Aims
Brazil ranks high in the number of coronavirus disease 19 (COVID‐19) cases and the COVID‐19 mortality rate. In this context, autopsies are important to confirm the disease, determine associated ...conditions, and study the pathophysiology of this novel disease. The aim of this study was to assess the systemic involvement of COVID‐19. In order to follow biosafety recommendations, we used ultrasound‐guided minimally invasive autopsy (MIA‐US), and we present the results of 10 initial autopsies.
Methods and results
We used MIA‐US for tissue sampling of the lungs, liver, heart, kidneys, spleen, brain, skin, skeletal muscle and testis for histology, and reverse transcription polymerase chain reaction to detect severe acute respiratory syndrome coronavirus 2 RNA. All patients showed exudative/proliferative diffuse alveolar damage. There were intense pleomorphic cytopathic effects on the respiratory epithelium, including airway and alveolar cells. Fibrinous thrombi in alveolar arterioles were present in eight patients, and all patients showed a high density of alveolar megakaryocytes. Small thrombi were less frequently observed in the glomeruli, spleen, heart, dermis, testis, and liver sinusoids. The main systemic findings were associated with comorbidities, age, and sepsis, in addition to possible tissue damage due to the viral infection, such as myositis, dermatitis, myocarditis, and orchitis.
Conclusions
MIA‐US is safe and effective for the study of severe COVID‐19. Our findings show that COVID‐19 is a systemic disease causing major events in the lungs and with involvement of various organs and tissues. Pulmonary changes result from severe epithelial injury and microthrombotic vascular phenomena. These findings indicate that both epithelial and vascular injury should be addressed in therapeutic approaches.
The tensile strength of open-hole fibre reinforced composite laminates depends on in-plane, thickness and ply lay-up scaling. Translaminar (fibre direction) mode I fracture toughness has recently ...been experimentally determined to be thickness dependent. This paper presents a computational study of the tensile strength prediction of open-hole laminates using a cohesive zone model. To the authors’ knowledge, it is for the first time in the literature that the thickness-dependence of translaminar fracture toughness is accounted for in the numerical modelling of composites. The thickness size effect in the strength of open-hole composite laminates failed by pull-out is accurately predicted for the first time by a deterministic model. It is found that neglecting delamination in the numerical models will lead to mesh-dependency and over-estimation on the predicted strength. Smeared crack model with cohesive elements to model delamination is able to predict the correct failure mode; but it is found not suitable for accurate strength predictions for laminates failed by delamination.

Chayote (Sechium edule) has gained widespread consuming acceptance and recognized by its nutritional and bio-functional properties. The present review surveys and describes the current findings about the nutritional, phytochemical and pharmacological properties of chayote and identifies opportunities for further research. It also discusses chayote’s versatile utility in nutrition, as a functional ingredient in food, cosmetic and pharmaceutical industries, as well as in nanotechnology and biotechnological processes. It was concluded that although the pharmacological properties of chayote are currently well-established, only a few reports have been conducted on the isolation and identification of individual chemical constituents, and similarly, only a few in vivo studies have been conducted to assess their biological efficacy. In addition, the valorisation of the underutilized chayote by-products can be an important aspect in waste management from both economic and environmental standpoints. Thus, the recovery and utilization of valuable compounds from chayote is an important challenge for scientists.

Grilling muscle foods involves high temperatures that lead to production of cooking toxicants, such as heterocyclic aromatic amines (HAs) and polycyclic aromatic hydrocarbons (PAHs). To obtain realistic exposure levels of these two groups of mutagens analyses of the same samples using similar separation/detection techniques were performed. HAs and PAHs were quantified in well-done meat and fish samples grilled with wood and coconut shell charcoal at 200°C. Quantitative HAs and PAHs profiles were different for beef and salmon using the same type of charcoal. Higher levels of HAs and PAHs were found in salmon samples. No significant differences were observed for HAs and PAHs in beef samples grilled with both charcoal types, whereas salmon grilled with coconut shell charcoal presented significantly lower amounts of HAs and PAHs than salmon grilled with usual wood charcoal. Continuous barbecuing with the same charcoal shown that combustion of fat that dropped along the grilling period contributed to higher formation of HAs and PAHs. Special attention must be given to the intake of barbecued foods since high amounts of HAs and PAHs can be taken in a single meal.

Research on gastric lipolysis of commercial cow’s milk with different fatty acid (FA) compositions is scarce. Gastric lipase exhibits specificity for the sn-3 chain position of triacylglycerols, ...whose structure is influenced by milk FA composition. Therefore, during gastric digestion of conventional (C) vs pasture-based (P) milk, differences may occur on lipolysis, which has impact on free FA available, influencing their absorption/metabolism rate and physiological hormonal responses. Those two milk types were subjected to the INFOGEST semi-dynamic digestion model. Five gastric emptying points were analyzed for oxidative degradation of polyunsaturated fatty acids (PUFA) and individual free FA. The relative release of medium-chain FA (C8:0–C12:0) was higher than that of longer-chain FA (C14:0–C18:0), and a linear increase in markers of PUFA oxidative degradation occurred along gastric digestion. Quantitatively, C8:0, C18:2n-6, C18:3n-3, and CLAc9t11 were higher (P < 0.001) in P milk when compared with C milk.

This paper presents a new method suitable for modelling multiple discontinuities within a finite element. The architecture of the proposed method is similar to that of the phantom node method (which is equivalent to XFEM with Heaviside enrichment), and the solution of it is equivalent to local remeshing within the cracked element. The new method shows several advantages over the phantom node method, such as avoiding errors in the mapping of the crack geometry from the physical to the natural space and avoiding performing integrations over only part of an element. Compared to remeshing, the proposed method enables the representation of discontinuities through relatively closed FE codes (such as user-defined elements) without modifying the initial mesh and geometry, thus making it computationally more efficient. Additionally, the proposed method is particularly suited for modelling weak and cohesive discontinuities and for the representation of complex crack networks; it can model multiple plies and interfaces of a composite laminate, and both matrix crack and delamination, within a user-defined element; the information is shared between the plies and interfaces within such an element, allowing the direct implementation of interactive mechanisms. Verification examples show that the floating node method can predict stress intensity factors and crack propagation accurately. An application example shows that the proposed method can predict well the transition from matrix cracking to delamination and the subsequent saturation of matrix crack density in a cross-ply laminate.
A multi-residue methodology based on a solid phase extraction followed by gas chromatography–tandem mass spectrometry was developed for trace analysis of 32 compounds in water matrices, including ...estrogens and several pesticides from different chemical families, some of them with endocrine disrupting properties. Matrix standard calibration solutions were prepared by adding known amounts of the analytes to a residue-free sample to compensate matrix-induced chromatographic response enhancement observed for certain pesticides. Validation was done mainly according to the International Conference on Harmonisation recommendations, as well as some European and American validation guidelines with specifications for pesticides analysis and/or GC–MS methodology. As the assumption of homoscedasticity was not met for analytical data, weighted least squares linear regression procedure was applied as a simple and effective way to counteract the greater influence of the greater concentrations on the fitted regression line, improving accuracy at the lower end of the calibration curve. The method was considered validated for 31 compounds after consistent evaluation of the key analytical parameters: specificity, linearity, limit of detection and quantification, range, precision, accuracy, extraction efficiency, stability and robustness.

The effect of heat treatment and the presence or absence of fish skin on the volatile composition of Senegalese sole muscle was studied. The volatile profile of Senegalese sole at different storage periods was also evaluated. All samples were analysed by HS-SPME–GC–IT/MS and subjected to sensory evaluation. As expected, cooking enhanced the production/liberation of volatile compounds. Fish with the skin present, after cooking, had higher levels of sulphur compounds, 2-nonanone, ethyl octanoate and lower contents of hexanol and heptanol than skinned fish; moreover, the samples with the skin had a better overall sensory acceptability. During storage, changes on the volatile composition of Senegalese sole samples were found. The major differences were obtained after 2 weeks of storage. Compounds such as hexanal, heptanal, octanal, decanal, (E)-2-hexenal, (E)-2-decen-1-al, (E,Z)-2,6-nonadienal, benzaldehyde, 4-ethyl-benzaldehyde, 1-penten-3-ol, heptanol and (E)-2-octen-1-ol decreased after 2 weeks of storage, and other compounds, such as 3-methyl-1-butanal, 2-methyl-1-butanal, 2-heptanone, dimethyl trisulphide, dimethyl tetrasulphide and 2-methyltetrahydrothiophen-3-one increased. These differences were confirmed by sensory evaluation. Principal component analysis was applied to the chemical data.
Prostate cancer (PCA) is the second most common cancer diagnosis in men and the fifth leading cause of death worldwide. The conventional treatments available are beneficial to only a few patients ...and, in those, some present adverse side effects that eventually affect the quality of life of most patients. Thus, there is an urgent need for effective, less invasive and targeted specific treatments for PCA. Photothermal therapy (PTT) is a minimally invasive therapy that provides a localized effect for tumour cell ablation by activating photothermal agents (PTA) that mediate the conversion of the light beam's energy into heat at the site. As tumours are unable to easily dissipate heat, they become more susceptible to temperature increases. In the PTT field, gold nanoparticles (AuNPs) have been attracting interest as PTA. The aim of this study was to formulate AuNPs capable of remaining retained in the tumour and subsequently generating heat at the tumour site. AuNPs were synthesized and characterized in terms of size, polydispersity index (PdI), zeta potential (ZP), morphology and the surface plasmon resonance (SPR). The safety of AuNPs and their efficacy were assessed using in vitro models. A preliminary in vivo safety assessment of AuNPs with a mean size lower than 200 nm was confirmed. The morphology was spherical-like and the SPR band showed good absorbance at the laser wavelength. Without laser, AuNPs proved to be safe both in vitro (>70% viability) and in vivo. In addition, with laser irradiation, they proved to be relatively effective in PCA cells. Overall, the formulation appears to be promising for use in PTT.
