The location of and threats to biodiversity are distributed unevenly, so prioritization is essential to minimize biodiversity loss. To address this need, biodiversity conservation organizations have ...proposed nine templates of global priorities over the past decade. Here, we review the concepts, methods, results, impacts, and challenges of these prioritizations of conservation practice within the theoretical irreplaceability/vulnerability framework of systematic conservation planning. Most of the templates prioritize highly irreplaceable regions; some are reactive (prioritizing high vulnerability), and others are proactive (prioritizing low vulnerability). We hope this synthesis improves understanding of these prioritization approaches and that it results in more efficient allocation of geographically flexible conservation funding.

To overcome this limitation, we herein report a simple oxidative deprotection protocol. This transformation rapidly converts the robust amide to a labile imide, allowing subsequent cleavage in a simple one‐pot fashion to rapidly access carboxylic acids or amides as final products.

Our aim has been to evaluate the effect of cryoprotective agents (CPAs) on the exposure, vitrification (VIT), and in vitro culture (IVC) of ovarian tissue with regard to the expression and ...immunolocalization of aquaporins (AQPs) 3 and 9 in ovine preantral follicles. Tissues were treated as follows: Experiment I: (1) control (without exposure to CPAs), (2) e-EG (exposure to ethylene glycol), (3) er-EG (exposure to and removal of EG), (4) e-DMSO (exposure to dimethyl sulfoxide), (5) er-DMSO (exposure to and removal of DMSO), (6) e-EG+DMSO (exposure to EG+DMSO), (7) er-EG+DMSO (exposure to and removal of EG+DMSO); Experiment II: (1) control, (2) VIT, (3) IVC, (4) VIT-IVC. In Experiment I, following er-EG or er-DMSO, tissue showed the down-regulation (

AQP3
mRNA. The mRNA transcript levels were reduced (

< 0.05) for
AQP9
in tissue following er-EG+DMSO. Immunolocalization was positive for both proteins (AQP3 and AQP9) on ovine preantral follicles following all treatments, except in the e-EG+DMSO group. In Experiment II, the mRNA levels of
AQP3
and
AQP9
following VIT treatment were similar (

> 0.05) to that of the control group. Nevertheless, VIT-IVC treatment led to the down-regulation of mRNA of
AQP3
and
AQP9
. Thus, AQP3 and AQP9 act in a mutually dependent way, maintaining the cell homeostasis that is essential for the ovary cryopreservation process. Furthermore, the changes in the expression profiles of mRNA and protein after culture are a strong indicator that in vitro conditions have to be strictly controlled to ensure follicle viability and functionality.
Parkinson’s disease (PD) is the second most common neurodegenerative disorder, mainly characterized by motor deficits correlated with progressive dopaminergic neuronal loss in the substantia nigra ...pars compacta (SN). Necroptosis is a caspase-independent form of regulated cell death mediated by the concerted action of receptor-interacting protein 3 (RIP3) and the pseudokinase mixed lineage domain-like protein (MLKL). It is also usually dependent on RIP1 kinase activity, influenced by further cellular clues. Importantly, necroptosis appears to be strongly linked to several neurodegenerative diseases, including PD. Here, we aimed at identifying novel chemical inhibitors of necroptosis in a PD-mimicking model, by conducting a two-step screening. Firstly, we phenotypically screened a library of 31 small molecules using a cellular model of necroptosis and, thereafter, the hit compound effect was validated in vivo in a sub-acute 1-methyl-1-4-phenyl-1,2,3,6-tetrahydropyridine hydrochloride (MPTP) PD-related mouse model. From the initial compounds, we identified one hit—Oxa12—that strongly inhibited necroptosis induced by the pan-caspase inhibitor zVAD-fmk in the BV2 murine microglia cell line. More importantly, mice exposed to MPTP and further treated with Oxa12 showed protection against MPTP-induced dopaminergic neuronal loss in the SN and striatum. In conclusion, we identified Oxa12 as a hit compound that represents a new chemotype to tackle necroptosis. Oxa12 displays in vivo effects, making this compound a drug candidate for further optimization to attenuate PD pathogenesis.
This study aimed to compare four constructs from the three-minute all-out test (AO3)-end power (EP), the area above EP (WEP), maximum power (Pmax), and attained Formula: see text-to those derived ...from the classical CP model in tethered running. Seventeen male recreational runners underwent two experiments to test for reliability and agreement of AO3 parameters with those obtained from the classical CP model (Wꞌ and CP), a graded exercise test (Formula: see text) and a 30-second all-out test (AO30s; Pmax); all performed on a non-motorized treadmill (NMT). Significance levels were set at p<0.05. There were no significant differences between test-retest for Pmax (p = 0.51), WEP (p = 0.39), and EP (p = 0.64), showing generally close to zero bias. Further, retest ICC were high for Pmax and EP (ICC > 0.86) but moderate for WEP (ICC = 0.69). Pmax showed no difference between AO3 and AO30s (p = 0.18; CV% = 9.5%). EP and WEP disagreed largely with their classical critical power model counterparts (p = 0.05; CV%>32.7% and p = 0.23; CV%>39.7%, respectively), showing greater error than their test-retest reliability. Formula: see text from AO3 was not different (p = 0.13) and well related (CV% = 8.4; ICC = 0.87) to the incremental test Formula: see text. Under the studied conditions, the agreement of EP and WEP to CP and Wꞌ was not strong enough to assure their use interchangeably. Pmax and Formula: see text were closer to their criterion parameters.
Bioindicators have been widely used to assess the contamination of lead/zinc (Pb/Zn) in smelter areas, mining waste disposal areas and in areas containing slags from Pb ore smelting. In this context, ...the analysis involving microorganisms has gained prominence as a complementary tool in studies aimed at assessing contaminated sites. Therefore, the objective of this study was to assess the contamination of Pb, Zn and cadmium (Cd) in an area where a PbZn smelter operated, using the integration of geochemical and microbial data. The experimental analysis was conducted to characterize the soil collected at three different sites (samples NS, EW and EP). The analysis conducted was physicochemical, metal concentration, metal speciation and analysis of the microbial community through high-throughput sequencing technique. Through the results it was observed that the high concentrations of metals altered the bacterial community present in the soil. Differences were noted between the microbial communities according to the sampling site, especially in sample EP, collected at the margin of the dirt road, which presented higher metal concentrations and microbial diversity. The main phyla detected in the samples were: Proteobacteria, Bacteroidetes and Acidobacteria. Bacteria tolerant to the presence of potentially toxic metals (PTM), such as Rhodoplanes, Kaistobacter, Sphingomonas and Flavisolibacter were identified in the analyzed samples. The phylogenetic groups identified in the study area are similar to those obtained in other studies in metal contaminated areas. The differences between the bacterial communities in each sample indicate that the concentration of PTM may have influenced the microbial community in the soil. Thus, it is noted the importance of the integration of geochemical and microbial data to evaluate the impact of the improper disposal of high PTM concentrated slags in natural soils.

Large-scale restoration programmes in the tropics require large volumes of high quality, genetically diverse and locally adapted seeds from a large number of species. However, scarcity of native ...seeds is a critical restriction to achieve restoration targets. In this paper, we analyse three successful community-based networks that supply native seeds and seedlings for Brazilian Amazon and Cerrado restoration projects. In addition, we propose directions to promote local participation, legal, technical and commercialisation issues for up-scaling the market of native seeds for restoration with high quality and social justice. We argue that effective community-based restoration arrangements should follow some principles: (i) seed production must be based on real market demand; (ii) non-governmental and governmental organisations have a key role in supporting local organisation, legal requirements and selling processes; (iii) local ecological knowledge and labour should be valued, enabling local communities to promote large-scale seed production; (iv) applied research can help develop appropriate techniques and solve technical issues. The case studies from Brazil and principles presented here can be useful for the up-scaling restoration ecology efforts in many other parts of the world and especially in tropical countries where improving rural community income is a strategy for biodiversity conservation and restoration.
The incidence of infection by the dengue virus (DENV) has grown dramatically, reaching 128 countries in tropical and subtropical regions worldwide, with a pattern of hyper-endemicity. DENV is a ...mosquito-borne disease having four serotypes, one or two circulating in epidemic outbreaks. The diagnosis of DENV is challenging mainly due to the circulation of new viruses with remarkable similarities, such as Zika (ZIKV) that may cause fetal microcephaly. DENV affects 390 million people per year, but these numbers may be higher due to the underreported and misclassified cases. Recently, the NS1 nonstructural protein has been described in serum and urine of DENV and ZIKV patients, suggesting its use as a biomarker for screening since a negative NS1 sample confirms the absence of these infections. Herein, a label-free immunosensor comprising an assembled nanostructured thin film of carbon nanotube-ethylenediamine is described. The advantage of in situ electrosynthesis of polymer film is to allow major control of thickness and conductivity, in addition to designing the reactive groups for functionalization. A quartz crystal microbalance system was used to estimate the thickness of the polymeric film obtained. The anti-NS1 monoclonal antibodies were immobilized to carbon nanotubes by covalent linkage, permitting a high stability during measurements. Analytical responses to NS1 were obtained by differential pulse voltammetry (DPV), showing a linear range from 20 to 800 ng mL
−1
and reproducibility of 3.0%, with a limit of detection (LOD) of 6.8 ng mL
- 1
. This immunosensor was capable of detecting ZIKV and DENV NS1 in spiked urine and real serum in a clinical range.

Cosmetic products play an essential role in everyone's life. People everyday use a large variety of cosmetic products such as soap, shampoo, toothpaste, deodorant, skin care, perfume, make-up, among ...others. The cosmetic industry encompasses several environmental, social and economic impacts that are being addressed through the search for more efficient manufacturing techniques, the reduction of waste and emissions and the promotion of personal hygiene, contributing to an improvement of public health and at the same time providing employment opportunities. The current trend among consumers is the pursuit for natural ingredients in cosmetic products, as many of these products exhibit equal, better or additional benefits in comparison with the chemical-based products. In this sense, biosurfactants are natural compounds with great potential in the formulation of cosmetic products given by their biodegradability and impact in health. Indeed, many of these biosurfactants could exhibit a "prebiotic" character. This review covers the current state-of-the-art of biosurfactant research for cosmetic purposes and further discusses the future challenges for cosmetic applications.
