line antifungal treatment with liposomal or lipid-complex amphotericin B with a minimum dose of 5 mg/kg/day. Amphotericin B deoxycholate is better avoided because of severe adverse effects. For salvage treatment we strongly recommend posaconazole 4 × 200 mg/day. Reversal of predisposing conditions is strongly recommended, i.e. using granulocyte colony-stimulating factor in haematological patients with ongoing neutropenia, controlling hyperglycaemia and ketoacidosis in diabetic patients, and limiting glucocorticosteroids to the minimum dose required. We recommend against using deferasirox in haematological patients outside clinical trials, and marginally support a recommendation for deferasirox in diabetic patients. Hyperbaric oxygen is supported with marginal strength only. Finally, we strongly recommend continuing treatment until complete response demonstrated on imaging and permanent reversal of predisposing factors.

Context.
The ESA PLAnetary Transits and Oscillations of stars (PLATO) mission will search for terrestrial planets in the habitable zone of solar-type stars. Because of telemetry limitations, PLATO ...targets need to be pre-selected.
Aims.
In this paper, we present an all sky catalogue that will be fundamental to selecting the best PLATO fields and the most promising target stars, deriving their basic parameters, analysing the instrumental performances, and then planing and optimising follow-up observations. This catalogue also represents a valuable resource for the general definition of stellar samples optimised for the search of transiting planets.
Methods.
We used
Gaia
Data Release 2 astrometry and photometry and 3D maps of the local interstellar medium to isolate FGK (
V
≤ 13) and M (
V
≤ 16) dwarfs and subgiant stars.
Results.
We present the first public release of the all-sky PLATO input catalogue (asPIC1.1) containing a total of 2 675 539 stars including 2 378 177 FGK dwarfs and subgiants and 297 362 M dwarfs. The median distance in our sample is 428 pc for FGK stars and 146 pc for M dwarfs, respectively. We derived the reddening of our targets and developed an algorithm to estimate stellar fundamental parameters (
T
eff
, radius, mass) from astrometric and photometric measurements.
Conclusions.
We show that the overall (internal+external) uncertainties on the stellar parameter determined in the present study are ∼230 K (4%) for the effective temperatures, ∼0.1
R
⊙
(9%) for the stellar radii, and ∼0.1
M
⊙
(11%) for the stellar mass. We release a special target list containing all known planet hosts cross-matched with our catalogue.
Polar bears (
Ursus maritimus
Phipps, 1774) depend on sea ice for catching marine mammal prey. Recent sea-ice declines have been linked to reductions in body condition, survival, and population size. ...Reduced foraging opportunity is hypothesized to be the primary cause of sea-ice-linked declines, but the costs of travel through a deteriorated sea-ice environment also may be a factor. We used movement data from 52 adult female polar bears wearing Global Positioning System (GPS) collars, including some with dependent young, to document long-distance swimming (>50 km) by polar bears in the southern Beaufort and Chukchi seas. During 6 years (2004–2009), we identified 50 long-distance swims by 20 bears. Swim duration and distance ranged from 0.7 to 9.7 days (mean = 3.4 days) and 53.7 to 687.1 km (mean = 154.2 km), respectively. Frequency of swimming appeared to increase over the course of the study. We show that adult female polar bears and their cubs are capable of swimming long distances during periods when extensive areas of open water are present. However, long-distance swimming appears to have higher energetic demands than moving over sea ice. Our observations suggest long-distance swimming is a behavioral response to declining summer sea-ice conditions.
Tri-axial accelerometers have been used to remotely identify the behaviors of a wide range of taxa. Assigning behaviors to accelerometer data often involves the use of captive animals or surrogate ...species, as their accelerometer signatures are generally assumed to be similar to those of their wild counterparts. However, this has rarely been tested. Validated accelerometer data are needed for polar bears Ursus maritimus to understand how habitat conditions may influence behavior and energy demands. We used accelerometer and water conductivity data to remotely distinguish 10 polar bear behaviors. We calibrated accelerometer and conductivity data collected from collars with behaviors observed from video-recorded captive polar bears and brown bears U. arctos, and with video from camera collars deployed on free-ranging polar bears on sea ice and on land. We used random forest models to predict behaviors and found strong ability to discriminate the most common wild polar bear behaviors using a combination of accelerometer and conductivity sensor data from captive or wild polar bears. In contrast, models using data from captive brown bears failed to reliably distinguish most active behaviors in wild polar bears. Our ability to discriminate behavior was greatest when species- and habitat-specific data from wild individuals were used to train models. Data from captive individuals may be suitable for calibrating accelerometers, but may provide reduced ability to discriminate some behaviors. The accelerometer calibrations developed here provide a method to quantify polar bear behaviors to evaluate the impacts of declines in Arctic sea ice.
The extent of increasing anthropogenic impacts on large marine vertebrates partly depends on the animals’ movement patterns. Effective conservation requires identification of the key drivers of ...movement including intrinsic properties and extrinsic constraints associated with the dynamic nature of the environments the animals inhabit. However, the relative importance of intrinsic versus extrinsic factors remains elusive. We analyze a global dataset of ∼2.8 million locations from >2,600 tracked individuals across 50 marine vertebrates evolutionarily separated by millions of years and using different locomotion modes (fly, swim,walk/paddle). Strikingly,movement patterns show a remarkable convergence, being strongly conserved across species and independent of body length and mass, despite these traits ranging over 10 orders of magnitude among the species studied. This represents a fundamental difference between marine and terrestrial vertebrates not previously identified, likely linked to the reduced costs of locomotion in water. Movement patterns were primarily explained by the interaction between species-specific traits and the habitat(s) they move through, resulting in complex movement patterns when moving close to coasts compared with more predictable patterns when moving in open oceans. This distinct difference may be associated with greater complexity within coastal microhabitats, highlighting a critical role of preferred habitat in shaping marine vertebrate global movements. Efforts to develop understanding of the characteristics of vertebrate movement should consider the habitat(s) through which they move to identify how movement patterns will alter with forecasted severe ocean changes, such as reduced Arctic sea ice cover, sea level rise, and declining oxygen content.
Context.
Gas giants orbiting close to hot and massive early-type stars can reach dayside temperatures that are comparable to those of the coldest stars. These ‘ultra-hot Jupiters’ have atmospheres ...made of ions and atomic species from molecular dissociation and feature strong day-to-night temperature gradients. Photometric observations at different orbital phases provide insights on the planet’s atmospheric properties.
Aims.
We aim to analyse the photometric observations of WASP-189 acquired with the Characterising Exoplanet Satellite (CHEOPS) to derive constraints on the system architecture and the planetary atmosphere.
Methods.
We implemented a light-curve model suited for an asymmetric transit shape caused by the gravity-darkened photosphere of the fast-rotating host star. We also modelled the reflective and thermal components of the planetary flux, the effect of stellar oblateness and light-travel time on transit-eclipse timings, the stellar activity, and CHEOPS systematics.
Results.

< 0.48. We also find that the eclipse depth can only be explained by thermal emission alone in the case of extremely inefficient energy redistribution. Finally, we attribute the photometric variability to the stellar rotation, either through superficial inhomogeneities or resonance couplings between the convective core and the radiative envelope.
Conclusions.
Based on the derived system architecture, we predict the eclipse depth in the upcoming Transiting Exoplanet Survey Satellite (TESS) observations to be up to ~165 ppm. High-precision detection of the eclipse in both CHEOPS and TESS passbands might help disentangle reflective and thermal contributions. We also expect the right ascension of the ascending node of the orbit to precess due to the perturbations induced by the stellar quadrupole moment

(oblateness).
In B-cell chronic lymphocytic leukemia (B-CLL) cells, Lyn, a tyrosine kinase belonging to the Src family, is overexpressed and atypically localized in an aberrant cytosolic complex in an active ...conformation, contributing to the unbalance between cell survival and pro-apoptotic signals. In this study, we demonstrate that Lyn constitutively phosphorylates the immunoreceptor tyrosine inhibitory motifs of the inhibitory cell surface co-receptor CD5, a marker of B-CLL. As a result, CD5 provides an anchoring site to Src homology 2 domain-containing phosphatase 1 (SHP-1), a known negative regulator of hematopoietic cell function, thereby triggering the negative B-cell receptor (BCR) signaling. The subsequent segregation of SHP-1 into two pools, one bound to the inhibitory co-receptor CD5 in an active form, the other in the cytosol in an inhibited conformation, proves crucial for withstanding apoptosis, as shown by the use of phosphotyrosine phosphatase-I-I, a direct inhibitor of SHP-1, or SHP-1 knockdown. These results confirm that Lyn exhibits the unique ability to negatively regulate BCR signaling, in addition to positively regulating effectors downstream of the BCR, and identify SHP-1 as a novel player in the deranged signaling network and as a potential attractive target for new therapeutic strategies in B-CLL.
