The article delves into communication between service sector professionals and their customers, utilizing transcripts of authentic conversations sourced from the Russian "One Day of Speech" sound ...corpus. Its primary aim is to unearth recurring speech patterns that typify customer-service interactions. To achieve this, the conversation transcripts are segmented into individual speech acts. Each segment is then labeled, highlighting the speaker's role and the specific type and subtype of the speech act. By scrutinizing these subtypes, the study juxtaposes the discourse styles of both clients and service representatives, thereby discerning patterns influenced by the domain of communication. The findings from the study are beneficial for scientific research in speech communication as well as for optimizing automatic answering systems or chatbots.
Prime editing is a rapidly developing method of CRISPR/Cas-based genome editing. The increasing number of novel PE applications and improved versions demands constant analysis and evaluation. The ...present review covers the mechanism of prime editing, the optimization of the method and the possible next step in the evolution of CRISPR/Cas9-associated genome editing. The basic components of a prime editing system are a prime editor fusion protein, consisting of nickase and reverse transcriptase, and prime editing guide RNA, consisting of a protospacer, scaffold, primer binding site and reverse transcription template. Some prime editing systems include other parts, such as additional RNA molecules. All of these components were optimized to achieve better efficiency for different target organisms and/or compactization for viral delivery. Insights into prime editing mechanisms allowed us to increase the efficiency by recruiting mismatch repair inhibitors. However, the next step in prime editing evolution requires the incorporation of new mechanisms. Prime editors combined with integrases allow us to combine the precision of prime editing with the target insertion of large, several-kilobase-long DNA fragments.
Circulating cancer markers are metabolic products found in body fluids of cancer patients, which are specific for a certain type of malignant tumors. Cancer marker detection plays a key role in ...cancer diagnosis, treatment, and disease monitoring. The growing need for early cancer diagnosis requires quick and sensitive analytical approaches to detection of cancer markers. The approach based on the photonic crystal surface mode (PC SM) detection has been developed as a label-free high-precision biosensing technique. It allows real-time monitoring of molecular and cellular interactions using independent recording of the total internal reflection angle and the excitation angle of the PC surface wave. We used the PC SM technique for simultaneous detection of the ovarian cancer marker cancer antigen 125 and two breast cancer markers, human epidermal growth factor receptor 2 and cancer antigen 15-3. The new assay is based on the real-time flow detection of specific interaction between the antigens and capture antibodies. Its particular advantage is the possibility of multichannel recording with the same chip, which can be used for multiplexed detection of several cancer markers in a single experiment. The developed approach demonstrates high specificity and sensitivity for detection of all three biomarkers.
Living in times of increasing complexity is hard; it becomes even harder with the realisation of diminishing control. How do we adapt our governance to this complexity to ensure peaceful cohabitation ...of the established and emergent order regimes? This paper contends that it is important to embrace complexity in full, conceptually and practically, by shifting from ‘the global’ to ‘the local’, to understand the pressure of transformational change and to prepare the ground for the emergence of more resilient and cooperative orders. We apply this complexity-thinking, using a 3P analysis, to Central Eurasia, presently a battleground of three competing order-making regimes—the EU, China and Russia. We argue that for more resilient and cooperative orders to emerge, it is essential to understand and enable ‘the local’ and embrace the region in is diversity, to facilitate a more joined-up and bottom-up governance in managing the complexity of a changing world.
Glycogen storage disease type 1 (GSD1) is a rare hereditary monogenic disease characterized by the disturbed glucose metabolism. The most widespread variant of GSD1 is GSD1a, which is a deficiency of ...glucose-6-phosphatase-ɑ. Glucose-6-phosphatase-ɑ is expressed only in liver, kidney, and intestine, and these organs are primarily affected by its deficiency, and long-term complications of GSD1a include hepatic tumors and chronic liver disease. This article is a brief overview of existing animal models for GSD1a, from the first mouse model of 1996 to modern CRISPR/Cas9-generated ones. First whole-body murine models demonstrated exact metabolic symptoms of GSD1a, but the animals did not survive weaning. The protocol for glucose treatment allowed prolonged survival of affected animals, but long-term complications, such as hepatic tumorigenesis, could not be investigated. Next, organ-specific knockout models were developed, and most of the metabolic research was performed on liver glucose-6-phosphate-deficient mice. Naturally occuring mutation was also discovered in dogs. All these models are widely used to study GSD1a from metabolic and physiological standpoints and to develop possible treatments involving gene therapy. Research performed using these models helped elucidate the role of glycogen and lipid accumulation, hypoxia, mitochondrial dysfunction, and autophagy impairment in long-term complications of GSD1a, including hepatic tumorigenesis. Recently, gene replacement therapy and genome editing were tested on described models, and some of the developed approaches have reached clinical trials.

bivalve mollusk widespread in the Northwest Pacific was purified. The isolation procedure included ammonium sulfate precipitation followed by affinity chromatography on pectin-Sepharose. The full-length MkC1qDC sequence was assembled using de novo mass-spectrometry peptide sequencing complemented with N-terminal Edman's degradation, and included 176 amino acid residues with molecular mass of 19 kDa displaying high homology to bivalve C1qDC proteins. MkC1qDC demonstrated antibacterial properties against Gram-negative and Gram-positive strains. MkC1qDC binds to a number of saccharides in Ca
-dependent manner which characterized by structural meta-similarity in acidic group enrichment of galactose and mannose derivatives incorporated in diversified molecular species of glycans. Alginate, κ-carrageenan, fucoidan, and pectin were found to be highly effective inhibitors of MkC1qDC activity. Yeast mannan, lipopolysaccharide (LPS), peptidoglycan (PGN) and mucin showed an inhibitory effect at concentrations three orders of magnitude greater than for the most effective saccharides. MkC1qDC localized to the mussel hemal system and interstitial compartment. Intriguingly, MkC1qDC was found to suppress proliferation of human adenocarcinoma HeLa cells in a dose-dependent manner, indicating to the biomedical potential of MkC1qDC protein.
The article examines societal fragilities and local resilience strategies in Belarus with a particular focus on the notion of peoplehood. Premised on the idea of evolving forms of agency under the ...Anthropocene, and the emergent complexity-thinking in International Relations, the article draws on these approaches to societal fragilities and community resilience to understand and explain the unprecedented levels of mobilization occurring in Belarus since the disputed presidential election in August 2020. To this end, the article zooms onto the local communities to provide an analytical perspective on the study of resilience as self-organization. In line with complexity-thinking, it argues in favor of history-specific processual identities, shaped by the aspirations of a “good life,” and realized via local support infrastructures which lie at the heart of societal resilience in Belarus. Yet, the potential of all these elements to actualize into a sweeping transformative force, referred to as “peoplehood” in this article, is rare, and comes at a time of unprecedented crises and existential threats to the life of a community. The Belarusian society seems to be undergoing such a moment that not only makes it more resilient and adaptive to change; it also transforms it into a new form of societal being, self-aware of its worth, self-organized, and self-reliant on its inner capabilities to fight for a life of excellence. The article traces these moments of becoming with, and societal being, via a critical discussion of fragilities and the elements of resilience, actualized into peoplehood.
The rate of hydrolysis–condensation reaction of phenyltrichlorosilane in water-acetone solutions and the product yields were shown to significantly depend on the concentration of HCl (CHCl) in the ...solutions. The main product of the reaction was all-cis-(tetrahydroxy)(tetraphenyl)cyclotetrasiloxane. This was different from the earlier published results of analogous reactions of m-tolylSiCl3, m-ClPhSiCl3, and α-naphtylSiCl, in which some products of other types were formed. For example, trans-1,1,3,3-tetrahydroxy-1,3-di-α-naphtyldisiloxane was obtained in the case of α-naphtylSiCl3. All-cis-(tetrahydroxy)(tetraphenyl)cyclotetrasiloxane was treated in acetone with HCl to give the other three geometric isomers (cis-cis-trans-, cis-trans-, and all-trans-). The thermal self-condensation of these four isomers under “pseudo”-equilibrium conditions (under atmospheric pressure) was investigated in different solvents, in quartz or molybdenum glass flasks. The compositions of the products were monitored by APCI-MS and 29Si NMR spectroscopy. It was shown that all-cis- and cis-cis-trans-isomers in toluene or anisole mostly gave the cage-like Ph-T8,10,12,14 and uncompleted cage-like Ph-T10,12OSi(HO)Ph compounds. In contrast to these two isomers, the cis-trans–isomer in toluene mainly formed dimers with the loss of one or two molecules of water. However, in acetonitrile, significant amounts of Ph-T10,12 and Ph-T10,12OSi(HO)Ph species were formed along with the dimers. All-trans-isomer did not enter into the reaction at all.

under the forest canopy based on the composition and age of the forest stand. The studies conducted in the most widespread forest type of the South Ural Mountains confirmed our null hypothesis about the possibility of modelling the regeneration of
P. obovata
and
A. sibirica
by the structure of the stand. The number of young generations of
P. obovata
and
A. sibirica
showed high sensitivity to the age and composition of the stand. The graphical models developed by us can be used to combine with forest management data in order to obtain large-scale data on the regeneration of woody plants in the primary spruce small-grass-green mossy forests and secondary birch and aspen forests of the South Ural Mountains. In order to expand the scope of application of the developed models, it is planned to conduct additional studies for other forest types of the Urals.
