ABSTRACT
Characterization of very high cycle fatigue (VHCF) performance is of significant issue for ensuring long‐term durability and reliability of machinery and structural components due to the ...growing industrial demands and significant requirements of the advanced systems. In this study, VHCF characteristics of nanocrystallized skins (nanoskin) on JIS SCM435 (AISI 4137) steels were investigated as three different nanoskins on the surface, which was fabricated by altering the static load of ultrasonic nanocrystal surface modification (UNSM) treatment. The fatigue characterization, which shows linearly proportional correlation in the range of 80–120 µm depth of subsurface, was subjected to severe plastic deformation by altering the static loads of UNSM treatment to 40, 70 and 100 N, respectively. The fatigue strength increased up to 30% in the regime of VHCF. The improved strength mainly resulted from the generation of nanocrystalline structure, the enhanced surface uniformity, hardness and residual stress.
Stacking fault energies (SFE) were determined in additively manufactured (AM) stainless steel (SS 316 L) and equiatomic CrCoNi medium-entropy alloys. AM specimens were fabricated via directed energy ...deposition and tensile loaded at room temperature. In situ neutron diffraction was performed to obtain a number of faulting-embedded diffraction peaks simultaneously from a set of (hkl) grains during deformation. The peak profiles diffracted from imperfect crystal structures were analyzed to correlate stacking fault probabilities and mean-square lattice strains to the SFE. The result shows that averaged SFEs are 32.8 mJ/m
for the AM SS 316 L and 15.1 mJ/m
for the AM CrCoNi alloys. Meanwhile, during deformation, the SFE varies from 46 to 21 mJ/m
(AM SS 316 L) and 24 to 11 mJ/m
(AM CrCoNi) from initial to stabilized stages, respectively. The transient SFEs are attributed to the deformation activity changes from dislocation slip to twinning as straining. The twinning deformation substructure and atomic stacking faults were confirmed by electron backscatter diffraction (EBSD) and transmission electron microscopy (TEM). The significant variance of the SFE suggests the critical twinning stress as 830 ± 25 MPa for the AM SS 316 L and 790 ± 40 MPa for AM CrCoNi, respectively.
ABSTRACT We present the X-ray spectral analysis of the 1855 extragalactic sources in the Chandra COSMOS-Legacy survey catalog having more than 30 net counts in the 0.5-7 keV band. A total of 38% of ...the sources are optically classified type 1 active galactic nuclei (AGNs), 60% are type 2 AGNs, and 2% are passive, low-redshift galaxies. We study the distribution of AGN photon index Γ and of the intrinsic absorption based on the sources' optical classification: type 1 AGNs have a slightly steeper mean photon index Γ than type 2 AGNs, which, on the other hand, have average times higher than type 1 AGNs. We find that ∼15% of type 1 AGNs have cm−2, i.e., are obscured according to the X-ray spectral fitting; the vast majority of these sources have 1044 erg s−1. The existence of these objects suggests that optical and X-ray obscuration can be caused by different phenomena, the X-ray obscuration being, for example, caused by dust-free material surrounding the inner part of the nuclei. Approximately 18% of type 2 AGNs have cm−2, and most of these sources have low X-ray luminosities (L 1043 erg s−1). We expect a part of these sources to be low-accretion, unobscured AGNs lacking broad emission lines. Finally, we also find a direct proportional trend between and host-galaxy mass and star formation rate, although part of this trend is due to a redshift selection effect.
This study investigates the influence of ultrasonic nanocrystal surface modification (UNSM) technique on the mechanical characteristics and fatigue life of two types of annealed and aged Inconel 718 ...alloys. The mechanical properties of both UNSM‐treated alloys were compared with the untreated specimens. The fatigue life of the untreated and UNSM‐treated Inconel 718 alloy specimens was investigated by ultrasonic fatigue and rotary‐bending fatigue tests. Results revealed that the UNSM‐treated specimens exhibited longer fatigue life than those of the untreated specimens. The improvement in fatigue life is mainly attributed to the induced compressive residual stress induced in the subsurface by UNSM technique.
Background. Invasive candidiasis is an important cause of morbidity and mortality among patients with health care–associated infection. The echinocandins have potent fungicidal activity against most ...Candida species, but there are few data comparing the safety and efficacy of echinocandins in the treatment of invasive candidiasis. Methods. This was an international, randomized, double-blind trial comparing micafungin (100 mg daily) and micafungin (150 mg daily) with a standard dosage of caspofungin (70 mg followed by 50 mg daily) in adults with candidemia and other forms of invasive candidiasis. The primary end point was treatment success, defined as clinical and mycological success at the end of blinded intravenous therapy. Results. A total of 595 patients were randomized to one the treatment groups and received at least 1 dose of study drug. In the modified intent-to-treat population, 191 patients were assigned to the micafungin 100 mg group, 199 to the micafungin 150 mg group, and 188 to the caspofungin group. Demographic characteristics and underlying disorders were comparable across the groups. Approximately 85% of patients had candidemia; the remainder had noncandidemic invasive candidiasis. At the end of blinded intravenous therapy, treatment was considered successful for 76.4% of patients in the micafungin 100 mg group, 71.4% in the micafungin 150 mg group, and 72.3% in the caspofungin group. The median time to culture negativity was 2 days in the micafungin 100 mg group and the caspofungin group, compared with 3 days in the micafungin 150 mg groups. There were no significant differences in mortality, relapsing and emergent infections, or adverse events between the study arms. Conclusions. Dosages of micafungin 100 mg daily and 150 mg daily were noninferior to a standard dosage of caspofungin for the treatment of candidemia and other forms of invasive candidiasis.
ABSTRACT We present the catalog of optical and infrared counterparts of the Chandra COSMOS-Legacy Survey, a 4.6 Ms Chandra program on the 2.2 deg2 of the COSMOS field, combination of 56 new ...overlapping observations obtained in Cycle 14 with the previous C-COSMOS survey. In this Paper we report the i, K, and 3.6 m identifications of the 2273 X-ray point sources detected in the new Cycle 14 observations. We use the likelihood ratio technique to derive the association of optical/infrared (IR) counterparts for 97% of the X-ray sources. We also update the information for the 1743 sources detected in C-COSMOS, using new K and 3.6 m information not available when the C-COSMOS analysis was performed. The final catalog contains 4016 X-ray sources, 97% of which have an optical/IR counterpart and a photometric redshift, while 54% of the sources have a spectroscopic redshift. The full catalog, including spectroscopic and photometric redshifts and optical and X-ray properties described here in detail, is available online. We study several X-ray to optical (X/O) properties: with our large statistics we put better constraints on the X/O flux ratio locus, finding a shift toward faint optical magnitudes in both soft and hard X-ray band. We confirm the existence of a correlation between X/O and the the 2-10 keV luminosity for Type 2 sources. We extend to low luminosities the analysis of the correlation between the fraction of obscured AGNs and the hard band luminosity, finding a different behavior between the optically and X-ray classified obscured fraction.
Increasing antimicrobial resistance and medical device-related infections have led to a renewed interest in phage therapy as an alternative or adjunct to conventional antimicrobials. Expanded access ...and compassionate use cases have risen exponentially but have varied widely in approach, methodology, and clinical situations in which phage therapy might be considered. Large gaps in knowledge contribute to heterogeneity in approach and lack of consensus in many important clinical areas. The Antibacterial Resistance Leadership Group (ARLG) has convened a panel of experts in phage therapy, clinical microbiology, infectious diseases, and pharmacology, who worked with regulatory experts and a funding agency to identify questions based on a clinical framework and divided them into three themes: potential clinical situations in which phage therapy might be considered, laboratory testing, and pharmacokinetic considerations. Suggestions are provided as answers to a series of questions intended to inform clinicians considering experimental phage therapy for patients in their clinical practices.
Background We have previously shown that oncostatin M (OSM) levels are increased in nasal polyps (NPs) of patients with chronic rhinosinusitis (CRS), as well as in bronchoalveolar lavage fluid, after ...segmental allergen challenge in allergic asthmatic patients. We also showed in vitro that physiologic levels of OSM impair barrier function in differentiated airway epithelium. Objective We sought to determine which hematopoietic or resident cell type or types were the source of the OSM expressed in patients with mucosal airways disease. Methods Paraffin-embedded NP sections were stained with fluorescence-labeled specific antibodies against OSM, GM-CSF, and hematopoietic cell-specific markers. Live cells were isolated from NPs and matched blood samples for flow cytometric analysis. Neutrophils were isolated from whole blood and cultured with the known OSM inducers GM-CSF and follistatin-like 1, and OSM levels were measured in the supernatants. Bronchial biopsy sections from control subjects, patients with moderate asthma, and patients with severe asthma were stained for OSM and neutrophil elastase. Results OSM staining was observed in NPs, showed colocalization with neutrophil elastase (n = 10), and did not colocalize with markers for eosinophils, macrophages, T cells, or B cells (n = 3-5). Flow cytometric analysis of NPs (n = 9) showed that 5.1% ± 2% of CD45+ cells were OSM+ , and of the OSM+ cells, 56% ± 7% were CD16+ Siglec-8− , indicating neutrophil lineage. Only 0.6 ± 0.4% of CD45+ events from matched blood samples (n = 5) were OSM+ , suggesting that increased OSM levels in patients with CRS was locally stimulated and produced. A majority of OSM+ neutrophils expressed arginase 1 (72.5% ± 12%), suggesting an N2 phenotype. GM-CSF levels were increased in NPs compared with those in control tissue and were sufficient to induce OSM production ( P < .001) in peripheral blood neutrophils in vitro . OSM+ neutrophils were also observed at increased levels in biopsy specimens from patients with severe asthma. Additionally, OSM protein levels were increased in induced sputum from asthmatic patients compared with that from control subjects ( P < .05). Conclusions Neutrophils are a major source of OSM-producing cells in patients with CRS and severe asthma.

A series of axial tensile fatigue tests (R = 0.1) was carried out to investigate the initiation and the growth behaviours of very small surface fatigue cracks under two different surface ...conditions (viz. smooth and pitted surfaces) of AISI 304 stainless steel at room temperature. This paper deals with both of the two approaches regarding the analysis of fatigue: the approach based on the concept of fracture mechanics and low cycle fatigue. In particular, both the initiation and growth of cracks and the coalescence of small cracks by fatigue in the specimen have been investigated by the methods of surface replicas and photomicrographs. Quantitative information such as the initiation period, growth and coalescence behaviours of small cracks, and crack growth properties were systematically obtained. The results show that the accurate determination of these parameters is critical for the application of fracture mechanics to fatigue life assessment.
Claudins (CLDNs) are a family of integral membrane proteins central to the formation of tight junctions, structures that are involved in paracellular transport and cellular growth and ...differentiation, and are critical for the maintenance of cellular polarity. Recent studies have provided evidence that CLDNs are aberrantly expressed in diverse types of human cancers, including hepatocellular carcinomas (HCCs). However, little is known about how CLDN expression is involved in cancer progression. In this study, we show that CLDN1 has a causal role in the epithelial-mesenchymal transition (EMT) in human liver cells, and that the c-Abl-Ras-Raf-1-ERK1/2 signaling axis is critical for the induction of malignant progression by CLDN1. Overexpression of CLDN1 induced expression of the EMT-regulating transcription factors Slug and Zeb1, and thereby led to repression of E-cadherin, β-catenin expression, enhanced expression of N-cadherin and Vimentin, a loss of cell adhesion, and increased cell motility in normal liver cells and HCC cells. In line with these findings, inhibition of either c-Abl or ERK clearly attenuated CLDN1-induced EMT, as evidenced by a reversal of N-cadherin and E-cadherin expression patterns, and restored normal motility. Collectively, these results indicate that CLDN1 is necessary for the induction of EMT in human liver cells, and that activation of the c-Abl-Ras-Raf-1-ERK1/2 signaling pathway is required for CLDN1-induced acquisition of the malignant phenotype. The present observations suggest that CLDN1 could be exploited as a biomarker for liver cancer metastasis and might provide a pivotal point for therapeutic intervention in HCC.
