HOX transcript antisense RNA (HOTAIR) is a well-known long non-coding RNA (lncRNA) whose dysregulation correlates with poor prognosis and malignant progression in many forms of cancer. Here, we ...investigate the expression pattern, clinical significance, and biological function of HOTAIR in serous ovarian cancer (SOC). Clinically, we found that HOTAIR levels were overexpressed in SOC tissues compared with normal controls and that HOTAIR overexpression was correlated with an advanced FIGO stage and a high histological grade. Multivariate analysis revealed that HOTAIR is an independent prognostic factor for predicting overall survival in SOC patients. We demonstrated that HOTAIR silencing inhibited A2780 and OVCA429 SOC cell proliferation in vitro and that the anti-proliferative effects of HOTAIR silencing also occurred in vivo. Further investigation into the mechanisms responsible for the growth inhibitory effects by HOTAIR silencing revealed that its knockdown resulted in the induction of cell cycle arrest and apoptosis through certain cell cycle-related and apoptosis-related proteins. Together, these results highlight a critical role of HOTAIR in SOC cell proliferation and contribute to a better understanding of the importance of dysregulated lncRNAs in SOC progression.

Clayey silt reservoirs bearing natural gas hydrates (NGH) are considered to be the hydrate-bearing reservoirs that boast the highest reserves but tend to be the most difficult to exploit. They are ...proved to be exploitable by the first NGH production test conducted in the South China Sea in 2017. Based on the understanding of the first production test, the China Geological Survey determined the optimal target NGH reservoirs for production test and conducted a detailed assessment, numerical and experimental simulation, and onshore testing of the reservoirs. After that, it conducted the second offshore NGH production test in 1225 m deep Shenhu Area, South China Sea (also referred to as the second production test) from October 2019 to April 2020. During the second production test, a series of technical challenges of drilling horizontal wells in shallow soft strata in deep sea were met, including wellhead stability, directional drilling of a horizontal well, reservoir stimulation and sand control, and accurate depressurization. As a result, 30 days of continuous gas production was achieved, with a cumulative gas production of 86.14 ×104 m3. Thus, the average daily gas production is 2.87 ×104 m3, which is 5.57 times as much as that obtained in the first production test. Therefore, both the cumulative gas production and the daily gas production were highly improved compared to the first production test. As indicated by the monitoring results of the second production test, there was no anomaly in methane content in the seafloor, seawater, and atmosphere throughout the whole production test. This successful production test further indicates that safe and effective NGH exploitation is feasible in clayey silt NGH reservoirs. The industrialization of hydrates consists of five stages in general, namely theoretical research and simulation experiments, exploratory production test, experimental production test, productive production test, and commercial production. The second production test serves as an important step from the exploratory production test to experimental production test.
Trimethylamine-N-oxide (TMAO) is a gut microbial metabolite that promotes Alzheimer's disease (AD) progression. Given that probiotics can alleviate AD symptoms by inhibiting the synthesis of TMAO, ...here we investigated the correlation between TMAO and cognitive deterioration by measuring TMAO levels in the plasma of choline-treated APP/PS1 mice (an AD mouse model) with and without probiotic treatments. We found that declines in
in the gut were associated with cognitive impairment. Moreover, 12-weeks of treatment with memantine plus
ameliorated cognitive deterioration, decreased Αβ levels in the hippocampus, and protected neuronal integrity and plasticity. These effects were accompanied by reductions in TMAO synthesis and neuroinflammation. These experiments demonstrate that
augments the beneficial therapeutic effects of memantine treatment in APP/PS1 mice by remodeling the intestinal microbiota, inhibiting the synthesis of TMAO, and reducing clusterin levels. Our results thus highlight intestinal microbiota as a potential therapeutic target to decrease the risk of AD.
Extant research on workplace ostracism has investigated a victimization perspective to understand ostracism at the cost of examining the perpetrator-centric view of ostracism. The current study draws ...on the self-categorization theory and the social exchange theory to investigate the harmful effects of workplace romance in cultivating workplace ostracism from the perpetrator's perspective to combat concerns for victim blaming. The study further proposes that workplace ostracism triggered by workplace romance provokes knowledge sabotage. Besides, the study investigates the moderating role of parochial altruism in the underlying linkages. The study utilizes a multisource, time-lagged research design to collect data from employees working in service sector organizations in Pakistan. The study analyzes 343 responses using SmartPLS (v 4.0). The findings of this study reveal that workplace romance elicits workplace ostracism, which, in turn, fosters knowledge sabotage. In addition, the study finds that parochial altruism strengthens the associations between (a) workplace romance and workplace ostracism and (b) workplace romance and knowledge sabotage, mediated by workplace ostracism such that the associations are more potent at higher levels of parochial altruism and vice versa. This is the first study that examines workplace romance as the perpetrator-centric antecedent of workplace ostracism, and parochial altruism exaggerates outgroup ostracism and knowledge sabotage.

A Zinc-loaded montmorillonite (Zn-MMT) coating was hydrothermally prepared using Zn2+ ion intercalated sodium montmorillonite (Na-MMT) upon magnesium (Mg) alloy AZ31 as bone repairing ...materials. Biodegradation rate of the Mg-based materials was studied via potentiodynamic polarization curves, electrochemical impedance spectroscopy (EIS) and hydrogen evolution tests. Results revealed that both Na-MMT and Zn-MMT coatings exhibited better corrosion resistance in Dulbecco’s modified eagle medium (DMEM) + 10% calf serum (CS) than bare Mg alloy AZ31 counterparts. Hemolysis results demonstrated that hemocompatibility of the Na-MMT and Zn-MMT coatings were 5%, and lower than that of uncoated Mg alloy AZ31 pieces. In vitro MTT tests and live-dead stain of osteoblast cells (MC3T3-E1) indicated a significant improvement in cytocompatibility of both Na-MMT and Zn-MMT coatings. Antibacterial properties of two representative bacterial strains associated with device-related infection, i.e. Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus), were employed to explore the antibacterial behavior of the coatings. The measured inhibitory zone and bacterial growth rate confirmed that Zn-MMT coatings exhibited higher suppression toward both E. coli and S. aureus than that of Na-MMT coatings. The investigation on antibacterial mechanism through scanning electron microscopy (SEM) and lactate dehydrogenase (LDH) release assay manifested that Zn-MMT coating led to severe breakage of bacterial membrane of E. coli and S. aureus, which resulted in a release of cytoplasmic materials from the bacterial cells. In addition, the good inhibition of Zn-MMT coatings against E. coli and S. aureus might be attributed to the slow but sustainable release of Zn2+ ions (up to 144 h) from the coatings into the culture media. This study provides a novel coating strategy for manufacturing biodegradable Mg alloys with good corrosion resistance, biocompatibility and antibacterial activity for future orthopedic applications.
The significance of the current work is to develop a corrosion-resistant and antibacterial Zn-MMT coating on magnesium alloy AZ31 through a hydrothermal method. The Zn-MMT coating on magnesium alloy AZ31 shows better corrosion resistance, biocompatibility and excellent antibacterial ability than magnesium alloy AZ31. This study provides a novel coating on Mg alloys for future orthopedic applications.
Natural gas hydrates (NGH) is one of key future clean energy resources. Its industrialized development will help remit the huge demand of global natural gas, relieve the increasing pressure of the ...environment, and play a vital role in the green sustainable growth of human societies. Based on nearly two decades’ studying on the reservoir characteristics in the South China Sea (SCS) and the knowledge of reservoir system, the China Geological Survey (CGS) conducted the first production test on an optimal target selected in Shenhu area SCS in 2017. Guided by the “three-phase control” exploitation theory which focused on formation stabilization, technologies such as formation fluid extraction, well drilling and completing, reservoir stimulating, sand controlling, environmental monitoring, monitoring and preventing of secondary formation of hydrates were applied. The test lasted for 60 days from May 10th when starting to pump, drop pressure and ignite to well killing on July 9th, with gas production of 3.09×105 m3 in total, which is a world record with the longest continuous duration of gas production and maximal gas yield. This successful test brings a significant breakthrough on safety control of NGH production.
Scope
Short‐peptide‐based enteral nutrition (SPEN) is absorbed more efficiently in patients with severe acute pancreatitis (SAP). More importantly, SPEN decreases SAP‐induced enterogenous infection ...risk. This study aims to investigate whether SPEN alleviates intestinal bacterial translocation in mice with SAP, and the underlying mechanisms.
Methods and results
The SAP model is established after pre‐treatment with SPEN or intact‐protein‐based enteral nutrition. Although there is no improvement in pancreas injury, as evaluated through Hematoxylin‐Eosin staining or serum amylase, SPEN obviously attenuates intestinal bacterial translocation after SAP. To unveil the mechanisms, it is found that the intestinal mechanical barrier destroyed by SAP is significantly relieved by SPEN, which presents with recovered ZO‐1 expression, mucus layer, and goblet cell function. Additionally, SPEN alleviates local CCR6/CCL20 induced CD11c+ dendritic cell infiltration, systemic immunosuppression, and inhibits the secretion of luminal secretory immunoglobulin A. Possibly responsible for SAP‐induced mucosal dysfunctions, destroyed intestinal mucosal microcirculation and local hypoxia are largely improved in SAP+SPEN group.
Conclusion
SPEN can improve downregulated intestinal mucosal microcirculation secondary to SAP, which may be responsible for mucosal inflammation relief, maintenance of the mechanical barrier and mucosal immunity, the correction of systemic immunosuppression, and play a protective role in defending commensal bacterial translocation after SAP.
Enteral nutrition can decrease the risk of enterogenous infection after severe acute pancreatitis (SAP), especially short‐peptide‐based enteral nutrition (SPEN). This is the first study to report that SPEN improves downregulated intestinal mucosal microcirculation secondary to SAP, which may be responsible for mucosal inflammation relief, maintenance of mechanical barrier and mucosal immunity, and defends against commensal bacterial translocation.

This paper presents the design and development of a three-phase 10-kV/400-V 500-kVA electronic power transformer (EPT). The power circuit is designed in a modular fashion, i.e., the main circuit ...consists of many identical ac-dc-dc-ac modules (abbreviated as power modules). Each power module consists of a high-voltage power cell, a low-voltage power cell (LVPC), a medium-frequency isolation transformer, and a filter. The corresponding control and protection system is developed. A special three-stage startup strategy is designed to shorten the startup time and reduce the startup inrush current. The negative-sequence current compensation is introduced in the input stage to handle the unbalanced loads. To keep the dc-link voltages balanced, an individual dc voltage balancing controller based on regulating the output power of each parallel LVPC is proposed. The detailed control hardware design and software implementation are discussed. The functions of this 10-kV EPT prototype are verified through the laboratory and field tests. The results are shown in this paper. Currently, the prototype is operating in the industrial power grid.
Two-dimensional heterojunctions exhibit many unique features in nanoelectronic and optoelectronic devices. However, heterojunction engineering requires a complicated alignment process and some ...defects are inevitably introduced during material preparation. In this work, a laser scanning technique is used to construct a lateral WSe2 p–n junction. The laser-scanned region shows p-type behavior, and the adjacent region is electrically n-doped with a proper gate voltage. The laser-oxidized product WO x is found to be responsible for this p-type doping. After laser scanning, WSe2 displays a change from ambipolar to unipolar p-type property. A significant photocurrent emerges at the p–n junction. Therefore, a self-powered WSe2 photodetector can be fabricated based on this junction, which presents a large photoswitching ratio of 106, a high photoresponsivity of 800 mA W–1, and a short photoresponse time with long-term stability and reproducibility. Therefore, this selective laser-doping method is prospective in future electronic applications.
