An update is provided of Northern Hemisphere (NH) spring (March, April) snow cover extent (SCE) over the 1922–2010 period incorporating the new climate data record (CDR) version of the NOAA weekly ...SCE dataset, with annual 95% confidence intervals estimated from regression analysis and intercomparison of multiple datasets. The uncertainty analysis indicates a 95% confidence interval in NH spring SCE of ±5–10% over the pre-satellite period and ±3–5% over the satellite era. The multi-dataset analysis shows larger uncertainties monitoring spring SCE over Eurasia (EUR) than North America (NA) due to the more complex regional character of the snow cover variability and larger between-dataset variability over northern Europe and north-central Russia. Trend analysis of the updated SCE series provides evidence that NH spring snow cover extent has undergone significant reductions over the past ~90 yr and that the rate of decrease has accelerated over the past 40 yr. The rate of decrease in March and April NH SCE over the 1970–2010 period is ~0.8 million km2 per decade corresponding to a 7% and 11% decrease in NH March and April SCE respectively from pre-1970 values. In March, most of the change is being driven by Eurasia (NA trends are not significant) but both continents exhibit significant SCE reductions in April. The observed trends in SCE are being mainly driven by warmer air temperatures, with NH mid-latitude air temperatures explaining ~50% of the variance in NH spring snow cover over the 89-yr period analyzed. However, there is also evidence that changes in atmospheric circulation around 1980 involving the North Atlantic Oscillation and Scandinavian pattern have contributed to reductions in March SCE over Eurasia.
Natural capital and ecosystem service concepts are embodied in the ecosystems approach to sustainable development, which is a framework being consistently adopted by decision making bodies ranging ...from national governments to the United Nations. In the Millennium Ecosystem Assessment soils are given the vital role of a supporting service, but many of the other soil goods and services remain obscured. In this review we address this using and earth-system approach, highlighting the final goods and services soils produce, in a stock-fund, fund-service model of the pedosphere. We also argue that focusing on final goods and services will be counterproductive in the long run and emphasize that final goods and services are derived from an ecosystem supply chain that relies on ecological infrastructure. We propose that an appropriate ecosystems framework for soils should incorporate soil stocks (natural capital) showing their contribution to stock-flows and emergent fund-services as part of the supply chain. By so doing, an operational ecosystems concept for soils can draw on much more supporting data on soil stocks as demonstrated in a case study with soils data from England and Wales showing stocks, gaps in monitoring and drivers of change. Although the focus of this review is on soils, we believe the earth-system approach and principles of the ecosystem supply chain are widely applicable to the ecosystems approach and bring clarity in terms of where goods and services are derived from.

This paper describes the long-term, satellite-based visible snow cover extent National Oceanic and Atmospheric Administration (NOAA) climate data record (CDR) currently available for climate studies, ...monitoring, and model validation. This environmental data product is developed from weekly Northern Hemisphere snow cover extent data that have been digitized from snow cover maps onto a Cartesian grid draped over a polar stereographic projection. The data have a spatial resolution of 190.6 km at 60° latitude, are updated monthly, and span the period from 4 October 1966 to the present. The data comprise the longest satellite-based CDR of any environmental variable. Access to the data is provided in Network Common Data Form (netCDF) and archived by NOAA's National Climatic Data Center (NCDC) under the satellite Climate Data Record Program (doi:10.7289/V5N014G9). The basic characteristics, history, and evolution of the data set are presented herein. In general, the CDR provides similar spatial and temporal variability to its widely used predecessor product. Key refinements included in the CDR improve the product's grid accuracy and documentation and bring metadata into compliance with current standards for climate data records.
Static strain in complex oxide heterostructures has been extensively used to engineer electronic and magnetic properties at equilibrium. In the same spirit, deformations of the crystal lattice with ...light may be used to achieve functional control across heterointerfaces dynamically. Here, by exciting large-amplitude infrared-active vibrations in a LaAlO3 substrate we induce magnetic order melting in a NdNiO3 film across a heterointerface. Femtosecond resonant soft X-ray diffraction is used to determine the spatiotemporal evolution of the magnetic disordering. We observe a magnetic melt front that propagates from the substrate interface into the film, at a speed that suggests electronically driven motion. Light control and ultrafast phase front propagation at heterointerfaces may lead to new opportunities in optomagnetism, for example by driving domain wall motion to transport information across suitably designed devices.
Broad scale population estimates of declining species are desired for conservation efforts. However, for many secretive species including large carnivores, such estimates are often difficult. Based ...on published density estimates obtained through camera trapping, presence/absence data, and globally available predictive variables derived from satellite imagery, we modelled density and occurrence of a large carnivore, the jaguar, across the species' entire range. We then combined these models in a hierarchical framework to estimate the total population. Our models indicate that potential jaguar density is best predicted by measures of primary productivity, with the highest densities in the most productive tropical habitats and a clear declining gradient with distance from the equator. Jaguar distribution, in contrast, is determined by the combined effects of human impacts and environmental factors: probability of jaguar occurrence increased with forest cover, mean temperature, and annual precipitation and declined with increases in human foot print index and human density. Probability of occurrence was also significantly higher for protected areas than outside of them. We estimated the world's jaguar population at 173,000 (95% CI: 138,000-208,000) individuals, mostly concentrated in the Amazon Basin; elsewhere, populations tend to be small and fragmented. The high number of jaguars results from the large total area still occupied (almost 9 million km2) and low human densities (< 1 person/km2) coinciding with high primary productivity in the core area of jaguar range. Our results show the importance of protected areas for jaguar persistence. We conclude that combining modelling of density and distribution can reveal ecological patterns and processes at global scales, can provide robust estimates for use in species assessments, and can guide broad-scale conservation actions.
Castration-resistant prostate cancer (CRPC) continues to pose a significant clinical challenge with new generation second-line hormonal therapies affording limited improvement in disease outcome. As ...the androgen receptor (AR) remains a critical driver in CRPC, understanding the determinants of its transcriptional activity is important for developing new AR-targeted therapies. FOXA1 is a key component of the AR transcriptional complex yet its role in prostate cancer progression and the relationship between AR and FOXA1 are not completely resolved. It is well established that FOXA1 levels are elevated in advanced prostate cancer and metastases. We mimicked these conditions by overexpressing FOXA1 in the androgen-responsive LNCaP prostate cancer cell line and observed a significant increase in AR genomic binding at novel regions that possess increased chromatin accessibility. High levels of FOXA1 resulted in increased proliferation at both sub-optimal and high 5α-dihydrotestosterone (DHT) concentrations. Immunohistochemical staining for FOXA1 in a clinical prostate cancer cohort revealed that high FOXA1 expression is associated with shorter time to biochemical recurrence after radical prostatectomy (hazard ratio (HR) 5.0, 95% confidence interval (CI) 1.2-21.1, P=0.028), positive surgical margins and higher stage disease at diagnosis. The gene expression program that results from FOXA1 overexpression is enriched for PTEN, Wnt and other pathways typically represented in CRPC gene signatures. Together, these results suggest that in an androgen-depleted state, elevated levels of FOXA1 enhance AR binding at genomic regions not normally occupied by AR, which in turn facilitates prostate cancer cell growth.
SuperCDMS SNOLAB will be a next-generation experiment aimed at directly detecting low-mass particles (with masses ≤10 GeV/c2) that may constitute dark matter by using cryogenic detectors of two types ...(HV and iZIP) and two target materials (germanium and silicon). The experiment is being designed with an initial sensitivity to nuclear recoil cross sections ∼1×10−43 cm2 for a dark matter particle mass of 1 GeV/c2, and with capacity to continue exploration to both smaller masses and better sensitivities. The phonon sensitivity of the HV detectors will be sufficient to detect nuclear recoils from sub-GeV dark matter. A detailed calibration of the detector response to low-energy recoils will be needed to optimize running conditions of the HV detectors and to interpret their data for dark matter searches. Low-activity shielding, and the depth of SNOLAB, will reduce most backgrounds, but cosmogenically produced H3 and naturally occurring Si32 will be present in the detectors at some level. Even if these backgrounds are 10 times higher than expected, the science reach of the HV detectors would be over 3 orders of magnitude beyond current results for a dark matter mass of 1 GeV/c2. The iZIP detectors are relatively insensitive to variations in detector response and backgrounds, and will provide better sensitivity for dark matter particles with masses ≳5 GeV/c2. The mix of detector types (HV and iZIP), and targets (germanium and silicon), planned for the experiment, as well as flexibility in how the detectors are operated, will allow us to maximize the low-mass reach, and understand the backgrounds that the experiment will encounter. Upgrades to the experiment, perhaps with a variety of ultra-low-background cryogenic detectors, will extend dark matter sensitivity down to the “neutrino floor,” where coherent scatters of solar neutrinos become a limiting background.
Animal experiments remain essential to understand the fundamental mechanisms underpinning malignancy and to discover improved methods to prevent, diagnose and treat cancer. Excellent standards of ...animal care are fully consistent with the conduct of high quality cancer research. Here we provide updated guidelines on the welfare and use of animals in cancer research. All experiments should incorporate the 3Rs: replacement, reduction and refinement. Focusing on animal welfare, we present recommendations on all aspects of cancer research, including: study design, statistics and pilot studies; choice of tumour models (e.g., genetically engineered, orthotopic and metastatic); therapy (including drugs and radiation); imaging (covering techniques, anaesthesia and restraint); humane endpoints (including tumour burden and site); and publication of best practice.
Background & Aims Sinusoidal obstruction syndrome (SOS) following oxaliplatin based chemotherapy can have a significant impact on post-operative outcome following resection of colorectal liver ...metastases. To date no relevant experimental models of oxaliplatin induced SOS have been described. The aim of this project was to establish a rodent model which could be utilised to investigate mechanisms underlying SOS to aid the development of therapeutic strategies. Methods C57Bl/6 mice, maintained on a purified diet, were treated with intra-peritoneal FOLFOX (n = 10), or vehicle (n = 10), weekly for five weeks and culled one week following final treatment. Sections of the liver and spleen were fixed in formalin and paraffin embedded for histological analysis. The role of oxidative stress on experimental-induced SOS was determined by dietary supplementation with butylated hydroxyanisole and N-acetylcysteine. Results FOLFOX treatment was associated with the development of sinusoidal dilatation and hepatocyte atrophy on H&E stained sections of the liver in keeping with SOS. Immunohistochemistry for p21 demonstrated the presence of replicative senescence within the sinusoidal endothelium. FOLFOX induced endothelial damage leads to a pro-thrombotic state within the liver associated with upregulation of PAI-1 ( p <0.001), vWF ( p <0.01) and Factor X ( p <0.001), which may contribute to the propagation of liver injury. Dietary supplementation with the antioxidant BHA prevented the development of significant SOS. Conclusions We have developed the first reproducible model of chemotherapy induced SOS that reflects the pathogenesis of this disease in patients. It appears that the use of antioxidants alongside oxaliplatin based chemotherapy may be of value in preventing the development of SOS in patients with colorectal liver metastases.
